Weighted Gene Co-expression Network Analysis in Identification of Endometrial Cancer Prognosis Markers

  • Objective: Endometrial cancer (EC) is the most common gynecologic malignancy. Identification of potential biomarkers of EC would be helpful for the detection and monitoring of malignancy, improving clinical outcomes. Methods: The Weighted Gene Co-expression Network Analysis method was used to identify prognostic markers for EC in this study. Moreover, underlying molecular mechanisms were characterized by KEGG pathway enrichment and transcriptional regulation analyses. Results: Seven gene co-expression modules were obtained, but only the turquoise module was positively related with EC stage. Among the genes in the turquoise module, COL5A2 (collagen, type V, alpha 2) could be regulated by PBX (pre-B-cell leukemia homeobox 1)1/2 and HOXB1(homeobox B1) transcription factors to be involved in the focal adhesion pathway; CENP-E (centromere protein E, 312kDa) by E2F4 (E2F transcription factor 4, p107/p130-binding); MYCN (v-myc myelocytomatosis viral related oncogene, neuroblastoma derived [avian]) by PAX5 (paired box 5); and BCL-2 (B-cell CLL/lymphoma 2) and IGFBP-6 (insulin-like growth factor binding protein 6) by GLI1. They were predicted to be associated with EC progression via Hedgehog signaling and other cancer related-pathways. Conclusions: These data on transcriptional regulation may provide a better understanding of molecular mechanisms and clues to potential therapeutic targets in the treatment of EC.

Magnesium vs. machined surfaced titanium - osteoblast and osteoclast differentiation

  • PURPOSE. This study focused on in vitro cell differentiation and surface characteristics in a magnesium coated titanium surface implanted on using a plasma ion source. MATERIALS AND METHODS. 40 commercially made pure titanium discs were prepared to produce Ti oxide machined surface (M) and Mg-incorporated Ti oxide machined surface (MM). Surface properties were analyzed using a scanning electron microscopy (SEM). On each surface, alkaline phosphatase (ALP) activity, alizarin red S staining for mineralization of MC3T3-E1 cells, and quantitative analysis of osteoblastic gene expression, were evaluated. Actin ring formation assay and gene expression analysis of TRAP and GAPDH performing RT-PCR were performed to characterize osteoclast differentiation on mouse bone marrow-derived macrophages (BMMs). RESULTS. MM showed similar surface morphology and surface roughness with M, but was slightly smoother after ion implantation at the micron scale. M was more hydrophobic than MM. No significant difference between surfaces on ALP activity at 7 and 14 days were observed. Real-time PCR analyses showed similar levels of mRNA expression of the osteoblast phenotype genes; osteopontin (OPN), osteocalcin (OCN), bone sialoprotein (BSP), and collagen 1 (Col 1) in cell grown on MM at 7, 14 and 21 days. Alizarin red S staining at 21 days showed no significant difference. BMMs differentiation increased in M and MM. Actin ring formation assay and gene expression analysis of TRAP showed osteoclast differentiation to be more active on MM. CONCLUSION. Both M and MM have a good effect on osteoblastic cell differentiation, but MM may speed the bone remodeling process by activating on osteoclast differentiation.

Increased interleukin-6 and TP53 levels in rotator cuff tendon repair patients with hypercholesterolemia

