한국산 홍화씨분말 보충식이의 급여가 골절된 흰쥐의 골대사지표에 미치는 영향 (The Effects of Korean Safflower(Carthamus tinctorious L.) Seed Powder Supplementation Diet on Bone Metabolism Indices in Rats during the Recovery of Rib Fracture)

  • This study was carried out to investigate the effects of Korean safflower(Carthamus. tinctorious L.) seed powder supplementation on bone metabolism during the recovery of rib-fracture induced by surgical operation in rats. Male Sprague-Dawley rats of 10 weeks old, weighing 370$\pm$5g, were divided into two groups after arrival : the control group(C group, AIN-76 semifurified diet) and safflower seed group(S group, AIN-76 semifurified diet+10% safflower seed powder). They were fed experimental diets for 12 days before the rib fracture operation and for 30 more days after the operation. A number 9 rib was fractured surgically and sham-operation was also performed. After the rib-fracture operation, rats were sacrificed on the 8th, 11th, 16th, 21st, and 30th day, respectively. The body weights were decreased on the 8th day after operation in both groups but recovered gradually thereafter. The fractured sections of the S group were more strongly adhered and more rapidly repaired than those of C group. Effects of safflower seed powder supplementation on indices of bone metabolism during the repairing of rib-fracture can be summarized as following. There were no differences between C and S group. In the levels of PTH and calcitonin, which regulate the concentration of plasma calcium. However, PTH level on the 8th day was significantly higher than that on the 16th day in control group. There were also no differences between groups or within groups in the levels of calcitonin. Osteocalcin levels, one of the bone-formation indices, were similar in two groups. Activities of total-ALP in plasma were highest at the 8th day in both groups and decreased thereafter, except on the 30th day in control group. The activities of bone-specific ALP that is directly related to bone calcification, showed higher levels in the S group than in the C group throughout the experimental period. They were significantly increased in the S group on the 11th, 16th, and 21st days. In comparisons of plasma calcium and phosphate levels, Ca levels were tended to be lower in the S group than in the C group and P levels showed the opposite trend. It seems that the fractured ribs were more rapidly repaired in rats supplemented with safflower seed powder than in control rats, possibly due to stimulation of bone calcification from increased activity of bone-specific ALP. 신경병증성 통증을 유발한 흰쥐에서 신경손상부위에 따른 배근신경절 및 척수의 신경전달물질의 변동 (The Changes of Immunoreactivity for CGRP and SP in the Spinal Cord and DRG According to the Distance between the DRG and Injury Site of a Peripheral Neuropathic Rat)

  • Peripheral nerve injury sometimes leads to neuropathic pain and depletion of calcitonin gene related-peptide (CGRP) and substance P (SP) in the spinal cord. However, the pathophysiological mechanisms for depletion of CGRP and SP following the neurorathic injury are still unknown. This study was performed to see whether the distribution of immunoreactivity for CGRP and SP in the superficial dorsal horn and dorsal root ganglia(DRG) was related to the distance between the DRG and injury site. To this aim, we compared two groups of rats; one group was subjected to unilateral inferior and superior caudal trunk transections at the level between the S3 and S4 spinal nerves (S34 group) and the other group at the levels between the S1 and S2, between S2 and S3 and between S3 and S4 spinal nerve (S123 group). The transections in both groups equally eliminated the inputs from the tail to the S1-3 DRG, but the distance from the S1/S2 DRG to the injury site was different between the two groups. Immunostaining with SP and CGRP antibody was done in the S1-S3 spinal cord and DRG of the two groups 1 and 12 weeks after the injury. The results obtained are as follows: 1. The immunoreactivity for CGRP and SP in the ipsilateral superficial dorsal horn and DRG decreased 1 and 12 weeks after neuropathic nerve injury. 2. The immunoreactive area of SP and CGRP in the S1 dorsal horn was smaller in the S123 group than in the S34 group, whereas that in the S3 dorsal horn was not significantly different between the two groups. The number of SP-immunoreactive DRG cells decreased on the neuropathic side as compared to the sham group's in all DRGs of experimental groups except the S1 DRG of the S34 group. These results suggest that the amounts of SP and CGRP in the dorsal horn and DRG following neuropathic injury inversely decrease according to the distance between the DRG and injury site.

