Background: Epstein Barr virus (EBV) infected B cells are transformed into lymphoblastoid cell lines. Some researchers suggested some a few similarities between this process and carcinogenesis. We observed the expression of CD80 and CD86, co-stimulatory molecules on EBV-transformed B cells and changes of CD54 expression after stimulation of CD80 and CD86. Methods: CD80 and CD86 were stimulated using anti-CD80 and anti-CD86 monoclonal antibodies. To assess apoptosis and surface protein expression, flow cytometric analysis was performed. Intracellular signal molecules were evaluated by RT-PCR and immunoblot. Morphology and localization of proteins were examined using inverted or confocal microscope. Results: Cross-linking of CD80 and CD86 induced apoptosis and interfered with proliferation of EBV-transformed B cells, and dispersion of clumped cells. We also examined that their stimulation induced ROS accumulation and reduced CD54 expression. Interestingly, we observed that CD80 and CD86 diminished the expression of CD54 in different methods. Both CD80 and CD86 downregulated activation of focal adhesion kinase. CD80 stimulus inhibited CD54 expression through mainly RhoA inactivation, while CD86 down-regulated Ras and JNK phosphorylation. Conclusion: These results suggest that co-stimulatory CD80 and CD86 molecules, expressed EBV-transformed B cells, may play a role in apoptosis and cell adhesion.
The determination of Cd2+ and Cu2+ with ammonium pyrrolidinedithiocarbamate (APDC) in Tween 80 micellar media has been studied. The UV-visible spectrum of Cd(PDC)2 complex in Tween 80 media had more sensitivity tha n in chloroform. Although the UV-visible spectrum of the Cu(PDC)2 complex in Tween 80 media had somewhat less sensitivity than that in chloroform, absorbance data of Cu2+ were more reproducible in Tween 80 media. The Cd(PDC)2 and Cu(PDC)2 complexes were very stable at pH 7.0 for up to 100 minutes and could be quantitatively chelated if APDC were added to the sample solution more than 30 times the moles of Cd2+ and Cu2+ . The optimum concentration of Tween 80 was 0.1%. The calibration curves of Cd(PDC)2 and Cu(PDC)2 complexes with good linearity were obtained in 0.1% Tween 80 media. The detection limits of Cd2+ and Cu2+ were 0.0493 ㎍mL-1 and 0.0393 ㎍mL-1 , respectively. Recovery yields of Cd2+ and Cu2+ ions in the spiked real samples were almost 100%. Based on experimental results, this proposed method could be applied to the rapid and simple determination of Cd2+ and Cu2+ in real samples.
We fabricated gamma radiation detector using high resistive p-Cd$_{80}$Zn$_{20}$Te grown by high pressure bridgman method and forming au thin film electrode by chemically electroless deposition method. The device of Au/Cd$_{80}$Zn$_{20}$Te/Au is a typical MIM structure. The characteristic of current-voltage showed good linearity to 3kV/cm but it depend on the square of electric field over 3kV/cm. As the results of rutherford backscattering spectroscope(RBS) and auger spectroscope on the Au/Cd$_{80}$Zn$_{20}$Te, Au penetrated to the surface of Cd$_{80}$Zn$_{20}$Te detector absorbed slightly high energy radiation like a few hundred keV and showed good performance to detect low energy gamma ray.mma ray.
생쥐 小腸의 deoxycytidine-2-$^14 C$ (CdR-2-$^14 C$)와 deoxyuridine-2-$^14 C$ (UdR-2-$^14 C$)의 代謝를 관계酵素의 熱處理에 대한 영향과 관련해서 in vitro에서 고찰하였다. CdR-2-$^14 C$는 CdR-aminohydrolase의 作用에 의해서 먼저 nucleoside level에 급속히 deamination된 후, nucleosidase의 作用에 의해 uracil로 分解된다. 생쥐 小腸에서는 nucleosidase 가 CdR과 CR에는 親和力이 없기 때문에 이들 cytosine nucleoside의 N-pentose 結合을 分解하지 못하는 것으로 보인다. CdR-aminohydrolase는 $80^\\circ C$ 높은 不活性化溫度를 나타냈으며, 이에 반해서 nucleosidase는 $60^\\circ C$에서 不活性化가 되었다. 品種이 다른 생쥐의 여러 組織에 있는 CdR-aminohydrolase는 모두 $80^\\circ C$에서 不活性化됨이 관찰되었으나, 토끼 組織에서는 $80^\\circ C$에서도 不活性化가 일어나지 않는 점으로 미루어 不活性化溫度에는 "屬"特異性이 있는 것으로 짐작된다. 哺乳類의 分化된 組織에서 CdR-aminohydrolase 가 出現하는 生物學的 意味는 주로 分解過程과 有關한 것으로 생각된다. 것으로 생각된다.
