Calcium Uptake in Mouse Oocyte Matured in Vitro (배양액 내에서 성숙한 생쥐 난자의 $Ca^{2+}$ Uptake)

  • 배양중인 생쥐 난자의 성숙에 미치는 배양액 내 calcim의 영향을 알아보기 위하여 1.71mM $Ca^{2+}$을 처리한 배양액과 $Ca^{2+}$이 존재하지 않는 배양액에서 난자를 배양하여 불꽃 원자 흡수 분광 광도계를 이용하여 배양된 난자의 $Ca^{2+}$농도를 측정하였다. 1) $Ca^{2+}$처리한 배양액에서 배양된 cumulus-cell이 제거된 (denuded oocyte)난자들은 시간이 지남에 따라 $Ca^{2+}$농도가 높게 나타났고, 2) $Ca^{2+}$처리하지 않은 배양액에서는 denuded난자는 3시간째 배양될때 부터 $Ca^{2+}$양이 줄어 들었다. Cumulus-enclosed난자는 $Ca^{2+}$존재하에서는 GVBD가 일어났다고 생각되는 4시간까지 계속 증가를 보인 반면 $Ca^{2+}-free$에서는 배양되지 않은 난자와 거의 차이가 없게 나타났다. 3) 핵막붕괴가 일어난 후부터 15시간까지 배양시컸을 때에는 CEO와 denuded oocyte에서 공히 $Ca^{2+}$의 농도는 다시 증가된 상태로 계속 되었다. 이런 결과로 미루어 보아 난자 성숙시 부터 성숙과정이 끝날때가지 exteral $Ca^{2+}$이 요구되고 있음을 증명해 주고있다. 그러나 이러한 세포질내의 $Ca^{2+}$및 bound calcium이 난자 성숙시부터 어떤 역활을 하고 있는 기작에 대해서는 좀 더 연구가 있어야겠다.

Effects of Aminoethoxyvinylglycine Application and Heat Treatment on Fruit Quality of 'Fuji' Apples during CA Storage (Aminoethoxyvinylglycine 및 열처리가 '후지' 사과의 CA 저장 동안 과실품질에 미치는 영향)

  • This study was carried out to investigate the effects of aminoethoxyvinylglycine (AVG) application and heat treatment on fruit quality of 'Fuji' apples during CA storage. AVG ($75mg{\cdot}L^{-1}$) was applied to 'Fuji' tree at 150 and 175 days after full bloom (DAFB) and the fruit were harvested at 185, 195, and 205 DAFB, respectively. Respiration rate and ethylene production of fruit applied with AVG were greatly reduced more than those in control. Flesh firmness and acidity of fruit applied with AVG were higher than those in control. Flesh browning occurred in all of the control fruit regardless of harvest date. However, AVG-treated fruits were free of flesh browning except for late harvested fruits. At 185 DAFB, 'Fuji' apples were harvested and prestorage heat treatment was done for 3 days at $38^{\circ}C$ and 6 hours at $46^{\circ}C$. Heat treatment at $38^{\circ}C$ and $46^{\circ}C$ prior to CA storage greatly reduced respiration rate and ethylene production. The incidence of flesh browning were 35%, 14%, and 5%, in control fruit, heating at $38^{\circ}C$ and $46^{\circ}C$, respectively. The titratable acidity was lower at heated apple than at controlled one.

Combined Foliar Spray of Boron, Calcium, and Silicon can Influence Quality and Shelf Life of Cherry Tomato in Modified Atmosphere Packaging (붕소, 칼슘, 규소의 복합 엽면시비가 방울토마토의 품질과 MAP 조건에서 저장성에 미치는 영향)

  • This study was analyzed the effects of boron (B), calcium (Ca), silicon (Si) on quality and shelf life of 'Unicorn' cherry tomato at the light red maturity-stage. The storage conditions were modified atmosphere packaging (MAP) by oxygen transmission rate (OTR) packaging film at $5^{\circ}C$, $11^{\circ}C$, and $24^{\circ}C$. Respiration and ethylene production were the lowest in B + Ca + Si -treated tomato fruits. The lowest fresh weight loss and the longest shelf life resulted from the B + Ca + Si treatment. And the firmness was enhanced by B + Ca + Si treatment at harvest time, and it was retained after storage at $5^{\circ}C$, $11^{\circ}C$, and $24^{\circ}C$. Significantly lower soluble solids, lycopene, and color development were found at B + Ca + Si-treated tomato fruits compared with control after storage. Moreover, the highest titratable acidity and vitamin C content were observed in B + Ca + Si-treated tomato fruits after storage. From the above results, it was concluded that B + Ca + Si combined treatment can delay the maturity of cherry tomato after harvesting, and retained the firmness and prolong the shelf life.