산란종계 사료 내 이소플라본 및 Casein Phosphopeptide의 첨가가 종란 생산성 및 후기 난각질에 미치는 영향 (Effects of Dietary Isoflavone and Casein Phosphopeptide on Hatching Egg Production and Eggshell Quality in Aged Egg-Type Breeder Hens)

  • 본 실험은 산란종계 노계에서 이소플라본과 CPP의 첨가 급여가 종란 생산성 및 후기 난각질에 미치는 영향과 Ca 대사에 미치는 영향을 검토하기 위한 목적으로 실시하였다. 56주령의 Hy-Line Brown 산란종계 암탉 360수와 수탉 36수를 공시하여 사료 내 Ca을 3.3% 및 3.6% 수준으로 하고 이소플라본과 CPP를 각각 0.2% 및 0.5% 수준으로 첨가한 총 6개의 실험사료(3반복)를 5주간 급여하였다. 주별로 사료섭취량을 조사하였고, 산란율과 난중은 매일 조사하였다. 주별로 생산된 종란으로부터 종란율과 부화율을 조사하였다. 실험사료 급여 후 생산된 계란을 반복구별로 수집하여 난각질 및 난각막 내 glycosaminoglycan과 uronic acid 분석에 이용하였다. 실험 종료 시 반복구별로 평균체중이 비슷한 개체를 선발하여 채혈 후 희생시킨 다음 경골을 적출하였다. 채취한 경골은 화학적 조성의 분석을 위한 시료로 이용하였고, 얻어진 혈액에서 화학성분 및 호르몬 분석을 실시하였다.사료섭취량, 난생산성 및 종란율에서는 처리간에 유의한 차이가 없었다. 사료 내 Ca 수준 및 이소플라본 첨가는 난각강도를 각각 유의하게 개선시킨 것으로 나타났으며, CPP의 첨가에 의해서는 특별한 변화가 관찰되지 않았다. 수정율 및 부화율에서도 처리간에 유의한 차이는 없는 것으로 나타났다. 경골강도 및 경골 내 성분 조성에서 사료 내 Ca 수준 및 이소플라본과 CPP의 첨가 효과는 발견되지 않았다. 혈중 Ca, P, estrogen 및 calcitonin 농도 역시 처리간에 차이가 없는 것으로 나타났다. 본 실험에서는 사료 내 Ca 수준의 증가 및 이소플라본 같은 Ca의 이용성에 영향을 미치는 성분을 적절히 활용함으로서 후기 난각질의 개선을 통해 생산성을 향상시킬 수 있을 것으로 사료된다. 그러나 종란 생산성에 대해서는 이소플라본 및 CPP의 첨가가 크게 영향을 미치지 않았다.

Effect of remifentanil on pre-osteoclast cell differentiation in vitro

  • Background: The structure and function of bone tissue is maintained through a constant remodeling process, which is maintained by the balance between osteoblasts and osteoclasts. The failure of bone remodeling can lead to pathological conditions of bone structure and function. Remifentanil is currently used as a narcotic analgesic agent in general anesthesia and sedation. However, the effect of remifentanil on osteoclasts has not been studied. Therefore, we investigated the effect of remifentanil on pre-osteoclast (pre-OCs) differentiation and the mechanism of osteoclast differentiation in the absence of specific stimulus. Methods: Pre-OCs were obtained by culturing bone marrow-derived macrophages (BMMs) in osteoclastogenic medium for 2 days and then treated with various concentration of remifentanil. The mRNA expression of NFATc1 and c-fos was examined by using real-time PCR. We also examined the effect of remifentanil on the osteoclast-specific genes TRAP, cathepsin K, calcitonin receptor, and DC-STAMP. Finally, we examined the influence of remifentanil on the migration of pre-OCs by using the Boyden chamber assay. Results: Remifentanil increased pre-OC differentiation and osteoclast size, but did not affect the mRNA expression of NFATc1 and c-fos or significantly affect the expression of TRAP, cathepsin K, calcitonin receptor, and DC-STAMP. However, remifentanil increased the migration of pre-OCs. Conclusions: This study suggested that remifentanil promotes the differentiation of pre-OCs and induces maturation, such as increasing osteoclast size. In addition, the increase in osteoclast size was mediated by the enhancement of pre-OC migration and cell fusion.

