우리 나라에 서식하는 뱀목 중 뱀과의 유혈목이, 능구렁이 및 무자치와 살모사과의 살모사, 까치살모사 및 쇠살모사를 재료로 하여 그 위장관에 출현하는 장내분비세포들 중 bombesin, CCK-8, gastrin, glucagon, insulin, PP, serotonin, somatostatin, substance P 및 VIP 등 10종류의 세포를 면역세포화학적 방법으로 염색한 후 광학현미경하에서 세포의 분포, 출현빈도, 모양 및 특징 등을 관찰하였던 바 다음과 같은 결론을 얻었다. 1. 능구렁이, 유혈목이, 무자치, 까치살모사 및 쇠살모사의 위장관 점막상피에서는 CCK-8, gastrin, PP 및 serotonin 등 4종류의 분비세포가 관찰되었으며 동물의 종 및 위장관 각 부위별에 따라 내분비세포의 다과의 차이는 다소 있었다. 2. 살모사의 위장관 점막상피에서는 CCK-8, gastrin 및 serotonin 등 3종류의 분비세포가 관찰되었다. CCK-8, gastrin 및 serotonin 세포는 위장관 전반에 걸쳐 관찰되었으며 gastrin세포는 유문부와 소장 점막에 출현하였으며 각 부위에 따른 수의 차이가 있음을 알게 되었다. 3. 위장관 점막상피에서 관찰된 장내분비세포들의 모양은 위에서는 추체형 및 난원형을 띠었고 폐쇄형이었으며 소장 및 대장 전부에서는 주로 방추형을 띤 개구형이었으나 소수는 폐쇄형도 있었다. 대장 후부 및 직장에서는 난원형의 폐쇄형이었다. 4. 본 실험에서 사용된 10종류의 항혈청 중에서 bombesin, glucagon, insulin, somatostatin, subsatance P 및 vasoactive intestinal polypeptide 항체 등에 양성반응을 보인 장내분비세포들은 관찰되지 않았다.
dopamine이 십이지장구를 포함한 소장의 운동성에 미치는 영향, cholecystokinin이 dopamine의 작용에 미치는 영향 그리고 이들 작용의 신경성 기전을 알아보고자 다음과 같은 실험을 실시하였다. ether로 마취한 토끼 54마리에서 십이지장구, 십이지장, 공장 그리고 회장을 적출하고 절편(길이 1cm)을 만들어 Krebs-Ringr 용액이 채워진 기록 용기에 넣고 자발적인 등장성 수축을 기록하였다. Krebs-Ringr 용액에는 5% $Co_2$를 함유하는 $O_2$를 계속 공급하였으며, 용액의 온도가 $37^{\circ}C$를 유지하도록 하였다. 자발적 수축이 시작하고 20분이 경과한 다음 dopamine($10^{-4}M$), CCK-8($10^{-8}M$), dopamine($10^{^6}M$)등을 투여하면서 수축성을 관찰하여 다음과 같은 결과를 얻었다. 1) dopamine은 소장의 모든 부위에서 자발적 수축성을 억제하였으며, 이러한 dopamine의 작용은 회장을 제외한 다른 부위에서 tetrodotoxin에 의하여 유의하게 감소하였다. 2) domperidone은 소장의 모든 부위에서 dopamine의 억제작용에 길항적으로 작용하였으며, tetrodotoxin을 전처치하면 회장을 제외한 다른 부위에서 domperidone의 길항작용은 완전히 소실되었다. 3) CCK-8는 소장의 모든 부위에서 dopamine의 작용을 감소시켰으며, tetrodotoxin을 전처치하면 CCK-8의 작용은 유의하게 감소되었다. 이상의 결과로 미루어 보아 dopamine은 십이지장구를 포함한 모든 소장의 수축성을 억제하며, CCK-8는 dopamine의 억제 작용을 감소시키는데, 이러한 작용들은 장관내 신경계를 거쳐서 간접적으로 일어나는 것으로 생각된다.