Background: Our previous study showed that purified protein derivative (PPD)-stimulated pleural mononuclear cells (PMC) from tuberculous pleurisy (Tbp) produced significantly more $IFN-{\gamma}$ (10- to 70-fold) after in vitro PPD stimulation than freshly isolated pleural cells from malignant pleurisy. The present study was designed to determine whether blocking the CD40-CD40 ligand (CD40L) interaction decreases $IFN-{\gamma}$ production by altering IL-12 levels. Methods: IL-12 and $IFN-{\gamma}$ production after neutralizing anti-CD40L antibody treatment was compared to the efficacy of anti-CD80, anti-CD86, and a combination of anti-CD80 and CD86 (CD80+86) monoclonal antibodies (mAb). These activities were measured by enzyme-linked immunosorbent assays (ELISAs) and reverse transcription-polymerase chain reaction (RT-PCR), after in vitro stimulation with PPO antigen (Ag). Results: Neutralization of CD80, CD86 and CD80+86 did not decrease $IFN-{\gamma}$ and IL-12 production in Tbp-PMC, whereas neutralization of CD40L significantly depressed IL-12 p40 and $IFN-{\gamma}$. In addition, neutralization of CD40L completely inhibited IL-12 p40 and $IFN-{\gamma}$ mRNA expression. Conclusion: The CD40-CD40L interaction might play a major role in IL-12 and $IFN-{\gamma}$ production in Tbp-PMC, thus contributing to protective immunity in human tuberculosis.
Background: CD40-activated B (CD40-B) cells might be an attractive source of autologous antigen-presenting cells (APCs) for immunotherapy due to the convenience to obtain from peripheral blood and expand in vitro. Moreover, CD40-B cells were found to be comparable with DCs in their capacity to raise antigen-specific CD8+ T cells. Here, we have established K562 cells expressing CD40L to expand CD40-activated B cells used for APCs. Methods: After activation of B cell by K562/CD40L, CD40-B cells were examined by counting B cell numbers. Surface expression of CD54, CD80, CD86 and HLA class II was measured by flow cytometry. The CD40-B cells were tested for its function as APC by mixed lymphocyte reactions (MLR) and by induction of T cell responses specific for pp65 peptide in vitro. Results: The expansion of B cells by K562/CD40L increased about 6-folds compared with anti-CD40 or K562. Furthermore, the expression of CD54, CD80, CD86 and HLA class II was up-regulated by K562/CD40L. B cells by K562/CD40L showed comparable antigen presentation activity with mature DCs as shown in MLR, INF-${\gamma}$ ELISPOT assay. Conclusion: These results suggest that K562/CD40L could be used to generate activated B cells as potent APCs which could be useful for cellular vaccination and adoptive immunotherapy.

Granulocyte-macrophage colony stimulating factor (GM-CSF) has a role in inducing emergency hematopoiesis upon exposure to inflammatory stimuli. Although GM-CSF generated murine bone marrow derived cells have been widely used as macrophages or dendritic cells in research, the exact characteristics of each cell population have not yet been defined. Here we discriminated GM-CSF grown bone marrow derived macrophages (GM-BMMs) from dendritic cells (GM-BMDCs) in several criteria. After C57BL/6J mice bone marrow cell culture for 7 days with GM-CSF supplementation, two main populations were observed in the attached cells based on MHCII and F4/80 marker expressions. GM-BMMs had $MHCII^{low}F4/80^{high}$ as well as $CD11c^+CD11b^{high}CD80^-CD64^+MerTK^+$ phenotypes. In contrast, GM-BMDCs had $MHCII^{high}F4/80^{low}$ and $CD11c^{high}CD8{\alpha}^-CD11b^+CD80^+CD64^-MerTK^{low}$ phenotypes. Interestingly, the GM-BMM population increased but GM-BMDCs decreased in a GM-CSF dose-dependent manner. Functionally, GM-BMMs showed extremely high phagocytic abilities and produced higher IL-10 upon LPS stimulation. GM-BMDCs, however, could not phagocytose as well, but were efficient at producing $TNF{\alpha}$, $IL-1{\beta}$, IL-12p70 and IL-6 as well as inducing T cell proliferation. Finally, whole transcriptome analysis revealed that GM-BMMs and GM-BMDCs are overlap with in vivo resident macrophages and dendritic cells, respectively. Taken together, our study shows the heterogeneicity of GM-CSF derived cell populations, and specifically characterizes GM-CSF derived macrophages compared to dendritic cells.
