Effects of 14 Chung-bu Medicinal Materials Described in the Dongui Bogam on Inflammatory Cytokines Production in HaCaT Keratinocytes (피부각질형성세포에서 동의보감 충부약재 14 종이 염증성 사이토카인 생성에 미치는 영향)

  • The purpose of this study was to investigate the effects of 14 Chung-bu medicinal materials described in the Dongui Bogam on inflammatory cytokine production in HaCaT human keratinocyte cells. In order to confirm this possibility, we screened inhibition activity of 17 cytokines using Bio-Plex ProTM Human Cytokine 17-plex assay in HaCaT cell lines. Of the 14 Chung-bu medicinal materials, Holotrichia (Ho) and Scorpio (Sc) exerted inhibitory effects on interleukin (IL)-5 production; Ho, Mantidis Ootheca (MO), and Hirudo (Hi) exerted inhibitory effects on IL-6 production; Ho, MO, Lumbricus (Lu), Hi, and Meretricis Concha (MC) showed significant inhibitory effects on IL-8 production; Gecko (Ge), Bombycis Faeces (BF), Cicadidae Periostracum (CP), and MC showed significant inhibitory effects on IL-13 production; and Testudinis Chinemis Plastrum et Carapax (TCPC), BF, and Lu exerted significant inhibitory effects on MIP-1β production. Results indicated that the Chung-bu medicinal materials might be a good candicate as potential anti-inflammatory agents for inhibition of skin inflammation. However, further investigations on these materials, including mechanistic studies, should be carried out to validated the effects in human skin equivalent models of dermatitis.

Ethanol Extracts of Citrus Peel Inhibits Adipogenesis through AMPK Signaling Pathway in 3T3-L1 Preadipocytes (진피 에탄올 추출물의 AMPK signaling pathway를 통한 3T3-L1 지방전구세포의 adipogenesis 억제에 관한 연구)

  • Citrus peel (CP) is used as a traditional herb with diverse beneficial pharmacological activities, such as anti-inflammatory, anti-oxidant, and anti-allergic effects. However, the anti-obesity effects of citrus peel are poorly defined. The aim of this study was to evaluate ethanol extracts of citrus peel (EECP) for its adipocyte differentiation and adipogenesis in 3T3-L1 preadipocytes. The aim of this study was to evaluate an EECP for its adipocyte differentiation and adipogenesis in 3T3-L1 preadipocytes. Treatment with EECP significantly suppressed the terminal differentiation of 3T3-L1 preadipocytes in a dose-dependent manner, as confirmed by a decrease in lipid droplet number and lipid content and an accumulation of cellular triglyceride. EECP exhibited potential adipogenesis inhibition and downregulated the expression of pro-adipogenic transcription factors, such as sterol regulatory elementbinding protein-1c (SREBP-1c), peroxisome proliferator-activated receptor-γ (PPARγ), CCAAT/enhancerbinding proteins α (C/EBPα) and C/EBPβ, and adipocyte expressed genes, such as adipocyte fatty acid binding protein (aP2) and Leptin. In addition, EECP treatment effectively activated the AMP-activated protein kinase (AMPK) signaling pathway; however, compound C, a specific inhibitor of AMPK, significantly reduced the EECP-induced inhibition of adipogenesis. Taken together, these results indicate EECP showed strong anti-obesity effects through the AMPK signaling pathway, and further studies will be needed to identify the active compounds that confer the anti-obesity activity of EECP.

Helium dielectric barrier discharge-cold plasma treatment for microbiological safety and preservation of onion powder (유전체 방벽 방전 콜드 플라즈마 기술을 이용한 양파 분말 미생물 안전성 향상 및 품질 보존)

  • Efficacy of dielectric barrier discharge-cold plasma treatment (DBD-CPT) for microbial decontamination of onion powder was evaluated. Onion powder, inoculated with Escherichia coli O157:H7, Salmonella Enteritidis, or Listeria monocytogenes, was treated with helium DBD-CPT. DBD-CPT (9 kV, 20 min) inhibited E. coli O157:H7, S. Enteritidis, and L. monocytogenes by $1.4{\pm}0.5$, $2.3{\pm}0.3$, and $1.2{\pm}0.0log\;CFU/cm^2$, respectively. The inactivation levels of E. coli O157:H7, S. Enteritidis and L. monocytogenes increased by $2.2{\pm}0.1$, $2.5{\pm}0.1$ and $1.9{\pm}0.3log\;CFU/cm^2$, respectively, as water activity increased from 0.4 to 0.8, and increased by $2.3{\pm}0.4$, $2.1{\pm}0.1$ and $1.6{\pm}0.1log\;CFU/cm^2$, respectively, as the particle size increased from 0.3 to $1.0cm^2$. Neither the ascorbic acid and quercetin concentrations nor the color of onion powder was changed by DBD-CPT (p>0.05). These results demonstrate the potential for application of DBD-CPT in improving microbiological safety of onion powder while preserving the physicochemical properties.

