관절6호방이 Type II collagen으로 유발된 관절염에 미치는 영향 (Suppressive Effect of Kwanjulbang-6 on Collagen-Induced Arthritis in Mice)

  • This study was carried out to know the effects of Kwanjulbang-6(GJB-6) on the inhibition of arthritis. GJB-6 was orally administered to mouse with arthritis induced by collagen II . Cytotoxicity, hepatotoxicity, arthritis index, value of immunocyte in draining lymph node and paw joint, rheumatoid factor in serum were measured in vivo. The incidence of arthritis was significantly decreased. Total cell number of draining lymph node was significantly increased compared with control. Total cell number of paw joint was significantly decreased compared with control. The absolute number of $CD19^+$, $CD8^+$, $CD3^+/CD69^+$, $CD3^+/CD49b^+$, $CD4^+/CD44^+$, $CD3^+$, $CD4^+$, $CD4^+/CD25^+$ and $CD3^+/CD8^+$ cells in draining lymph node were significantly increased compared with control. The absolute number of $CD3^+$, $CD4^+$, $CD4^+/CD25^+$ and $CD11b^+/Gr-1^+$ cells in paw joint were significantly decreased compared with control. The absolute number of $B220^+/CD23^+$ and $MHCII^+/CD11c^+$ cell in draining lymph node were significantly decreased compared with control. The levels of IgG was decreased and The levels of IgM was significantly decreased compared with control. Anti-collagen II in serum was significantly decreased compared with control. With the hematoxylin and eosin stain, the cartilage destruction and synovial cell proliferation were decreased compared with control. With the Masson's trichrome stain, the expression of collagen fibers was decreased compared with control. Results showed that GJB-6 had immunomodulatory effects. So we expect that GJB-6 should be used as a effective drugs for not only rheumatoid arthritis but also another auto-immune disease.

배독환(排毒丸)의 아토피피부염 병태모델에서의 면역 억제 효능 (Effects of Baedokhwan on Immune Modulation in Atopic Dermatitis Model of NC/Nga Mice)

  • Atopic dermatitis induced NC/Nga mice were used to investigate the efficacy of BDH(Baedokhwan) on the recovery of dermatitic symptoms through its influence on the immune related factors and histological changes. First of all, BDH treated group showed improvement of atopic dermatitis with naked eye observation, and significant decrease of clinical index(CI) was observed after 14 weeks. And Infiltration of leukocytes was suppressed in BDH treated group, and the thickness of hypertrophied epidermis and dermis were decreased. In dorsal skin, BDH treated group showed significant decrease of the ratio of CD3+, CD11b+/Gr-1+ immune cells by 52.8%, 25.2, respectively. And also significant decrease the level of IL-5 mRNA and IL-13 mRNA by 44.4%, 28.0, respectively. In PBMC and serum, BDH treated group showed an decrease of CD4+/CD45+, B220+/CD23+, CD4+, CD8+, CD4+/CD25+ immune cells by 35.0%, 12.6%, 42.7%, 31.6% and 55.6%, respectively, and the level of histamine was decreased by 39.0%. The results above indicated that BDH clinically used for atopic dermatitis treatment has objective validity, and therefore can be provided as the basic data for anti-allergic and anti-inflammatory studies.

  • 카드뮴 1 mM를 함유한 배지에서 배양된 고도 카드뮴 내성 효모 Hansenula anomala B-7으로부터 분리한 malate dehydrogenase(카드뮴 적응 MDH)와 카드뮴을 함유하지 않은 배지에서 배양된 사용균주의 malate dehydrogenase(카드뮴 비적응 MDH)활성에 미치는 금속 이온의 영향을 비교 검토했다. 카드뮴 적응 MDH와 비적응 MDH 활성은 1 mM $Cd^{2+}$에 의하여 대조구에 비하여 450%와 150%가 각각 증가되었으며, 카드뮴 적응 MDH 활성은 1 mM $Zn^{2+}$, $Mn^{2+}$$Cu^{2+}$에 의하여 각각 147%, 150%와 135% 증가되었다. 더욱이, 카드뮴 적응 MDH 활성은 2 mM $Cd^{2+}$와 2.5 mM $Zn^{2+}$에 의하여 각각 925%와 250% 증가되었다. 카드뮴에 의한 카드뮴 적응 MDH의 활성 촉진은 효소반응 4분으로 최대값에 도달했으나, 카드뮴 비적응 MDH의 활성 촉진은 효소 반응 초기인 1분으로 최대값에 도달했다. 카드뮴 적응 MDH와 비적응 MDH의 Km값은 두 효소 모두 6.9 mM L-malate였다. 그러나, 카드뮴 비적응 MDH 활성은 카드뮴에 의하여 Km값이 변화되지 않았고 Vmax만 증가되었으나 카드뮴 적응 MDH 활성은 카드뮴에 의하여 Km값과 Vmax값 모두 변화되었다.

