Microarray analysis of gene expression has become a powerful approach for exploring the biological effects of drugs, particularly at the stage of toxicology and safety assessment. Acetaminophen (APAP) has been known to induce necrosis in liver, but the molecular mechanism involved has not been fully understood. In this study, we investigated gene expression changes of APAP using microarray technology. APAP was orally administered with a single dose of 50 mg/kg or 500 mg/kg into ICR mice and the animals were sacrificed at 6, 24 and 72 h of APAP administration. Serum biochemical markers for liver toxicity were measured to estimate the maximal toxic time and hepatic gene expression was assessed using high-density oligonucleotide microarrays capable of determining the expression profile of >30,000 well-substantiated mouse genes. Significant alterations in gene expression were noted in the liver of APAP-administered mice. The most notable changes in APAP-administered mice were the expression of genes involved in apoptosis, cell cycle, and calcium signaling pathway, cystein metabolism, glutatione metabolism, and MAPK pathway. The majority of the genes upregulated included insulin-like growth factor binding protein 1, heme oxygenase 1, metallothionein 1, S100 calcium binding protein, caspase 4, and P21. The upregulation of apoptosis and cell cycle-related genes were paralleled to response to APAP. Most of the affected gene expressions were returned to control levels after 72 hr. In conclusion, we identified potential hepatotoxicity makers, and these expressions profiling lead to a better understanding of the molecular basis of APAP-induced hapatotoxicity.
The purpose of this study was to investigate the correlations among the anthropometry, serum lipid levels and nutrient intake in Korean female university students. The subjects were 119 female students at a university located in Incheon. This study was conducted using a self-administered questionnaire. Anthropometric data were measured and blood lipid levels were analyzed. Nutrient intake collected from 3 day-recalls was analyzed by the Computer Aided Nutritional Analysis Program. The data were analyzed by SPSS 10.0 program. Average age, height and weight of the subjects were 20.9 years, 160.1cm and 54.3kg, respectively. Average serum TG (triglyceride), total cholesterol, HDL-C (high density lipoprotein-cholesterol) and LDL-C (low density lipoprotein-cholesterol) levels of the subjects were 69.47mg/dl, 146.85 mg/dl, 50.49mg/dl and 82.52mg/dl, respectively. Average AI (atherogenic index) of the subjects was 2.03, which was in the normal range based on risk values. Average intake of most nutrients except protein, vitamin B$_1$, vitamin C and phosphorus were lower than the Korean RDA. Especially calcium and iron intakes of the subjects were under 65% of the Korean RDA. Serum TG, total cholesterol and LDL-C levels were negatively correlated with DBP (diastolic blood pressure). HDL-C/LDL-C and HDL-C/total cholesterol were positively correlated with height. Age was positively correlated with phosphorus intake. DBP of the subjects was positively correlated with calcium and iron intakes. Serum TG level was positively correlated with total cholesterol, HDL-C, LDL-C and AI, while negatively correlated with HDL-C/total cholesterol. Total cholesterol level was positively correlated with HDL-C, LDL-C and AI, while negatively correlated with HDL-C/LDL-C, HDL-C/total cholesterol. HDL-C level was positively correlated with LDL-C, HDL-C/LDL-C and HDL-C/total cholesterol, while negatively correlated with AI. LDL-C level was negatively correlated with HDL-C/LDL-C and HDL-C/total cholesterol, while positively correlated with AI HDL-C/LDL-C ratio was positively correlated with HDL-C/total cholesterol and AI. HDL-C/total cholesterol was negatively correlated with AI. Fat intake was positively correlated with total cholesterol, HDL-C level, and vitamin B$_2$ intake was positively correlated with TG, HDL-C/LDL-C. 우리나라 석회암지대는 한반도에서 그다지 넓지 않은 좁은 지역에 집중적으로 분포하고 있고, 독특한 생태계 지역이므로 석회암지대 산림관리를 위한 기초자료를 제공하고자 산림식생의 유형분류, 산림토양의 이화학적 특성, 환경요인과 식생유형의 상관관계 등을 구명한 결과, 식생은 굴참나무군락군과 신갈나무군락군의 2개 군락군으로 크게 분류되었고, 기타 하위식생단는 4개 군락, 6개 군, 2개 소군으로 세분되어 총 8개의 식생단위로 나타났다. 석회암지대 조사지의 토성은 미사질양토로 나타났고, A층의 토양산도는 7.55로 나타났으며, 치환성 양이온인 칼슘($Ca^{2+}$)과 마그네슘($Mg^{2+}$) 함량은 각각 평균 $26.04cmol_{c}/kg$와 $2.93cmol_{c}/kg$으로서 매우 높게 나타났다. 굴참나무군락군은 pH가 높고, 사면경사가 급하고, 노암율이 높으며, Ca 이온의 농도가 높은 입지에 주로 분포하는 경향이었고, 신갈나무군락군은 표고가 높은 입지에 분포하는 경향이었다.
