Examination was made of the urinary metabolite(s) of CKD-712, which is a chiral compound, named S-YS49 derived from higenamine (one component of Aconite spp.) derivatives. First of all, to analyze the metabolite(s) of CKD-712, a simple and sensitive detection method for CKD-712 was developed by using gas chromatography-mass spectrometry GC/MS). Urine was collected from adult male Sprague-Dawley rats 250${\pm}$10g) in metabolic cage for 24hr after oral administration of 100 mg/kg of CKD-712. The recovery of CKD-712 after extraction and concentration with AD-2 resin column was above 90 % from rat urine. The detection limits of CKD-712 in urine was approximately 0.1 ng/mL. It has well been suggested that isoquinoline possessing catechol moiety such as CKD-712 should be subjected to the catechol-O-methyl kransferase activity in vivo. We detected three major peaks of presumed CKD-712 metabolites in the total ion chromatogram obtained from the rat urine sample after oral administration of CKD-712. From these results, it is assumed that the urinary metabolites are mono-methylation in the naphthyl moiety (metabolite I ), methylation at the C-6 or 7 hydroxy group in the isoquinoline moiety and hydroxylation at in the naphthyl moiety (metaboliteII), and methylation at the C-6 or 7 hydroxy group in the isoquinoline moiety (metaboliteIII).
Background: Subcutaneous injection of 5% formalin into the hind paw of the rat produces a biphasic nociceptive response. The second phase depends on changes in the dorsal horn cell function that occur shortly after an initial C-fiber discharge, spinal sensitization, or windup phenomenon. This study was performed to investigate the role of glutamate during spinal sensitization. Methods: Sprague-Dawley rats weighing 200 to 250 g were used for this study. Under light anesthesia (0.5% isoflurane) the rats were segregated in a specially designed cage and $50{\mu}l$ 0.5% formalin was injected subcutaneously in the foot dorsum of right hindlimb. Forty minutes after the formalin injection, the rat was quickly decapitated and spinal cord was removed. The spinal segments at the level of L3 (largest area) was collected and stored in a deep freezer ($-70^{\circ}C$). The mRNA gene expression of N-methyl-D-aspartate receptor (NMDAR) and the metabotropic glutamate receptor subtype 5 (mGluR5) were determined by the polymerase chain reaction. Results: The number of flinches was $19.8{\pm}2.3/min$. at one minute after formalin injection and decreased to zero after then. The second peak appeared at 35 and 40 minutes after formalin injection. The values were $17.8{\pm}2.2$ and $17.2{\pm}3.0/min$. The mRNA gene expressions of NMDAR and mGluR5 were increased by $459.0{\pm}46.8%$ (P < 0.01) and $111.1{\pm}4.8%$ (P > 0.05) respectively at 40 minutes after formalin injection. The increased rate of NMDAR was significantly higher than that of mGluR5 (P < 0.01). Conclusions: From these results it suggested that NMDAR partly contributed to the mechanism of central sensitization after the formalin test but mGluR5 did not.
위 내용물 분석을 위하여 경남 통영시 바다목장 해역내 한국해양연구원 가두리 설치 지역에서 2004년 7월부터 8월까지 채집을 실시하였으며, 분석에 사용된 볼락은 점등, 비점등 총 200개체였다. 점등에 의해 유집된 동물플랑크톤중 볼락의 가장 중요한 먹이생물은 요각류(Copepoda), 단각류(Amphipoda)순으로 나타났으며, 비점등구간에서도 역시 요각류(Copepoda), 단각류(Amphipoda)순으로 나타났다. 계절에 따른 먹이조성의 변화를 살펴보면 점등구간의 경우 비점등구간에 비해 비교적 다양한 먹이생물을 고루 섭이하였으며, 특히 점등에 의해 요각류와 단각류의 출현량이 많아 중간육성 기간중 볼락의 위내용물 중 이들이 차지하는 비율이 비교적 높았으며, 비점등구에서는 전조사기간중 요각류의 출현량이 대부분을 차지하였다. 중간육성 기간중 점등, 비점등구간에서 먹이공급 조건에 따른 성장관계를 살펴보면 동일한 조건에서 비점등구에 비해 점등구에서 사육한 볼락의 성장이 양호한 것으로 나타났다.
