수용액 중에서 비이온성 계면활성제와 요오드간의 상호작용에 미치는 $Ca^{2+}$의 영향 (The Effects of $Ca^{2+}$on the Interaction of Nonionic Surfactant with Iodine in Aqueous Solution)

  • 수용액 중에서 요오드와 $4-(C_9H_{19})C_6H4_O(CH_2CH_2O)_{40} [NP-(EO)_40]$간의 charge transfer(CT) 상호작용에 미치는 $Ca^{2+}$이온의 영향을 UV-visible spectrophotometer를 이용하여 조사하였다. CT 상호작용에 따른 최대 흡수는 390 nm 부근에서 나타났으며, $Ca^{2+}$ 이온 존재하에서는 370 nm 영역으로 이동되었다. 계면활성제 농도를 CMC 이상에서 고정시켰을 때, CT 상호작용에 따른 최대흡수곡선의 강도가 $Ca^{2+}$이온 첨가 농도에 따라 증가하다가 감소하는 변곡점을 보였다. CT 상호작용에 기인한 흡수곡선의 강도가 증가하는 것은 $Ca^{2+}$이온 존재하에서 미셀 구조가 더욱 조밀해진데 다른 요오드와의 donor-acceptor overlap 증가로 볼 수 있다. 이러한 현상들은 수용액 중에서 상대적으로 자유로이 여러가지 배향을 가질 수 있는 비이온성 계면활성제의 선형 oxyethylene(EO) 사슬이 유사 크라운 에테르 구조를 형성하여 $Ca^{2+}$이온과 착물을 형성할 수 있는 가능성을 보여준다.

Studies on the Oocytes Activation Regimed for Nuclear Transfer and Co-culture of Nuclear Transferred Embryos

  • This study was carried out to investigate the optimal activation condition for parthenogenetic development. In order to activate oocytes at 24 hrs post onset of maturation, the oocytes were cultured 3 - 13 μM Ca for 5 min., 5-8 ㎍/㎖ cytoclacin(CH) for 6 hrs, 0.5-2.0 mM 6-dimethylaminopurine(DMAP) for 3 hrs alone or combination. The activated oocytes were cultured in TCM-199 media at 5% CO₂, 95% N₂, 38℃. 1. The cleavage rate after 48 hrs culture of oocytes treated with 3-13 μM Ca for 5 min. were 9.6%-20.0% and 3.8-7.3%, respectively. When oocyte were treated with 10 μM Ca, the blastocyst formation rate was significantly higher than other group. 2. The cleavage rate after 48 hrs culture of oocytes treated with 5-8 ㎍/㎖ cytoclacin(CH) for 6 hrs, were 9.4%-21.8% and 0.0-7.3%, respectively. When oocyte were treated with 10㎍/㎖ CH, the blastocyst formation rate was significantly higher than other group. 3. The cleavage rate after 48 hrs culture of oocytes treated with 0.5-2.0 mM 6-dimethylaminopurine(DMAP) for 3 hrs were 9.1%-21.8% and 0.0-7.3%, respectively. When oocyte were treated with 2.0mM DMAP, the blastocyst formation rate was significantly higher than other group. 4. The cleavage rate after 48 hrs culture of oocytes treated with Ca+CH, Ca+DMAP, CH+DMAP were 75.9%-93.5% and 9.7 -13.3%, respectively. When oocytes were treated with Ca followed by DMAP, the blastocyst formation rate was significantly higher than other group(p〈0.05). 5. When necleus transferred embryos co-cultured with BSA, EGF and CS, the developmental rate to blastocyst were higher than control group.

