The effect of octanol on the intracellular Ca2+ increase in submandibular acinar cells evoked by β-adrenergic activation (악하선 선포세포에서 β-아드레날린계 활성화 후 세포내 Ca2+ 농도 증가에 미치는 옥탄올의 효과)

  • The concentration of intracellular $Ca^{2+}$ was measured by spectrofluorometer after rat submandibular acinar cells were loaded with fura-2/AM(fura-2). After isoproterenol and octanol were administered while letting submandibular gland acinar cell placed in a perfusion chamber flow through a standard solution, the changes of $Ca^{2+}$ concentration were measured. When they were administered separately, there showed little changes of intracellular $Ca^{2+}$ concentration. When they were administered at the same time, however the concentration of intracellar $Ca^{2+}$ was shown to increase. When forskolin, an adenylate cyclase activater, was administerd together with octanol the response looked similar to the response of isoproterenol. In case of the extracellular $Ca^{2+}$ was removed by omitting $Ca^{2+}$ in standard solution and treating EGTA, isoproterenol and forskolin does not affect to the concentration of intracellular $Ca^{2+}$. Therefore, it was certified that the increase of intracellular $Ca^{2+}$ was caused from outside the cell. In order to know that the $Ca^{2+}$ influx is related with capacitative entry pathway, godolinium, blocker of that pathway was treated. With the result of that experiment there was no complete control of the increase in the concentration of intracellular $Ca^{2+}$. However, speed and amount of $Ca^{2+}$ increase was comparatively diminished.

Effects of Delayed CA Treatment on Fruit Quality of 'Fuji' Apples During Storage (지연 Controlled Atmosphere(CA)저장이 저장 중 '후지' 과실의 품질에 미치는 영향)

  • This study was conducted to investigate the effect of delayed controlled atmosphere (CA) storage on fruit quality and the incidence of storage disorders during CA storage of 'Fuji' apples (Malus x domestica Borkh.). In the first year of experiment, 'Fuji' apples showed 40% of watercore disorder at the commercial harvest time (Oct. 22). Twenty days delayed CA storage caused to develop the incidence of flesh browning while 30 days delayed CA storage did not. In the second year of experiment, watercore was increased with delaying harvest time. As the estabilishment of delayed CA storage was delayed, the incidence of flesh browning increased at 10 to 30 days delayed CA storage but 40 days delayed CA storage did not have any flesh browning incidence. Respiration rate increased with extending the storage duration. Delayed CA storage reduced respiration rate but was not different compared to the rapid CA. Ethylene production rate was lower in delayed CA storage than in cold storage but higher in delayed CA storage, compared to rapid CA storage. Titratable acidity was lower in delayed CA storage as CA storage progressed. Firmness was not different between rapid CA storage and delayed CA storage after 8 months of CA storage. Therefore, the results indicate that delayed CA storage should be a promising approach to reduce the risk development of flesh browning and watercore during CA storage of 'Fuji' apples.

Enhancement of Stem Firmness in Standard Chrysanthemum 'Baekma' by Foliar Spray of Liquid Calcium Compounds (액상 칼슘 화합물 엽면살포에 의한 스탠다드 국화 '백마'의 줄기 경도 강화)

  • This study was conducted to enhance the stem firmness of standard chrysanthemum 'Baekma' bred in Korea for commercial quality improvement and inhibition of stem breaking during transportation through foliar spray with calcium agents. Calcium agent screening 'Baekma' was examined using $CaCl_2{\cdot}2H_2O$, $Ca(NO_3)_2{\cdot}4H_2O$, and OS-Ca (natural liquid calcium compounds extracted from oyster shell) depending on each concentration (0, 0.001, 0.01, 0.1, and 1.0%, respectively). All calcium agents sprayed with 1.0% caused chemical injury such as stem bending or leaf burn. OS-Ca also showed more sensitive response to chemical injury than the other calcium agents because OS-Ca was absorbed very well by 'Baekma' leaves. Maximum stem firmness measured during the final harvest was greater in OS-Ca than in the other calcium agents. Especially, maximum stem firmness was greatest in 0.01% OS-Ca. However, elastic strength and maximum bending stress were greater in 0.001% OS-Ca than in the others. Thus, OS-Ca ranged from 0.005 to 0.05%, which did not show any chemical injury, was finally selected as the first candidate for hardening the stem of 'Baekma'. The next experiment using OS-Ca was conducted with the concentrations of 0, 0.005, 0.01, and 0.05%, respectively. From the results, 0.05% OS-Ca showed better plant growth and parameters such as plant height, stem diameter (upper and middle part), the number of leaves, and dry weights of each part than the other concentrations of OS-Ca and control. As for stem firmness depending on OS-Ca concentration, the Ca content within stem, maximum firmness, elastic strength, and maximum bending stress of stem in 'Baekma' sprayed with 0.05% OS-Ca showed the highest values among all the treatments and it turned out to be very high level of significance between control and OS-Ca treatments. However, the area and percentage of the inside cavity within horizontal stem section in 'Baekma' did not show any significance between any treatments including control. Thus, stem firmness of 'Baekma' did not show any correlation with the inside cavity area of stem. In conclusion, we recommend foliar sprays with 0.05% OS-Ca at vegetative growth stage to enhance stem firmness of 'Baekma' during transportation.