BACKGROUND: Globally, large amounts of phosphogypsum (PG), which is a by-product of the phosphate fertilizer production, is deposited in open areas. As PG contains calcium, phosphate, and sulphate, it can be used as a soil amendment in farmlands. This study was conducted to investigate the effects of PG application on properties of field soil and yield and quality of garlic (Allium sativum L.), and to seek appropriate level of PG application into the field. METHODS AND RESULTS: This experiment was conducted by applying PG at four different levels that were adjusted based on 65% calcium base saturation in the field soil: 0% (control), 50% (PG50, 100 kg/10a), 100% (PG100, 200 kg/10a), and 150% (PG150, 300 kg/10a). Following cultivation, soil electrical conductivity (EC), organic matter, available P and exchangeable Ca increased, whilst soil pH decreased. With increase in PG application level, soil EC and exchangeable Ca increased. PG application increased concentrations of water soluble Ca and SO4 across the soil profile, especially in PG150. The highest yield of garlic was found in PG100 treatment. The plant's uptake for N, P, Ca, and S increased by PG applications, but that for K decreased. Moreover, concentrations of S-related amino acids such as cysteine and methionine in garlic increased by PG applications. The increased content of nutrients and amino acids with PG supply might improve yield, quality, and favor of the crop. CONCLUSION: Overall, PG application at 200 kg/10a into a field had the best effect on improving soil fertility as well as yield and quality of garlic. Further studies are required to maximize efficiencies of PG supply in soil management and production of various crops.
As the consumption of wheat has increased recently, the number of people who have digestive problems resulting from gluten in wheat has also increased. Teff has an attractive nutritional profile, as it not only gluten-free but also high in dietary fiber, protein, iron, and calcium. Seven samples were prepared for this study. The quality characteristics of gluten-free noodles were evaluated based on pH, salinity, water absorption, turbidity, color, texture properties, tensile strength, and SEM. The pH value was the highest in TF100 with a pH of 6.66 and the lowest in the control with a pH of 6.42. Salinity showed no significant difference among all samples, and it ranged from 0.02~0.04% (p<0.05). Water absorption was the highest in TFX with a value of 66.11%, and the lowest in the control with a value of 44.81%. Turbidity showed no significant difference among all samples, and it ranged from 0.14~0.21 O.D. (p<0.05). While the lightness and yellowness values decreased with an increase in teff flour content, the redness value tended to decrease. The color difference value was the highest in the sample group without gluten. Based on the texture profile analysis, the hardness was highest in the control with a value of 46.74 N and lowest in TF100 with a value of 18.34 N. The springiness showed no significant difference among all samples. The cohesiveness was highest in the control with a value of 0.92 N. The chewiness decreased with an increase in teff flour content. Although the control with gluten had the highest tensile strength at $3.42kg/cm^2$, TFX had considerable tensile strength at $2.30kg/cm^2$. This study demonstrated the processability of gluten-free noodles using teff flour.
The molecular understanding of resistance and susceptibility of host plants to scab, a most threatful disease to pome fruit production worldwide, is very limited. Comparing resistant line '93-3-98' to susceptible one 'Sweet Skin' at seven time points of 0, 0.5, 1, 2, 3, 4, 8 days post inoculation, RNA-sequencing data derived from infected and mock-inoculated young leaves were analyzed to evaluate the tolerant response and to mine candidate genes of pear to the scab pathogen Venturia nashicola. Analysis of the mapped reads showed that the infection of V. nashicola led to significant differential expression of 17,827 transcripts with more than 3-fold change in the seven pairs of libraries, of which 9,672 (54%) are up- and 8,155(46%) are down-regulated. These included mainly receptor (NB-ARC domains-containing, CC-NBS-LRR, TIR-NBS-LRR, seven transmembrane MLO family protein) and transcription factor (ethylene responsive element binding, WRKY DNA-binding protein) related gene. An arsenal of defense response of highly resistant pear accessions derived from European pear was probably supposed no sooner had V. nashicola infected its host than host genes related to disease suppression like Polyketide cyclase/dehydrase and lipid transport protein, WRKY family transcription factor, lectin protein kinase, cystein-rich RLK, calcium-dependent phospholipid-binding copine protein were greatly boosted and eradicated cascade reaction induced by pathogen within 24 hours. To identify transcripts specifically expressed in response to V. nashicola, RT-PCRs were conducted and compare to the expression patterns of seven cultivars with a range of highly resistant to highly susceptible symptom. A DEG belonging to the PR protein family genes that were higher expressed in response to V. nashicola suggesting extraordinary role in the resistance response were led to the identification. This study provides the first transcriptional profile by RNA-seq of the host plant during scab disease and insights into the response of tolerant pear plants to V. nashicola.
