Whole genome sequence analyses of thermotolerant Bacillus sp. isolates from food

  • The Bacillus cereus group, also known as B. cereus sensu lato (B. cereus s.l.), is composed of various Bacillus species, some of which can cause diarrheal or emetic food poisoning. Several emerging highly heat-resistant Bacillus species have been identified, these include B. thermoamylovorans, B. sporothermodurans, and B. cytotoxicus NVH 391-98. Herein, we performed whole genome analysis of two thermotolerant Bacillus sp. isolates, Bacillus sp. B48 and Bacillus sp. B140, from an omelet with acacia leaves and fried rice, respectively. Phylogenomic analysis suggested that Bacillus sp. B48 and Bacillus sp. B140 are closely related to B. cereus and B. thuringiensis, respectively. Whole genome alignment of Bacillus sp. B48, Bacillus sp. B140, mesophilic strain B. cereus ATCC14579, and thermophilic strain B. cytotoxicus NVH 391-98 using the Mauve program revealed the presence of numerous homologous regions including genes responsible for heat shock in the dnaK gene cluster. However, the presence of a DUF4253 domain-containing protein was observed only in the genome of B. cereus ATCC14579 while the intracellular protease PfpI family was present only in the chromosome of B. cytotoxicus NVH 391-98. In addition, prophage Clp protease-like proteins were found in the genomes of both Bacillus sp. B48 and Bacillus sp. B140 but not in the genome of B. cereus ATCC14579. The genomic profiles of Bacillus sp. isolates were identified by using whole genome analysis especially those relating to heat-responsive gene clusters. The findings presented in this study lay the foundations for subsequent studies to reveal further insights into the molecular mechanisms of Bacillus species in terms of heat resistance mechanisms.

부산지역 사업체급식소 식품공급업자의 위생인식조사 (A Study on the Sanitary Perception of the Food Suppliers for the Business and Industry Foodservice in Busan Area)

  • It is thought that the sanitary perception and hygienic food treatment of food suppliers as the first handlers of foodstuffs are extremely significant for the safe and sanitary management of food in group meal services. So in this study, a survey of 103 food suppliers who provide raw materials for 80 meal services in business places around Busan area was conducted on general matters, participation in sanitation education, sanitary perception, sanitary treatment of foodstuffs, knowledge of sanitation, etc. The results of the survey are as follows: 1. 42.7% of the subjects have worked for the food supply companies for one to five years and the main job of 36.9% of them is delivery service. Food suppliers who handle over two foodstuffs are 6839%. 2. Concerning the participation in sanitation education, 43.7% of the subjects have not experienced any such education, The reason of 23.3% of them for not having the education is that there have been no opportunity for them at all. And 83.5% of them regard the education on hygiene as necessary. 3. In the sanitary perception, 93.1% of the subjects considered the temperature control of the food delivery vehicles as important and 82.5% of them replied on of the leading causes of food poisoning as foodstuffs. 64.0% thought of their knowledge of food sanitation as not very good, but moderate. 4. Concerning sanitary treatment of foodstuffs, 50.5% of delivery vehicles were wearing sanitary uniforms and 24.3% of them washed their hands while supplying food. 5. In the score of hygienic knowledge, the average score of all food suppliers was 60.6 point. In the score of hygienic knowledge on general matters, managing supervisors got 6.31$\pm$1.70. In the score of hygienic knowledge based on the perception of food sanitation, the food suppliers with the experience of sanitation education scored 6.42$\pm$1.93 point and the point was significant(p<0.01), compared with that of the food suppliers without the experience of sanitation education. The food suppliers who answered their knowledge was very good scored the highest point, 8.00$\pm$1.41. The food suppliers who replied that sanitation education was truly necessary recorded the hygienic knowledge score of 6.75$\pm$1.77, significantly(p<0.01) high. In the score of hygienic knowledge on the basis of the practical degree of sanitary handling of food materials, the food suppliers who answered they cleaned their delivery vehicles everyday scored 6.48$\pm$1.93, the food suppliers who answered they sterilized their delivery vehicles everyday scored 6.29$\pm$1.89, the food suppliers who answered they controlled the temperature of their delivery vehicles irregularly scored 6.58$\pm$1.79 and the food suppliers who answered they washed their hands every time when they were working scored 6.86$\pm$2.24, significantly(p<0.05) high in comparison with every item in other factors. And the food suppliers who answered they were wearing sanitary uniforms irregularly while supplying foodstuffs scored the highest point, 6.66$\pm$2.92.

