Influence of UV Irradiation Interval on Mineral Content in Metatarsus of Broiler Chicks (자외선 조사간격이 브로일러 병아리의 중족골 광물질 함량에 미치는 영향)

  • A study was conducted to investigate the concentrations of Ca, P and ash in metatarsal bone of broiler chicks exposed to UV light in different Interval. Day-old Hubbard broiler chicks (199=10 control+3 irradiation interval $\times$ 9 elapsed time $\times$ 7 replicate) were fed vitamin D3 deficient diet for 3 wk in a windowless subdued-light room and exposed to 297 nm UVB light by 0.068 mJ/$\textrm{cm}^2$ three times In 0, 12 or 24 h interval. The metatarsal bones were taken at 0, 6, 12, 18, 24, 48, 96, 144 or 240 h after last irradiation, separated from adhering tissue, ether extracted, dried and ashed. The Ca concentration was measured by atomic absorption spectrophotometry and P by ammonium metavanadate colorimetry. When the birds were continuously exposed to UVB light for 30 min without interval, the Ca content in metatarsus increased gradually according to the time after irradiation and reached the highest value 16.75% at 240 h after exposure. The P content also increased gradually until 144 h, where it was 9.75%. The ash content in metatarsus increased continuously until 240 h, the final time in this research, where 42.75% was shown. As 10 min three times irradiation in 12 h interval was applied to the chicks, the metatarsal Ca presented a small peak(13.31%) at 12 h after irradiation and a large peak(16.91%) at 144 h. P content showed a small peak(7.18%) at 12 h and a large level(8.34%) at 240 h. Ash content increased continuously until 240 h, where it was 46.53%. The small peaks in Ca and P concentration were thought to be derived from preirradiation at 12 and 24 h before final irradiation for 10 min. When 24 h interval system was treated, the peak value of Ca content(24.18%) occurred earlier(96 h) than those in 0 and 12 h interval systems. P content also showed the maximum value at 96 h(7.29%). Ash content presented an increasing trend until 240 h, where 45.75% was appeared. In respecting the method of UVB irradiation, the peak value of Ca content in metatarsus appeared earlier in 24 h interval system than in other systems. Meanwhile the ash contents in metatarsus of birds exposed to UVB light in 12 and 24 h interval procedures were higher than those in 0 h interval one. Therefore, it was concluded that a daily 10 min irradiation of UVB light would be desirable for increasing the Ca and ash content in metatarsus of brolier chicks.

  • In this paper, CSMA/CA protocol is modified for aeronautical VHF communication through collision alleviation and the change of retransmission slot selection method in flag used transmission. When collision occurred in the contention period of CSMA/CA process, it is generally waiting for retransmission delay and it has the double size of CW(Contention Window). To solve this problem, this paper modifies the change procedure of original contention window size and reduces the state transition in collision among the whole of it. Also, in this paper we reduces the second collision probability through the station has to enter Backoff mode with increased contention window in the first collision. In the result of simulation, it is verified that it has good property in throughput and delay, So the proposed protocol is suitable for aeronautical VHF communication.

  • Anoctamin 1 (ANO1) is an anion channel that is activated by changes in cytosolic $Ca^{2+}$ concentration and noxious heat. Although the critical roles of ANO1 have been elucidated in various cell types, the control of its gating mechanisms by $Ca^{2+}$ and heat remain more elusive. To investigate critical amino acid residues for modulation of $Ca^{2+}$ and heat sensing, we constructed a randomized mutant library for ANO1. Among 695 random mutants, reduced $Ca^{2+}$ sensitivity was observed in two mutants (mutant 84 and 87). Consequently, the E143A mutant showed reduced sensitivity to $Ca^{2+}$ but not to high temperatures, whereas the E705V mutant exhibited reduced sensitivity to both $Ca^{2+}$ and noxious heat. These results suggest that the glutamic acids (E) at 143 and 705 residues in ANO1 are critical for modulation of $Ca^{2+}$ and/or heat responses. Furthermore, these findings help to provide a better understanding of the $Ca^{2+}$-mediated activation and heat-sensing mechanism of ANO1.

