Optimized Production of Poly(γ-Glutamic acid) By Bacillus sp. FBL-2 through Response Surface Methodology Using Central Composite Design

  • In the present study, the optimization of poly(${\gamma}$-glutamic acid) (${\gamma}$-PGA) production by Bacillus sp. FBL-2 was studied using a statistical approach. One-factor-at-a-time method was used to investigate the effect of carbon sources and nitrogen sources on ${\gamma}$-PGA production and was utilized to select the most significant nutrients affecting the yield of ${\gamma}$-PGA. After identifying effective nutrients, response surface methodology with central composite design (CCD) was used to obtain a mathematical model to identify the optimum concentrations of the key nutrients (sucrose, $\text\tiny{L}$-glutamic acid, yeast extract, and citric acid) for improvement of ${\gamma}$-PGA production. The optimum amount of significant medium components appeared to be sucrose 51.73 g/l, $\text\tiny{L}$-glutamic acid 105.30 g/l, yeast extract 13.25 g/l, and citric acid 10.04 g/l. The optimized medium was validated experimentally, and ${\gamma}$-PGA production increased significantly from 3.59 g/l (0.33 g/l/h) to 44.04 g/l (3.67 g/l/h) when strain FBL-2 was cultivated under the optimal medium developed by the statistical approach, as compared to non-optimized medium.

Visual Tracking Technique Based on Projective Modular Active Shape Model (투영적 모듈화 능동 형태 모델에 기반한 영상 추적 기법)

  • Visual tracking technique is one of the essential things which are very important in the major fields of modern society. While contour tracking is especially necessary technique in the aspect of its fast performance with target's external contour information, it sometimes fails to track target motion because it is affected by the surrounding edges around target and weak egdes on the target boundary. To overcome these weak points, in this research it is suggested that PDMs can be obtained by generating the virtual 6-DOF motions of the mobile robot with a CCD camera and the image tracking system which is robust to the local minima around the target can be configured by constructing Active Shape Model in modular base. To show the effectiveness of the proposed method, the experiment is performed on the image stream obtained by a real mobile robot and the better performance is confirmed by comparing the experimental results with the ones of other major tracking techniques.

Antioxidant activity and MMP-1 inhibitory activity of Panax Ginseng Sprout Extracts (새싹인삼 추출물의 항산화 활성 및 MMP-1 저해 활성 )

  • Objectives : As a substitute for high-price ginseng, this study attempted to examine a possibility of the ferment extract of Panax ginseng sprout whether leaves and roots can be used together as a cosmetic ingredient with anti-oxidative and wrinkle-care effects. Methods : In terms of a test method, antioxidant activities were confirmed through total polyphenol contents, total flavonoid contents, DPPH radical scavenging activity and ABTS radical scavenging activity using the Panax ginseng sprout. In addition, to assess wrinkle-care effectiveness, the cytotoxicity of the extract was analyzed through MTT assay, and inhibition of collagenase activity in the cells was tested using the Panax ginseng sprout fermented by Saccharomyces cerevisiae. Resuits : The content of polyphenols and flavonoids in natural plants was highest in Panax Ginseng Sprout Extract at 100℃, which also demonstrated high DPPH, ABTS radical scavenging activity. MTT assay demonstrated that the Panax Ginseng Sprout Ferment Extract did not have a cytotoxic effect in CCD-986SK cell. Also, Panax Ginseng Sprout Ferment Extract was found to inhibit MMP-1 expression by 51.85±6.09% at a concentration of 10%. Conclusions : Therefore, this study has confirmed a possibility of Panax ginseng sprout ferment extract as a cosmetic ingredient with MMP-1-inhibitory effects.

