Functional Characterization of cAMP-Regulated Gene, CAR1, in Cryptococcus neoformans

  • The cyclic AMP (cAMP) pathway plays a major role in growth, sexual differentiation, and virulence factor synthesis of pathogenic fungi. In Cryptococcus neoformans, perturbation of the cAMP pathway, such as a deletion in the gene encoding adenylyl cyclase (CAC1), causes defects in the production of virulence factors, including capsule and melanin production, as well as mating. Previously, we performed a comparative transcriptome analysis of the Ras- and cAMP- pathway mutants, which revealed 163 potential cAMP-regulated genes (38 genes at a 2-fold cutoff). The present study characterized the role of one of the cAMP pathway-dependent genes (serotype A identification number CNAG_ 06576.2). The expression patterns were confirmed by Northern blot analysis and the gene was designated cAMP-regulated gene 1 (CAR1). Interestingly, deletion of CAR1 did not affect biosynthesis of any virulence factors and the mating process, unlike the cAMP-signaling deficient cac1$\Delta$ mutant. Furthermore, the car1$\Delta$ mutant exhibited wild-type levels of the stress-response phenotype against diverse environmental cues, indicating that Car1, albeit regulated by the cAMP-pathway, is not essential to confer a cAMP-dependent phenotype in C. neoformans.

Analysis and Small Scale Model Expriment on the Vertical Vibration of the KT-23 Type Passenger Vehicle (KT-23형 여객 차량의 상하 진동 해석 및 축소모형 실험)

  • The purpose of this study is to obtain the effects of the parameters of the suspension system in railway rolling-stock for KT-23 type Passenger vehicle. According to the analysis and the small scale model car test. optimal condition was obtained for the stiffness ratio of secondary spring to primary spring of the suspension system and the mass ratio of the bogie frame to the car body. The analysis of the study shows that if the car body mass is increased or secondary stiffness Is lowered, the vertical vibration level is reduced and the passenger comfort can be improved. Especially, strong peaks are occurred in the frequencies corresponding to the rotational speed of driving axle and vehicle wheel. Hence, in order to obtain the dynamic characteristics through the small scale model car, the driving method of the vehicle on the test bench, rotational characteristics of the wheel and the natural modes of vehicle should be investigated and be modified.

  • Through the multibody dynamic simulation, the analysis model of the modalohr freight car of the DMT freight car was developed. By using the developed analysis model, the running dynamic characteristics was inquired through the dynamic analysis about the modalohr freight car. As the running speed and the primary suspension were increased, the lateral and vertical vibration accelerations of the car-body and the bogie were also increased. In case of the lateral vibration acceleration of the car-body, however, review should be considered since it can be influenced by the nonlinear characteristic of the primary suspension. The lateral and vertical vibration of the car-body were generated at the frequency of $2{\sim}3\;Hz$ and $7{\sim}8\;Hz$. And the lateral and vertical vibration of the bogie were generated at the frequency of $25{\sim}35\;Hz$ at the low speed section, $40{\sim}50\;Hz$ at the high speed section.

  • In this paper, the crush characteristic of the tilting train express M-car was estimated under a head-on collision scenario. The car body was divided into three parts - front, middle, and rear. For each part, crush-force relation was evaluated numerically using LS-DYNA 3-diementional shell element analysis. This result will be used for one-dimensional collision analysis of the full train rake.

A Study on the Assessment Factors for Car Audio Sound Quaility (자동차용 오디오의 실내음질 평가인자에 관한 연구)

  • It is getting higher the demand on a good sound in a car. Like some other sensory qualities, achieving a good sound is still treated as a black-box followed by the individual taste of tuning engineers. This study aims to define the attributes of a good car audio sound and the assessment factors affecting the sound quality. With a variety of surveys on professional listening groups and car audio installers, 26 most frequently used vocabularies are selected as typical presentations of car audio sound quality. After repetitions of listening tests with deliberately selected test CD, pre-recorded 15 different digital car-recordings, headphones replay system, and three different listening groups, it is proved that there are at least 5 different assessment factors which significantly affect the car audio sound quality. They are high fidelity, spectral balance, tonal accuracy, sound stage and acoustic fault.

Characterization of Carnation mottle camovirus(CarMV) Isolated from Lilium spp. in Korea

  • Carnation mottle carmovirus(CarMV) was isolated from Lilium spp. in Korea. This isolate, CarMV, was done bioassay, which plants were Dianthus caryophyllus, Gomphrena globosa, Chenopodium amaranticolor, Dianths chinensis. CarMV was propagated on the leaves of Chenopodium amaranticolor with the crude-sap inoculation method and purified by Mossops method(1976). We produced antiserum against CarMV and analyzed the antiserum specificity with ELISA, Gel diffusion method, and Rapid Immunofilter Paper Assay (RIPA). From these results of the assay, RIPA method was simple and rapid for CarMV detection. We have established successfully the CarMV detection system. CarMV coat protein gene was amplified by RT-PCR with specific primers and sequencing analysis was done.

Impact Analysis for Vehicle Accident Reconstruction (자동차 사고 재고성을 위한 충돌 해석)

  • We have developed a planar impact model with a capability of reverse calculation to reconstruct various types of automobile collisions. This topic is the main part of what is referred to as accident reconstruction. The model uses the principle of impulse and momentum, and introduces a restitution coefficient and an impulse ratio at the impact center. Based on the car-to-car collision test results, we present how to estimate the restitution coefficient and the impulse ratio from some impact conditions. To validate the model and improve its reliability in accident analysis, the collision analysis has been performer with the estimated parameters. The analysis and experimental results agree well in the kinetic energy loss and the post-impact velocity.

Interior Noise Reduction of a Passenger Car using Panel Contribution Analysis (패널 기여도 분석에 의한 승용차의 실내 소음 저감)

  • The panel contribution analysis to reduce interior booming noise of a passenger car is carried out using both experimental method and numerical one. The accelerations of panels are measured on the outer surface of car body during operation. The acoustic characteristic of cavity is represented by two different ways. One is the acoustic transfer function obtained by experiment with reciprocal manner. The other is the boundary element model and numerical results of the model are calculated using SYSNOISE. The results from numerical method show more good agreement with measured sound pressure levels than the experimental one. Contributions of panels for interior noise are ranked and structure of the car is reinforced according to the results, which shows that the panel contribution analysis is a powerful tool to lessen structure-borne noise of passenger vehicle.

  • The purpose of this study is to analyse the main factors and attributes of psychological evaluation to noise. For this purpose, a psycho-acustic experiment was conducted by using the method of rating scale. 52 subjects were participated in the experiment, 20 stimuli were presented to subjects in random order with 23 adjectives. As a result of factor analysis, it was found that the primary factors for evaluating the quality of noise subjectively are four factors of unpleasantness, intensity, irregularity, and sharpness. And, as a result of MDPREF(multi-dimensional analysis of preference data), it was found that the noise sources including the factors of unpleasantness, intensity, and irregularity are related to the sounds of hammering in construction field, car horn, road traffic, idling of car, and printing by computer printer, and the factor of sharpness are the sounds of car siren, claps of thunder, car horn, and snowstorm etc.

  • Modelling and crashworthiness analysis of simplified vehicle structures with beam element and nonlinear spring element to which axial and bending collapse mecha- nisms are applied are carried out. And on the basis of these analyses, two types of full car modelling and crahworthiness analyses with nonlinear spring and beam element are accomplished. The one is the full car model of which 30% of the structures are modelled with nonlinear spring and beam element, and the other 75% of whole structures. And the results are compared with those of full car analysis with shell element.
