Growth Characteristics of Enterobacter sakazakii Used to Develop a Predictive Model

  • A mathematical model was developed for predicting the growth rate of Enterobacter sakazakii in tryptic soy broth medium as a function of the combined effects of temperature (5, 10, 20, 30, and $40^{\circ}C$), pH (4, 5, 6, 7, 8, 9, and 10), and the NaCl concentration (0, 1, 2, 3, 4, 5, 6, 7, 8, 9, and 10%). With all experimental variables, the primary models showed a good fit ($R^2=0.8965$ to 0.9994) to a modified Gompertz equation to obtain growth rates. The secondary model was 'In specific growth $rate=-0.38116+(0.01281^*Temp)+(0.07993^*pH)+(0.00618^*NaCl)+(-0.00018^*Temp^2)+(-0.00551^*pH^2)+(-0.00093^*NaCl^2)+(0.00013^*Temp*pH)+(-0.00038^*Temp*NaCl)+(-0.00023^*pH^*NaCl)$'. This model is thought to be appropriate for predicting growth rates on the basis of a correlation coefficient (r) 0.9579, a coefficient of determination ($R^2$) 0.91, a mean square error 0.026, a bias factor 1.03, and an accuracy factor 1.13. Our secondary model provided reliable predictions of growth rates for E. sakazakii in broth with the combined effects of temperature, NaCl concentration, and pH.

  • Background The prenatal ultrasound detection of cleft lip with or without cleft palate (CL/P) and its continuous management in the prenatal, perinatal, and postnatal periods using a multidisciplinary team approach can be beneficial for parents and their infants. In this report, we share our experiences with the prenatal detection of CL/P and the multidisciplinary management of this malformation in our institution's Congenital Disease Center. Methods The multidisciplinary team of the Congenital Disease Center for mothers of children with CL/P is composed of obstetricians, plastic and reconstructive surgeons, pediatricians, and psychiatrists. A total of 11 fetuses were diagnosed with CL/P from March 2009 to December 2013, and their mothers were referred to the Congenital Disease Center of our hospital. When CL/P is suspected in the prenatal ultrasound screening examination, the pregnant woman is referred to our center for further evaluation. Results The abortion rate was 28% (3/11). The concordance rate of the sonographic and final diagnoses was 100%. Ten women (91%) reported that they were satisfied with the multidisciplinary management in our center. Conclusions Although a child with a birth defect is unlikely to be received well, the women whose fetuses were diagnosed with CL/P on prenatal ultrasound screening and who underwent multidisciplinary team management were more likely to decide to continue their pregnancy.

  • Steady-state sublimation vapour pressures of anhydrous bismuth trichloride have been measured by the continuous gravimetric Knudsen effusion method from 370.6 to 488.8 K. Additional effusion measurements have also been made from 438.2 to 495.7 K by the torsion effusion method. Based on a correlation of ${\Delta}_H0298$.15 and ${\Delta}_H0298$.15, a recommended p(T) equation has been obtained for BiCl3(s); log(p/Pa)=-C/(T/K)-2.838log(T/K)-8.097${\times}$10-2(K/T)2+22.588, where p is in Pa, T in Kelvin and ${\Delta}_H0298$.15 in kJmo1-1 and C=(${\Delta}_H0298$.15+5.9988)/1.9146${\times}$10-2. Condensation coefficients and their temperature dependence have been derived from the effusion measurements.

  • In order to establish informations important to the measurement of lipoxygenase (LOX) activity, providing conditions most favorable for its action and determining factors that inhibit activity, the influence of extraction buffer, substrate, pH, storage, temperature, NaCl, $CaCl_2$, other cations and antioxidants on LOX activity, and localization of LOX in cucumber tissues were carried out. The most favored substrate for LOX was linolenic acid followed by linoleic and arachidonic acids. LOX activity in both peel and mesocarp tissue extracts was maximum at pH 5.5 and relatively stable at $40^{\circ}C\;and\;50^{\circ}C$ temperature. The condition of 0.2 M NaCl with pH 5.0 was observed to provide optimum LOX stability. The enzyme activity was reduced by addition of cations, $Mn^{2+},\;Cu^{2+}\;or\; Al^{3+}$, except $Ca^{2+}$ which stimulated activity of LOX. Butylated hydroxy anisole (BHA) and propyl gallate decreased LOX activity with increasing concentration. Cucumber peel had higher activity than other tissues, locule or mesocarp, of cucumber.