Effect of Postharvest Calcium Solution Dipping and Vacuum Infiltration on Calcium Content and Quality of Chojuro Pear Fruit (배 장십랑품종의 저장 전 $CaCl_2$처리에 따른 과실 칼슘함량과 과실특성의 변화)

  • Postharvest dipping of vacuum infiltration treatments of 'Chojuro' pear fruit in $CaCl_2$ resulted in increased calcium content, especially in fruit peel and outer flesh such as just below peel. As $CaCl_2$ concentrations increaced from 2% to 8% in dipping treatment, calcium content became higher. But vacuum infiltration under 200-600 mmHg did not affect and dipping and vaccum infiltration did not have any difference in fruit calcium content. Decreasing of fruit firmness determined at 4 weeks during storage was slower in fruits with $CaCl_2$ treatment than control.

Postharvest Quality of Satsuma Mandarin Fruit Affected by Controlled Atmosphere (CA 저장이 온주밀감의 수확 후 품질에 미치는 효과)

  • This research was conducted to find out whether controlled atmosphere(CA) can maintain Satsuma Mandarin fruit fresh longer compared to cold storage. Fruit harvested in Nov. 1998 at Jeju Province were stored in three different CA condition including cold storage. The CA treatments consisted of combinations of 3%, 5% and 10.9% $CO_2$ with 1% and 3-3.9% $O_2$, respectively. Weight and firmness were maintained higher in static CA chambers than in the cold storage. Soluble solid content in fruit of cold storage increased until 2 months of storage and thereafter sharply decreased, whereas that of CA-stored fruit increased slowly throughout the storage period of 120 days. Citric acid decreased obviously in all the treatments with a partially longer retention in CA chambers during storage. Sensory evaluation clearly showed that CA could extend market period of Satsuma Mandarin fruit.

Freshness of 'Fuji' Apples Packed MA Film Treated with Acid and Anti-fogging Agents (산 및 결로방지 처리한 기능성 MA포장으로 포장한 '후지' 사과의 신선도)

  • This study investigated the effect of functional MA films (FMA) masterbatched by zeolite powder treated with 1 N HCl solution and anti-fogging agent (CA) on the freshness extension of 'Fuji' Apples. Preference, weight loss, total ascorbic acid, sugar content, acidity, change of gas composition in package were evaluated during storage at $15^{\circ}C$. After 150 days of storage, the weight loss of LLDPE film (control, L) apples was 0.4%, and that of apple in FMA film (CA) was 1.1%. Total ascorbic acid content of apples in control after 150 days was 2.09 mg%, and those of apple in FMA film was 2.94 mg%. The titratable acidity of apple in FMA film was higher than that in control, while soluble solids content of packages was lower than that in control after 150 days. Ethylene gas adsorbability in control package was 192.2 ppm and those in FMA film was 165.6 ppm. Overall, apples in FMA film was better than that of control. It was verified that apples packed with LLDPE film(control) lasted about 130 days, while those with FMA film lasted about 170 days. Also, FMA film treated with anti-fogging agent achieved no significant effect on freshness extension compared to non-treated film, but quality of 'Fuji' Apples was improved by anti-fogging agent when compared to that of non-treated anti-fogging agent.

Effect on Shoot Growth Inhibition by Prohexadion-Calcium in 'Sato Nishiki' Sweet Cherry (Prohexadion-Calcium이 체리 '좌등금'의 신초생장 억제에 미치는 영향)

  • This research was conducted to get the basic information for improving production amount as well as fruit quality by using Prohaxadion-Calcium (Pro-Ca) used as 'Sato Nishiki', cherry cultivar, which is most frequently cultivated in Korea. In fruiting trees, we used Pro-ca and concentration of treatment selected 150, 200 and $250\;mg{\cdot}L^{-1}$. When treating Pro-Ca $200\;mg{\cdot}L^{-1}$, we could get the highest effect on inhibition of shoot growth. All treatments of Pro-Ca were increased chlorophyll a and b, respectively compared with control. Although treatments of Pro-Ca treatment showed the decreased effect on leaf area and there was no difference in fruit quality among treatments. Also, Pro-Ca $250\;mg{\cdot}L^{-1}$, sprayed 3 times, and pclobutrazol ($PP_{333}$) 500 and $1,000\;mg{\cdot}L^{-1}$, applicated soil, were treated to 1 year old plnted pot. Pro-Ca and $PP_{333}$ showed inhibition of shoot growth, especially, Pro-Ca showed the more effect than $pp_{333}$ on growth inhibition. Soil applications of $pp_{333}$ showed less effect to enlargement of tree trunk compared to Pro-Ca and control. Pro-Ca and $pp_{333}$ were not difference in number of inter-nodes when comparing to control, but they on decreased leaf area and increased chlorophyll.