천연물을 이용한 살균 및 살바이러스 효과에 관한 연구 (A Study of Antimicrobial & Antiviral Effect of Natural Product)

  • 식중독 원인균인 S. aureus, E. coli O157:H7, S. typhimurium, S. enteritidis, L. monocytogenes, 장티프스 원인균 S. typhi, 패혈증 원인균 V. parahaemolyticus, 세균성 이질 원인균 S. sonnei를 $20^{\circ}C$에서 30분간 $그리존^{TM}$과 접촉시킨 결과 최소 3배 희석액부터 최대 24배 희석액에서 최대 24배 희석액에서 균에 대한 살멸 효과를 나타내었다. $그리존^{TM}$ 3배 희석액을 이용하여 30초, 1분, 5분간 균과 접촉시킨 결과 S. aureus만 제외하고 모두 30초에 100% 살멸하는 효과를 나타내었다. $그리존^{TM}$을 이용하여 사스의 원인체와 동일한 바이러스인 코로나 바이러스에 대한 살바이러스 효과를 본 결과 제품의ㅣ 5배 희석액까지 유효간 살바이러스 효과를 보였다. 사람과 접촉이 많은 애완견 바이러스인 파보바이러스 (CPV), 디스템퍼바이러스 (CDV)에 대하여 시험한 결과 유기물과 경수 등의 악조건에서도 바이러스를 살멸하는 효과를 나타내었다. $그리존^{TM}$을 이용한 회와 냉장육 등에서의 적용 시험결과 미생물 생육이 현저히 저해됨을 알 수 있었다. 체소내의 미생물에 대한 살멸효과 시험에서도 미생물의 수가 현저하게 감소하였고, 특히 대장균의 수가 현저하게 감소하였다. $그리존^{TM}$은 세균, 바이러스의 살멸에 탁원한 효과를 보이며, 음식물에 직접 처리할 시에도 그 안전성과 효능이 입증되었다.

  • 알칸화합물(alkanes)에서 탄소-수소결합을 활성화시켜서 더욱 값이 비싸고 더 유용한 알켄화합물(alkenes)로 만들 수 있는 촉매를 만들고자 지난 수 십 년간 많은 화학자들이 연구해왔다 이러한 목적의 하나로서 두개의 수소를 가지는 이리디움 P-C-P핀서(pincer) 착물 $(IrH_2{C_6H_3-2,6-(CH_2PBu_2^t)_2})$을 성공적으로 합성하였다. 이 착물은 알칸의 탈수소화반응(dehyrogenation)에 아주 효과적인 촉매로 알려졌다 알칸의 탈수소화반응에 촉매독으로 작용하는 질소, 물, 이산화탄소 및 일산화탄소와 같은 작은 화합물들과 직접 반응시켜서 어떻게 촉매독으로 작용하는지를 알아봤다. 촉매독으로 작용할 수 있는 화합물들을 합성하여 핵자기공명분광법(NMR)과 적외선분광법(IR)으로 확인하였고 분자구조를 알아보기 위해서 단결정X-ray 회절법을 통하여 확인하였다. 본 논문에서는 촉매 및 촉매독물질의 합성과 분자구조와 각각의 화합물의 반응성과 특이성을 알아보았다.