  • Background: A previous study reported that hyperlipidemia increases the incidence of tears in the rotator cuff tendon and affects healing after repair. The aim of our study was to compare the gene and protein expression of torn rotator cuff tendons in patients both with and without hypercholesterolemia. Methods: Thirty patients who provided rotator cuff tendon samples were classified into either a non-hypercholesterolemia group (n=19, serum total cholesterol [TC] <200 mg/dL) and hypercholesterolemia group (n=11, serum TC ≥240 mg/dL) based on their concentrations of serum TC. The expression of various genes of interest, including COL1A1, IGF1, IL-6, MMP2, MMP3, MMP9, MMP13, TNMD, and TP53, was analyzed by real-time quantitative reverse transcription polymerase chain reaction (qRT-PCR). In addition, Western blot analysis was performed on the proteins encoded by interleukin (IL)-6 and TP53 that showed significantly different expression levels in real-time qRT-PCR. Results: Except for IGF1, the gene expression levels of IL-6, MMP2, MMP9, and TP53 were significantly higher in the hypercholesterolemic group than in the non-hypercholesterolemia group. Western blot analysis confirmed significantly higher protein levels of IL-6 and TP53 in the hypercholesterolemic group (p<0.05). Conclusions: We observed an increase in inflammatory cytokine and matrix metalloproteinase (MMP) levels in hypercholesterolemic patients with rotator cuff tears. Increased levels of IL-6 and TP53 were observed at both the mRNA and protein levels. We suggest that the overexpression of IL-6 and TP53 may be a specific feature in rotator cuff disease patients with hypercholesterolemia.

  • This paper is intended to analyze and investigate the versity libraries in Korea. 16종광 도비직기에서 네트워크조직의 디자인발전에 관한 연구 (A Study on Development of network draft design on 16 shaft dobby loom)

  • Through network draft, it′s possible to describe curve draft with main motive in a lobby loom and to fulfill draft design more conveniently thanks to the development of computer device. Network draft was introduced by Alice Schlein, who is an American weaving artist, and I had ever published research paper on "The unfolding and development of network draft using computer dobby system" . The purpose of the next study was to develop the design of network draft while do make a design network draft in a dobby loom with 16 shafts, and could reach follow conclusion as a result of designing a variety of drafts. The initial of 4-end in a loom with 16 shafts was a basic condition to describe more perfect shape in comparison with draft in 8 shafts through the development of network. The development of draft line was essential to deride the pattern of fabric, and the pattern of draft is decided according to selecting key peg plan. Thereby, could get a variety of draft patterns derive from mix key peg plan with initial selected by developing the kind of draft line and applying diverse key peg plan. As for the variation and diversification of draft line, the shape of patters varied depending col the curve extent and connectivity of draft line and the size of curve. The pattern of network draft can be changed infinitely by free round curve of draft line. In addition, a variety of draft designs shall be developed by increasing the number of shaft, enlarging the scale of draft line, and developing more creative draft line.

A Korean Patient with Kniest Syndrome associated with Lipomeningomyelocele

  • Kniest syndrome (OMIM #156550) is a rare autosomal dominant disorder caused by a dysfunction of type II collagen, which is encoded by the COL2A1 gene (OMIM +120140) mapped to chromosome 12q13.11. Type II collagen, a molecule found mostly in the cartilage and vitreous tissues, is essential for the normal development of bones and other connective tissues. Kniest syndrome is a type II collagenopathy that presents as skeletal abnormality associated with disproportionate dwarfism, kyphoscoliosis, enlarged joints, visual loss, hearing loss, and cleft palate. This report describes a Korean patient with Kniest syndrome who was diagnosed with typical clinical features and radiologic findings. The patient presented with disproportionately short stature and kyphoscoliosis from birth. A skeletal survey revealed fused lamina in the thoracic spine, hemivertebrae, flexion deformities in multiple joints, and plagiocephaly.

공동확장개념에 의한 사질토에서의 말뚝의 선단지지각 해석 (End Bearing Capacity of a Single Pile in Cohesionless Soils using Cavity Expansion Concept)

  • 사질토에 있어서의 말뚝지지력을 해석하기 위하여 아세톤 고형, 수지충진기법을 사용한 모형말뚝 실험을 하여 흙의 변형을 조사하였다. 이 결과로부터 새로운 변형형태가 가정되었으며, 구형공동확장이론을 적용하여 새로운 해석법을 제시하였다. 새로운 해석법에 의한 말뚝지지력은 극한 구형공동확장압력과 지지력 상관계수로 표시된다. 본 해석법의 결과는 공동확장이론을 근거로 한 타 해석법과 비교되었다. 비교 결과, 본 해석법은 공동확장이론에 의한 말뚝지지력 해석의 결점으로 지적되는 실제보다 높은 지지력을 주는 문제점을 해결할 수 있을 것으로 기대된다.