마산만과 낙동강 하구역 해양 퇴적토의 중금속 오염도 산정 연구 (Estimation of Heavy Metal Contamination Level in Masan Bay and Nakdong Estuary Sediments)

  • 본 연구는 마산만과 낙동강 하구 인근의 해양퇴적토에 포함된 중금속 농도를 측정하여 기존 오염도 산정방법을 이용하여 6개 중금속에 대한 오염도를 평가하고자 하였다. 미국 환경청 기준에 따르면 Cu의 경우 B1지역은 심한오염으로 분류되었고 Ni과 Zn의 경우 일부지역에서 중간정도 오염으로 분류되었다. Igeo, EF, PERF의 분류에 따르면, A와 B 지역은 Cd에 의해 오염이 된 것으로 나타났다. 특히, B1 지역의 경우 Igeo, EF, PERF 값이 전 지역을 통틀어 제일 높은 값을 나타내어 심각한 오염으로 평가되었다. 6개 중금속 모두의 영향을 고려한 mean PEL quotient 분류에 따르면 모든 지역에서 중금속으로 인해 독성이 발생할 확률은 21%로 파악되었다. B1 지역의 Cd 농도는 1.5mg/kg으로 가장 높은 값을 보였고 이는 기존의 연구와 비교해서도 가장 높은 값임을 알 수 있었다. 따라서, 마산만 인근의 퇴적토에 포함된 Cd의 오염이 심각하므로 이에 대한 원인을 밝히고 향후 처리방안에 대해 신중한 접근이 필요할 것으로 사료된다.

마행감석양가감방(麻杏甘石揚加減方)이 천식모델생쥐의 면역세포 및 사이토카인에 미치는 영향 (Effects of Mahaenggamseok-tang-gagambang on Immune Cells and Cytokines in OVA-Induced Asthmatic Mice)

  • The purpose of this research is to evaluate the effect of Mahaenggamseok-tang-gagambang (MGTG) on airway hyper- responsiveness (AHR), immune cells, cytokines and lung tissue in OVA-induced asthmatic mice. C578L/6 mice were injected, inhaled and sprayed with OVA for 12 weeks (3times a week) for asthma sensitization and challenge. Two experimental groups were treated with different concentrations of MGTG (400 mg/kg and 200 mg/kg) extract and cyclosporin A (10 mg/kg) for the later 8 weeks. Enhanced pause (Penh) levels were measured by whole body plethysmography. Immune cells were analyzed by flow cytometer in peripheral blood monocyte cell (PBMC) and lung cells. The IL-1b, IL-12, IFN-${\gamma}$, OVA-lgE, IL-4, IL-5, TNF-${\alpha}$ were analyzed by ELISA kit in serum and splenocyte+a-cCDS/a-CD28. Enhanced pause (Penh) levels of the MGTG groups (400 mg/kg and 200 mg/kg) were decreased significantly compared with that of control group. The numbers of MGTG groups (400 mg/kg and 200 mg/kg) on lung total cells were decreased significantly compared with that of control group. The numbers of MGTG groups (400 mg/kg and 200 mg/kg) on $CD3^+/CD69^+$, $B220^+/CD22^+$, $B220^+/CD23^+$, $B220^+/lgE^+$, $CCR3^+$ cells were decreased significantly compared with that of control group. The number of MGTG group (400 mg/kg) on $CD3^+/CD49b^+$ cells was decreased significantly compared with that of control group. The level of MGTG groups (400 mg/kg and 200 mg/kg) on IL-4, IL-5, IL-12, TNF-${\alpha}$, OVA-lgE were decreased significantly compared with that of control group. The level of MGTG group (400 mg/kg) on IL-1b, IL-1S, OVA-lgE were decreased significantly compared with that of control group. These results demonstrate that MGTG could be a desirable alternative therapy for allergic asthma by inhibiting the expression of immune cells, the activation of inflammatory mediator.

