Anti-inflammatory activity of a sulfated polysaccharide isolated from an enzymatic digest of brown seaweed Sargassum horneri in RAW 264.7 cells

  • BACKGROUND/OBJECTIVES: Sargassum horneri is an edible brown alga that grows in the subtidal zone as an annual species along the coasts of South Korea, China, and Japan. Recently, an extreme amount of S. horneri moved into the coasts of Jeju Island from the east coast of China, which made huge economic and environmental loss to the Jeju Island. Thus, utilization of this biomass becomes a big issue with the local authorities. Therefore, the present study was performed to evaluate the anti-inflammatory potential of crude polysaccharides (CPs) extracted from S. horneri China strain in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. MATERIALS/METHODS: CPs were precipitated from S. horneri digests prepared by enzyme assistant extraction using four food-grade enzymes (AMG, Celluclast, Viscozyme, and Alcalase). The production levels of nitric oxide (NO) and pro-inflammatory cytokines, including tumor necrosis factor (TNF)-${\alpha}$ and interleukin (IL)-$1{\beta}$ were measured by Griess assay and enzyme-linked immunosorbent assay, respectively. The levels of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2), nuclear factor (NF)-${\kappa}B$, and mitogen-activated protein kinases (MAPKs) were measured by using western blot. The IR spectrums of the CPs were recorded using a fourier transform infrared spectroscopy (FT-IR) spectrometer. RESULTS: The polysaccharides from the Celluclast enzyme digest (CCP) showed the highest inhibition of NO production in LPS-stimulated RAW 264.7 cells ($IC_{50}$ value: $95.7{\mu}g/mL$). Also, CCP dose-dependently down-regulated the protein expression levels of iNOS and COX-2 as well as the production of inflammatory cytokines, including TNF-${\alpha}$ and IL-$1{\beta}$, compared to the only LPS-treated cells. In addition, CCP inhibited the activation of NF-${\kappa}B$ p50 and p65 and the phosphorylation of MAPKs, including p38 and extracellular signal-regulated kinase, in LPS-stimulated RAW 264.7 cells. Furthermore, FT-IR analysis showed that the FT-IR spectrum of CCP is similar to that of commercial fucoidan. CONCLUSIONS: Our results suggest that CCP has anti-inflammatory activities and is a potential candidate for the formulation of a functional food ingredient or/and drug to treat inflammatory diseases.

Market-Based Transmission Expansion Planning Under Uncertainty in Bids by Fuzzy Assessment

  • In this paper, by a simple example it is shown that existing market-based criteria alone cannot completely and correctly evaluate the transmission network expansion from market view. However criteria congestion cost (CC) and social welfare (SW) together are able to correctly evaluate transmission network from market view and so they are adopted for the market-based transmission expansion planning. To simply indicate the limits of CC and SW social welfare percentage (SWP) and congestion cost percentage (CCP) are defined. To consider uncertainty in bids of market producers and consumers, and also indeterminacy in the acceptable boundaries of the SWP and CCP and their priorities, fuzzy assessment approach is used. In this approach, appropriate fuzzy sets and a fuzzy rule base are provided to evaluate the acceptability of an expansion plan. Then, the least-investment cost plan, which is acceptable in all probable scenarios, is searched. The proposed method is applied to an 8-bus system.

Fault Detection of Plasma Etching Processes with OES and Impedance at CCP Etcher

  • Fault detection was carried out in a etcher of capacitive coupled plasma with OES (Optical Emission Spectroscopy) and impedance by VI probe that are widely used for process control and monitoring at semiconductor industry. The experiment was operated at conventional Ar and Fluorocarbon plasma with variable change such as pressure and addition of N2 and O2 to assume atmospheric leak, RF power and pressure that are highly possible to impact wafer yield during wafer process, in order to observe OES and VI Probe signals. The sensitivity change on OES and Impedance by VI probe was analyzed by statistical method including PCA to determine healthy of process. The main goal of this study is to find feasibility and limitation of OES and Impedances for fault detection by shift of plasma characteristics and to enhance capability of fault detection using PCA.

  • Dry etcher, PECVD등의 플라즈마 공정 장비의 구조물에는 유전체로 절연된 금속판들이 사용된다. 플라즈마 이론을 보면 이들 표면의 전위는 양전하 플럭스와 음전하 플럭스가 같아서 순전류가 0이 되는 부유 전위를 갖는다. 금속과 같은 전도체의 표면은 모두 같은 전위를 가져야 한다. 일반적인 플라즈마 시뮬레이션 소프트웨어에는 이런 경계 조건이 선택 가능하지 않다. ESI사의 CFD-ACE+의 사용자 루틴 개발 기능을 이용하여 Fortran90문법으로 경계면 최인접 셀의 중심에서 구한 전자 온도와 경계면의 이온 입사 플럭스로 가중평균을 구한 이온 질량을 맥스웰분포를 가정한 부유 전위식에 대입하여 시뮬레이션을 CCP에서 구현하였다. 원형 챔버의 가장자리에 떠 있는 사각 링 전극을 가정하고 이 전극 표면이 접지 전위일때, 유전체 일때, 본 연구에서 개발한 루틴을 적용한 결과를 Ar CCP에 대해서 비교 분석하였다.

$CF_4/O_2$ Plasma에 Ar첨가에 따른 $SiO_2/Si_3N_4$ 에칭 특성 변화 (Study on the Etching Profile and Etch Rate of $SiO_2/Si_3N_4$ by Ar Gas Addition to $CF_4/O_2$ Plasma)

  • CCP방식의 식각에 있어서 CF4/O2 Plasma Etch에 Ar을 첨가함으로써 Etch특성이 어떻게 변화하는지를 조사하였다. FE-SEM를 이용하여 Etch Profile를 측정하였다. 또한 Elipsometer와 Nanospec을 이용하여 Etch rate를 측정하였다. Ar의 비율이 전체의 47%정도를 차지하였을 때까지 Etch Profile이 향상되었다가 그이후로는 다시 감소하는 것을 볼 수 있었다. Ar을 첨가할수록 etch rate은 계속 향상되었다. Ar을 첨가하는 것은 물리적인 식각으로 반응하여 Etch rate의 향상과 적정량의 Ar을 첨가했을 때 Etch profile이 향상되는 결과를 얻었다.