  • This study is for know chemistry of the representative soils from Keumsan, its relationships with dissolved compositions, and its absorption relationships within the flora. It has been done for two comparisons: 1) phyllite, shale and granite(PSG), and 2) granite and coal bearing shale(GC). Among the soil leachate of the PSG, cations are mainly low in the phyllite area while anions are mainly low in the granite area. In the soil compositions, Ca and Mg of the beans are high and Na is high in the granite area while Ca and Na of the perilla are high in the granite area and Mg is high in the phyllite area. In th both species, Na for the granite area and Mg for the phyllite area are high while Ca for the shale area is low. Among the flora, shale area shows low Ca and Mg contents while granite area shows slightly high Na contents, regardless the species. Compared with beans, perilla is low in the Mg and high in the Na contents. These relationships show that the contents of the soil leachate do not reflect absorption within the flora. Among the soil leachate of the GC, shale area is high in the most of the elements. Especially, SO$_4$ is over 15 times high In the phyllite area. In the soil, granite area shows high Ca, Na and low Mg contents. In the flora, the Miscanthus sinensis shows high Ca and Mg contents in the granite area while the Artemisia vulgaris shows high Ca, Ca and Mg contents in the shale area. These relationships for contents of the flora and soil leachate suggest that the flora has a different absorption according different species.

  • Calcium alginate fiber were prepared by wet spinning of various conditions, including different concentrations of sodium alginate solution and $CaCl_2$ concentrations for coagulating the fiber through an absorption of calcium ion. The absorption of calcium ion during the coagulating step lead to solidify the fibers by the replacement of sodium ion with calcium ion to produce some crosslinking. The concentration of calcium ion in the calcium alginate fiber seems to be well related to the mechanical and physical property of the fiber, such as fiber strength moisture regain, and degree of swelling. The tensile strength of calcium alginate fiber was increased along with the increasing amount of sodium alginate solution. According to EDS analysis, 7 wt% $CaCl_2$ coagulation bath resulted in more calcium ion in the fiber compared to 3 wt% $CaCl_2$ coagulation bath. The decomposition temperature of calcium alginate fiber was $199^{\circ}C$, which $14^{\circ}C$ higher than that of sodium alginate.

Xenografted Tumorigenesis in the oral vestibule of nude mice by Snail transfection: Histological and immunohistochemical study

  • Purpose: The purpose of this study is to investigate the epithelial-mesenchymal transition (EMT) induced by Snail transcription factor and Snail-transfected in vivo tumors with histopathological features. Materials and methods: We induced in vivo xenografted tumorigenesis in the oral vestibules of nude mice by a Snail transfected HaCaT cell line and investigated morphological and immunohistochemical features in Snail expressive tumors. Results: We identified tumor masses in 14 out of 15 nude mice in the HaCaT-Snail cell inoculation group, but no tumors were present in any of the HaCaT cell inoculation group. Induced tumors showed features of poorly differentiated carcinoma with invasion to neighboring muscles and bones. The HaCaT-Snail tumors showed decreased expressions of E-cadherin and cytokeratin, but showed increased expressions of vimentin and N-cadherin. Discussion: The Snail transfected xenograft can improve productivity of malignant tumors, show various histopathological features including invasive growth, and aid in the investigation of tumor progression and the interaction with surrounding tissues.

Effect of Magnesium Ion on the Contractility of the Isolated Rat Uterine Smooth Muscle (마그네슘 이온이 적출한 흰쥐 자궁근의 수축성에 미치는 효과)

  • The effects of $Mg^{++}$ upon the spontaneous contraction activated by 1 IU/l oxytocin were studied in the isolated rat uterine muscle. Longitudinal muscle strips u·ere prepared from the rat uteri at the estrous stage. All experiments were performed in tris-buffered Tyrode solution which was aerated with 100% $O_2$ and kept at $35^{\circ}C$. The results obtained were as follows: 1) In the uterine strips contracting spontaneously, as $Mg^{++}$ concentration increased in the Tyrode solution the amplitude of peak tension decreased in all the experimental solutions containing the various concentrations of $Ca^{++}\;(0.5{\sim}4 mM)$. And the amplitude of peak tension increased in inverse proportion to the $[Mg^{++}]/[Ca^{++}]\; ratio$. It is suggested that the tension-lowering effect of $Mg^{++}$ would be developed through decreasing intracellular ionized free calcium ion concentration by uncertain mechanism. 2) The frequency of the uterine contraction activated by oxytocin increased as the $[Mg^{++}]/[Ca^{++}]\;ratio$ ratio increased up to 1/2, but the frequency decreased above this ratio. It is speculated that $Mg^{++}$ would influence the excitability control action of $Ca^{++}$.