고양이 담낭근에서 효소학적으로 분리한 평활근 세포는 cholecystokinin octapeptide (CCK-8), acetylcholine (ACh) 및 KCl에 의하여 용량에 의존하여 수축하였다. 이들 효현제 (CCK-5, ACh 및 KCl)에 의한 평활근 세포의 최대수축은 각각$10^{-9}M$, $10^{-5}M$ 및 20mM 농도에서 야기되었다. CCK-8에 의하여 야기되는 이들 평활근 세포의 수축은 HEPES 완충액에 $Ca^{2+}$을 제거시킴에 의하여 영향을 받지 아니하였으나, $Ca^{2+}$ 대신에 strontium을 첨가시켰을때 수축반응이 완전하게 억제되었다 (p<0.001). 이와는 반대로 KCl에 의한 수축반응은 strontium 치환에 의하여 영향을 받지 아니하고 HEPES 완충액에 $Ca^{2+}$을 제거시킴에 의하여 억제되었다 (p<0.01). ACh에 의하여 야기되는 수축반응은 세포 외액의 $Ca^{2+}$을 제거시킴에 의하여 중등도의 억제반응이 야기되었으나 (p<0.05) strontium에 의하여 영향을 받지 아니하였다. Saponin으로 세포 투과성 변동을 야기시킨 근세포에서 inositol 1,4,5-trisphosphate $(IP_3)$와 CCK-8은 수축반응을 일으켰고, 이러한 수축반응은 calmodulin 길항제인 CGS 9343B에 의하여 차단되었으며 (p<0.001), heparin은 CCK-8 및 $IP_3$의 작용을 완전하게 봉쇄하였다 (p<0.001). 그러나 이러한 수축반응에 있어서 protein kinase C 길항제인 H7은 아무런 작용을 나타내지 못하였다. 이러한 결과로 볼 때 CCK-8에 의하여 야기된 고양이 담낭근 세포의 수축반응은 $IP_3$에 의하여 세포내 저장소로부터 유리된 $Ca^{2+}$과 calmodulin에 의존적인 과정에 의하여 매개되어 지는 것으로 생각된다. 또한 ACh는 세포외액의 $Ca^{2+}$ 뿐만 아니라 세포내 저장소의 $Ca^{2+}$ 모두를 이용하며, KCl은 전적으로 세포외액의 $Ca^{2+}$에 의존적인 형태로 calmodulin과는 무관하게 고양이 담낭근 세포의 수축반응을 야기시키는 것으로 사료된다.
In this study, we investigated the characteristics of CCK-producing cells and mucus-secreting goblet cells with respect to stomach fish and stomachless fish of the Gobiidae in order to provide a basis for understanding the digestive physiology. Hairychin goby (Sagamia geneionema), which is stomachless fish, the numbers of mucus-secreting goblet cells is highest in the posterior intestine portion (P<0.05), while CCK-producing cells are scattered throughout the intestine. Gluttonous goby (Chasmichthys gulosus), which is stomach fish, mucus-secreting goblet cells are most abundant in the mid intestine portion (P<0.05), whereas CCK-producing cells are observed only in the anterior and mid intestine portion. Trident goby (Tridentiger obscurus) which is stomach fish, mucus-secreting goblet cells were most abundant in the mid intestine portion (P<0.05). CCK-producing cells are found in the anterior and mid intestine portion. Giurine goby, Rhinogobius giurinus which is also stomach fish, the largest number of mucus-secreting goblet cells showed in anterior intestine portion except for esophagus (P<0.05). CCK-producing cells are present only in the anterior and mid intestine portion. In S. geneionema, digestive action occurs in the posterior intestine portion to protect and functions to activate digestion. In contrast, in C. gulosus, T. obscurus and R. giurinus, their digestive action occurs in the anterior and mid intestine portion to protect and functions to activate digestion. Further studies of the modes of food ingestion by these fish, the contents of their digestive tracts, and the staining characteristics of the goblet cells need to be carried out.
Quantitative analysis of gallbladder emptying is important in diagnosis of motility disorder of gallbladder and in studies of biliary physiology. However, the normal range of gallbladder ejection fraction (GBEF) has not been determined yet and the best method for stimulating the gallbladder to contract has not been elucidated adequately. The purpose of this study was to compare the gallbladder emptying effect of the fatty meal ingestion with that of the continuous infusion of cholecystokinin (CCK) and to establish the normal GBSF values of normal subjects. Quantitative hepatobiliary scan with $^{99m}Tc-DISIDA$ after a fatty meal was performed for 22 normal healthy volunteers. Among them, 10 subjects repeated the test with a fatty meal. Again, for 7 subjects quantitative heaptobiliary scan with an infusion of CCK (sincalide) at a rate of 20 ng/kg/hr for 45 minutes was performed repeatedly. The results were as follows. 1) With a fatty meal, the mean GBEF was $89.6{\pm}8.2%$ in 22 normal subjects, and there was no difference between subjects. 2) With a continuous infusion of CCK, the mean GBEF was $62.4{\pm}16.6%$ in 7 normal subjects, and there was a significant difference between subjects(p<0.05). 3) The reproducibility of GBEF by a fatty meal was significantly higher than by an infusion of CCK (p < 0.05). 4) The mean GBEF by a fatty meal was significantly higher than that by an infusion of CCK (p < 0.05). We concluded that a fatty meal is superior to a continuous infusion of CCK for inducing gall-bladder contraction because that induces more complete emptying and the response is more reproducible and constant.