Fresh-cut사과 전처리에 따른 물성학적 품질 특성

  • 편의와 fresh-like quality를 선호하는 소비 경향에 맞추어 가공한 fresh-cut사과의 선도 연장을 위한 저해제를 탐색하였다. 실험에 사용된 사과는 박피하여 속심을 제거한 사과를 8조각으로 절단하여 저해제에 3분간 침지한 후 물기를 제거하고 각각 LDPE 포장지에 포장하여 실온에서 저장하면서 colorimeter와 rheometer를 이용하여 색도와 물성을 측정하였다. 저해제로는 1% ascorbic acid, citric acid, NaCl, MgCl$_2$, CaCl$_2$, KCl, CaSO$_4$, MgSO$_4$, $Na_2$SO$_4$, oxalic acid, calcium lactate, calcium citrate를 사용하였으며 천연저해제로는 건칡, 맥문동, 목향, Rhubarb, 당귀, 오약, 시체, 지각, 천문동, 청피, 계피, 구기자, 당귀, 진피, 감초, 녹차, 후추 등을 열수추출하여 사용하였다. 물리적인 변화는 시간이 경과함에 따라 감소하는 추세를 보였으며 일부 건조로 인해 texture의 증가를 나타내기도 했다. 처리에 따른 물리적인 변화로는 CaCl$_2$, NaSO$_4$, Cl, 시금치를 처리하였을 때 그 변화가 가장 작았으나 시금치의 경우 시금치로 인한 사과의 색변화로 관능적으로 좋지 않았다. 한약재는 목향, 당귀, 지각, 계피를 처리하였을 때 물리적인 변화가 가장 작았으나 시금치와 마찬가지로 처리 초기에 한약재 자체의 향과 색으로 인하여 관능적으로 좋지 못한 결과를 나타냈다. 갈변 정도를 나타내는 ΔE의 변화는 KCl과 CaCl$_2$, CaSO$_4$를 처리한 구에서 변화가 가장 작았으며 한약재를 처리하였을 경우 당귀, 천문동을 처리한 구에서 효과를 보였다. oxalic acid는 갈변저해에는 효과적이었으나 시간이 경과함에 따라 수분의 용출이 증가하였으며 물성의 변화도 컸다. 또한 ZnCl$_2$를 처리한 구에서는 이취 발생과 수분 용출이 컸으며 색의 변화도 컸다.

Effects of Dipping and Preheating Treatments on Susceptibility to Browning of Potato Slices During Cold Storage (침지용액과 예열 처리가 감자 슬라이스의 냉장 중 갈변정도에 미치는 영향)

  • Potato slices were dipped in solution of 2% CaCl$_2$, and 1% chitosan or preheated in each of these solutions for refrigeration for 4 weeks at 5$^{\circ}C$. Changes in L value, content of total phenol and chlorogenic acid and po lyphenol oxidase activity were determined. During refrigeration, it was found that L values of potato slices treated with CaCl$_2$, (CaPS) and those treated with chitosan (ChPS) increased. Contents of total phenol and chlorogenic acid of potato slices decreased, While potato slices preheated in CaCl, solution (Hcaps) showed much decreases. And polyphenol oxidase (PPO) activity of potato slices decreased except CaPS and decreases in PPO activities were much larger in preheated potato slices. From these results, of those treatments used, chitosan treatment combined with preheating have shown to be effective to control enzymatic browning of potato slices during refrigeration.

  • In an effort to investigate the role of calmodulin (CaM) as a modulating molecule in the signal transduction system in plant cells, we established methods for introduction of purified CaM into cultured soybean cells. CaM was purified from bovine testis, and was labelled with fluorescein isothiocyanate (FITC). Suspension -cultured cells were healed with saponin (0.1 mg/mL) to permeabilize the plasma membrane and coincubated with FITC-CaM complex. Saponin pretreatment was found to increase the fluorescence in the suspension cultured cells, indicating that the FITC-CaM complex could be incorporated into the cytoplasm. Optimal conditions for introducing FITC-CaM complex into protoplasts by electroporation were established with various electric pulses. With increasing field strength, the fluorescence in the protoplase was increased, while the viability of the protoplase decreased. FITC-CaM complex was successfully introduced into the protoplasts by electroporation and the amount of FITC-CaM complex in the protoplase was estimated.

Increase of Strawberry Fruit Shelf-life through Preharvest Spray of Calcium-chitosan and Post-harvest Treatment with High Pressure CO2 (수확 전후 칼슘-키토산 및 고농도 CO2 단기처리에 의한 '설향' 딸기 과실의 저장성 증진)

  • This experiment examined the effects of preharvest Ca-chitosan spraying and short-term post-harvest treatment with high $pCO_2$ on the shelf-life of 'Seolhyang' strawberry fruit. Fruit firmness decreased when harvested late in the season and was little affected by Ca-chitosan spray treatments. Short-term treatment with high $pCO_2$ after harvest, however, significantly increased fruit firmness regardless of preharvest treatment. The combined treatment of Ca-chitosan spray and high $pCO_2$ led to an additional increase in shelf-life through delay of skin disorder, fruit decay incidence and firmness loss during simulated marketing. The residual effects of both treatments, however, were reduced when fruit were harvested late in the season. Fruit firmness was more affected by high $pCO_2$ treatment whereas decay and visual quality were affected by preharvest spraying with Ca-chitosan. Although more research is required to determine the optimum concentration of calcium and chitosan, and the optimal spray interval, our results show that the combined treatment of preharvest Ca-chitosan spray and postharvest high $pCO_2$ is effective in improving strawberry fruit shelf-life by increasing firmness and delaying decay.

