Preliminary construction of a chimeric cellulose operon containing two structural genes coding for CMCase and cellobiase

  • CMCase, a member of cellulose decomposing enzymes, hydrolyze cellulose up to cellobiose. Cellobiase splits cellobiose to glucose units. Therefore, a linkage of the twogenes coding for CMCase and cellobiase on the same plasmid is needed to produce a cellulase complex which can produce glucose from cellulose. A genetic operon in which the two structural genes are under the control of a single promoter would be ideal for this purpose. The present report is on the linking of the two cellulase genes in one plasmid as a preliminary step of the operon construction.

  • This study was carried out to select the useful bacteria which secret extracellula enzymes for enzymatic deinking agent of old newspaper. CMCase, FPase and xylanase activities of the bacteria liquid culture were measured at optimal growth conditions. Clear zone test was checked on the solid culture. The results of this study were as follow: Eight strains of 28 bacteria isolated from a paper mill soil ground were shown strong CMCase and xylanase activity with the clear zone test. The optimal pH and temperature for culture growth were 6~8 and 26~$34^{\circ}C$, respectively and optimal culture period were less than 60 hours. Based on CMCase, FPase and xylanase activity, strain No. 18, 21, 22 and 28 which were relatively higher than the other strains, were selected for further enzymatic deinking research.

  • Caboxymethyl cellulase (CMCase) I was purified from the induced culture filtrate of Penicllium verruculosum F-3 by ammonium sulfate precipitation, DEAE-Sephadex A-50 chromatography and Bio-gel P-150 filtration. The purified enzyme was assumed to be a glycoprotein consisting of 8.5% carbohydrate and having a molecular weight of 70.000 in SDS-polycrylamide gel electrophoresis (SDS-PAGE). The purified enzyme-specific anti-CMCase I IgG was obtained by rabbit immunization and protein A-sepharose CL-4B chromatography. The fungal poly($A^+$) RNA was isolated from the total RNA of the mycelium grown under cellulase induction conditions by oligo(dT)-cellulosse chromatography. The translation products in vitro were prepared by translating the isolated poly ($A^+$) RNA in rabbit reticulocyte lysate and analyzed by SDS-PAGE and fluorography. Of the translation products, CMCase I was identified by the immunoprecipitation against anti-CMCase I IgG.

Properties of the CMCase produced by Pseudomonas sp. YD-15 (Pseudomonas sp. YD-15가 생산하는 CMCase의 특성)

  • A bacterium having CMCase activity was isolated form soil and identifed as a Pseudomonas sp YD-15. The optimum conditions for the production of CMCase were avicel 1.2%, yeast extract 0.5%, $KNO_3$ 0.06%, $K_2HPO_4$ 0.2%, $MgSO_4$ $7H_2O$ 0.15%, pH 8.0, $30^{\circ}C$ and 60 hours cultivation. The CMCase was purified 15.2 folds with 14ft yield through ammonium sulfate precipitation, DEAE-sepharose column chromatography and sephadex G-100 gel filtration chromatography. The optimum pH and temperature for the enzyme activity were 6.0 and $50^{\circ}C$, respectively. The enzyme was stable between pH 5.0 and 8.0, below $50^{\circ}C$. The molecular weight was calculated about 100,000 by SDS-polyacrylamide gel electrophoresis. $K_m$ value for CMC used as a substrate was 40 mg/ml.

Isolation and Characterization of Thermophilic Bacillus licheniformis YJ09 from Spent Mushroom (Pleurotus ostreatus) Substrates (느타리 수확후배지로부터 분리된 고온성 Bacillus licheniformis YJ09의 특성)

  • In order to isolate thermophilic bacteria with high activity of CMCase and xylanase, spent mushroom substrates was collected from an oyster mushroom cultivation farm in Jinju, Gyeongnam, Korea. Among the isolates, one strain designated as YJ09 was selected by agar diffusion method. The isolate YJ09 was identified as a member of Bacillus licheniformis based on biochemical characteristics using Bacillus ID kit and MicroLog system. Comparative 16S rDNA sequence analysis showed that isolate YJ09 formed a distinct phylogenetic tree within the genus Bacillus and was most closely related to Bacillus licheniformis with sequence similarity of 98.9%. Based on its physiological properties, biochemical characteristics and phylogenetic distinctiveness, the isolate YJ09 was classified as Bacillus licheniformis. The CMCase and xylanase activity of B. licheniformis YJ09 was slightly increased corresponding to the bacterial population from exponential phase to stationary phase in the growth curve of B. licheniformis YJ09.