A SURVEY OF INTERSTELLAR LINES: RADIAL VELOCITY PROFILES AND EQUIVALENT WIDTHS

  • An atlas of high resolution (${\lambda}/{\Delta}{\lambda}$=45,000) profiles of interstellar atomic lines of K I (7665, 7699 ${\AA}$), Na I (D 1, D2), Ca II (H, K), Ca I (4227 ${\AA}$), molecular structures of CH, CH+, CN and the major diffuse interstellar bands at 5780 and 5797 ${\AA}$ based on ${\~}$300 echelle spectra of ${\~}$200 OB stars is presented. Relationships between the reddenings, distances and equivalent widths of NaI, CaII, KI, CH, CH+, CN and diffuse bands are discussed. The equivalent width of K I (7699 ${\AA}$) as well as of CH4300 ${\AA}$ / correlate very tightly with E(B- V) in contrast to the features of neutral sodium, ionized calcium and the molecular ion CH+. The equivalent widths of the Hand K lines of Call grow with distance at a rate ${\~}$250m${\AA}$ per 1 kpc. A similar relation for NaI is much less tight. The strengths of neutral potassium lines, molecular features and diffuse interstellar bands do not correlate practically with distance. These facts suggest that ionized calcium fills the interstellar space quite homogeneously while the other carriers mentioned above, especially K I, CH and these of diffuse bands occupy more and more compact volumes, also filled with dust grains. Apparently the carriers of narrow diffuse bands are spatially correlated with simple molecules and dust grains - all abundant in the so-called 'zeta' type clouds. The same environment seems to be hostile to the carriers of broad diffuse interstellar bands (DIEs) (like 5780 or 6284) and -to a certain extent - also to CaII, NaI and CH+.

Protective effect of Lycium barbarum leaf extracts on atopic dermatitis: in vitro and in vivo studies

  • BACKGROUND/OBJECTIVES: Atopic dermatitis (AD) is a chronic disease with an increasing incidence globally; therefore, there is a growing demand for natural compounds effective in treating dermatitis. In this study, the protective effects of Lycium barbarum leaves with and without chlorophyll (LLE and LLE[Ch-]) on AD were investigated in animal models of AD and HaCaT cells. Further, we investigated whether LLE and LLE(Ch-) show any differences in physiological activity. MATERIALS/METHODS: AD was induced by 2,4-dinitrochlorobenzene (DNCB) for three weeks, while NC/Nga mice were fed LLE or LLE(Ch-) extracts for 7 weeks. Serum immunoglobulin E (IgE) and cytokine (tumor necrosis factor [TNF]-α, interleukin [IL]-6, and IL-4) concentrations and the degree of DNA fragmentation in lymphocytes were examined. A histopathological examination (haematoxylin & eosin staining and blue spots of toluidine) of the dorsal skin of mice was performed. To elucidate the mechanism of action, the expression of the thymus and activation-regulated chemokine (TARC) and macrophage-derived chemokine (MDC) were measured in HaCaT cells. RESULTS: Serum IgE and cytokines (TNF-α and IL-6) levels as well as DNA fragmentation of lymphocytes were significantly decreased in AD-induced mice treated with LLE or LLE(Ch-) compared to those of the control group. The epidermal thickness of the dorsal skin and mast cell infiltration in the LLE group significantly reduced compared to that in the control group. The LLE extracts showed no cytotoxicity up to 1,000 ㎍/mL in HaCaT cells. LLE or LLE(Ch-)-treated group showed a reduction of TARC and MDC in TNF-α-and IFN-γ-stimulated HaCaT cells. CONCLUSIONS: These results suggest that LLE potentially improves inflammation by reducing the expression of chemokines that inhibit T helper 2 cell migration. LLE(Ch-) showed similar effects to LLE on blood levels of IgE, TNF-α and IL-6 and protein expression in HaCat cells, but the ultimate effect of skin improvement was not statistically significant. Therefore, both LLE and LLE(Ch-) can be used as functional materials to alleviate AD, but LLE(Ch-) appears to require more research to improve inflammation.

알파형 반수석고 기반 결합재의 pH 및 강도발현 평가 (Evaluation of pH and Compressive Strength Development of Alpha-Calcium Sulfate Hemihydrate-based Binder)

