Redox Chemistry and Valence Tautomerism of Cobalt-Quinone Complexes in Nonaqueous Solvents

  • The electrochemistry and valence tautomerism of CoⅢ(N-N)(SQ)(Cat), [N-N=N,N,N',N'-tetramethylethylenediamine (TMEDA); SQ=3,5- or 3,6-di-tert-butyl-semiquinone, Cat=3,5- or 3,6-di-tert-butyl-catechol], have been investigated by spectroscopic, electrochemical, spectroelectrochemical methods in nonaqueous solvents under anaerobic condition. The transition temperature between tautomers is dependent upon the donation effect of substituted quinone ligand and solvent. It increases with the increase of donation effect of solvent and quinone ligand. CoⅢ(TMEDA)(SQ)(Cat) is reduced to [CoⅡ(TMEDA)(SQ)(Cat)]-, and then reduces to [CoⅡ(TMEDA)(Cat)2]2-. CoⅢ(TMEDA)(SQ)(Cat) is oxidized to [CoⅢ(TMEDA)(SQ)2]+, but the stability of the oxidized form in DMF is dependent upon the solution temperature. With the increase of solution temperatrue the oxidized form may be converted to [CoⅡ(TMEDA)(SQ)(BQ)]+ by intramolecular electron transfer from SQ ligand to CoⅢ.

  • In order to gain insight into the mechanism of the regulation of cytochrome P450IAl by arylhydrocarbon, the 5'-flanking region of a trout CYP450IAl 5'flanking DNA was cloned into pCAT-basic vector and it was transfected into Hepa-1 cells. 3MC treatment to hepa Ⅰ cells transfected with fish CYP450IAl-CAT construct results in mRNA increased by 2.81 fold when it was compared with that of control This increase of mRNA was decreased by concomitantly treated flavonoids such as morin. The levels of CAT mRNA that was treated with morin was 29.2-58.0% of 3MC stimulated CAT mRNA. Further investigation to find out if there are DRE, XRE or negative regulatory cis element in CYP450IA1 gene was undertaken. Results of the deletion study of 5'flanking DNA of trout P450IA indicate the existance of the negative(-1600 ~ -1300). CAT mRNA was about two-fold higher in deleted trout CYP450IAl-CAT construct transfected cells compared to the wi Id type trout CYP450IAl-CAT construct transfected cells. Effects of Resistance Training on Serum Inflammatory Markers and CatSper 1-4 Protein Expression in Testis of OLETF Rats (저항성 운동이 OLETF 쥐의 혈청 염증반응지표 및 고환조직의 CatSper 1-4 단백질 발현에 미치는 영향)

  • PURPOSE: This study was to investigate the effects of resistance exercise on serum inflammatory markers and CatSper 1-4 expression in testis of OLETF rats. METHODS: Male OLETF rats were divided into two groups; control group (n=12), resistance exercise group (n=12). The exercise group performed a total of 8 weeks of moderate intensity resistance exercise on a 1.35 m vertical ladder with weights secured to their tails. RESULTS: The results of this study was following; The exercise group showed a significant decreased in the inflammatory ($IL-1{\beta}$, IL-6, $TNF-{\alpha}$) levels as compared to the control group. But CRP was no significant difference between control and exercise groups. Also, CatSper 1 and 2 were significantly increased in the exercise group compared to the control group, whereas CatSper 3, 4 were no significant difference between both groups. CONCLUSIONS: This study suggests that resistance exercise training can contribute to reduce pro-inflammatory responses in whole body and it affects male reproductive function by improve sperm quality and CatSper protein expression.