Effects of hot melt extrusion processed nano-iron on growth performance, blood composition, and iron bioavailability in weanling pigs

  • This study was conducted to investigate the effects of hot melt extrusion (HME) nano-iron as an alternative for the common ferrous sulfate on iron (Fe) bioavailability, growth performance, nutrient digestibility, intestinal morphology, and intestinal microbiota of weanling pigs. A total of 200 piglets (Landrace ${\times}$ Yorkshire ${\times}$ Duroc) were randomly allotted to seven treatments on the basis of initial body weight (BW) and sex. Treatments were the INO100 (100 ppm Fe as $FeSO_4$), HME-Fe levels (50, 75, and 100 ppm nano-Fe as $FeSO_4$). ORG100 (100 ppm Fe as iron methionine). In phase 1, the HME50 pigs showed the lowest Fe content in feed and feces. Plasma Fe concentration was increased in HME100 and ORG100 pigs. In phase 2, there were significantly lower concentration of Fe in feed and feces of HME50 pigs (p < 0.01). A lower Fe concentration in the plasma and liver were observed in HME50 pigs compared with HME100 pigs. Concentration of red blood cell (RBC) was the lowest (p < 0.01) for HME50 pigs. During phase 2, the HME100, HME75, and ORG100 pigs showed a higher RBC and hemoglobin values compared with HME50 pigs. Digestibility of gross energy (GE) and crude protein (CP) were significantly higher in HME100 pigs compared with HME50 pigs. There was an increased (p < 0.01) villus height in the duodenum and jejunum of HME100 pigs compared with HME50 pigs. It is concluded that dietary Fe does not improve growth performance of weanling pigs; however, increasing the dietary iron concentration in weanling piglets increased the RBC and hemoglobin. In addition, the potential ability of HME to be used at a lower level (HME75) was observed.

  • Objectives The purpose of this study is to investigate differences in brain activities when Neurodevelopmental treatment (NDT) is used alone compare to NDT is combined with intradermal acupuncture (IDA) treatment, using functional infrared spectroscopy (fNIRS) Methods Three children less than 7 year-old with cerebral palsy were participated. On their first visit, only NDT was used. After a week, they were treated with both NDT and IDA. During the treatment, fNIRS was used to measure any changes in their brain activities. Results In first patient with NDT, oxyhemoglobin level was increased during Standing exercise and Gait training compared to resting state. When the patient was treated with NDT and IDA, oxyhemoglobin level was decreased during Standing exercise and Gait training compared to resting state, and the result was significant (p<0.05). In second patient, oxyhemoglobin level was decreased in Gait training compared to resting state when NDT was used, but the level was increased when NDT and IDA were used in Gait training compared to resting state (p<0.05). In third patient, the difference in oxyhemoglobin levels between Gait training and resting state was significant (p<0.05). Conclusions Treatment involving both NDT and IDA has more potential to improve brain activities compared to that of NDT alone, and no adverse effect was reported. In order to confirm the finding, larger scale randomized controlled trials are needed.

Apoptosis of Kinetin Riboside in Colorectal Cancer Cells Occurs by Promoting β-Catenin Degradation