가금티푸스 감염에 대한 키토산의 면역반응 (Immunomodulatory effect on chitosan against Fowl typhoid infection)

  • Chitosan is similar in structure to cellulose and are the second most abundant polysaccharides in nature, comprising the horny substance in the exoskeletons of crabs, shrimp and insects as well as fungi. This study was conducted to access the effect of immunomodulation responses of chitosan(N-acetyl-${\beta}$-D-glucosamine) chicken infected with in Fowl typhoid(Salmonella gallinarum). One-day-old broiler chicks were divided into eight groups: The 1st group was inoculated intra-peritoneally with chitosan and challenged intra-peritoneally with S. gallinarum. The 2nd group was inoculated intra-peritoneally with chitosan. The 3rd group was feeding with chitosan and intra-peritoneally inoculated with cyclophosphamide and challenged intra-peritoneally S. gallinarum. The 4th group was feeding with chitosan and intra-peritoneally with cyclophosphamide. The 5th group was feeding with chitosan and challenged intra-peritoneally with S. gallinarum. The 6th group was feeding with chitosan. The 7th group was challenged intra-peritoneally with S. gallinarum. The 8th group was nontreated-uninfected control group. The results shows that $CD4^+$, $CD8^+$ and B lymphocyte in lymphoid organs of chickens treated with chitosan increased in especially $CD4^+$, $CD8^+$ lymphocytes (p<0.05). The group of feeding chitosan showed the significantly increased $CD4^+$, $CD8^+$ and B lymphocyte than inoculated intra-peritoneally with chitosan. As the result suggests that the feeding of chitosan induced immunostimulatant effect than the inoculation intra-peritoeally of chitosan.

IMMUNOREGULATORY EFFECTS OF A MONOCLONAL ANTIBODY TO HUMAN 4-1 BB MOLECULE ON ALLOANTIGEN-MEDIATED IMMUNE RESPONSES.

  • 4-lBB molecule is expressed on the surface of activated CD4$\^$+/ and CD8$\^$+/ T cells. We generated a panel of anti-4-1 B5 murine mAbs using a fusion protein consisting of the extracellular domain of human 4-1 BB fused to Glutathione S-transferase. The binding activity against cell surface 4-1 BB molecule was assessed by flow cytometry analysis. These studies showed that several anti-4-1 BB mAbs bound to 10-30% of CD4$\^$+/ and CD8$\^$+/T cells in PHA or Con A stimulated PBLs, although these mAbs interacted with only, l-2% of CD4$\^$+/ and CD8$\^$+/ T cells in normal PBLs, indicating the specificity of mAbs to the 4-l BB molecule on activated CD4$\^$+/ and CD8$\^$+/ T cells. Next, we examined the effect of an anti-4-l BB mAb (4B4-1-1) on allogeneic mixed lymphocyte reactions (MLRs). The data indicated that the antibody significantly inhibited the proliferative response at higher concentrations. When tested with several T cell mitogens, the antibody had no stimulatory or inhibitory effects on the mitogen-mediated T cell proliferation. These data suggest that 4-1 BB molecule may play a role in the regulation of antigen-mediated immune response.