  • 이 연구에서는 중성(pH 6~7) 수준의 식생용 콘크리트 결합재를 개발하기 위해 알파형 반수석고 기반의 결합재들의 pH 값과 압축강도 발현을 평가하였다. 식생용 콘크리트를 위한 결합재의 경제성 및 강도발현을 고려하여, 알파형 반수석고는 GGBS, FA 및 보통포틀랜드 시멘트를 사용하여 25% 및 50% 치환하였다. 알파형 반수석고를 100% 사용한 모르타르의 압축강도는 시멘트 100% 모르타르에 비해 약 57% 수준으로 있었다. GGBS 및 FA 치환율 증가와 함께 알파형 반수석고 기반 결합재의 압축강도는 감소하였지만 pH 값은 재령에 관계없이 6.5~7.5 수준으로 일정하게 있었다. GGBS 및 FA가 치환된 알파형 반수석고 결합재의 주요 수화생성물은 석고($CaSO_4$)이었으며, 수산화칼슘 [$Ca(OH)_2$]은 나타나지 않았다. 반면 시멘트가 치환된 알파형 반수석고의 pH 값은 $Ca(OH)_2$의 생성으로 인해 약 11.5 이상이었는데, 생성된 $Ca(OH)_2$ 양은 단위 시멘트 양의 약 10% 수준이었다.

유기수은의 신경독성에 대한 셀레늄의 보상작용 (Interaction of Sodium Selenite on Neurotoxicity Induced by Methylmercuric Chloride)

  • This study was conducted to investigate the mechanism of protective effect by sodium selenite in methylmercuric chloride neurotoxicity, increasing intracellular $Ca^{2+}$concentration of the neuron. Methylmercuric chloride of 3mg/kg of body weight was administered simultaneously with sodium selenite of 5mg/kg and pretreatment of sodium selenite via intraperitoneal injection to rats. Also, effect of methylmercuric chloride($25{\mu}M,\;50{\mu}M,\;100{\mu}M$) and sodium selenite($200{\mu}M$) on free intrasynaptosomal $Ca^{2+}$ concentration were studied using the fluorescent $Ca^{2+}$ indicator fura -2 in vitro. After the treatment, at 6, 24, and 48 hours later, mercury in the cerebral cortex, liver and kidney tissues, succlnic dehydrogenase activities, adenosin-5'-triphosphate concentration, acetylcholinesterase activities, and intracellular $Ca^{2+}$ concentration in the cerebral cortex were determined in vivo. Cerebral synaptosomes of rats were incubated with methylmercuric chloride and sodium selenite in Hepes buffer for 10 minutes and free intrasynaptosomal $Ca^{2+}$ concentration were measured with fura-2 in vitro. The results were summarized as follows ; 1. The combined administration of $CH_3HgCl$ and $Na_2SeO_3$ and pretreatment of $Na_2SeO_3$ according to time significantly more increased in the cerebral cortex and decreased in the liver, kidney mercury concentrations compared to the administration of $CH_3HgCl$ only. 2. The combined administration of $CH_3HgCl$ and $Na_2SeO_3$ and pretreatment of $Na_2SeO_3$ increased more succinic dehydrogenase and acetylcholinesterase activities compared to the administration of $CH_3HgCl$ only. Particularly pretreatment of $Na_2SeO_3$ significantly more compared to the administration of $CH_3HgCl$ only. The concentration of adenosine-5'-triphosphate in $Na_2SeO_3$ treatment groups revealed a favourable effect compared to the administration of $CH_3HgCl$ only. 3. Intracellular $Ca^{2+}$ concentration in administration of $CH_3HgCl$ only was increased significantly more than control group in all test hours but was increased significantly more at 48 hours only after treatment in combined administration of $CH_3HgCl$ and $Na_2SeO_3$ and pretreatment of $Na_2SeO_3$ according to time interval more decreased significantly intracellular $Ca^{2+}$ concentration compared to the administration of $CH_3HgCl$ only. 4. Free intrasynaptosomal $Ca^{2+}$ concentration in the combined administration of $CH_3HgCl$ and $Na_2SeO_3$ was decreased ($24%{\sim}40%$) significantly more than the administration of $CH_3HgCl$ only. From the above results, the specific dosage of $Na_2SeO_3$ decreased increment of intracellular $Ca^{2+}$ concentration induced by administration of $CH_3HgCl$. These findings suggest the protective mechanism of $Na_2SeO_3$ on the neurotoxicity of $CH_3HgCl$.