  • A bacterium producing the extracellular xylanase and CMCase was isolated from soil and has been identified as Bacillus sp. The isolate, named Bacillus sp. A-7, was shown to be very similar to Bacillus licheniformis on the basis of its biochemical and physiological properties. The maximum xylanase and CMCase production were obtained when 2.0% (w/v) glucose and 0.3% (w/v) yeast extract were used as carbon source and nitrogen source, respectively. The best mineral conditions for xylanase and CMCase production were 0.1%(w/v) $CaC1_2$. Among the various feedstuffs, 1.0%(w/v) soybean meal was selected for the best xylanase and CMCase production.

Production of Cellulase from Cellulomonas sp. KL-6 (Cellulomonas sp. KL-6에 의한 섬유소 분해효소의 생산)

  • Among the cellulases by Cellulomonas sp. KL-6. CMCase and filter paperase, which were produced as the out enzymes of cell, had been much produced, but very small amounts of ${\beta}-glucosidase $, the enzyme of which is cell bound form, was produced by this organism. The optimal culture times for CMCase and filter paperase productions were 5 days, while that of ${\beta}-glucosidase$ was 4 days. When this strain was cultured under the optimal medium for enzyme production, CMCase, FPase and ${\beta}-glucosidase$ were $82\;units/m{\ell},\;80\;units/m{\ell}\;and\;1.2\;units/m{\ell}$, respectively. Thus these results were showed to increase enzyme productivities as about $60{\sim}70%$ than those produced in basal medium. $CaCO_3$ injected to the medium as the ratio of 0.1% was not only enhanced cellulase activities but also effective as acid neutralizing agent. The production effects of lignase and lactase by this bacterium in filter paper medium was not appeared.

  • Production of alkaline carboxymethyl cellulase (CMCase) and xylanase by batch and fed-batch cultures of alkalophilic Cephalosporium sp. RYM-202 was investigated. Of carbon sources tested, wheat bran gave the highest production of those enzymes. The high levels of CMCase on carboxymethyl cellulose and xylanase on birchwood xylan suggest that the biosynthesis of CMCase and xylanase in Cephalosporium sp. RYM-202 is regulated separately at the level of enzyme induction. The temperature and pH for maximal production of those enzymes was $20^{\circ}C$ and 9.0, respectively. High concentration of wheat bran in batch fermentation resulted in the lower and delayed production of the enzymes by catabolite repression. In fed-batch fermentation with controlled feeding of 5% final wheat bran concentration, the highest activities of CMCase and xylanase were 0.39 and 9.2 units/ml, respectively, and 1.22 and 1.36 times higher respectively than those in batch fermentation on 5% wheat bran.

Studies on the cellulase properties of Aspergillus clavatus from the Cellulose-Cultural Properties (섬유질 문화재로부터 분리된 Aspergillus clavatus의 섬유소분해호소에 관한 연구)

  • Cellulolytic mocrooganisms were isolated from the cellulose-cultural properties. Among them, Aspergillus clavatus with the highest cellulase activity was identified by the morphological characteristics and it's enzyme activities were compared on the various cultural conditions. It was found that the induction of carboxymethylcellulase(CMCase), avicelase and salicinase in CMC liquid media showed the highest enzyme acitivity on five day's cultivation at $30^{\circ}C$ and thereafter their activities were gradually decreased with time. After crude extracellular enzymes precipitated with the 70% saturated ammonium sulfate solution were dialyzed with 20 mM acetate buffer pH 6.0, each crude enzyme was examined. The optimal activities of CMCase and avicelase were both found to be at $50^{\circ}C$ and pH 6.0. Their thermal stability was appeared at the $50^{\circ}C$. CMCase and avicelase had the maxima activities with 1.5% and 2.2% substrate concentration, respectively. The concentration of 5 mM $Mg^{++}$ or $Ca^{++}$ was found to have a maximum cellulase activity and its activity was inhibited with more than 5 mM $Cu^{++}$ and $Zn^{++}$ concentration. Cellulase activity was also inhibited sensitively by the inhibitors such as 2-mercaptoethanol, iodine and sodium azide.