ROLE OF 5'FLANKING DNA FOR THE REGULATION OF CYP450IA1 GENE EXPRESSION

  • To investigate the mechanism of the regulation of cytochrome P450IAl, the 5'-flanking region of a trout cytochrome P4501Al was cloned into the CAT basic expression vector at HindⅢ site. This trout Cytochrome P450IAl upstream DNA containing CAT construct was transfected into Hepa-1 cells .3MC treatment to hepa I cells transfected with trout P450IAl-CAT construct increased CAT protein and mRNA by 2.81 fold when it was compared with that of control. This increase CAT protein and mRNA was decreased by concomitantly treated flavonoids and aminopyrine. The level of CAT protein was 29.2-58.0% of 3MC stimulated CAT protein.

  • As the major methanol fueled vehicle exhaust components, formaldehyde & methanol conversion over the existing commercial 3-way catalyst was examined in a labolatory tains different Ag loadings on commercial 3-way catalyst, and german commercial catalysts for methanol engine exhaust manufactured by a commercial manufacturer. Silver catalysts were prepared by the wet impregnation of silver nitrate solution on commercial 3-way catalyst. These catalysts were characterized with BET Surface area and pore size distribution. In general, the formaldehyde(HCHO) conversion of the tested catalysts was similar to that of methanol$(CH_3OH)$. At 100$^\circ$C, which is equivalent to the cold start condition, 5wt% Ag cat. showed the most excellent HCHO and $CH_3OH$ conversion. The order of activity for conversion of HCHO & $CH_3OH$ to carbon dioxide and water vapor was as follows ; 5wt% Ag/3-way cat.>2wt% Ag/3-way cat.>german cat. front(1) > german cat. rear(2) > 10wt% Ag/3-way cat.> commercial 3-wat catalyst. However there was no significant activity difference between those tested catalysts in the hot run condition of 400$^\circ$C. Therefore, it could be concluded that the Ag-modified 3-way catalyst was the most effective and practical catalyst system which could be capable of removal the HCHO and methanol at the special condition of low temperature such as cold start condition.

Development of RTOS Based LinuxCNC 3-axis Control System with EhterCAT Communication (RTOS기반 LinuxCNC에서 EtherCAT 통신이 적용된 3축 CNC 제어 시스템 개발)

  • In this paper, we proposed a PC-based CNC control system using EtherCAT-based servo drive and I/O device. The default communication of LinuxCNC is a parallel port, and data processing with high bandwidth is impossible. However, it is possible to apply various bandwidth devices through the application of EtherCAT, one of the industrial Ethernet communications with high bandwidth. Therefore, the hardware control method of LinuxCNC was applied through EtherCAT communication from the existing parallel port. Finally, through HAL configuration, I/O device operation check and 3-axis motion control proved the LinuxCNC system with EtherCAT.

Expression and Activity of Catalases Is Differentially Affected by GpaA (Ga) and FlbA (Regulator of G Protein Signaling) in Aspergillus fumigatus

  • Vegetative growth signaling of the opportunistic human pathogenic fungus Aspergillus fumigatus is mediated by GpaA ($G{\alpha}$). FlbA is a regulator of G protein signaling, which attenuates GpaA-mediated growth signaling in this fungus. The flbA deletion (${\Delta}flbA$) and the constitutively active GpaA ($GpaA^{Q204L}$) mutants exhibit enhanced proliferation, precocious autolysis, and reduced asexual sporulation. In this study, we demonstrate that both mutants also show enhanced tolerance against $H_2O_2$ and their radial growth was approximately 1.6 fold higher than that of wild type (WT) in medium with 10 mM $H_2O_2$. We performed quantitative PCR (qRT-PCR) for examination of mRNA levels of three catalase encoding genes (catA, cat1, and cat2) in WT and the two mutants. According to the results, while levels of spore-specific catA mRNA were comparable among the three strains, cat1 and cat2 mRNA levels were significantly higher in the two mutants than in WT. In particular, the ${\Delta}flbA$ mutant showed significantly enhanced and prolonged expression of cat1 and precocious expression of cat2. In accordance with this result, activity of the Cat1 protein in the ${\Delta}flbA$ mutant was higher than that of $gpaA^{Q204L}$ and WT strains. For activity of the Cat2 protein, both mutants began to show enhanced activity at 48 and 72 hr of growth compared to WT. These results lead to the conclusion that GpaA activates expression and activity of cat1 and cat2, whereas FlbA plays an antagonistic role in control of catalases, leading to balanced responses to neutralizing the toxicity of reactive oxygen species.