Relationship between the Methane Production and the CNCPS Carbohydrate Fractions of Rations with Various Concentrate/roughage Ratios Evaluated Using In vitro Incubation Technique

  • The objective of the trial was to study the relationship between the methane ($CH_4$) production and the Cornell Net Carbohydrate and Protein System (CNCPS) carbohydrate fractions of feeds for cattle and the suitability of CNCPS carbohydrate fractions as the dietary variables in modeling the $CH_4$ production in rumen fermentation. Forty-five rations for cattle with the concentrate/roughage ratios of 10:90, 20:80, 30:70, 40:60, and 50:50 were formulated as feed samples. The Menke and Steingass's gas test was used for the measurement of $CH_4$ production. The feed samples were incubated for 48 h and the $CH_4$ production was analyzed using gas chromatography. Statistical analysis indicated that the $CH_4$ production (mL) was closely correlated with the CNCPS carbohydrate fractions (g), i.e. CA (sugars); $CB_1$ (starch and pectin); $CB_2$ (available cell wall) in a multiple linear pattern: $CH_4=(89.16{\pm}14.93)$ $CA+(124.10{\pm}13.90)$ $CB_1+(30.58{\pm}11.72)$ $CB_2+(3.28{\pm}7.19)$, $R^2=0.81$, p<0.0001, n = 45. Validation of the model using 10 rations indicated that the $CH_4$ production of the rations for cattle could accurately be predicted based on the CNCPS carbohydrate fractions. The trial indicated that the CNCPS carbohydrate fractions CA, $CB_1$ and $CB_2$ were suitable dietary variables for predicting the $CH_4$ production in rumen fermentation in vitro.

Pt 전극을 사용한 $SnO_{2}(Ca)/Pt$ 후막소자의 탄화수소계가스에 대한 감응특성 (Gas Sensing Characteristics of $SnO_{2}(Ca)/Pt$ Thick Film Using Pt Electrode for Hydrocarbon Gases)

  • 공침법을 이용하여 Ca 및 Pt 가 첨가된 $SnO_{2}$ 미세 분말을 제조하였다. 제조한 미세분말에 대해 TEM, XRD 및 BET 분석으로 결정크기 및 비표면적을 분석하였으며, 제조한 원료분말을 이용하여 스크린 인쇄법으로 후막형 가스 감지 소자를 제작하고, 그 특성과 탄화수소계가스에 대한 감지 특성을 조사하였다. 첨가된 Ca 및 Pt에 의해 하소와 소성의 열처리 과정 중에 $SnO_{2}$ 결정 성장이 억제되었으며, 탄화수소계가스에 대한 응답특성의 향상을 가져왔다. 한편, 소자의 전극으로써 Au 와 Pt를 사용하여 비교 분석하였는데, Au를 전극으로 사용할 경우에 Pt에 비해 가스 감응 특성 면에서 큰 저하를 가져왔다. 이는 Pt 전극도 $CH_{4}$의 산화에 기여했기 때문이다. Ca 및 Pt를 각각 0.1, 1 wt% 첨가한 $SnO_{2}$ 후막소자의 경우 2000 ppm의 $CH_{4}$에 대해 약 83%의 감도를 보였다.

칼슘급원별 수관살포가 사과 과실의 칼슘축적에 미치는 영향 (Effects of Various Calcium Salt Spray on Calcium Accumulation into Apple Fruits (Malus domestica Borkh.))

  • 사과나무에 대한 칼슘용액의 수관살포 시 칼슘염의 종류에 따른 과실 내 칼슘함량증가효과를 구명하고 자 본 연구를 수행하였다. $CaCl_2$, $CaCO_3$, Ca$(CH_3COO)_2$ 및 Ca$(NO_3)_2$ 등의 칼슘염을 수관살포한 결과 "쓰가루" 품종에는 Ca$(NO_3)_2$, $CaCl_2$$CaCO_3$, 그리고 "후지" 품종에는 $CaCl_2$$CaCO_3$가 과실의 칼슘함량증가에 효과적이었다. 수관살포에 의하여 과실로 흡수된 칼슘은 과피와 과피직하의 과육에 국한되었다. Ca$(CH_3COO)_2$를 살포한 과실은 저장 중에 에틸렌 발생량이 감소하는 경향이었다.