According to reports, it is impossible to make Ohmic Contact with high resistivity p type CdTe or CdZnTe semiconductor theoretically. But it is in need of making Ohmic Contact to fabricate semiconductor radiation detector By electroless deposition method using gold chloride solution, we made Ohmic Contact of Au and p-Cd$_{80}$Zn$_{20}$Te which grown by High Presure Bridgman Method in Aurora Technologies Corporation. We investigated the interface with Rutherford Backscattering Spectrometry and Auger electron spectroscopy. And we evaluated the degree of Ohmic Contact for the Au/CdZnTe interface by the I/V characteristic curve. As a result, we concluded that it showed excellent Ohmic Contact property by tunneling mechanism through the interface.e.
Cd을 처리하여 기내 배양한 담배 식물의 생장과 엽록소 함량, rubisco와 rubisco activase의 함량과 활성에 미치는 질산염의 영향을 연구하였다. Cd에 의해 억제되었던 담배 식물의 생장과 엽록소의 함량은 질산염에 의해 회복되었으며, 80 mM 질산염에서 회복 효과가 가장 높았다. 80 mM 질산염에서의 rubisco의 함량이 타 농도에서 보다 현저하게 증가하였으며, rubisco의 활성 또한 rubisco의 함량과 같은 변화를 보였다. 이 결과들은 Cd에 의해 감소된 rubisco의 함량과 활성이 질산염에 의해 회복되었음을 의미한다. SDS-PAGE의 결과, 55 kD와 15 kD의 large subunit와 small subunit의 강도는 타 농도에서 보다 80 mM 질산염에서 현저하게 증가하였다. Rubisco activase의 함량과 활성을 측정한 결과, 80 mM 질산염에서의 rubisco activase의 함량은 타 농도에서 보다 현저하였으며, 이의 활성은 함량과 같은 양상을 나타내어, rubisco에 대한 질산염에 의한 회복 현상이 rubisco activase와 관련 있음을 추측하게 한다.
본 연구에서는 해양오염퇴적물의 유기 오염물질을 제거하기 위한 세척기반처리 공정 중 중금속 특히 생물이용이 가능한 중금속 부분(Bioavailable Heavy Metal)의 동시 처리 가능성에 대해 평가하였다. 해양오염퇴적물 정화사업 대상인 N해역에서 퇴적물을 채취하여 본 연구의 시료로 사용하였다. 세척기반처리 공정에서 첨가제로는 산화제인 과산화수소($H_2O_2$)와 비이온 계면활성제(Tween-80)를 사용하였다. 퇴적물과 해수의 고액비는 1 : 3이며 과산화수소의 농도 1 M, 3 M, 4 M, 5 M과 0.05% Tween-80을 첨가하여 각 반응 시간(10, 20, 30, 40, 50, 60, 70, 80분, 24시간) 조건에서 시험하였다. 그 결과 총유기탄소(Total Organic Carbon, TOC) 제거 효율은 5 M + 0.05% Tween-80 24시간에서 최대 55.2%로 나타났다. 중금속 총 함량(Total Heavy Metals)은 구리(Cu), 아연(Zn), 카드뮴(Cd) 각각 29.5, 42.3, 73.0%의 제거 효율로 5 M + 0.05% Tween-80 10분에서 가장 좋은 제거효율을 보였다. 생물이용이 가능한 중금속 부분의 경우도 Cu, Zn, Cd에서 각각 60.0, 77.7, 90.2%로 5 M + 0.05% Tween-80 10분에서 가장 좋은 효율을 나타내어 유기물 처리 공정에서의 Cu, Zn, Cd 등 일부 중금속의 동시 제거 가능성을 확인하였다. 총유기탄소 함량과 생물이용이 가능한 중금속 부분의 상관분석 결과 제거 된 Cu, Zn, Cd은 $r^2$값이 0.94, 0.85, 0.69로 좋은 상관관계를 보여주었다.