폐흡충 감염 마우스에 있어 Anti-lgE 단일크론 항체 처치시 혈청내 총 IgE, 비장 B 세포표면의 $Fc{\varepsilon}RII/CD23$ 발현 및 충체수에 미치는 영향 (Effects of antii-IgE mAb on serum IgE, $Fc{\varepsilon}RII/CD23$ expression on splenic B cells and worm burden in mice infected with Paragonimus westermani)

  • 윤충 감염시 증가되는 기생충 특이 IgE 항체는 항기생충 작용을 하는 것으로 알려져 있으나 총 IgE 항체의 90% 이상을 차지하는 비특이성 IgE 항체의 역할에 대해서는 잘 알려져 있지 않다 이 실험에서는 폐흡충 감염 마우스에 anti-lgE 단일크론 항체를 처치한 후 혈청내 IfgE. 비장 B 세포 의 $Fc{\varepsilon}RII/CD23$의 발현 및 충체수를 관찰하여 비특이성 IgE가 숙주의 방어면역에 미치는 역할을 알아보았다. Anti-lgE를 처치한 폐흡충 감염 마우스의 혈청내 총 IgE 항체의 양은 실험 기간 동안 억제되어(($<{\;}1{\;}{\mu\textrm{g}/ml}$) PBS로 처치한 감염군에 비해 유의한 수준(P < 0.05)으로 감소되었다 또 한 Anti-IgE를 처치한 폐흡충 감염 마우스에서의 $Fc{\varepsilon}RII/CD23$ 양성인 비장 B 세포수 및 그 발현도 PBS로 처치한 감염군에 비해 실험 기간 동안 유의한 수준(P < 0.05)으로 감소되었다. 충체 회수율에 있어서도 anti-lgE를 처치한 감염 마우스는 6주에 44%로 PBS로 처치한 감염군(75%)에 비해 유의한 수준(P < 0.05)으로 감소하였다. 이상의 결과를 종합할 때 폐흡충 감염 마우스에서 높게 증가되는 비특이성 IgE는 숙주의 방어면역을 방해하여 오히려 충체를 보호하는 역할을 하는 것으로 생각된다.

A Comparative Study of Two Different SnO2:F-coated Glass Substrates for CdTe Solar Cells

  • Two different fluorine-doped tin oxide (FTO)-coated glass substrates were investigated to find better suitability for CdTe solar cells. Substrate A consisted of FTO (300 nm)/$SiO_2$ (24 nm)/intrinsic $SnO_2$ (30 nm)/borosilicate glass (2.2 mm), and substrate B consisted of FTO (700 nm)/intrinsic $SnO_2$ (30nm)/borosilicate glass (1.8 mm). The overall thickness of the FTO/glass substrates was about 2.5 mm. The total light transmittance of substrate B was much higher than that of substrate A throughout the whole spectral region, even though the thickness of the FTO in substrate B was twice larger than that of the FTO in the substrate A. The short-circuit current greatly increased in substrate B and the external quantum efficiency (EQE) increased over the whole wavelength range. This study shows that the diffuse optical transmittance played a key role in the large EQE value in the blue wavelength region, and the direct transmittance played a key role in the large EQE value in the red wavelength region. The higher transmittance is due to the rough surface generated by the thicker FTO on glass. The conversion efficiency of the CdTe solar cell increased from 12.4 to 15.1% in combination of rough FTO substrate and Cu solution back contact.

감초(Glycyrrhiza uralensis Fisch, GLU)가 천식모델 생쥐의 BALF 내 면역세포 및 Cytokine에 미치는 효과 (Effects of Glycyrrhiza uralensis Fisch on Immunocyte and Cytokine Production in Asthma Model Mouse)

  • Objective : The aim is to identify the effects of Glycyrrhiza uralensis Fisch on immunocyte and cytokine production in asthmatic laboratory mice. This experiment was designed to investigate the antiallergic and antiinflamatary the effects of Glycyrrhiza uralensis Fisch on asthma. Materials and Methods : We measured the eosinophil, IL-4, IL-5, IL-13, $IFN-{\gamma}$, CD4, CDS, CD69, CCR3, CD11b, Gr-1 in bronchoalveolar lavage fluid of ovalbumin induced asthmatic mice. Results : Glycyrrhiza uralensis Fisch increased the proliferation of eosinophils, IL-4, IL-5, IL-13, IgE, granulocyte, CCR3, CD4, IgE, CD69. Glycyrrhiza uralensis Fisch increased the proliferation of $IFN-{\gamma}$. Conclusion : Results suggest that Glycyrrhiza uralensis Fisch extract is useful in treatment and prevention allergic asthma.