Optimizing Nitrobenzene Synthesis Catalyzed by Sulfated Silica (SO4/SiO2) through Response Surface Methodological Approach

  • Today, the principles of green chemistry are being fundamentally applied in the chemical industry, such as the nitrobenzene industry, which is an essential intermediate for various commercial products. Research on the application of response surface methodology (RSM) to optimize nitrobenzene synthesis was conducted using a sulfated silica (SO4/SiO2) catalyst and batch microwave reactor. The nitrobenzene synthesis process was carried out according to RSM using a central composite design (CCD) design for three independent variables, consisting of sulfuric acid concentration on the silica (%), stirring time (min), and reaction temperature (℃), and the response variable of nitrobenzene yield (%). The results showed that a three-factorial design using the response surface method could determine the optimum conditions for obtaining nitrobenzene products in a batch microwave reactor. The optimum condition for a nitrobenzene yield of 63.38 % can be obtained at a sulfuric acid concentration on the silica of 91.20 %, stirring time of 140.45 min, and reaction temperature of 58.14 ℃. From the 20 experiments conducted, the SO4/SiO2 catalyst showed a selectivity of 100 %, which means that this solid acid catalyst can potentially work well in converting benzene to nitrobenzene.

Adaptive Vehicle License Plate Recognition System Using Projected Plane Convolution and Decision Tree Classifier (투영면 컨벌루션과 결정트리를 이용한 상태 적응적 차량번호판 인식 시스템)

  • In this paper, an adaptive license plate recognition system which detects and recognizes license plate at real-time by using projected plane convolution and Decision Tree Classifier is proposed. And it was tested in circumstances which presence of complex background. Generally, in expressway tollgate or gateway of parking lots, it is very difficult to detect and segment license plate because of size, entry angle and noisy problem of vehicles due to CCD camera and road environment. In the proposed algorithm, we suggested to extract license plate candidate region after going through image acquisition process with inputted real-time image, and then to compensate license size as well as gradient of vehicle with change of vehicle entry position. The proposed algorithm can exactly detect license plate using accumulated edge, projected convolution and chain code labeling method. And it also segments letter of license plate using adaptive binary method. And then, it recognizes license plate letter by applying hybrid pattern vector method. Experimental results show that the proposed algorithm can recognize the front and rear direction license plate at real-time in the presence of complex background environments. Accordingly license plate detection rate displayed $98.8\%$ and $96.5\%$ successive rate respectively. And also, from the segmented letters, it shows $97.3\%$ and $96\%$ successive recognition rate respectively.

Optimization of Bleaching Conditions for Stain Removal in Japanese Hackberry (Celtis sinensis Persoon) Using Response Surface Methodology (반응표면분석법을 이용한 팽나무(Celtis sinensis Persoon)의 최적 변색제거조건 결정)

  • This research was performed to investigate the effect of hydrogen peroxide on the stain removal in japanese hackberry. Response surface method (RSM) was used to optimize the bleaching conditions such as reaction temperature, reaction time and the concentration of hydrogen peroxide. Fifteen different bleaching conditions were selected according to $2^3$ factorial central composite design (CCD). The bleaching effect were evaluated by lightness differences of wood surface before and after the bleaching. The RSM model was determined and its $R^2$ values were 0.93, showing it well represented the bleaching effect. The most affecting factor on the stain removal was the concentration of hydrogen peroxide, followed by reaction time and reaction temperature. Second degree of concentration was proved to have an effect on the bleaching. Bleaching rates above 3% concentrations of hydrogen peroxide were tended to be slightly decreased, and low bleaching effect was found at $20^{\circ}C$. The determined RSM model may offer very practical ways to obtain the desired levels of bleaching because it offers multiple solutions.

  • Recently many efforts on the development of automatic processing system for delivery sequency sorting have been performed in ETRI, which requires the use of postal 4-state bar code system to encode delivery points. This paper addresses the issue on the extension of read range and the improvement of image processing method. For the improvement of image processing procedure, applied information acquisition method through basic two thresholds onto the horizontal axial line of gray image based on reference information of 4-state bar code symbology. Symbol values are computed after creating two threshold values based on the obtained information through search of horizontal axial values. The implementation result of 4-state bar code reader are obtained the symbol values within 30~60 msec (58,000~116,000 mail item/hour)without noise removal or image rotation in spite of the incline $\pm 45^{\circ}$.

