The Ca and P balance studies were carried out with two different diets varying Ca levels : the current Korean RDA level(normal Ca diet) and the high level (High Ca diet) which was determined by adding 2SD to mean Ca intake of all subjects. The subjects were sever healthy adult woman, aged from 25 to 32 years old. The metabolic studies were conducted for two weeks with a days recess in between : during the fist week with normal Ca diet and during the second week with high Ca diet. The composition of nutrients excepting Ca of both diets was similar to subjects usual intake. The result were summarized as follows: Mean daily Ca intake was 728.8mg from norma Ca diet and 945.5mg from high Ca diet. Fecal excretion of Ca increased significantly(p<0.05) on high Ca diet, but urinary excretion of Ca did not show any differences between the two diet periods. There were also no significant differences in Ca retention between the two diet periods but it tended to be greater during high Ca diet period: 112.1mg/day during normal Ca diet period vs 208.2mg/day during high Ca diet period. Mean apparent Ca absorption was 41.2% on normal Ca diet and 42.1% on high Ca diet, indicating it was not affected by high Ca intake level used in this study. On the contrary, P retention was significantly increased up to 109.4mg/day with high Ca diet as compared to- 41mg/day with normal Ca diet. There were no significant differences in fecal and urinary excretion of P but those to be lower during high Ca diet period. The above results showed that higher Ca intake more than current RDA(700mg/day), in the level of 945.5mg/day, could increase Ca retention through Ca absorption comparable to the rate appeared on RDA level intake. P retention was also improved by high Ca intake. Therefore, higher Ca intake than the current RDA level seemed to produce favorable effects on bone health in adult women. However, the current RDA level seemed to be relatively appropriate, considering the results that all the subjects but one maintained positive Ca balance with normal Ca diet. In order to clarify the depositional environment and sea-level change at Ilsan area including Gawaji and Saemal valley plains, which is located at the right side in downstream of the Han River, boring data, radiocabon dating and diatom analysis were comprehensively investigated. As a result, the palaeogeographies fo this area altered by the transgressions and regressions after 7,000 y. BP could be restored. The high tide sea-level(mean high water level of spring tide) was arrived ca. 7,000y. BP at the valley plain and risen to ca. 5.5m at ca. 5000y. BP. Since then, the sea-level was kept up the same level to ca. 3,200 BP. The descended sea-level to ca. 2,300 BP was risen up to ca. 5.8m in ca. 1,800 y. BP. It is presumed that such a sea-level change as well as the different sediments in quantity supplied from the river basin of the valley plain could be effected to change diversely the depositional environment of the study area and eventually to the prehistoric human life.
To study the effects of the age and the dietary protein content on Ca metabolism male rats of 1 month 6 month 12 month of age were fed experimental diets containing 5%, 15% or 50% casein for 4 weeks. Food and ca intake were higher in old rats and in high protein groups. The weight ash and Ca contents of femur and tibia were higher in old rats. The higher dietary protein level resulted in higher skeletal weigh ash and Ca contents. But high protein diet(50% casein) lead to reduced bone mineral density(ash/dry bone weight) and Ca density(Ca/dry bone weight) in 1 month old rats. Low protein diet(5% casein) on the other hand reduced the bone growth even though the bone density was higher in this group. The ill effect of low protein diet was not evident in 12 month old rats. Glomerular filteration rate(GFR) and urinary Ca excretionincreased with age and with dietary protein level especially in 12 month old rats. Serum immunoreactive parathyroid hormone(iPTH) level tended to be higher in aged rats but was not affected by dietary protein level except 1 month old rats where 50% protein group showed significantly higher value. This study showed that the dietary protein level seemed to have different effect on Ca metabo-lism in rats of different age., The low bone density in the high protein group of growing rats may be due to the higher iPTH level and increased urinary Ca. The dietary protein level however had no effects on the bone composition in aged rats even though the higher urinary Ca excretion. In conclusion this study suggests that high protein intake from young may lead to less peak bone mass and to increase the bone loss in later years, which would increase the risk for osteporosis.