  • Effects of sodium hypochlorite (NaOCl) concentration, temperature and pH on oxidation mechanism of corn starch were investigated. The rate of oxidation was dependent on the concentration of hypochlorite, pH and temperature of oxidation. The reaction was either first or second order depending on the concentration of NaOCl. At oxidant concentration of $0.75{\sim}3.0%$ active Cl/g starch, the reaction was first-order and it was second-order at $3.75{\sim}4.5%$ active Cl/g starch. The first-order rate constants were increased with increasing oxidant concentration. The rate of oxidation of starch was highest at pH 7 and decreased with increasing acidity or alkalinity of the medium. As the reaction temperature increased, the rate of oxidation was increased.

  • This study was to investigate the effect of salt stress on physiological characteristics such as plant growth, photosynthesis, solutes related to osmoregulation of Beta vulgaris. A significant increase of dry weight was observed in 50 mM and 100 mM NaCl. The contents of Chl a, b and carotenoid were lower in NaCl treatments than the control. On 14 day after NaCl treatment, photosynthetic rate (PN), the transpiration rate (E) and stomatal conductance of CO2 (gs) were reduced by NaCl treatment. On 28 day after NaCl treatment, the significant reduction in gs and E was shown in NaCl 200 mM. However, PN and water use efficiency (WUE) in all NaCl treatments showed higher value than that of control. Total ion contents (TIC) and osmolality were higher than the control. On 14 day after treatment, the contents of proline (Pro) increased significantly in 200 mM and 300 mM NaCl concentration compared with control, whereas on 28 day in all treatments it was lower than that of the control. The contents of glycine betaine (GB) increased with the increase of NaCl concentration. The contents of Na+, Cl-, GB, osmolality and TIC increased with the increase of NaCl concentrations. These results suggested that under severe NaCl stress conditions, NaCl treatment did not induce photochemical inhibition on fluorescence in the leaves of B. vulgaris, but the reduction of chlorophyll contents was related in a decrease in leaf production. Furthermore, increased GB as well as Na+ and Cl- contents resulted in a increase of osmolality, which can help to overcome NaCl stress.

Inhibition and Chemical Mechanism of Protocatechuate 3,4-dioxygenase from Pseudomonas pseudoalcaligenes KF707 (Pseudomonas pseudoalcaligenes KF707에서 유래한 protocatechuate 3,4-dioxygenase 의 저해 및 화학적 메커니즘)

  • We carried out pH stability, chemical inhibition, chemical modification, and pH-dependent kinetic parameter assessments to further characterize protocatechuate 3,4-dioxygenase from Pseudomonas pseudoalcaligenes KF707. Protocatechuate 3,4-dioxygenase was stable in the pH range of 4.5~10.5. L-ascorbate and glutathione were competitive inhibitors with $K_{is}$ values of 0.17 mM and 0.86 mM, respectively. DL-dithiothreitol was a noncompetitive inhibitor with a $K_{is}$ value of 1.57 mM and a $K_{ii}$ value of 8.08 mM. Potassium cyanide, p-hydroxybenzoate, and sodium azide showed a noncompetitive inhibition pattern with $K_{is}$ values of 55.7 mM, 0.22 mM, and 15.64 mM, and $K_{ii}$ values of 94.1 mM, 8.08 mM, and 662.64 mM, respectively. $FeCl_{2}$ was the best competitive inhibitor with a $K_{is}$ value of $29{\mu}M$. $FeCl_{3}$, $MnCl_{2}$, $CoCl_{2}$, and $AlCl_{3}$ were also competitive inhibitors with $K_{is}$ values of 1.21 mM, 0.85 mM, 3.98 mM, and 0.21 mM, respectively. Other metal ions showed noncompetitive inhibition patterns. The pH-dependent kinetic parameter data showed that there may be at least two catalytic groups with pK values of 6.2 and 9.4 and two binding groups with pK values of 5.5 and 9.0. Lysine, cysteine, tyrosine, carboxyl, and histidine were modified by their own specific chemical modifiers, indicating that they are involved in substrate binding and catalysis.