Survey of Perception of Complementary and Alternative Therapies (CAT) (암환자를 돌보는 일부 종합병원 간호사의 보완대체요법에 대한 인식도)

  • Purpose: This study was done to investigate oncology nurses' perception, attitude, knowledge and practice of CAT and to describe nurses' needs for education on CAT Method: A cross-sectional descriptive study was conducted. A total of 270 oncology nurses working at four medical centers in Seoul and Kyungki participated in this study. They completed the questionnaire between December 2005 and February 2006. Results: Oncology nurses perceived CAT as possible nursing interventions, but felt that there was no conclusive evidence on effectiveness/safety. Of the participants 44.4% have used CAT for themselves, and 73.9% of them were satisfied after using CAT. Two biggest reasons for CAT use were symptom control and emotional support. However, 80.8% of the participants had not had any training related to CAT Both nurses who have used CAT and those who have been trained to practice CAT represented a more positive perception toward CAT (p=.000). The biggest perceived obstacles to the practice of CAT were concerns about side effects (71.2%) and lack of knowledge and skill (69.6%). The most favorable CAT modalities in both practice and training were mind-body medicine. Conclusion: Overall oncology nurses' perception toward CAT was encouraging. However a high number of the nurses reported lack of knowledge and skill in CAT Education and training strategy for CAT need to be developed.

Nucleotide Sequence and Inducibility Analysis of Chloramphenicol Acetyltransferase Gene from Staphylococcus aureus R-plasmid pSBK203 (Staphylococcus aureus에서 분리된 R-plasmid pSBK203상의 chloramphenicol acetyltransferase 인자의 염기서열 및 유발성 분석)

  • The nucleotide sequence of inducible chloramphenicol acetyl-transferase(CAT) gene isolated from a small plasmid pSBK203 of Staphylococcus aureus was determined. The base sequence shows that structural gene of pSBK203-CAT encodes a protein of 213 amino acids and has a leader region which encodes a short polypeptide of 9 amino-acids in its upstream. vertical bar /sup 35/S vertical bar-Methionine labelled CAT gene product in minicell showed almost same mobility with pC194-CAT of which molecular weight is 24Kdal on polyacrylamide gel electrophoresis. Predicted amino acid sequence of pSBK203-CAT has revealed a high degree of homology with the CATs of pC194 and pC221 than those of cat-86, Tn9 and proteus mirabilis PM13.

Application of Complementary Alternative Therapies(CAT) and Influence of Health Locus of Control on CAT in Patients with Chronic Liver Disease (만성 간 환자의 보완대체요법 시행 실태와 건강 통제위의 영향)

  • Purpose: This study was done to investigate the application of complementary and alternative therapies (CAT) in patients with chronic liver disease. and to analyze the Influence of health locus of control (HLOC) on CAT. Method: Questionnaires on CAT and a multidimensional health locus of control (MHLOC) scale developed by Wallston and Wallston were completed by 141 patients. Result: Analysis showed that 44.7% of the patients had experience with CAT and 49.2% of the patients reported being willing to use CAT. Reliability scores for the scale were HLOC-I 0.7376, HLOC-P 0.6383, HLOC-C 0.7351. The mean scores for the HLOC were HLOC-I 24.86, HLOC-P 22.86, HLOC-C 16.00. There were no significant differences in mean scores for HLOC between the patients who had used CAT and those who had not. Eight response Patterns for HLOC were identified. Among them, 'yea sayer' was the largest group (27.0%). A significant difference was found in the 'yea sayer' pattern between the group who had used CAT and the group who had not. Conclusion: CAT relates to various cognitive factors such as HLOC. Further study is needed to examine the influence of HLOC on CAT and its response patterns.