전통 명태식해(식해)의 기능성 (Physiological Functionalities of Traditional Alaska Pollack Sik-hae)

  • 본 연구에서는 전통적 명태식해를 제조한 다음 저장.숙성조건에 따른 명태식해의 항균성, 항산화성, 항고혈압성 및 항콜레스테롤성 등의 생리적 기능특성에 대해 조사하였다. 항균성에서는 9종의 균주에 대해 모두 항균작용이 있는 것으로 나타났으며 , 이중 gram 양성균에 대해 Listeria monocytogenes를 제외하고 저장 직후에 항균활성이 나타났으며, 모든 gram 양성균에 대해 전 저장기간 동안 항균활성이 확인되었고, Staphylococcus aureus에 대해 강한 항균활성을 나타내었다. 반면에 gram 음성균 및 진균류에 대해서는 항균활성 이 gram 양성균에 비해 약하였다. 항산화성은 저장.숙성조건에 따라 모두 관능적으로 맛이 우수한 15~16일 경에 가장 강한 항산화활성을 나타내었으며, 그 후로는 저장기간이 증가함에 따라 감소하는 경향을 나타내었다. 항고혈압성은 명태식해 제조직후를 제외하고는 모든 저장.숙성조건에서 항고혈압활성이 나타났으며, 항콜레스테롤성은 2$0^{\circ}C$ 15일과 변온 16일에서만 항콜레스테롤활성이 검출되었다. 이러한 결과를 바탕으로 할 때 전통적인 방법에 따라 제조된 명태식해는 김치류와 같은 우수한 생리적 기능성을 가지고 있었으며, 명태식해의 유통기간 연장을 위한 변온조건에서도 그 생리적 기능성이 계속해서 유지됨을 알 수 있었다.

중학교 교과과정에 따른 보건교육 내용 분석 연구 (A study on health education content analysis in middle school curriculum)

  • The study reserches the related contents of the health education in the existing gymnastics and the domestics textbook, catches learning-Ulaching contents by the health-control function and compares the contents with the health-education curriculum of the Texas State in America. This study is questioned and reserched the general features of the teachers who is change in the health education, their attitudes about the teaching of it, and the recognition degree about their health education. The results above is like the following. 1. Learning-teaching contents by the health-control function. The domestics and gymnastics which contains the health education contents, is not discripted continually and at the same time is short of the contents in the health-control function according to the students' individual domains. Also the recent serious problems, (that is, Smoking, Drinking and Medical poisoning, Emotion, and Psychic domains) were no mentioned. 1) In gymnastics textbook, the physical domains are mentioned 8, 9 in juinor, and senior class, the affective, psychic domains only senior class and the social domains only 1 grade except junior and senior class. Growth-development function is concentratedly stated only 1 grade in physical domains and there is no stated in. The social domains are referred a little in senior class. In cure-recovery functions, the physical domains is being taught in 2, and 3 grade, but not 1 grade. Specially, the social domains are being taught in all grade, but, the affective domains are not at all 2) In the subject of domestics, the physical domains of the daily life functions are taught in all the years, but the emotional, mental domains are taught a little in 1 and 2 grades. The growth-development functions are being taught about the nutrition-health, in the physical parts of all graries, and the emotional-mental, social domains are being taught some in 7 and 8 grade except 3 grade, he cure-recovery functions ere taught in only 1 grade, the contents of rehabilitative functions are not being taught like gymnastics. 2. The general characteristics of the health-education teachers and the health-education. In the calss field, the average age of gymnastics-teachers is from 35 to 40 years old, and it of domestics teachers is from 24 to 29 years old. 1) In the class-hours a week, the gymnastics teachers take lessons 3 hours (75.6%) and the domestics teachers 4 hours (65.6%). In "the health-lessons hours alloted"in a semister, 3 or 4 hours is 38.7% and no hours is 18.0% in gymnastics teachers. On the other hand, there is no hours to teach the health-education in domestics teachers. About the opinion: the health subject must be taught as a independent subject, 42.9% of the gymnastics teachers agreed and 45.3 % of the domestics teachers did. 2) The factors which have an influence upon the health-teachers #. In age, the class lesson hours of a week(r= .0014), the confident manner about the lessons(r= .0485) and the necessity of the lessons (r= .0316) are closely related one another. #. In sexes, the lesson hours a class(r= .0000), the alloted hours of health lesson a semister(r= .0000) the confident manner about the class(r= .0001). the quantity of lesson(r= .0001): and, the teacher's belief(r= .0264) are co-related one another. #. In subject in change. the class hours a week(r= .0000), the alloted hours of health-lesson a semister(r= .0000) the quantity of the lesson(r= .0114) are co-related each other. #. In majority, the class lesson hours a week(r= .0000) the alloted hours of health lesson a semister(r= .0000) the quantity of the lesson are co-related each other. The quantity of lesson and the confidence about the lesson (r= .3223) and the quantity of lesson and the interest of lesson (r= .2450) are co-related each other.