개방철심형 고온초전도한류기의 동작 특성 (Operational Characteristics of a Superconducting Fault Current Limiter with an Open Core)

  • Recently. the high-tc superconducting fault col-rent limiters (SFCL) are studied worldwide to be classified as a resistive type or an inductive type such as a magnetic shielding type and a inductive type. The high-tc SFCL wish an open core belongs to the magnetic shielding type SFCL. Unlike conventional magnetic shielding type SFCLS it uses the open core to reduce the mechanical vibrations and installation space, The high-tc SFCL with an open core was designed and manufactured by stacking three BSCCO 2212 tubes. It was tested in the maximum source voltage of 400 Vrms. The results such as the reduction of fault current and impedance of the SFCL are described in this paper. The results show that the fault current in the source voltage of 400 Vrms was reduced to be about 123 Apeak. about 3.9 times greater than the normal state current. Also, the impedance of the high-tc SFCL was about 9${\Omega}$ about 9 times greater than the normal state impedance. The impedance of the SFCL appears just after the fault, and its size is dependent on the source voltage. From the impedance, the inductance of the SFCL was calculated.

급식형태와 우리나라 영양사의 직무만족도의 관련성에 관한 연구 (Relationships Between Characteristics of Foodservice and Job Satisfaction of the Korean Dietitian)

  • This study was carried out to investigate relationships between characteristics of foodservice and job satisfaction of Korean dietitians. The subjects were 2987 dietitians who responded to the questionnaire distributed at the annual education program of the Korean Dietetic As-sociation in 1995. The results were followings; 1)The level of jov satisfaction of Korean dietitians was low for payment work environment and value achievement as a dietitian while it was relatively high for the relationship with col-leagues. The other parts of job satissatisfaction such as work load work contents supervision by the superiors participation of policy decision and selp disccertion were just below average. 2)The type of operating style menu style and serving time in foodservise were not sig-nificantly related to all fields of job satisfaction but the kinds of meals served a day and the to-tal number of persons served a day were significantly correlated. 3) The dietitians who had to provide meal plans such as breakfast+dinner and + snack or all day serving had significantly lower job satisfaction in all fields. 4)Job satisfaction was significantly higher when the total number of persons served a day was between 500-700.(Korean J Community Nutrition 2(2) : 232~245, 1997)

Platelet-Rich Plasma Enhances Proliferation and Migration and Inhibits Inflammatory Processes in Canine Chondrocytes

  • This study was performed to assess the anti-inflammatory and cartilage regenerative effects of platelet-rich plasma (PRP) on canine chondrocytes. Proliferation and migration assays under both normal and lipopolysaccharide (LPS)-induced inflammatory conditions were performed with various concentrations of PRP (1% to 10%). The expression levels of genes related to osteoarthritis were evaluated in the following groups: PRP group, LPS group and LPS + PRP group. mRNA expression levels were detected using real-time polymerase chain reaction (RT-PCR). Proliferation assays showed significantly enhanced proliferation in all PRP-treated groups compared with the no serum group. Compared with 10% fetal bovine serum (FBS), PRP concentrations above 3% in the normal condition and 1% to 7% PRP in the LPS-induced inflammatory condition were found to significantly promote chondrocyte proliferation. In the normal condition, all PRP-treated groups showed significantly increased cell migration compared with the no serum group. Chondrocyte migration was decreased with LPS-induced inflammation, but PRP treatment resulted in significantly enhanced migration compared with the other groups in this condition. According to RT-PCR, the LPS + PRP group showed significantly higher levels of COL1A1, IL-6, aggrecan and lower levels of $TNF-{\alpha}$, MMP-1, MMP-3 mRNA expression compared to the LPS group. The results of this study suggest that PRP application can enhance the proliferation and migration of canine chondrocytes and improve canine articular cartilage regeneration.