In order to evaluate the effect of habitual Na and Ca intake on blood pressure regulation, we measured the habitual dietary intakes of Na and Ca, urinary excretion of Ca, Na and K, and plasma level of renin activity, aldosterone, and indices of Ca metabolism in 27 untreated hypertensive women and 30 age-matched normal women on a free diet. Hypertensive and total subjects were divided into four groups according to habitual dietary intakes of Na and Ca as low Na-low Ca(LNLC), low Na-high Ca(LNHC), high Na-low Ca(HNLC), and high Na-high Ca(HNHC). HNLC hypertensive group showed the lowest level of plasma renin activity, 25-(OH) Vit D$_3$, calcitonin and serum total Ca, and presented the highest level of PTH and urinary excretions of Na/K and Ca/Cr. There were no significant difference in plasma level of aldosterone and urinary excretion of Na and K among four hypertensive groups. When all subjects were divided into four groups according to the same method, HNLC group showed the highest level of blood pressure with no statistical significance and the lowest level of calcitonin and total serum Ca. The above results indicated that renin-aldosterone system and Ca regulating hormone has a mutual relationship in hypertension. Na and Ca may interact each other, rather than affecting independently blood pressure control. As a result, considering the fact that daily balance of Na and Ca intakes affects Na and Ca regulating hormones and urinary excretion of Na and Ca, it may be involved in blood pressure control. These results suggest that maintaining an adequate intake of Ca with less intake of Na may prevent from the risk of hypertension. (Korean J Nutrition 34(4) : 409~416, 2001)
This study was designed to investigate the effects of aerobic dancing and/or Ca supplemen-tation for six months on Ca metabolism in postmenopausal women. The subjects were healthy 29 women aged from 60 to 70 years old. They were divided into four groups ; placebo and Ca supplementation group with and without exercise. The amount of Ca supplemented was 500mg a day. The frequency of doing exercise was three times a week and it took 40 minutes to complete once all the course of aerobic dancing programmed for old women. The results were summarized as follows : 1) Bone density of the second lumbar spine and ward's triangle measured after experiment decreased significantly in control group as compared with pre-experimental level(P<0.05). On the contrary it showed a tendency to increase or maintained the pre-experimental level after experiment in other groups. 2) Urinary Ca/creatinine and P/creatinine ratios did not show any significant differences among groups before and after experiment. But urinary Ca/creatinine ration decreased signifi-cantly in Ca group after experiment(P<0.05) 3) Serum Ca and P levels did not show any significant differences among groups before and after experiment. But serum Ca level in creased significantly in Ca group after experiment(P<0.05) 3) Serum Ca and P levels did not show any significant differences among groups before and after experiment. But serum Ca level increased significantly in Ca group after experiment (P<0.05). 4) Serum PTH level also did not show any significant differences among groups before and after experiment. But serum calcitonin level decreased significantly in Ca group after experiment(P<0.05). The above results showed that it will be difficult to prevent degenerative bone loss without regular exercise and/or Ca supplementation in postmenopausal women having Korean usual diets.
To determine relationships of biochemical parameters involved in Ca metabolism with eggshell quality, serum Ca level, duodenal and uterine calcium binding protein (CaBP) and uterine carbonic anhydrase (CA) activities were measured using 102-week old hens. Three groups of chickens were selected, those showing high quality (HQ) and low quality (LQ) eggshells and non-laying activity (NE). NE hens exhibited significantly (p < 0.05) lower serum Ca levels than laying hens. HQ and LQ hens were not different in the Ca level, indicating that serum Ca level was not good indicator of hen's ability to produce different quality eggshells. Duodenal CaBPs was highest in HQ and lowest in NE (p < 0.10). Uterine CaBPs of LQ and NE were not different and lower significantly (p<0.10) than that of HQ, suggesting that CaBP played an important role in determining eggshell quality. Uterine CA activities of the three groups were significantly different (p < 0.01) ; highest in HQ and lowest in NE, suggesting intimate relationship between CA and Eggshell quality.
To investigate the effect of dietary Ca levels on metabolic changes of Ca and skeleton in postmenopausal women, 10-month-old ovariectomized female rats were compared with 2 month old rats. The rats were fed either 0.2% or 1.2% Ca diets for 16 weeks. Food intake and weight gain as higher in rats fed high Ca diets and in ovariectomized rats. Apparent Ca absorption as higher, and Ca balance was lower in the low Ca groups. Vertebrae density was higher in old rats or those fed a high Ca diets. The old rats and ovariectomized rats showed decreased bone formation, increased bone resorption and kidney function deterioration resulting in increased urinary Ca excretion. Contradictory to the above observation, old rats and ovariectomized rats still showed higher bone mass and bone ash content. Therefore aging was not fully onging in 10-month-old rats. Bone weights, mineral contents, and mineral/wt ratio were lower in ovariectomized rats. Dietary Ca level did not affect urinary Ca excretion, urinary protein excretion, GFR, serum alkaline phosphatase, or urinary hydroxyporline excretion. This means that dietary Ca level did not influence kidney function or bone turnover. However Ca content and the ash content of femur, 4th vertebra, and scapula were increased in high Ca groups. Therefore, it is considered that decreased bone formation and accelerated bone resorption may account for the increased osteoporotic risk in women in menopause after middle age. However, Ca metabolism can be improved and bone components can be maintained if Ca is supplemented.