Effect of Submerged Culture of Ceriporia lacerata Mycelium on Insulin Signaling Pathway in 3T3-L1 Cell (Ceriporia lacerata 균사체 배양물이 3T3-L1 세포에서 인슐린 신호 전달에 미치는 영향)

  • In this study, we evaluated the antidiabetic effect of submerged culture of Ceriporia lacerata mycelium (CL01) on glucose uptake and the expression of mRNA and protein of major signal markers of insulin signaling pathway in 3T3-L1 adipocytes. After 3T3-L1 adipocytes were pre-treated by CL01 (0, 2, 10 mg/ml) for 8 hours, followed with treatment of insulin, the glucose uptake levels significantly increased by more 55.1%, 94.4% than negative control respectively (p<0.01, 0.001) in a dose-dependent manner. However, in case of CL01 pre-treatment without insulin, the glucose uptake did not increase compared with insulin-treated 3T3-L1. Also we demonstrated that the protein expression levels of pIR β, pAkt, pPI3K and pAMPK and the mRNA expression levels of GLUT4 in adipocytes inducing insulin resistance increased in CL01-treated group compared with negative control. These results demonstrated that CL01 affected glucose metabolism and the protein and gene expression through insulin signaling pathway, and increased glucose uptake levels effectively. More than 90% of those who have suffered for type 2 diabetes are more likely to have from hyperinsulinemia, hypertension, obesity and etc. because of altered insulin signaling pathway. So, it is probably considered that intake of CL01 may treat type 2 diabetes by normalization of insulin signaling pathway, and it will provide useful evidences regarding a mechanism for cure of type 2 diabetes.

A Study on pH and Soluble Reactive Phosphorus (SRP) from Litter Using Various Poultry Litter Amendments During Short-Term: A Laboratory Experiment (다양한 깔짚 첨가제 이용이 단 기간 깔짚내 pH와 수용성 인에 미치는 영향에 관한 연구)

  • The objective of this study was conducted to evaluate the effects of poultry litter amendments on pH and soluble reactive phosphorus (SRP) in poultry litter. Two laboratory studies were conducted for 42 d in Exp. 1 and for 10 d in Exp. 2, respectively. The poultry litter was treated with various amendments which included 4 g fly ash and 4 g $AlCl_3\;(AlCl_36H_2O)/100g$ litter in Exp. 1 and 4 g alum$(Al_2(SO_4){_3}\;14H_2O)$, 8 g alum, 8.66 g liquid alum, and 17.3 g liquid alum/100 g litter in Exp. 2; untreated litter served as controls. There were no differences in pH between control and T1(4 g fly ash) and SRP contents between T1(4 g fly ash) and T2(4 g $AlCl_3$) in Exp. 1. A significant difference in pH and SRP contents in Exp. 2 was observed among all treatments(P< 0.05). In experiment 1, T1(4 g fly ash) and T2(4 g $AlCl_3$) at 42 d decreased SRP in litter by 47.1% and 62.6% of that from litter alone, respectively. In experiment 2, T1(4 g alum), T2(8.66 g liquid alum), T3(8 g alum), and T4(17.3 g liquid alum) treatments at 10 days reduced SRP contents by up to 36.2%, 62.9%, 87.0%, and 83.9%, respectively, when compared with the controls. Decrease in SRP contents was chiefly associated with reduction in litter pH. These results indicate that use of various litter amendments to limit P solubility has potential and should be pursued as a means of reducing soluble reative phosphorus during short term.

Studies on the Infection Condition and Formaldehyde Inactivation of Human Rotavirus Wa (사람 로타바이러스 Wa의 감염 조건 및 formaldehyde에 의한 inactivation에 대한 연구)

  • Effects of protease, $CaCl_2$ EGTA, polybrene, medium pH, and formaldehyde on the infection and inactivation of human rotavirus Wa were investigated using T-flask culture of monkey kidney MA-104 cells. Rotavirus titer was improved by the addition of trypsin or clostripain. Rotavirus titer was increaeed 8 and 10 time sin the infection medium supplemented with 300 $\mu\textrm{g}$/mL of $CaCl_2$ and in the medium adjusted its pH to 8, respectively. However, addition of EGTA or polybrene to the medium decreased rotavirus titer. Rotavirus titer was reduced to 53-95% of the initial value at 1 hr after formaldehyde treatment. Furthermore, rotavirus was inactivated more than 98% at 12 hrs after formaldehyde treatment.