좌귀환(左歸丸)이 산화적 손상, 염증 및 골절유합 관련 인자에 미치는 영향 (Effects of Joaguihwan (JGH, 左歸丸) Extract on Changes of Anti-oxidation, Anti-inflammatory in RAW 264.7 Cells and on Factors Related with Bone Metabolism in Skull Fractured Rat)

  • Objectives The study was designed to evaluate the healing effects of Joaguihwan (JGH) extract on Anti-oxidation, Anti-inflammatory in RAW 264.7 Cells and factors related with bone metabolism in skull fractured Rat. Methods The fracture healing effect of JGH was measured by scavenging activities of1,1-diphenyl-2-picryl-hydrazyl (DPPH), 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid (ABTS) and nitric oxide (NO) in RAW 264.7 cells. The inhibitory effect against the production of inflammatory mediators including interleukin-$1{\beta}$ (IL-$1{\beta}$), interleukin-6 (IL-6), tumor necosis factors-${\alpha}$ (TNF-${\alpha}$) expression was inhibited in RAW 264.7 cells was experimented using JGH. The effects of JGH on healing fractured rats was measured by osteocalcin, calcitonin, CTXII, TGF-${\beta}$, BMP-2, Insulin, ALP in the serum. and was checked every 3 weeks from 0 week to 6week using x-ray. Results 1. DPPH free radica and ABTS scavenging activity of JGH were increased according to concentration of JGH in RAW 264.7 Cells. 2. In the experiment, NO, IL-$1{\beta}$, IL-6, TNF-${\alpha}$ all showed decrease, in general. Especially NO and IL-$1{\beta}$ showed significantly decrease at a concentration of 10, 100 (${\mu}g/ml$). 3. In the production of osteocalcin in the serum, JGH 200, 400 mg/kg experimental group showed significant increased effect at 2 weeks. 4. In the production of calcitonin in the serum. JGH 200 mg/kg experimental group showed significant increased effect at 4, 6 weeks. JGH 400 mg/kg experimental group showed significant increased effect at 2, 4, 6 weeks. 5. In the production of CTX, TGF-${\beta}$, BMP-2 in the serum, experimental group showed increased effect. but no significant effect. 6. In the production of insulin in the serum. JGH 200, 400 mg/kg experimental group showed significant decrease effect at 2, 4, 6 weeks. 7. In the production of ALP in the serum. JGH 200 mg/kg experimental group showed significant increased effect at 2, 4, 6 weeks. JGH 400 mg/kg experimental group showed significant increased effect at 4, 6 weeks. 8. In the change of X-ray, the experimental group showed better healing effects on skull fractured rats than control group. Conclusions From above results, JGH showed healing effect on Anti-oxidation, Anti-inflammatory in RAW 264.7 Cells, factors related with bone metabolism in the serum of skull fractured rat and x-ray, which is expected to be applied in clinics.