In order to study the interrelationships of calcium (0.45 vs. 0.90%), phosphorus (0.40 vs. 0.70%) and protein (17, 20, 23%), $2{\times}2{\times}3$ factorial design was employed. A total of 480 broilers (Hisex-Hibro) aged 3 days were fed the experimental diets for a period of 28 days. Body weight gain, daily feed intake and feed efficiency were investigated for the simple effects, first order interaction and second order interaction of the dietary factors. These effects were also applied to bone ash retention, percent Ca in bone & ash, percent P in bone & ash, and protein utilizability. Results were as follows. 1) For body weight gain, simple effects of dietary levels of Ca, P, CP were found to be significant (p<0.05). Body weight gain at 0.90% Ca level was improved as the dietary CP levels increased. For the feed intake, single effects of dietary levels of both P and CP were found (p<0.05). Feed efficiency was improved as the dietary CP and P levels increased. Ca $\times$ P interaction was found to be significant for body weight gain, feed intake and feed efficiency (p<0.05), however, Ca $\times$ P $\times$ CP interaction effect was not found. 2) Protein utilizability decreased as the dietary CP level increased (p<0.01). 3) 0.90% Ca in diet showed less bone ash retention than 0.45% Ca level. And, increasing the dietary P level resulted in increased bone ash retention. Increasing the dietary P level resulted in increased bone Ca retention (p<0.01) and increased bone P retention (p<0.05). Dietary CP levels had significant (p<0.01) effect on bone Ca retention except for 23% CP level. Increasing the dietary Ca level resulted in wider Ca:P ratio of bone, but increasing the dietary P level resulted in narrower Ca:P ratio of bone. 4. Ca $\times$ P interaction effects were found to be significant (p<0.01) for bone ash, bone Ca & P, ash P content, and bone Ca:P ratio. Ca $\times$ P $\times$ CP interaction effects were found for bone ash (p<0.01), bone Ca (p<0.05) and bone P content (p<0.01).
Studies were carried out to explore the influence of dietary protein level on bone metabolism in uninephrectomized rat (experimental renal failure model) when dietary Ca and P contents were equal. Male rats were uninephrectomized or sham operated and fed 8%, 15% and 40% casein diets for 24 weeks. Ca and P contents of the all diet were 0.4% and 0.6% respectively. The results are summarized as follows. We did not found any significant difference in PTH and Ca level of the serum, Ca intake and Ca excretion among the experimental groups. There was significant positive correlation between the PTH and phosphate level. There was significant inverse correlation between serum Ca and creatinine level. The effect of the dietary protein level and renal mass loss on density and Ca contents of the bone were small and different according to the kinds of the bone. Low protein diet was associated with a significant enhancement of scapular density. Femur and vertebra density, however, were not influenced by dietary protein level and uninephrectomy. Light microscopic examination showed several calcified foci in the kidney in all experimental groups. Low protein diets have been used for a long time in the conservative management of chronic renal failure as they have a beneficial effect in preventing the appearance of symptoms. This study elucidated that part of beneficial effects of the low protein diet related to the suppression of the hyperphosphatemia. And these results, even though uninephrectomized rats fed high protein diet, the secondary hyperparathyroidism is supressed by the regulation of the P level. Therefore this study emphasized the need to pay more attention to the regulation of dietary P level as well as dietary protein content in chronic renal failure. The effect of N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) on the intracellular $Ca^{2+}$ level was studied in NIH3T3 fibroblast cells. A reduction of the intracellular $Ca^{2+}$ level was observed after exposure to 300 ${\mu}m$ MNNG. However, the intracellular level of $IP_3$, a well-known regulator of $Ca^{2+}$ release from internal storage, was not changed by MNNG treatment. Instead, a reduction of the intracellular NAD level was observed. NAD as well as $IP_3$ stimulated intracellular $Ca^{2+}$ release from permeabilized cells. The treatment of 3-aminobenzamide, which inhibited the MNNG-induced reduction of the NAD level, also prevented the MNNG-induced decrease of the $Ca^{2+}$ level. Our data suggest that MNNG reduces the intracellular $Ca^{2+}$ level by NAD depletion in NIH3T3 cells.