주요일간지(主要日刊紙)의 보건의료관계사설(保健醫療關係社說) 내용분석(內容分析) (An Analysis on Contents of Health-Concerned Editorials in Korean Neswspapers)

  • The editorials of the leading newspapers may reflect as well as help formulate the public opinions to a significant degree. Bearing this in mind, this study was carried out to provide information useful in formulating such public health policies that could practically meet the social interests and demands in health appearing in the editorials of the newspapers. This analysis covered the editorials of 5 leading newspapers such as Donga Ilbo, Chungang Ilbo, Hankook Ilto, Chosun Ilbo and Seoul Shinmoon for about 10 years from Jan. 1st, 1970 through Oct. 31th, 1979. The major findings are as follows: 1. The total number of health-concerned editorials in the five daily papers for the period was 1,768 or occupied 6.4% of the total editorials of the same sources. The increasing trend of frequnecy of the health-concerned editorials since 1976 indicates the increment of social interests and demands in health. Analysing the contents, environmental pollution received the greatest attention in those editorials, which was followed by medical affairs, environmental sanitation, and disease control in order in terms of frequency of appearance. However, there was a tendency that the interests in the environmental pollution, medical affairs and social welfare tended to increase year by year, whereas those in environmental hygiene and disease control comparatively decreased. 2. Motives of dealing with the health-concerned editorials were provided by announcement of the governmental policies and implementation for 25.6% and by out-breaks of the relevant events for 23.9%. This tendency coincides with the general characteristics of the editorials that reflect the timely issues. Closely analysing, however, the fact that the motives engendered by the out-breaks of the relevant events or by the season concerned comparatively tended to decrease as years pass by, indicates that the editorials tend more to seek the future-oriented demands in health rather than the current issue-oriented. 3. The editorials appeared to be more concerned with the governmental policies. 95.7% of all the editorials analysed were addressed to the government. This signifies the role of government in the field of public health and medical affairs. Their attitudes toward health-related policies of the government were much more negative than the other editorials that were addressed to the government in other fields. This suggests that the governmental interests in health were neither sufficient nor fair. 4. What the editorials most stressed were; (1) increment of governmental interests in health, (2) improvement of the governmental health administration, (3) enriching the basic statistics, and (4) development of various technologies pertinent to health affairs and disease control, and so forth. However, must of their suggestions were not concrete but rather abstract and conceptual. 5. The editorials also expressed strong interests in research area. The areas of the needed research most suggested by items are; (1) development of effective health care delivery system, (2) establishment of more practical system of calculating medical cost, (3) implementation of effective policies to control degenerative diseases, (4) division of medical care services and pharmaceutical services systems, (5) effective ways to prevent Co poisoning accidents, (6) changing status of environmental pollution and its effects upon health, and (7) status of occupational diseases, and so forth. 6. There were some editorials -not small in quantity -that have risk to mislead the public opinions as well as the health policies due to lack of professional knowledge of the writers. It is desirable to establish some kind of mechanism that screens the erroneous contents of the editorials to help prevent misleading opinions.

프로폴리스 추출물의 항균 활성에 대한 연구 (Studies on the Antimicrobial Effect of Extracts of Propolis)