A Novel Anticoagulant Protein from Scapharca broughtonii

  • An anticoagulant protein was purified from the edible portion of a blood ark shell, Scapharca broughtonii, by ammonium sulfate precipitation and column chromatography on DEAE-Sephadex A-50, Sephadex G-75, DEAE-Sephacel, and Biogel P-l00. In vitro assays with human plasma, the anticoagulant from 'S. broughtonii, prolonged the activated partial thromboplastin time (APTT) and inhibited the factor LX in the intrinsic pathway of the blood coagulation cascade. But, the fibrin plate assay did not show that the anticoagulant is a fibrinolytic protease. The molecular mass of the purified S. broughtonii anticoagulant was measured to be about 26.0kDa by gel filtration on a Sephadex G-75 column and SDS-PAGE under denaturing conditions. The optimum activity in the APTT assay was exhibited at pH 7.0-7.5 and $40-45^{\circ}C$ in the presence of $Ca^{2+}$.

Studies on the Oocytes Activation Regimed for Nuclear Transfer and Co-culture of Nuclear Transferred Embryos

  • This study was carried out to investigate the optimal activation condition for parthenogenetic development. In order to activate oocytes at 24 hrs post onset of maturation, the oocytes were cultured 3 - 13 μM Ca for 5 min., 5-8 ㎍/㎖ cytoclacin(CH) for 6 hrs, 0.5-2.0 mM 6-dimethylaminopurine(DMAP) for 3 hrs alone or combination. The activated oocytes were cultured in TCM-199 media at 5% CO₂, 95% N₂, 38℃. 1. The cleavage rate after 48 hrs culture of oocytes treated with 3-13 μM Ca for 5 min. were 9.6%-20.0% and 3.8-7.3%, respectively. When oocyte were treated with 10 μM Ca, the blastocyst formation rate was significantly higher than other group. 2. The cleavage rate after 48 hrs culture of oocytes treated with 5-8 ㎍/㎖ cytoclacin(CH) for 6 hrs, were 9.4%-21.8% and 0.0-7.3%, respectively. When oocyte were treated with 10㎍/㎖ CH, the blastocyst formation rate was significantly higher than other group. 3. The cleavage rate after 48 hrs culture of oocytes treated with 0.5-2.0 mM 6-dimethylaminopurine(DMAP) for 3 hrs were 9.1%-21.8% and 0.0-7.3%, respectively. When oocyte were treated with 2.0mM DMAP, the blastocyst formation rate was significantly higher than other group. 4. The cleavage rate after 48 hrs culture of oocytes treated with Ca+CH, Ca+DMAP, CH+DMAP were 75.9%-93.5% and 9.7 -13.3%, respectively. When oocytes were treated with Ca followed by DMAP, the blastocyst formation rate was significantly higher than other group(p〈0.05). 5. When necleus transferred embryos co-cultured with BSA, EGF and CS, the developmental rate to blastocyst were higher than control group.

The cleavage rate after 48 hrs culture of oocytes treated with $Ca^{2+}$ + cytoclacin, $Ca^{2+}$ + DMAP, cytoclacin + DMAP were 75.9%~93.5% and 9.7%~19.0%, respectively. When oocytes were treated with $Ca^{2+}$ followed by DMAP, the blastocyst formation rate was significantly higher than other group(p<0.05). When necleus transferred embryos co-cultured with bovine serum albumin(BSA), epithemal growth factor(EGF) and calf serum(CS), the developmental rate to blastocyst were higher than control group.
