Determination of Organic Acids in Tobacco Leaves by HPLC (HPLC를 이용한 잎담배 중 유기산 함량 분석)

  • This study was carried out to improve the analytical methods for determination of organic acids in tobacco leaf by HPLC. The samples for HPLC analysis were pre-treated by means of SPE. The calibration curve for each acid was linear and $R^2$ values ranged from 0.9990 to 1.0000. The limit of detection were obtained from the calibration curves and their values were between 1.39 to $4.87{\mu}g/mL$. Recovery rates of organic acids were between 88.6 % to 98.5 %. The concentrations of organic acids among the various tobacco leaves were compared to the concentration of organic acids, were in the order oriental, burley, flue-cured tobacco. In the case of flue-cured and oriental tobacco leaves, the order of concentration of organic acids was malic acid, oxalic acid, citric acid. But in the case of burley tobacco leaves, the order of concentration of organic acids was citric acid, oxalic acid, malic acid.

Calibration of Parameters for Predicting Hysteretic Behavior of Diagonally Reinforced Concrete Coupling Beams (반복하중을 받는 대각보강 콘크리트 연결보의 이력거동 예측을 위한 매개변수 결정방법)

  • The coupled shear wall system with coupling beams is an efficient structural system for high-rise buildings because it can provide excellent ductility and energy dissipation to the buildings. The objective of this study is to simulate the hysteretic behavior of diagonally reinforced concrete coupling beams including pinching and cyclic deteriorations in strength and stiffness using a numerical model. For this purpose, coupling beams are modeled with an elastic beam element and plastic spring element placed at the beam ends. Parameters for the analytical model was calibrated based on the test results of 6 specimens for diagonally reinforced concrete coupling beams. The analytical model with calibrated model parameters is verified by comparing the hysteretic curves obtained from analysis and experimental tests.

Measurement of Transient Current by using the Rogowski Coil (로고우스키코일을 이용한 과도전류의 측정)

  • This paper presents the operation principle and design rule of the Rogowski coil which can measure the transient current and describes the calibration and application experimental results for performance evaluation. It is obtained that the response curves of the Robowski coil with the turns of 300 and the passive integrator to sinusoidal input give a good linearity up to the frequency of 500 [kHz] and the current measurement system gaving the Rogowski coil is the frequency bandwidth of 40 [Hz]~700 [kHz]. As an application experiment for the fabricated modeling power transmission line, the impulse current, which limitates the direct lightning return stroke to overhead ground wire, is measured by the Rogowski coil and its fast Fourier transformation is carried out. The equivalent circuit of the Rogowski coil considering the stray capacitances is proposed, and the theoretical analysis is in good agreement with the measurement results. Also, it is found that for high frequency domain the stray capacitance such as a distributed capacitance to the shield and the capacitance between windings of coil should be considered in designing the Rogowski coils since the resonance originates from the stray capacitance and the self-inductance of the Rogowski coil.

Validation and measurement uncertainty of HPLC-UV method for quercetin quantification in various foods

  • The purpose of this study was to validate a high-performance liquid chromatography (HPLC) method for the quantitative analysis of quercetin in various foods. The method was based on HPLC-UV (360 nm). The method was validated using candy, beverage, and sausage which were tested for specificity, linearity, limit of detection (LOD), limit of quantification (LOQ), precision, and accuracy, and the measurement uncertainty was assessed. Matrix-matched calibration was also applied. The calibration curves (0.5-50 mg/L) showed good linearity (r2≥0.9998). LOD and LOQ ranged from 0.15 to 0.31 mg/kg and from 0.44 to 0.93 mg/kg, respectively. The average accuracy and precision at 0.5, 2.5, and 10 mg/kg ranged from 84.3 to 102.0% and 0.7 to 3.0 relative standard deviation (RSD%), respectively. This study confirmed the applicability of the proposed method by applying it to commercial products, such as teas and beverages. Thus, the proposed analytical method is suitable for quantifying quercetin in various foods.

Application of an Automated Time Domain Reflectometry to Solute Transport Study at Field Scale: Experimental Methodology and Calibration of TDR (시간영역 광전자파 분석기(Automatic TDR System)를 이용한 오염물질의 거동에 관한 연구: 실험방법 및 검정)

  • Field scale experiments using an automated 144-channel TDR system were conducted which monitored the movement of solute through unsaturated loamy soils. The experiments were carried out on two different field plots of 0.54 ha to study the vertical movement of solute plume created by applying a square pulse of $CaCl_2$ as a tracer. The residence concentration was monitored at 24 locations on a transect and 5 depths per location by horizontally-positioning 50 cm long triple wire TDR probes to study the heterogeneity of solute travel times and the governing transport concept at field scale. This paper describes details of experimental methodology and calibration aspects of the TDR system. Three different calibration methods for estimation of solute concentration from TDR-measured bulk soil electrical conductivity were used for each field site. Data analysis of mean breakthrough curves (BTCs) and parameters estimated using the convection-dispersion model (CDE) and the convective-lognormal transfer function model (CLT) reveals that the automated TDR system is a viable technique to study the field scale solute transport providing a normal distribution of resident concentration in a high resolution of time series, and that calibration method does not significantly affect both the shape of BTC and the parameters related to the peak travel time. Among the calibration methods, the simple linear model (SLM), a modified version of Rhoades' model, appears to be promising in the calibration of horizontally-positioned TDR probes at field condition.