  • The Wnt/β-catenin pathway plays essential roles in regulating various cellular behaviors, including proliferation, survival, and differentiation [1-3]. The intracellular β-catenin level, which is regulated by a proteasomal degradation pathway, is critical to Wnt/β-catenin pathway control [4]. Normally, casein kinase 1 (CK1) and glycogen synthase kinase-3β (GSK-3β), which form a complex with the scaffolding protein Axin and the tumor suppressor protein adenomatous polyposis coli (APC), phosphorylate β-catenin at Ser45, Thr41, Ser37, and Ser33 [5, 6]. Phosphorylated β-catenin is ubiquitinated by the β-transducin repeat-containing protein (β-TrCP), an F-box E3 ubiquitin ligase complex, and ubiquitinated β-catenin is degraded via a proteasome pathway [7, 8]. Colorectal cancer is a significant cause of cancer-related deaths worldwide. Abnormal up-regulation of the Wnt/β-catenin pathway is a major pathological event in intestinal epithelial cells during human colorectal cancer oncogenesis [9]. Genetic mutations in the APC gene are observed in familial adenomatous polyposis coli (FAP) and sporadic colorectal cancers [10]. In addition, mutations in the N-terminal phosphorylation motif of the β-catenin gene were found in patients with colorectal cancer [11]. These mutations cause β-catenin to accumulate in the nucleus, where it forms complexes with transcription factors of the T-cell factor/lymphocyte enhancer factor (TCF/LEF) family to stimulate the expression of β-catenin responsive genes, such as c-Myc and cyclin D1, which leads to colorectal tumorigenesis [12-14]. Therefore, downregulating β-catenin response transcription (CRT) is a potential strategy for preventing and treating colorectal cancer. Plant cytokinins are N6-substituted purine derivatives; they promote cell division in plants and regulate developmental pathways. Natural cytokinins are classified as isoprenoid (isopentenyladenine, zeatin, and dihydrozeatin), aromatic (benzyladenine, topolin, and methoxytopolin), or furfural (kinetin and kinetin riboside), depending on their structure [15, 16]. Kinetin riboside was identified in coconut water and is a naturally produced cytokinin that induces apoptosis and exhibits antiproliferative activity in several human cancer cell lines [17]. However, little attention has been paid to kinetin riboside's mode of action. In this study, we show that kinetin riboside exerts its cytotoxic activity against colon cancer cells by suppressing the Wnt/β-catenin pathway and promoting intracellular β-catenin degradation.

Development of PCR-based markers for selecting plastid genotypes of Solanum hjertingii (Solanum hjertingii 색소체 유전자형 선발을 위한 PCR 기반 분자마커 개발)

  • The tetraploid Solanum hjertingii, a wild tuber-bearing species from Mexico is a relative of potato, S. tuberosum. The species has been identified as a potential source of resistance to blackening for potato breeding. It does not exhibit enzymatic browning nor blackspot which are physiological disorders. However, due to their sexual incompatibility, somatic hybridization between S. hjertingii and S. tuberosum must be used to introduce various traits from this wild species into potato. After somatic hybridization, molecular markers are essential for selecting fusion products. In this study, the chloroplast genome of S. hjertingii was sequenced by next-generation sequencing technology and compared with those of other Solanum species to develop specific markers for S. hjertingii. The chloroplast genome has a total sequence length of 155,545 bp, and its size, gene content, order and orientation are similar to those of the other Solanum species. Phylogenic analysis including 15 other Solanaceae species grouped S. hjertingii with S. demissum, S. hougasii, and S. stoloniferum. After detailed comparisons of the chloroplast genome sequence with eight other Solanum species, we identified one InDel and seven SNPs specific to S. hjertingii. Based on these, five PCR-based markers were developed for discriminating S. hjertingii from other Solanum species. The results obtained in this study will aid in exploring the evolutionary aspects of Solanum species and accelerating breeding using S. hjertingii.

Evaluation of Growth Characteristics and Yield Potential of Summer Emergency Forage Crops (하계 응급 조사료 자원의 생육특성 및 조사료 생산성 평가)