The purpose of this study was to investigate self-perception of health and body image, blood lipid profiles and nutrient intake of adolescents. The subjects were 686 adolescent boys (379 middle school students, 307 high school students) and 613 adolescent girls (272 middle school students, 341 high school students) in Incheon. This cross-sectional survey was conducted by a self-administered questionnaire. Fasting blood samples were obtained and analyzed for triglyceride, total cholesterol and LDL- and HDL-cholesterol. Nutrient intakes collected from 3 day-recalls were analyzed by the Computer Aided Nutritional Analysis Program. Statistical analysis was conducted using SPSS 10.0 program. Average age, height and weight of male middle school students were 13.8 years, 163.5cm, 56.0kg and those of male high school students were 16.5 years, 171.7cm, 64.3kg. In female, average age, height and weight of middle school students were 13.7 years, 157.1cm, 51.5kg and those of high school students were 16.6 years, 159.7cm, 56.8kg. Female middle school students perceived their health status significantly better compared to female high school students (p < 0.001). Female high school students perceived themselves significantly fatter compared to female middle school students (p < 0.05). Average TG level of female middle school students was significantly higher compared to female high school students (p < 0.01). Nutrient intakes of the male and female students except phosphorus were lower than the Korean RDA. Especially, calcium and iron intakes of male and female students were under the 65% of the Korean RDA. Therefore, proper nutrition education is required for Korean adolescents to encourage desirable food habits and maintain nutritional lipids status. (J Community Nutrition 5(1) : 3∼12, 2003)

Severe hypercalcemia is rarely encountered in children, even though serum calcium concentrations above 15-16 mg/dL could be life-threatening. We present a patient having severe hypercalcemia and azotemia. A 14-year-old boy with no significant past medical history was referred to our hospital with hypercalcemia and azotemia. Laboratory and imaging studies excluded hyperparathyroidism and solid tumor. Other laboratory findings including a peripheral blood profile were unremarkable. His hypercalcemia was not improved with massive hydration, diuretics, or even hemodialysis, but noticeably reversed with administration of calcitonin. A bone marrow biopsy performed to rule out the possibility of hematological malignancy revealed acute lymphoblastic leukemia. His hypercalcemia and azotemia resolved shortly after initiation of induction chemotherapy. Results in this patient indicate that a hematological malignancy could present with severe hypercalcemia even though blast cells have not appeared in the peripheral blood. Therefore, extensive evaluation to determine the cause of hypercalcemia is necessary. Additionally, appropriate treatment, viz., hydration or administration of calcitonin is important to prevent complications of severe hypercalcemia, including renal failure and nephrocalcinosis.
본 연구에서는 분리대두단백질에 단백질 가수분해효소를 처리하여 대두단백질에 가수분해적 변성을 야기하여 그 기능성을 변화 고찰하고 실제 식품에의 이용방법으로 효소처리된 두유로 두부를 만들어 물성실험과 관능평가를 실시하였다. 장엽에서 SPI를 제조한 후 15분간 bromelain으로 처리하여 2.7% 가수분해된 MSPI를 만들어 기능특성을 측정 비교하였다. MSPI는 전 pH범위에서 용해도가 증가하였고, 특히 등전점에서 15% 정도 증가하였다. 유화형성력과 기포팽창력은 증가하였으나 이들의 안정성은 감소하였는데, 특히 기포안정성은 알칼리쪽에서 급격히 감소하였고, 등전점에서는 가장 큰 것으로 나타났다. 표준두유와 변형두유를 혼합하여 제조한 두부로 물성실험과 관능평가를 실시하였을 때 압착실험에서는 변형두부I이 연하고 탄력성 있는 두부를 형성하는 것으로 나타났다. 각 두부의 물성모형은 spring 한개와 Maxwell 모형 세개를 가진 7요소모형으로 동일한 물성모형을 나타내었다. 관능평가에서는 표준두유와 변형두유를 3 : 1의 비율로 혼합하여 제조한 두부의 품질이 가장 우수한 것으로 평가되었다.
본 연구는 phytic acid를 myo-inositol과 무기태인으로 분해시키는 효소인 phytase 생산균주의 분리 및 효소생산의 배지최적하에 관한 것이다. 1차로 calcium phytate를 기질로 함유한 phytase screening 배지를 이용하여 phytase 생산을 나타내는 균주 35가지를 이용하여 분리한 후, sodium phytate를 기질로 하여 재현성 있는 phytase 활성을 나타내는 균주 12가지를 선발하였다. 12개의 균주 중 BHI broth에서 배양한 조효소액의 phytase 활성이 가장 우수한 것으로 나타난 YH100을 선발하여 주사현미경 관찰, 16S rRNA sequence 분석, GC conten(mol%) 조성, 지방산 분석, API 20E kit를 이용한 test 결과 Enterobacter cloacae로 동정되어, 이 균주를 Enterobacter cloacae YH100이라 명명하였다. Enterobacter cloacae YH100에 의한 phytase 생산을 위한 최적 배지 조성을 파악한 결과 glucose 2.0%(w/v), peptone 1.0%(w/v), beef extract 1.0%(w/v), KCI 0.1%(w/v), sodium phytate 0.1%(w/v)로 나타났다.
