Retardation of axonal regeneration in the sciatic nerve after injury in streptozotocin-induced diabetes (좌골신경 손상 후 스트렙토조토신 유도성 당뇨병에 의한 축사재생의 지연 효과)

  • Objective: The goal of this study is to investigate whether peripheral axonal regeneration is affected by diabetes in experimental animals. Method: Sprague Dawely rat was injected with 45~50 mg/kg of streptozotocin (STZ) to generate an animal model of diabetes. Three months after STZ injection, sciatic nerve (2 cm length) was removed and the same length of nerve segments from STZ-injected animal or from control animal (CTL) was transplanted into STZ-injected animals (STZ-STZ and STZ-CTL respectively). Similarly, sciatic nerve segments from STZ-injected animal or from control animal were grafted into the control animals (CTL-STZ and CTL-CTL respectively). All animals were sacrificed 2 weeks after transplantation. Sciatic nerve sections were prepared and subjected to immunofluorescence staining analysis. Results: Immunofluorescence staining for NF-200 showed that distal elongation of regenerating axons reached 40~80% of proximal neve in both CTL-STZ and CTL-CTL groups. However, distal elongation in both STZ-STZ and STZ-CTL groups were 20~60% of proximal nerve. Furthermore, measurement of axonal regeneration after immuno-staining with SCG10 showed that the scores of distal elongation relative to proximal nerve were 50~90% in CTL-CTL and CTL-STZ groups and 10-60% in STZ-CTL and STZ-STZ. Conclusions: Our data showed that the levels of axonal regeneration were not affected irrespective of whether they were from STZ- or CTL graft, but were greatly reduced when the nerves were transplanted into the STZ host.

Therapeutic Effects of Curdrania tricuspidata Leaf Extract on Osteoarthritis (골관절염 실험모델에서 꾸지뽕나무 추출물의 골관절염 억제효과 연구)

  • The inhibitory effect of ethanol extracts from Curdrania tricuspidata leaves (CTL) on osteoarthritis was investigated in primary cultured rat cartilage cells and a monosodium-iodoacetate (MIA)-induced arthritis rat model. To identify the effects of CTL 80% ethanol extracts (CTL80) and CTL 10% ethanol extracts (CTL10) against $H_2O_2$ treatment in vitro, cell survival was measured by the MTT assay. Cell survival after $H_2O_2$ treatment increased with CTL80 and CTL10 close to normal up to $300{\mu}g/mL\;H_2O_2$. The mRNA expression of matrix metalloproteinases (MMPs) was determined MMP-7 and MMP-13 (known catabolic factors), were significantly inhibited by CTL 80 and CTL10; a $200{\mu}g/mL$ dose of CTL80 especially decreased MMP-13 expression. In vivo, osteoarthritis was induced by an intra-articular injection of MIA into the knee joints of rats, then CTL80 and CTL10 orally administered daily for 35 days. After the animals were sacrificed, histological evaluations of their knee joints revealed a reduction in polymorphonuclear cell infiltration and smooth synovial lining in the CTL80-500 group. Micro-CT analysis of hind paws from CTL80-500 and CTL10 showed a protection against osteophyte formation, soft tissue swelling, and bone resorption. In conclusion, CTL ethanol extracts are effective in ameliorating joint destruction and cartilage erosion in MIA-induced rats. CTL decreases and normalizes articular cartilage through preventing extracellular matrix degradation and chondrocyte injury, and could potentially serve as a therapeutic treatment for humans.

  • During the 2nd World War, several Coal-to-Liquid (CTL) plants were operated in Germany and England to convert coal to large volumes of liquid fuel. Big oil fields discovered in the Middle East after the war supplied crude oil at the low price and all CTL plants were forced to shut down. However, South Africa (Sasol) built a CTL plant in 1955 and 2 more plants afterward and the current production of coal-derived synfuel reached 150,000 bbl/day. Recently, the sustained high crude oil price and the fear of the "peak oil" rejuvenated the interest of CTL and several CTL projects are in progress. China established a plan to build CTL plants with the total capacity of 30 million tons of synfuel per year by 2030. China is building a direct coal liquefaction plant which is scheduled to produce 20,000bbl/day of synfuel in 2008. There are 8 CTL projects in USA either in the planning stage or in the ground-breaking stage. CTL projects are also carried out in Australia, Philippines, New Zealand, Indonesia and India. Korea needs to approach the CTL project in the perspective of the national energy security. In this paper, the history, the status, current activities and the prospect of CTL are described.