A possible potentiation between cholecystokinin (CCK) and secretin in amylase secretion from isolated rat pancreatic acini was investigated. Combined treatment of acini with secretin and CCK at low concentrations, which are known to be physiological, resulted in enzyme secretion larger than the arithmetic sum of their separate effects. Such a potentiating effect also occurred between secretin and A23187 (Ca ionophore), between forskolin (adenylate cyclase activator) and CCK, and between forskolin and A23187. Staurosporin (protein kinase C inhibitor) and W7 (calmodulin antagonist) inhibited markedly the potentiated amylase release induced by the agonists, but KT5720 (protein kinase A inhibitor) did not affect the potentiated amylase release. Therefore, we concluded that the action of CCK in a physiological concentration is potentiated by secretin in a physiological concentration range and vice versa, and that the intracellular mechanism necessary for the potentiation is associated with $Ca^{2+}$. However, it is uncertain what mechanisms are involved in potentiation of amylase release after CAMP and $Ca^{2+}$.
Aminoglycoside antibiotics are reported to enhance the amylase release from isolated slices of pancreas in vitro and the mode of action of aminoglycosides on amylase release is considered different from those of acetylcholine or cholecystokinin(CCK), i.e., electronmicroscopically intact zymogen granules are appeared in the lumen of pancreatic acini by treatment of aminoglycosides. It is known that atropine blocks the secretagogue effect of acetylcholine, and phenoxybenzamine is reported to block the effects of CCK or its analogue caerulein. Present study was undertaken to investigate the mode of action of aminoglycosides on the amylase release using atropine, phenoxybenzamine and propranolol as a membrane stabilizing agent in slices of chicken pancreas. The results are summarized as follows : 1) Streptomycin and kanamycin increased the amylase release significantly from slices of chicken pancreas. 2) The effect of streptomycin was inhibited by atropine but not by phenoxybenzamine or propranolol. 3) The amylase release by acetylcholine was blocked by atropine tut the effect of cholecystokinin octapeptide(CCK-8) was not influenced by atropine, phenoxybenzamine or propranolol. 4) Pretreatment of streptomycin enhanced the secretagogue effect of acetylcholine or CCK-8. From these results it is suggested that amylase releasing effects of aminoglycosides are mediated in part by cholinergic stimulation and in part by membrane alteration and these effects are enhanced by acetylcholine or cholecystokinin.
The isolated rabbit gall bladder strips were prepared according to the technique described by Amer and Becvar (1969). The strips were placed in a bath containing 100 ml of Locke-Ringer solution maintained at $38^{\circ}C$. Oxygen was continuously bubbled through the solution. The tension of the muscle strip was initially adjusted to 0.7g. The contractile response was measured isometrically by a force-displacement transducer connected to a polygraph. The effect of a number of autonomic drugs were studied for their interaction with caerulein (Prof. V. Erspamer, F.I. 6934 Caerulein, Farmitalia, Italia), a gastrin or CCK.PZ like peptide, on isolated rabbit gall bladder strips. In this preparation caerulein produced contractions of CCK-PZ type, but the relative potency on a weight basis was 40 times that of CCK-PZ. The response of caerulein was not modified by either cholinergic or alpha or beta adrenergic blockade. However, the response of caerulein and of barium on the strips were prevented by papaverine or aminophylline. Isoproterenol, papaverine or aminophylline alone relaxed the preparation whereas caerulein, CCK-PZ, acetylcholine, serotonin, histamine or barium chloride contracted the preparation. In summary, it is concluded that caerulein on the gall bladder strip seems to act independently of the autonomic nervous system and mediated via mechanisms apparently similar to those involved in the action of barium chloride.

In pancreatic cells, NO formation is associated with increased levels of cGMP and endocrine/exocrine secretion. In the present study, the role of NO in the regulation of exocrine secretion was investigated in rat pancreatic tissues. Treatment of rat pancreatic tissue with sholecystokinin-pancreozymin (CCK-PZ) resulted in an significant increase in arginine conversion to citruline, the amount of nitrite/nitrate, the release of amylase, and the level of cGMP. Furthermore, CCK-PZ stimulated increase of amylase release and conversion of arginine to citrulline transformation were counteracted by the inhibitor of NO synthase, $N^G-nitro-L-arginine$ methyl ester. The results on the time course of CCK-PZ-induced citrulline formation within the first seconds of simulation. The kinetics of citrulline accumulation correlate well with those of cGMP rise, which further confirms the conclusion that NO mediates the response to CCK-PZ by cGMP.
본 논문에서는 IEEE 802.11b/g 규격인 CCK/OFDM 방식의 54 Mbps 속도를 갖는 VoWLAN 단말기에서, RF 모듈을 통해 송수신되는 최종 전송선로(transmission line) 와 안테나 스위치단의 부정합으로 인해 쉽게 발생할 수 있는 입출력 사이의 I/Q 부정합 문제를 새로운 CPW 구조의 정합회로를 적용함으로써 해결할 수 있음을 보인다. I/Q 성능 개선에 대한 분석 방법으로 출력 Spectrum Mast 비선형 특성으로 출력된 CCK(complementary code keying)/OFDM(Orthogonal Frequency Division Multiplexing) 변조 데이터에 따른 EVM (Error Vector Magnitude) 특성 및 수신감도 특성을 분석하여 전체 I/Q 성능이 개선됨을 보인다.