출생후 성장발육에 따른 흰쥐 유곽유두의 CGRP 면역양성 신경분포변화 (POSTNATAL DEVELOPMENT OF CGRP-IMMUNOREACTIVE NERVE DISTRIBUTION OF RAT CIRCUMVALLATE PAPILLA)

  • 출생 후 성장 발육에 따른 흰쥐 유곽유두의 신경분포 및 변화를 관찰하기 위하여 생후 1, 5, 10, 15, 20, 25, 35일 및 완전히 성장한 흰쥐를 대상으로 calcitonin gene-related peptide (CGRP)를 이용한 면역조직화학적 방법으로 관찰하여 다음과 같은 결과를 얻었다. CGRP-면역양성 신경섬유들이 혀의 양측에서부터 유두 내로 진입하여 성장발육에 따라 그 분포밀도가 증가하다가 10일군부터는 상피의 직하방에 특히 풍부하게 분포하여 subepithelial plexus를 형성하고 있었다. 1일군에서는 CGRP-면역양성 신경섬유의 특징적인 양상인 염주알 모양이 명확히 관찰되었으나 그 이후로는 염주알 모양은 다소 감소하였고 하나의 선으로 보이는 경우도 있었다. 상피 내로 진입하는 CGRP-면역양성 신경섬유들은 5일군에서 드물게 관찰되었으며 taste pore를 가진 성숙한 미뢰가 처음으로 관찰되는 10일군에서 그 수가 증가하여 출생 후 15일군에서는 상피 전반에 걸쳐 풍부하게 관찰되었다. 그러나 그 이후 20일군부터는 미뢰가 존재하는 trench wall의 하방 2/3부위의 CGRP-면역양성 신경섬유는 감소하고 유두의 top surface와 trench wall의 상방 1/3에 CGRP-면역양성 신경섬유가 제한적으로 관찰되었다.

흰쥐대구치 치수노출후 치수조직내 CGRP함유 신경섬유의 분포에 관한 면역조직화학적 연구 (AN IMMUNOHISTOCHEMICAL STUDY ON THE DISTRIBUTION OF CGRP CONTAINING NERVE FIBERS AFTER PULP EXPOSURE IN RAT MOLAR)

  • The purpose of this study was to investigate the distribution of calcitonin gene-related peptide(CGRP) containing nerve fivers after pulp exposure in rats. The Spague-Dawley rats weighing about 250 - 300g were used. The animals were devided into normal control group and experimental groups. Experimental animals were sacrified on 2, 4, 7, 10 days after pulp exposure. The maxillary teeth and alveolar bone were removed and immersed in the 4% paraformaldehyde plus 0.1M phosphate buffer (pH 7.4). Serial frozen $50{\mu}m$ thick sections were cut with a cryostat. In the immunohistochemical staining procedure, the rabbit CGRP antibody was used as a primary antibody. The sections were incubated for 48 hours at $4^{\circ}C$, and placed into biotinylated anti-rabbit IgG as a secondary antibody and incubated in ABC (avidin-biotin complex), The sections were visualized by 0.05% 3.3 diaminobenzidine tetrahydrochloride. The results of this study were as follows: 1. In control group, CGRP containing nerve fibers ran parallel to the long axis of root and reached the coronal pulp. They were distributed on Raschkow plexus under the odontoblastic layer. 2. In 2 day group after pulp exposure, tissue necrosis and acute inflammation occurred and CGRP containing nerve fibers increased. In 4 day group, the necrotic tissue extended to the pulp and CGRP containing nerve fibers were distributed around the inflammation zone. 3. In 7 day group after pulp exposure, pulp necrosis occurred, and in 10 day group, the abscess under the necrotic pulp extended to the root apex area and CGRP containing nerve fibers were not observed in root canals. 4.The sprouting of CGRP nerve fibers was most remarkable at the pulp chamber under injury in 4 day group, and it was found at inflammation zone under the necrotic tissue in 7 day group and the remaining root pulp tissue in 10 day group. As mentioned above, CGRP nerve fibers had a tendency to increase around the inflammatory zone, especially around the acute inflammation tissue, when compared with control group. It is suggested that CGRP nerve fibers maybe related to the control of inflammatory response of pulp tissue.

