Astradalus membranaceus root extract (ARE) contains saponins, flavonoids, and their glycosides. Two flavonoids, calycosin and formononetin, were predominant constituents with the different amount by diverse extraction conditions. ARE showed the anti-oxidative activities, inhibited tyrosinase activity, and increased type I procollagen expression in proportion to calycosin and formononetin contents. The clinical study also showed that skin wrinkles were significantly reduced in the test group treated with a cream formulation containging 0.1% ARE compared with a group treated with a cream formulation without 0.1% ARE (p < 0.05). These results suggest that ARE may have potential as an anti-aging ingredient in cosmetics.

Background: Astragalus membranaceus belonging to the Leguminosae family is often utilized as a traditional medicine. The aim of this study was to elucidate the basic breeding information required to develop short stem A. membranaceus cultivars. Methods and Results: Roots of A. membranaceus advanced yield trial (AYT) lines were harvested in late October 2017. Root yield of six AYT lines were increased in a range of 8.9 - 74.8% compared with 'Aseong' as control (check variety). The height of seven AYT lines were shorter than that of 'Aseong'. In addition, stem diameter of nine AYT lines was thicker than that of 'Aseong'. Consequently, 1502-56, 1503-90, and 1510-80 were selected as elite lines for the development of short stem cultivars. HPLC analysis was perfromed to identify lines with high level active components such as calycosin-7-O-${\beta}$-D-glucoside and calycosin. The levels of both active components were higher in 1502-56, and 1503-90, but not in 1510-80 compared to 'Aseong'. In addition, 2,2-Diphenyl-1-picryl hydrazyl (DPPH) radical scavenging activity was higher in the 1502-56 compared to 'Aseong'. Considering these results, two AYT lines, 1502-56 and 1503-90 were selected as short stem lines with high calycosin-7-O-${\beta}$-D-glucoside and calycosin content. Conclusions: Taken together, Two short stem lines were identified in this study. In our future study, regional yield trial (RYT) will be conducted using these selected lines to develop new cultivars.
The present study was carried out in order to assess the protective effects of calycosin-7-O-$\beta$-D-glucopyranoside, isolated from Astragali radix (AR), on hyaluronidase (HAase) and the recombinant human interleukin-$1\beta$ (IL-$1\beta$)-induced matrix degradation in human articular cartilage and chondrocytes. We isolated the active component from the n-butanol soluble fraction of AR (ARBu) as the HAase inhibitor and structurally identified as calycosin-7-O-$\beta$-D-glucopyranoside by LC-MS, IR, ${1}^H$ NMR, and ${13}^C$ NMR analyses. The $IC_{50}$ of this component on HAase was found to be 3.7 mg/ml by in vitro agarose plate assay. The protective effect of ARBu on the matrix gene expression of immortalized chondrocyte cell line C28/I2 treated with HAase was investigated using a reverse transcription polymerase chain reaction (RT-PCR), and its effect on HAase and IL-$1\beta$-induced matrix degradation in human articular cartilage was determined by a staining method and calculating the amount of degraded glycosaminoglycan (GAG) from the cultured media. Pretreatment with calycosin-7-O-$\beta$-D-glucopyranoside effectively protected human chondrocytes and articular cartilage from matrix degradation. Therefore, calycosin-7-O-$\beta$-D-glucopyranoside from AR appears to be a potential natural ant-inflammatory or antii-osteoarthritis agent and can be effectively used to protect from proteoglycan (PG) degradation.
This study was carried out to obtain the basic information for isoflavonoid contents that can be used to index Astragalus membranaceus B. cultivated in the Republic of Korea and China. Isoflavonoid contents in Astragalus membranaceus B. which were cultivated in various areas (Jecheon, Jeongseon, Yeongju, and Taebaek in Korea, and China) and ages (1-year-old, 3-years-old) were determined. Calycosin and formononetin as major constituents were determinated by HPLC method in Astragalus membranaceus B. The results show that there were no statistically significant differences for the average contents of isoflavonoids among 1-year-old and 3-years-old. However, isoflavonoid contents were significant differences according to the cultivation areas. HPLC analysis showed that the calycosin content of 1-year-old at Jeongseon was the highest level of $0.090{\pm}0.002%$ and that of 1-year-old at Yeongju was the lowest level of $0.010{\pm}0.001%$. The highest level of formononetin content was $0.050{\pm}0.001%$ of 1-year-old at China, while the lowest level was $0.020{\pm}0.001%$ of 1-year-old at Yeongju. These results strongly suggest that contents of isoflavonoid in Astragalus membranaceus B. might be quite different with respect to the cultivation areas.
This study analyzed the physicochemical characteristics of Astragalus membranaceus (AM) fermented with seven different mushroom mycelia. Physicochemical characteristics, such as contents of moisture, pH, total reducing sugars, free sugar, and isoflavonoid, were investigated. The moisture content was increased in most of the samples. The pH values of AM fermented with Phellinus linteus and Flammulina velutipes were increased, while the pH of other samples were similar to that of non-fermented AM. The reducing sugar content was in the range of 211.69~391.74 mg/100 g. The extraction yield using water was higher than that when extracted with 80% ethanol. The free sugar content was increased through fermentation with mushroom mycelia. However, the glucose contents of the 80% ethanol and water extracts were decreased. Finally, the calycosin and formononetin contents in 80% ethanol and water extracts of AM fermented with Phellinus linteus were 2,549.24 mg/g, and 827.66 mg/g for calycosin, and 1,366.69 mg/g and 221.28 mg/g for formononetin, respectively. These results suggest that fermentation with mushroom mycelia could be used to increase the bioactivity of AM. The mycelium-fermented AM might be a valuable source of functional material and edible resource for industry.