The generation and characteristics of the dominant field in CTL cell (CTL cell에서의 우세장 발생과 특성)

  • CTL cell that is one of the standard electromagnetic generation equipment can measure the characteristic of the electromagnetic susceptibility and the electromagnetic interference. In case of being input the same magnitude signal with the phase difference of 0o or the phase difference of 180o at two input ports of CTL cell to be the fundamental resonant frequency(TE011) of 2.20GHz, the characteristics of the electric field and the magnetic field at the uniform area were measured. And, it measured the electric field characteristic due to the variation of the input power, the test position and the input frequency under the dominant E-field and the dominant H-field of CTL cell. Using these mesuremed data, it examined the operation characteristic and the available frequency band of CTL cell.

Quality Characteristics of Pork Patty Containing Lotus Root and Leaf Powder (연근 및 연잎 분말을 첨가한 돈육 Patty의 품질특성)

  • This study was carried out to investigate the effect of the addition of lotus (Nelumbo nucifera) root and leaf powder on the quality characteristics of pork patties. The pork patties were of four types: nothing added (CTL), 0.5% lotus root powder added (LRP), 0.25% lotus root and 0.25% lotus leaf powder added (LRLP), and 0.5% lotus leaf powder added (LLP). There was no significant difference in $L^*$ value (whiteness), cohesiveness, gumminess, chewiness, water holding capacity, VBN content, and total bacterial amount. The moisture was highest in CTL, protein was highest in LRLP, fat was highest in LRLP and LLP, and ash was highest in LRLP (p<0.05). The total polyphenol content of the CTL, LRP, LRLP, and LLP were 3.34, 3.64, 3.90 and 3.90 mg/100 g, respectively. The $a^*$ value was highest in CTL, and the $b^*$ value of LRLP and LLP were higher than those of CTL and LRP (p<0.05). The hardness of CTL and LRP were higher than those of LRLP and LLP, and springiness was lowest in LLP (p<0.05). The cooking loss, thickness change, diameter changes, and pH were highest in CTL (p<0.05). The TBARS was highest in CTL, and was lowest in LLP (p<0.05).

  • Employing the approach to isolate the genes expressed preferentially in cytolytic T cell (CTL) but not in other types of cell, 3 CTL-specific cDNAs were recently cloned. To characterize these cDNA clones in relation to CTL activation, their expression pattern after T cell antigen receptor (TCR) or interleukin 2 (IL-2) stimulation were investigated by RNA blot analysis of cloned CTL L3 cells. Transcripts level of two cDNA clones were markedly elevated by TCR stimulation but not by IL-2. In addition, transcripts expression of both clones were abrogated by cyclosporin A treatment. These results indicated that gene activation mediated by TCR is distinct from that mediated by IL-2 and imply that those two unidentified cDNA clones are related to TCR-mediated, IL-2-independent but cyclosporin A-sensitive pathway for CTL activation.

  • 모델체킹에 사용되는 논리는 CTL 과 LTL 이 있다. 이 두 논리식은 그 표현력과 효과적인 면에서 크게 차이가 있다. 현재 SPIN에서는 LTL 을 이용하여 모델체킹을 수행하고 있다. 본 논문에서는 LTL 모델체커인 SPIN 에 CTL 을 적용시킬 수 있는 가능성을 제시하고, LTL 모델 체커에 CTL 을 적용시킬 수 있는 방법에 대해 논한다.