The present study is concerned with the demonstration of the influence of long term treatment with caffeine and phenobarbital on pentobarbital sleeping time, gastric secretion, increase rate of body weight and brain and liver weight in rats. The experimental subjects were rats weighing about 140 to 150 g, each of them was isolated in a separate cage. Each group was given 1 ml normal saline solution as control, caffeine 10 mg/kg and phenobarbital 30 mg/kg as experimental groups. All drugs were injected intraperitoneally, daily for 4 weeks. The results obtained are summarized as follows; 1. There was significant difference between before and after injection of drugs (caffeine citrate 10 mg/kg and phenobarbital 30 mg/kg) on pentobarbital sleeping time. The sleeping time of caffeine treated group was delayed (22.4%, p<0.01) significantly compared with that of before injection. The sleeping time of phenobarbital treated group was markedly shortened (93.6%, p<0.001) compared with that of before injection of drugs. 2. The volume, free and total acidity and pH of gastric juice determined five hours after pyloric ligation in fasting rats were not significantly changed in experimental groups compared with control group. However the volume of gastric juice was increased 25% in both caffeine and phenobardital treated group. 3. The increased ratio of body weight revealed no remarkable difference compared with intial body weight. However, caffeine treated group showed markedly increased body weight after first and second week of injection. 4. The brain and liver weight in experimental group showed no significant difference compared with control group (as percentage of body weight).
The effects of barley and oat feeding in table duck were investigated. During a 49-day growing period a corn-based diet was supplemented by 45% barley and 45% oats (isonitrogenously and iso-energetically), respectively. Daily feed intake, FCR-, and weight gain were measured. Abdominal fat, liver, and gizzard weights were determined and dry matter, protein, fat content and fatty acid composition of femoro-tibial muscles and liver fat were measured on the $35^{th}$, $42^{nd}$ and $49^{th}$ days of age. Feeding 45% barley caused a decrease of growth rate ($p{\leq}0.05$) during the first 4 weeks, which was followed by a rapid, compensatory growth from the $6^{th}$ week of age ($p{\leq}0.05$). Both barley and oat supplementation increased protein ($p{\leq}0.05$), while decreasing fat ($p{\leq}0.05$) and dry matter ($p{\leq}0.05$) content of the liver. Feeding of 45% oats in the diet decreased the monounsaturated fatty acid ($p{\leq}0,05$) and increased the n-6 ($p{\leq}0,05$), n-3 ($p{\leq}0,05$) and total polyunsaturated ($p{\leq}0,05$) fatty acid content of the intramuscular fat owing to the high proportion of soluble non-starch polysaccharides (NSP) in the diet. This might be explained by the more pronounced decrease in digestibility of saturated than unsaturated fatty acids in birds fed a soluble NSP-enriched diet. This result might be caused by the "cage effect" of soluble NSP trapping the bile salts which are more important for the absorption of saturated than polyunsaturated fatty acids.
Fumaria indica Linn. (Syn: Fumaria parviflora, Fumariaceae) is a wildly grown weed, mentioned and recommended in classical Ayurvedic texts for treatments of variety of ailments including dermatological diseases, topical diseases, cardiovascular complaints, circulatory disease, fever and headache etc. The present pilot study was designed to experimentally verify the possibility that fumarates are the major bioactive principles of Fumaria indica extracts involved in their stress response modulating activities, and to estimate pharmacologicallyactive dose ranges of fumarates and standardized methanolic extract of Fumaria indica (MFI). Effect of single, 5 and 10 daily oral doses of pure fumaric acid (FA), monomethyl fumarate (MMF), dimethyl fumarate (DMF) and MFI was quantified in well validated rodent models viz. apomorphine induced cage climbing, stress induced hyperthermia, and elevated plus-maze tests. Obtained results reveal high efficacy of MFI and pure fumarates possess qualitatively analogous activity profiles in all the three tests. There were no significant difference in the potencies of pure FA, MMF and DMF in the three tests, whereas efficacy of MFI in the elevated plus maze test for anxiolytics was higher than in the other two tests. Efficacies of all the four test agents in all the three tests increased with increasing number of days of oral treatments. Results of these pilot experiments should be helpful for more rational selections of pharmacologically interesting dose ranges and treatment regimens of fumarates and Fumaria indica extracts for further more holistic explorations of their diverse therapeutic potentials.

Density-dependant growth and mortality rate of Manila clam Ruditapes philippinarum reared in net cages was investigated in Gomso Bay, Korea where unusually high mortality of clams has been reported. For the experiment, four groups of clam cages were set up with a density of $2,000clams/m^2$ (group A), $1,000clams/m^2$ (group B), $500clams/m^2$ (group C) and $100clams/m^2$ (group D). Mortality and growth of clams in each experimental cage was monitored biweekly from May 2001 to September 2001. Highest mortality in group A was observed in late August, while highest mortality of rest groups was observed in early September. In September, the cumulative mortality in group A was 99%, while it was 93.2% in group B, 91.2% in group C and 88% in group D. Shell growth rate of clams in thecages was found to be density dependent; monthly shell length increase was 0.67 mm in group A, 1.33 mm in group B, 1.63 mm in group C and 1.71 mm in group D. Our study indicated that clam growth and mortality in the Bay is density dependent and the growth and survival rate is negatively correlated with the density.