This study has been conducted to establish the optimal extraction process and HPLC analysis method for the determination of marker compounds as a part of the materials standardization for the development of health functional food materials from Astragali radix. Five extraction conditions including the shaking extraction at room temperature and the reflux extraction at $85^{\circ}C$ with 30%, 50% and 95% ethanol were evaluated. Reflux extraction with 50% ethanol showed the highest extraction yield as $27.27{\pm}2.27%$, while the extraction under reflux with 95% ethanol showed significantly the lowest yield of $10.55{\pm}0.24%$. The quantitative determination methods of calycosin-7-O-${\beta}$-D-glucoside and calycosin as marker compounds of Astragali radix extracts were optimized by HPLC analysis using a Thermo Hypersil column ($4.6{\times}250mm$, $5{\mu}m$) with the gradient elution of water and acetonitrile as the mobile phase at the flow rate of $0.8mLmin^{-1}$ and a detection wavelength of 230nm. The HPLC/UV method was applied successfully to the quantification of two marker compounds in Astragali radix extracts after validation of the method with the linearity, accuracy and precision. The contents of calycosin-7-O-${\beta}$-D-glucoside and calycosin in 50% ethanol extracts by reflux extraction were significantly higher as $1,700.3{\pm}30.4$ and $443.6{\pm}8.4{\mu}g-1$, respectively, comparing with those in other extracts. The results indicate that the reflux extraction with 50% ethanol at $85^{\circ}C$ is optimal for the extraction of Astragali radix, and the established HPLC method are very useful for the evaluation of marker compounds in Astragali radix extracts to develop the health functional material from Astragali radix.
Five isoflavones were isolated from the root of Caragana microphylla Lam. and the structures have been identified by means of chemical and physical methods. The isoflavones are 7-hydroxy-4'-methoxyisoflavone(formononetin), $form ononetin-7-O-{\beta}-glucoside(ononin)$, 7-hydroxy-3', 4'-methylenedioxyisoflavone(pseudobaptigenin), $7-O-{\beta}-glucuopyranosyloxy-3'- hydroxy-4'-methoxyisoflavone(calycosin-7-O-{\beta}-gluc-oside)$ and maackiain.

The major active components of Astragalus membranaceus (AM) are isoflavones, which exist in the form of various glycosides. Nuruk is a traditional fermentation starter in Korea, and is suitable for the biotransformation of isoflavone glycosides because it contains various microorganisms and enzymes. This study was performed to determine changes in the isoflavones and antioxidant properties of AM fermented (AF) with nuruk over 24 hours. AF was sampled after 0, 2, 4, 6, 12, 18, and 24 h of fermentation, and calycosin 7-glucoside, ononin, calycosin, and formononetin content, and the antioxidant properties of AF were analyzed. The total phenolic content increased with fermentation time, and the ABTS radical scavenging activity increased until 6 h of fermentation and then decreased. During fermentation, the isoflavone glycosides decreased significantly as fermentation time increased. The contents of calycosin and formononetin, which are aglycons of calycosin-7-glucoside and ononin, increased from 100.54 ㎍/g to 276.84 ㎍/g and from 56.29 ㎍/g to 123.04 ㎍/g, respectively, at 18 h of fermentation. Significant correlations were observed between fermentation time, isoflavone content, and antioxidant properties. The results of this study showed that fermentation with nuruk is suitable for the biotransformation of isoflavones in AM.
In order to isolate hyaluronidase (HAase) inhibitor from Astragali radix (AR), dried roots were extracted with ethanol, prior to sequential fractionations with n-hexane, chloroform, ethyl acetate, n-butanol, and aqueous fractions. The n-butanol soluble fraction was found to exhibit the most pronounced inhibitory effect (68%) on HAase, and the active components were separated using various chromatographic methods, including column chromatography and preparative HPLC. The active component was isolated from the n-butanol soluble fraction of AR and was structurally identified as calycosin-7-O-${\beta}$-D-glucopyranoside by LC-MS, IR, $^1H$ NMR, and $^{13}C$ NMR analysis. The $IC_{50}$ of calycosin-7-O-${\beta}$-D-glucopyranoside's HAase activity was found to be 3.7 mg/mL.
From the 70% EtOH extract of the roots of Astragalus membranaceus (Leguminosae), eleven flavonoid derivatives and a lignan, were isolated and identified as liquiritigenin (1), daidzein (2), formononetin (3), sophorophenolone (4), calycosin (5), methylnissolin (6), isomucronulatol (7), isomucronulatol 7-O-glucoside (8), methylnissolin 3-O-glucoside (9), calycosin 7-O-glucoside (10), (+)-syringaresinol O-${\beta}$-D-glucoside (11), and isomucronulatol 7,2'-di-O-glucoside (12), by spectroscopic methods. This is the second report of the isoflavonoid derivatives sophorophenolone (4) and isomucronulatol 7,2'-di-O-glucoside (12) from a natural source, as well as the first report of compounds liquiritigenin (1), daidzein (2) and (+)-syringaresinol O-${\beta}$-D-glucoside (11) from the species A. membranaceus.