Effects on Quality of Strawberry Fruit by Dipping of Calcium Solution and MA Packaging (칼슘처리와 MA포장이 딸기의 품질에 미치는 영향)

  • The strawberry was dipped into $1\%\;CaCl_2$ solution at $20^{\circ}C\;and\;45^{\circ}C$ for 15min. respectively, and then packaged with or without PE film (thickness: 0.011mm) before putting into the cold storage at $5^{\circ}C$. The results showed the lowest rate of rot incidence and the highest firmness with higher Vitamin C content and the best superficial appearance in the treatment of $CaCl_2$ dipping at $45^{\circ}C$ with packaging. It also showed lower pH and higher titratable acidity in the treatment However, no significant difference of SSC was found among various treatments. The results suggested that the combination of postharvest calcium dipping and packaging could result in more effective preservation for strawberry compared with the individual treatment.

Photosynthesis, Shoot Growth and Fruit Quality in 'Fuji'/M.9 Mature Apple Trees in Response to Prohexadione-calcium Treatments (Prohexadione-calcium 처리에 따른 성목기 '후지'/M.9 사과나무의 광합성, 신초생장 및 과실품질)

  • This study was conducted to elucidate the influence of prohexadione-calcium (Pro-Ca) application on shoot growth and fruit quality in 'Fuji'/M.9 mature apple trees. Pro-Ca was applied at concentrations ranging from 100 to $400mg{\cdot}L^{-1}$ one time at petal fall (PF; 5-10 cm terminal shoot growth) to the whole canopy of the tree, or at 100, 125, $150mg{\cdot}L^{-1}$ concentrations two times, at PF and then 4-8 weeks after PF. Pro-Ca treatment generally reduced mean shoot growth, with its effect being proportional to the application rate. Pro-Ca at $400mg{\cdot}L^{-1}$ significantly reduced the shoot growth by 15-22% compared to the control, while its effect was not significantly different from Pro-Ca $200mg{\cdot}L^{-1}$. Pro-Ca treatments induced higher occurrence of secondary growth compared to the control. Pro-Ca treatment increased the photosynthetic rate by 5-10% relative to the control, and also increased soluble solid concentration and fruit red color. However, fruit weight was significantly reduced by $400mg{\cdot}L^{-1}$ Pro-Ca, which was attributable to the greater secondary growth caused Pro-Ca $400mg{\cdot}L^{-1}$ treatment compared to the control and Pro-Ca $200mg{\cdot}L^{-1}$ treatment.

Effects of Foliar Application of CaCl2 on Ca Content and Occurrence and Alleviation of Blossom-end Rot of Paprika During High Temperature Season (고온기 CaCl2 엽면 살포가 파프리카 칼슘함량, 배꼽썩음과 발생과 경감에 미치는 영향)

  • This experiment was carried out to determine the effect of CaCl2 foliar spraying on the inhibition of blossom-end rot (BER) in hot summer paprika (Capsicum annum L. 'Special') cultivation. The effect of CaCl2 application was examined by the foliar application based on different fruit size, frequency, and spraying time. Also, it was investigated the occurrence and alleviation effect of BER symptom. Foliar application of CaCl2 (Ca 0.4%) was conducted by treating a fruit and leaf at 4 to 9 nodes above the crown flowers of each plant with 350 mL per week from June 3 to July 1. When the CaCl2 was sprayed at 7-day intervals for 4 weeks, the Ca content was the lowest in the fruit harvested with BER symptom in 11 to 20 mm of fruit width (FW). Four different regions in both BER symptom and normal fruits showed significant differences of Ca content, the highest was in pedicel, followed by stem-end, middle, and blossom-end. The Ca content increased sharply in normal paprika with 31-40 mm FW, in which Ca content was 78% higher than that of BER. Ca content in the middle and blossom-end of paprika over 21 mm in FW ranged 19.8% to 28.8% in normal fruits and 15.7% to 18.5% in BER, respectively. The incidence of BER increased rapidly by more than 60% in fruits with 31-40 mm FW. While there was no difference in fruit weight among the FW treatment, marketable yield rate was highest in the 21-30 mm FW, and the sugar content was high in the 11-30mm FW. When CaCl2 was applied three times for 7 days to a paprika having a 21- 30 mm FW, the cell wall-bound (CWB) Ca content was the highest and the rate of BER was lowest with 6.3%. After 10 days of CaCl2 foliar spray treatment, the CWB Ca content of paprika increased by 2.9 to 3.5 times compared to the control in all treatments. At 7 days after the CaCl2 foliar spraying once a day at varying spraying time, the leaf burn observed from 9:30 a.m. to 17:00 p.m. and the proline content increased as the spraying time was delayed. Therefore, the CaCl2 foliar spraying method for reducing of BER occurrence during paprika summer cultivation seems to be appropriate to spray 2-3 times at intervals of 3 days and before 8 a.m. at the time when the FW is 21-30 mm.