Current codes recommend large amounts of shear reinforcement for reinforced concrete beam-column joints that causes significant bar congestion. Increase in congestion of shear reinforcement in joint core (connection zone), leads to increase accomplishment problems. The congestion may also lead to diameter limitations on the beam bars relative to the joint dimensions. Using double headed studs instead of conventional closed hoops in reinforced concrete beam-column joints reduces congestion and ensures easier assembly of the reinforcing cage. The purpose of this research is evaluating the efficiency of the proposed reinforcement. In this way, 10 groups of exterior beam-column joints are modeled. Each group includes 7 specimens by different reinforcing details in their joint core. All specimens are modeled by using of ABAQUS and analyzed subjected to cyclic loading. After verification of analytical modeling with an experimental specimen, 3D nonlinear specimens are modeled and analyzed. Then, the effect of amount and arrangement of headed studs on ductility, performance, ultimate strength and energy absorption has been studied. Based on the results, all joints reinforced with double headed studs represent better performance compared with the joints without shear transverse reinforcement in joints core. The behavior of the former is close to joints reinforced with closed hoops and cross ties according to the seismic design codes. By adjusting the arrangement of double-headed studs, the decrease in ductility, performance, ultimate moment resistant and energy absorption reduce to 2.61%, 0.90%, 0.90% and 1.66% respectively compared with the joints reinforced by closed hoops on the average. Since the use of headed studs reduces accomplishment problems, these amounts are negligible. Therefore, use of double-headed studs has proved to be a viable option for reinforcing exterior beam-column joints.
This experiment examined the effects of feeding Aspergillus oryzae (AO) culture to laying hens, on fecal microbial populations, fecal pH and moisture content, egg quality, and metabolizabilities of several nutrients. Sixteen commercial 38-wk-old laying hens were randomly allotted to four diets: control; with 0.15% locally produced AO culture; with 0.3% locally produced AO culture, and; or with 0.3% imported AO. Each treatment consisted of four replicates (cages) containing one bird per cage according to a completely randomized design. After 4 wk, AO were recovered in the feces of birds fed the AO diets, indicating that AO might pass through the fore-gut alive and become active in the hind gut. The number of Lactobacillus spp. in feces was higher in all treated groups than that of the control, indicating that AO would provide a beneficial environment for the Lactobacillus spp. to proliferate in the intestine. The number of fecal E. coli was significantly reduced by the addition of AO. A similar trend was also found for aerobic bacteria. Although not significant, fecal moisture contents tended to be reduced by the addition of AO. Fecal pH was not significantly different among the treatments. The addition of AO did not affect the various economic traits of eggs. Metabolizabilities of gross energy and dry matter measured during the 5th wk were increased by the AO supplementation. It appears that AO culture alone could be used as a probiotic supplement for layers.
For many years beak trimming has been a controversial subject, particularly since the 1980's when the practice came under close scrutiny by animal welfare groups. In Australia it is considered an essential practice, averting losses of AUD$17.5m annually by reducing mortality from cannibalism. While mortality in flocks from cannibalism can be reduced from 25% of the flock to virtually nil, the beak trimming procedure is considered traumatic for the bird. This study examined if chronic pain in the beak was evident in birds 10, 20 and 60 weeks after being trimmed at hatch and in another group of birds, 8 and 52 weeks after being re-trimmed at 14 weeks. Chronic pain was assessed by measuring pecking behaviour and beak sensitivity responses. Pecking behaviour studies completed after beak trimming and re-trimming showed no evidence to indicate that birds were suffering severe chronic pain in the beak. Beak trimmed pullets pecked more at the cage and had more toe pecks, yet overall pecks made at the feed and the environment were no different than untrimmed controls. While the beak sensitivity studies provided evidence that the beak of birds trimmed at hatch and also re-trimmed at 14 weeks may be more sensitive there was no evidence that re-trimming resulted in a more sensitive beak than birds trimmed at hatch only. These studies have shown that birds which are beak trimmed and re-trimmed return to apparently normal feeding and pecking behaviour in the long term. However, there was limited evidence that beaks of trimmed birds have an altered threshold to potentially painful stimuli.