The Effects of CaCl2 Foliar Application on Inhibition of Abnormally Fermented Fruits and Chemical Composition of Oriental Melon(Cucumis melo L. var. makuwa Mak.) (CaCl2의 엽면처리(葉面處理)가 참외의 이상발효과 발생억제 및 과실성분(果實成分)에 미치는 영향)

  • The effects of $CaCl_2$ foliar application on inhibition of the occurrence of abnormally fermented fruit and chemical composition of oriental melon (Cucumis melo L. var. makuwa Mak. cv. Gumssaragi-eunchun) were examined. $CaCl_2$, applied at 0.3~0.7%, had an apparent inhibitory effect on occurrence of abnormally fermented fruit when sprayed three times at five days intervals from 10 days after flowering. The effect of $CaCl_2$ was diminished when the field was irrigated during the ripening period. Sugar content in the fruits was not affected by the $CaCl_2$ treatment. The Ca content was higher in the normal fruit than in the fermented ones, but no difference was noted in the K and Mg levels. The occurrence of fermented fruit was increased when the applying nitrogen levels were less (10kg) or higher (25kg/10a) than normal supply. The nitrogen application had no effect on the calcium content in fruits.

  • Mg-Zn-Ca합금의 잉곳트는 BN을 내벽에 바른 4x$10^{-5}$Torr의 진공의 석영관내에서 제조하였다. Mg-Zn합금계의 입자미세화를 위하여 0.5-5wt.%조성범위의 Ca를 첨가하였다. 제조된 합금을 용체화처리한 후 결정립크기와 경도를 측정하였다. Mg-6wt.%Zn합금의 입자미세화효과는 Ca가 1.5wt%첨가 될 때 최적의 조건이었다. Mg-6wt.% Zn과 Mg-6wt.% Zn-2wt.% Ca및 Mg-6wt.% Zn-1.5wt.% Si합금을 시효처리하여 시효거동을 조사하였다. 입자미세화에 의한 경도증가효과는 Mg-Zn-Ca합금계에서 크게 나타났으며 시효에 의한 경도증가효과는 Mg-Zn-Si합금계에서 크게 나타났다. Mg-6wt.%Zn합금의 부식전류밀도는 0.5wt.% Ca의 조성에서 감소되었다.

Changes in Mineral Uptake and Hormone Concentrations in Rice Plants Treated with Silicon, Nitrogen and Calcium Independently or in Combination (규소, 질소, 칼슘 단독 및 혼합처리가 벼 식물체 내 무기성분 흡수 및 식물호르몬 함량 변화에 미치는 영향)

  • To elucidate the physiological responses of rice plants to the essential mineral silicon (Si), we assessed the effects of treatments with Si, nitrogen ($NH_4NO_3$; ammonium nitrate), and calcium ($CaCl_2$; calcium chloride), independently or in combination on mineral uptake rates and levels of the hormones abscisic acid (ABA), gibberellin ($GA_1$) and jasmonic acid (JA). We found that nitrogen and calcium uptake was inhibited by Si application. However, solo application of nitrogen or calcium did not affect Si uptake. Compared to the untreated plants, the application of Si, $NH_4NO_3$ or $CaCl_2$ increased the endogenous hormone levels in treated plants. In particular, the concentrations of $GA_1$ and JA increased significantly after the application of Si or $NH_4NO_3$. The level of $GA_1$ observed after a treatment (solo or combine) with Si, and $NH_4NO_3$ was higher than that of the control. By contrast, independent application of $CaCl_2$ or a combined treatment with Si and $CaCl_2$ did not alter $GA_1$ levels. The highest level of $GA_1$ was present in plants given a combination treatment of Si and $NH_4NO_3$. This effect was observed at all time points (6 h, 12 h and 24 h). Endogenous JA contents were higher in all treatments than the control. In particular, a combination treatment with Si and $NH_4NO_3$ significantly increased the JA levels in plants compared to other treatments at all time points. A small increase in JA levels was observed after 6 h in plants given the $CaCl_2$ treatment. However, JA levels did not differ between plants given a $CaCl_2$ treatment and controls after 12 h or 24 h of exposure. We conclude that treatment with $CaCl_2$ alone does not affect endogenous JA levels in the short term. Endogenous ABA contents did not show any differences among the various treatments.