  • This study was carried out to evaluate the growth characteristics and forage yield potential for warm season grass as emergency forages. The experimental design was a randomized block design (RBD) with three replications. Two barnyard millet (Echinochloa species cv. Shirohie and Jeju native), a pearl millet (Pennisetum glaucum cv Feed milk 2) a proso millet (Panicum miliaceum cv Native), a teffgrass (Eragrostis tef cv. Tiffany) and a kleingrass (Panicum coloratum cv. Selection 75) were compared for forage production and quality at the Mid regions of Korea. Warm season forage crops were sown on May 21 and June 23 respectively, and in 2021, it was sown twice on May 21 and June 21 The number of days to seedling emergence for barnyard millet and teffgrass was observed approximately 10 and 3 days after seeding, respectively. The cultivation period from seeding to harvest was within 60 days for all entry spices except for the late-heading type barnyard millet (within 84 days). As for the dry matter yield by seeding date, the dry matter yield of the late-heading type barnyard millet in May seeding was the highest at 23,872 kg/ha, and the kleingrass was the lowest at 3,888 kg/ha. For the June seeding, the dry matter yield of the late-heading type barnyard millet was 17,032 kg/ha, the highest, and the proso millet, teffgrass and kleingrass showed the lowest at 5,468, 5,442, and 5,197 kg/ha, respectively. The crude protein (CP) content was varied by warm season grass species, but the early-heading type barnyard millet, teffgrass, and kleingrass showed the highest tendency, and the late-heading type barnyard millet showed the lowest at 5.7~5.9%. Neutral detergent fiber (NDF) and acid detergent fiber (ADF) content did not show a significant difference between the seeding in May, but kleingrass in June sowed lower than the others.

Characteristics of Fermented Hot Pepper Soybean Paste (Kochujang) Prepared by Liquid Beni-koji (액체홍국코지를 이용한 고추장의 제조)

  • Monascus anka produces biologically active materials, and liquid M. anka koji was employed for preparing fermented hot pepper soybean paste (kochujang). Three kinds of koji (M. anka, Aspergillus oryzae and mixed koji) were used to prepare kochujang, and changes of physicochemical characteristics were examined during fermentation. A. oryzae koji showed the highest ${\alpha}-amylase$ and protease activities; whereas, M. anka koji showed the highest ${\beta}-amylase$ activity in liquid koji. Water content of kochujang continuously decreased, whereas viscosity increased during fermentation. The water content and viscosity was 46% and $1.4{\times}10^5\;cP$, respectively after 40 days of fermentation. The pH of kochujang with M. anka and/or A. oryzae koji was $4.63{\sim}4.65$ in the beginning and was between 4.53 and 4.67 after 40 days of fermentation. L-, a- and b-values decreased rapidly during fermentation until 20 days of fermentation. After 40 days, L- and b-values of kochujang prepared with M. anka koji showed lower values than A. oryzae, and a-value showed higher values as the amount of M. anka koji increased. Ammoniacle nitrogen content continuously increased during fermentation. Amino nitrogen content was the highest, 241.2 ㎎%, and ammoniacle nitrogen was the lowest, 97.2 ㎎%, in kochujang prepared with A. oryzae. However, amino and ammoniacle nitrogen content showed no difference between M. anka and A. oryzae koji kochujang. Kochujang prepared with M. anka koji showed a potential that kochujang can be manufactured lower level of red pepper than previously used.

Investigation on the Amount of Water Evaporation from Composting Facilities Operated in Swine Farms (양돈농가에서 퇴비화시설별 수분변화량 분석에 관한 연구)

  • The results of the research on the amount of water evaporation from composting facilities operated in swine farms are below. The number of swine per a farm was 1433 head/farm for a Simple Composting Facility (SCF) and 3500 head/farm for a Escalator composting facility(ECF) system. The capacities of the SCF and the ECF were $0.33m^3/head$ and $0.25m^3/head$, respectively. The ECF had 24.2% less capacity than the SCF. The average water contents in the swine manure for the CP and the ECF of the surveyed farms were 86.8% and 85.7%, respectively, which revealed the ECF had 1.3% less average water content than the SCF. Daily water inputs into the SCF and the ECF were $4.1kg/m^3/day$ and $6.5kg/m^3/day$, respectively. The ECF had approximately 36.9% higher water input than the SCF. Fermentation temperatures during the composting period for the SCF and the ECF were up to $45^{\circ}C$ and $70^{\circ}C$, respectively. The decreases in water contents per each square meter for the SCF and the ECF were 3.7 kg and 5.2 kg, respectively. The ECF lost approximately 28.8% more water content than the ECF, which would be caused by the difference of fermentation temperature between two systems. Fertilizer components after composting were examined. Nitrogen contents of the SCF and the ECF were similar (0.84% and 0.86%, respectively) and ${P_2}{O_5}$ contents were 0.78% and 0.74%, respectively, showing the SCF had slightly higher content than the ECF. However, OM and OM/N did not show the difference between two systems. Hence, efforts to increase composting efficiency with considerations of the water content of swine manure, fermentation temperature, and water evaporation potential should be done when the SCF and the ECF were used in swine farms.