  • 기존에 식품의 저장성 향상을 우해 사용되어졌던 방법들의 문제점들을 최소화하고 보다 효과적인 식품저장을 위해 전통적으로 식용되어 왔으며 그 안전성이 확보된 프로폴리스를 대상으로 대표적인 식품 부패 미생물에 대한 생육억제활성을 식용되어 왔으며 그 안전성이 확보된 프로폴리스를 대상으로 대표적인 식품 부패 미생물에 대한 생육억제활성을 탐색하고 건강 지향적인 식품보존제로서 이를 식품에 적용하여 보다 좋은 품질의 가공식품개발에 활용코자 본 연구를 진행하였다. 프로폴리스 에탄올 추출물의 일반성분 분석 결과를 보면 19.41% 수분, 0.39% 조단백, 5.12% 조지방, 0.15% 조회분의 결과를 보여주었다. 프로폴리스는 에탄올 60%, 에탄올 80% 추출물에서는 높은 추출율을 보였다. 프로폴리스 에탄올 추출물에 의한 항균성은 100% 에탄올 추출물로 실험한 처리구에서 높은 항균 효과를 나타내어 이를 사용하여 유해 미생물들에 대한 농도별 항균 효과를 살펴보았다. 프로폴리스 추출물로 유해미생물을 처리했을 때 Bacillus subtilis, Micrococcus luteus는 1~2 mg/mL로 처리 시 생균수가 1~4log 감소하였고, E. coli는 3mg/mL로 처리 시, Bacillus cereus는 1~2 mg/mL의 노도로 처리 시 대조군에 비해 생균수가 2log 감소하였다. Staphylococcus aureus와 Shigella sonnei의 사멸효과는 0.1~3 mg/mL의 농도에서 생균수가 2~4log 감소하였다. Erwinia rhapontici의 사멸효과는 2 mg/mL 농도로 실험했을 때 2 log 정도 생ㅇ육저해효과를 보였고, Salmonella choleraesuis, Vibrio parahaemolyticus에서의 사멸효가는 크게 관찰할 수 없었다. 프로폴리스는 그램양성균에 대한 사멸효과가 그램음성균에 대한 효과 보다 더 우수하였다.

초정수압 처리에 의한 고등어(Scomber japonicus)육 중의 히스타민 생성 억제 효과 (Inhibitory Effect of High Hydrostatic Pressure Treatments on Histamine Production in Mackerel Scomber japonicus)

  • This study investigated the inhibitory effect of high hydrostatic pressure (HHP) treatments on histamine production in mackerel Scomber japonicus. Changes in viable cell counts, histamine contents, pH and VBN of mackerel fillet (stored at $4^{\circ}C$ for 25 days) were examined under HPP (200, 300, and 400 MPa). HPP treatments reduced viable cell counts by 2-3 log cycles during storage. Viable cells of mackerels treated with 400 MPa did not appear for 5 days. Histamine production was nearly suppressed by 300 and 400 MPa HPP treatments after 25 days. Furthermore, mackerels treated with HPP showed significantly lower VBN values compared with the control. Additionally, pH values were not affected by the treatments during storage periods. These results suggest that HPP treatment decreased histamine contents in mackerel muscles. Based on our results, HPP treatment may reduce scombroid fish poisoning by decreasing histamine production in mackerel during $4^{\circ}C$ storage.

한약재 및 해조류 추출물에 의한 고등어 육중의 Histamine 생성 억제 효과 (Inhibitory Effects of Histamine Production in Mackerel Muscle by Medicinal Herbs and Seaweed Extracts)

  • 한약재 및 해조류 추출물에 대한 Photobacterium phosphoreum의 항균력을 측정한 결과, 오미자 열수 및 에탄올 추출물과 감태 및 비틀대 모자반 에탄올 추출물이 항균력을 나타내었으며 비틀대 모자반과 감태 에탄올 추출물이 각각 0.015625 mg/mL, 0.125 mg/mL의 최소저해농도를 나타내었다. P. phosphoreum 유래 HDC의 저해 효과를 측정한 결과, 감태와 대황 에탄올 추출물이 1 mg/mL에서 각각 32%, 25%의 높은 저해활성을 나타내었다. P. phosphoreum에 대해 항균력과 HDC 저해 효과를 나타낸 천연물을 고등어육에 첨가한 후 균주를 접종하여 생균수 및 histamine 생성량을 측정한 결과, 저장일차가 증가함에 따라 처리구에서 균수가 크게 감소하였다. Histamine의 함량도 다소 감소하였지만 주로 항균활성에 의해 histamine이 저해된 것으로 나타났다.