Automatic Classification Algorithm for Raw Materials using Mean Shift Clustering and Stepwise Region Merging in Color (컬러 영상에서 평균 이동 클러스터링과 단계별 영역 병합을 이용한 자동 원료 분류 알고리즘)

  • In this paper, we propose a classification model by analyzing raw material images recorded using a color CCD camera to automatically classify good and defective agricultural products such as rice, coffee, and green tea, and raw materials. The current classifying agricultural products mainly depends on visual selection by skilled laborers. However, classification ability may drop owing to repeated labor for a long period of time. To resolve the problems of existing human dependant commercial products, we propose a vision based automatic raw material classification combining mean shift clustering and stepwise region merging algorithm. In this paper, the image is divided into N cluster regions by applying the mean-shift clustering algorithm to the foreground map image. Second, the representative regions among the N cluster regions are selected and stepwise region-merging method is applied to integrate similar cluster regions by comparing both color and positional proximity to neighboring regions. The merged raw material objects thereby are expressed in a 2D color distribution of RG, GB, and BR. Third, a threshold is used to detect good and defective products based on color distribution ellipse for merged material objects. From the results of carrying out an experiment with diverse raw material images using the proposed method, less artificial manipulation by the user is required compared to existing clustering and commercial methods, and classification accuracy on raw materials is improved.

  • As information-communication network has been developed rapidly, internet users' circumstances also have been changed for the better, in result, more information can be applied than before. At this moment, there are many differences between real color and reappeared color on the CRT. When we observe a material object, our eyes perceive the multiplied form of light sources and nature spectral reflection. However, when the photographed signal is reappeared, illumination at that time of photographing and spectral reflection of a material object are converted into signal, and this converted RGB signal is observed on the CRT under another illumination. At this time, RGB signal is the reflected result of illumination at that time of photographing Therefore, this signal is influenced by the illumination at present, so it can be perceived another color. To accord the colro reflections of another color source, the study has been reported by S.C.Ahn$^{[1]}$, which study is about the color reapperarance system using neuron network. Furthermore, color reappearing method become independent of its circumstances has been reported by Y.Miyake$^{[2]}$. This method can make the same illuminations even if the observe circumstances are changed. To assume the light sources of observe circumstances, the study about color reappearing system using CCD sensor also have been studied by S.C.Ahn$^{[3]}$. In these studies, a population is fixed, first, on ab coordinates of CIE L${\ast}$a${\ast}$b${\ast}$. Then, color reappearing can be possible using every population and existing digital camera. However, the color is changed curvedly, not straightly, according to value's changes on the ab coordinates of CIE L${\ast}$a${\ast}$b. To solve these problems in this study, first of all, Labeling techniques are introduced. Next, basis color-it is based on Munsell color system-is divided into 10 color fields. And then, 4 special color- skin color, grass color, sky color, and gray-are added to the basis color. Finally, 14 color fields are fixed. After analyzing of the principle elements of new-defined-color fields' population, utility value and propriety value are going to be examined in 3-Band system from now on.

RSM-based Practical Optimum Design of TMD for Control of Structural Response Considering Weighted Multiple Objectives (가중 다목적성을 고려한 구조물 응답 제어용 TMD의 RSM 기반 실용적 최적 설계)

  • In spite of bulk literature about the tuning of TMD, the effectiveness of TMD in reducing the seismic response of engineering structures is still in a row. This paper deals with the optimum tuning parameters of a passive TMD and simulated on MATLAB with a ten-story numerical shear building. A weighted multi-objective optimization method based on computer experiment consisting of coupled with central composite design(CCD) central composite design and response surface methodology(RSM) was applied to find out the optimum tuning parameters of TMD. After the optimization, the so-conceived TMD turns out to be optimal with respect to the specific seismic event, hence allowing for an optimum reduction in seismic response. The method was employed on above structure by assuming first the El Centro seismic input as a sort of benchmark excitation, and then additional recent strong-motion earthquakes. It is found that the RSM based weighted multi-objective optimized damper improves frequency responses and root mean square displacements of the structure without TMD by 31.6% and 82.3% under El Centro earthquake, respectively, and has an equal or higher performance than the conventionally designed dampers with respect to frequency responses and root mean square displacements and when applied to earthquakes.