Establishment of Target Water Quality for TOC of Total Water Load Management System (오염총량관리제도의 TOC 목표수질 설정 방안)

  • In this study, it was proposed that a method of setting the target water quality for TOC using the watershed model and the load duration curves to manage non-biodegradable organics in the total water load management system. To simulate runoff and water quality of the watershed, the HSPF model is used which is appropriate for urban and rural areas. Additionally, the load duration curve is used to reflect the variable water quality correlated with various river flow rates in preparing the TMDL plans in the U.S. First, the model was constructed by inputting the loads calculated from the pollutant sources in 2015. After the calibration and verification process, the water quality by flow conditions was analyzed from the BOD and TOC simulation results. When the BOD achieved the target water quality by inputting the target year loads for 2020, the median and average values of TOC were proposed for the target water quality. The provisional method of TOC target water quality for the management of non-biodegradable organics, which is one of the challenges of the total water load management system, was considered. In the future, it is expected to be used as basic data for the conversion of BOD into TOC in the total water load management system.

Purification of pivalic acid : its evaluation as a temperature standard reference material (온도표준 기준 물질로의 개발을 위한 pivalic acid의 고순도 정제)

  • The suitability of pivalic acid was confirmed as a secondary temperature standard material. Triple-point cells of pivalic acid were obtained by using combined process of vacuum distillation with zone refining. A detailed description of the purification process was given. The melting curves were used as criteria for determination of purities of pivalic acid. Triple points of these cells with purity of 99.9997% were measured to be $35.956\pm 0.003^{\circ}C$ by using the melting plateau curves. Thus the triple point cells of pivalic acid appear to be able to use for the calibration of thermistor thermometers with moderate precision.

Flare and Starspot-induced Variabilities of Red Dwarf Stars in the Open Cluster M37: Photometric Study on Magnetic Activity

  • Flare and rotational variabilities induced by stellar activity are important for studying the effect of magnetic fields on the evolution of red dwarf stars. The level and frequency of magnetic activity in these stars have a different aspect at every moment of the observations due to the effect of age-rotation relation. The use of both tracers is thus essential to have a relatively homogeneous set of stellar activity data for statistical studies. The archival light curves and imaging data of the open cluster M37 taken by MMT 6.5m telescope were used for this work. In order to achieve much more accurate photometric precisions and also to make the most efficient use of the data, the entire imaging database were re-analyzed with our new time-series photometry technique and carefully calibration procedures. Based on the new light curves, we study, for the first time, a variety of aspects of those two variabilities in red dwarfs and their relation to magnetic activity. In this talk, we present all observational evidences that support the idea that the strength of magnetic activity is closely connected with the rotation rate of a star and its evolutionary status (age-activity-rotation paradigm). In conclusion, we suggest future directions to improve our understanding of stellar activity in cool stars with photometric time-series data.

Analysis of Microtoxins in the Nakdong River Watershed

  • The different methods such as HPLC, indirect- and direct-ELISA were employed for the analysis of microtoxins and the results of each method were compared in terms of the detection limit and accurary. Three toxins, microcystin-RR, -LR and -YR were clearly separated by HPLC using 0.05 M methanol and phosphate buffer used as a solvent system. The calibration curves for the toxins were linear in the range of 5 ng to 50 ng. The standard curves for the immunoassay of microcystin obtained by direct and indirect ELISA are compared. The linear responses of inhibitions of binding by microcystin in the direct and indirect competitive ELISA were in the range of 10 ng to 1000 ng and 50 pg to 160 pg, respectively. Distribution of microtoxins at 11 sites in the Nakdong river and several lakes in Korea was also studied. The most dominant microcystin variant in the test sites was found to be microcystin-RR.

  • In this study, powder compaction of AZO (alumina doped zinc oxide) powder was performed with a MTS 810 test system using a cylindrical die having a diameter of 10mm. Pressure-density curves were measured based on the load cell and displacement of the punch. The AZO powder compacts with various densities were formed to investigate the mechanical properties such as fracture stress of the AZO powder as a function of the compact density. Two types of compression tests were conducted in order to estimate the fracture stress using different loading paths: a diameteral compression test and a uniaxial compression test. The pressure-density curves of the AZO powder were obtained and the fracture stress of the compacted powders with various densities was estimated. The results show that the compact pressure dramatically increases as the density increases. Based on the experimental results, calibration of the modified Drucker-Prager/Cap model of the AZO powder for use in FE simulations was developed.