시약 변경 시 모집단 분포 추이에 따른 결과 분석 (Result Analysis according to Population Distribution Trends when Reagents are Changed)

  • Purpose In this study, the monthly population distribution was calculated for statistical verification of 10 tests (CA19-9, CA15-3, testosterone, PTH, calcitonin, AFP, CEA, CA72-4, PSA, estradiol) with changed reagents, and the trend of change By comparing and analyzing the results, we want to verify the accuracy of the results and improve the reliability of the test. Materials and Methods From June 2021 to February 2023, for the 10 items in which the reagent was changed, the monthly percentage and standard deviation index before and after the reagent change were compared, respectively. At this time, the reference value was arbitrarily set for the comparison group in consideration of the reference value of the test item, and the groups were subdivided and compared, and the standard deviation index allowed range was -2.0 or more and 2.0 or less. Results For CA19-9, CA15-3, AFP, CEA, and calcitonin 5 test items, the change in monthly ratio in all test sections before and after the reagent change was kept constant. On the other hand, for CA72-4, PSA, testosterone, PTH, and estradiol items, the standard deviation index exceeded the acceptable standard after changing the reagent. CA72-4 test items showed an increasing trend in standard deviation index in the test section exceeding the reference value. In the case of the PSA test item, the maintenance ratio of 0.04 ng/mL was significantly decreased after changing the reagent. The testosterone test item had a standard deviation index of -2.5 in the test section exceeding 10.1 ng/mL after changing the reagent, and the standard deviation index of the PTH test item was out of the acceptable range in all test sections. It was confirmed that the estradiol test item showed an overall increase in the result value. Conclusion Through this study, the continuity and accuracy of the test results could be verified. It is considered that the stability of the test can be secured by analyzing the factors affecting the test result and solving the cause for the test item whose standard deviation index is out of the acceptable standard.

소아에서 발생한 갑상선 수질암종 - 1예 보고 - (Cytologic Features of Medullary Carcinoma of the Thyroid Occurring in a Child - A Case Report -)

  • Medullary carcinoma of the thyroid gland is a malignant neuroendocrine tumor arising from calcitonin producing-parafollicular cells. The tumor is clinically divided into sporadic and familial form, constituting about 80% and 20%, respectively. Recently, we experienced a case of unilateral and solitary sporadic medullary carcinoma of the left thyroid gland. The patient was a 9 year-old female, who presented with a palpable mass on the anterior lateral neck of 8 months duration without any familial and personal history of neuroendocrine disease. The cytopathologic findings showed spindle cells and plasmacytoid cells in the background of colloid-like materal. The nuclei were eccentrically located, mildly hyperchromatic and pleomorphic, showing speckled chromatin pattern without nuclear inclusion or prominent nucleoli. The cytoplasm was abundant and had a pale granular cyanophilic appearance. No amyloid materal could be identified.

통증유발 백서에서 미세전류자극이 척수 분절 내 c-fos 및 CGRP 발현에 미치는 영향 (Effects of Microcurrent Stimulation on c-fos and Calcitonin Gene-Related Peptide Expression in the Spinal Cord on Rats Induced Pain)

  • The purpose of study is that we will observe the change of c-fos and CGRP with the immunohistochemistry method and then we will study the effect of microcurrent stimulation following the frequency after inducing pain to rats with capsaicin. The experimental groups were divided by microcurrent application and pain induce. Normal control groups is used in experiment I, the group which we induce pain is used in experiment II, the application group which we induce pain and then the high frequency microcurrent stimulation is used in experiment III, the application group which we induce pain and then the low frequency microcurrent stimulation is used in experiment IV. c-fos was strongly expressed after pain induced 2 hours and positive neurons were decreased from 2 hours. At 7 days, positive neuron recovers to normal range, But c-fos positive neuron of microcurrent stimulation group were decreased from 2 hours. CGRP was strongly expressed after pain induced 24 hours, and positive neurons were decreased from 7 days. These results suggests that microcurrent stimulation therapy effect to control pain according to expression of c-fos and CGRP examined by immunohistochemistry. Also high frequency microcurrent stimulation is more effective than low frequency microcurrent stimulation for controling the pain.