Anti-wrinkle Activity of a Curdrania tricuspidata Extract on Ultraviolet-induced Photoaging (Hairless Mice를 이용한 광노화 모델에서 꾸지뽕나무 추출물의 피부주름 형성 억제효과)

  • We investigated the anti-wrinkle activity of an 80% ethanol extract of Curdrania tricuspidata leaves (CTL80) on ultraviolet-induced photoaging in hairless mice. Skin wrinkles were induced by 10 weeks of UVB-irradiation on the back of Skh-1 hairless mice three times a week. Mice were divided into ten groups; normal control (-UVB), UVB irradiated control group (+UVB), dietary groups (UVB+ascorbic acid 0.1%, UVB+CTL80 0.1%, UVB+CTL80 0.25%) and topical application groups (-UVB+base lotion (BL), UVB+BL, UVB+ascorbic acid 1%+BL, UVB+CTL80 1%+BL, UVB+CTL80 2%+BL). Wrinkle formation, histological changes, superoxide dismutase (SOD) activities, glutathione peroxidase (GSH-Px), and the expression of matrix metalloproteinases (MMP-1, MMP-3 and MMP-9) were analyzed. Wrinkles for the +UVB groups formed as a pattern of deep furrows and thick crests. Wrinkles with CTL80 treatment formed as a pattern of shallow furrows and thin crests, with wrinkle areas were lower than the +UVB group. In an antioxidant analysis of mouse blood, SOD and GSH-Px activities were significantly higher in the CTL80 topical application group compared to the +UVB group. The mRNA expression of MMPs in the +UVB group was significantly higher than the normal control group, and significantly lower in the CTL80-treated group. In conclusion, CTL80 exerted anti-wrinkle activity on ultraviolet-induced photoaging by regulating antioxidative defense systems and MMPs expression.

Molecular and Functional Characterization of Choline Transporter-Like Proteins in Esophageal Cancer Cells and Potential Therapeutic Targets

  • In this study, we examined the molecular and functional characterization of choline uptake in the human esophageal cancer cells. In addition, we examined the influence of various drugs on the transport of [$^3H$]choline, and explored the possible correlation between the inhibition of choline uptake and apoptotic cell death. We found that both choline transporter-like protein 1 (CTL1) and CTL2 mRNAs and proteins were highly expressed in esophageal cancer cell lines (KYSE series). CTL1 and CTL2 were located in the plasma membrane and mitochondria, respectively. Choline uptake was saturable and mediated by a single transport system, which is both $Na^+$-independent and pH-dependent. Choline uptake and cell viability were inhibited by various cationic drugs. Furthermore, a correlation analysis of the potencies of 47 drugs for the inhibition of choline uptake and cell viability showed a strong correlation. Choline uptake inhibitors and choline deficiency each inhibited cell viability and increased caspase-3/7 activity. We conclude that extracellular choline is mainly transported via a CTL1. The functional inhibition of CTL1 by cationic drugs could promote apoptotic cell death. Furthermore, CTL2 may be involved in choline uptake in mitochondria, which is the rate-limiting step in S-adenosylmethionine (SAM) synthesis and DNA methylation. Identification of this CTL1- and CTL2-mediated choline transport system provides a potential new target for esophageal cancer therapy.

The Status and Prospect of Technical Development of CTL (Coal-to-Liquid) in Korea (CTL (Coal-to-Liquid) 국내 기술 개발 현황)

  • 석탄을 합성석유로 전환시키는 석탄액화(CTL) 공장은 2차 세계대전시 독일 및 영국에서 가동되어 대량의 연료를 공급한 바 있다. 전후 대형 유전이 발견되어 값싼 석유가 공급되면서 CTL 공장의 운전은 중단되었다. 남아프리카공화국의 Sasol사만이 유일하게 1955년에 CTL공장의 조업을 시작하여 현재 하루 15만배럴의 석탄합성석유를 생산하고 있다. 최근 고유가가 지속되고 석유공급에 대한 불안감 때문에 여러 개의 석탄액화 프로젝트가 진행되고 있다. 중국은 2030년까지 석탄합성석유를 연간 3천만톤(60만배렬/일) 생산할 계획을 수립하였고, 2만배럴/일 규모의 석탄직접액화공장이 2008년 완공될 예정이다. 미국에서도 8개의 CTL 프로젝트가 진행되고 있다. 호주, 필리핀, 인도네시아, 인도 등에서도 석탄액화 프로젝트를 추진하고 있다. 석유를 전량 수입하는 우리나라도 에너지안보 차원에서 CTL에 대한 접근이 필요하다. 본고에서는 한국에너지기술연구원에서 추진되고 있는 석탄 기준 10톤/일급 석탄 합성석유 생산 공정 설계, 설비 시공 현황 및 향후 계획에 대하여 기술하였다.
