Gas Turbine Data Acquisition and Monitoring System for Combined Cycle Power Plant

  • This paper presents a data acquisition and monitoring system for a gas turbine. The proposed system entitled C-Tune DAS plays an important role to make an analysis of the real-time operation of the gas turbine under maintenance. The designed LabVIEW based software is divided into three parts according to their original functions, i.e., data acquisition, data analysis with display, and data storage. The data acquisition part receives data from a PMS (Plant Management System) server and two cFPs (Compact-Field Point). To verify the validity of the developed system, it is applied to gas turbines in the combined cycle power plant in Korea.

  • The purpose of this study is to define the role of a T. Q. C. engineer in the plant engineering activities. A "plant engineering" moves to complete a new project through a series of stages such as: 1. Research & Development stage. 2. Economic evaluation & engineering design stage. 3. Erection & start up stage. 4. Final production & control stage. This suggests that the plant engineering project should be carried out with a wide variety of tasks, as well as a central control. For a successful completion of the project, a T. Q. C. engineer is required, in practice, either as a central activity creator or as a key advisor in each task.

Molecular Cloning, Phylogenetic Analysis, Expressional Profiling and In Vitro Studies of TINY2 from Arabidopsis thaliana

  • A cDNA that was rapidly induced upon abscisic acid, cold, drought, mechanical wounding and to a lesser extent, by high salinity treatment, was isolated from Arabidopsis seedlings. It was classified as DREB subfamily member based on multiple sequence alignment and phylogenetic characterization. Since it encoded a protein with a typical ERF/AP2 DNA-binding domain and was closely related to the TINY gene, we named it TINY2. Gel retardation assay revealed that TINY2 was able to form a specific complex with the previously characterized DRE element while showed only residual affinity to the GCC box. When fused to the GAL4 DNA-binding domain, either full-length or its C-terminus functioned effectively as a trans-activator in the yeast one-hybrid assay while its N-terminus was completely inactive. Our data indicate that TINY2 could be a new member of the AP2/EREBP transcription factor family involved in activation of down-stream genes in response to environmental stress.

Molecular Cloning and Expression of Dihydroflavonol 4-reductase Gene in Tuber Organs of Purple-fleshed Potatoes

  • A full-length cDNA encoding dihydroflavonol 4-reductase (st-dfr) of potato was isolated by rapid amplification of cDNA ends, and their expression was investigated from purple-fleshed potato (Solanum tuberosum L. cv. Jashim). The st-dfr exists as a member of a small gene family and its transcripts was abundant in the order of tuber flesh, stem, leaf, and root. The expressions of st-dfr gene were light inducible and cultivar dependant. Transgenic potato plants harboring antisense st-dfr (AS-DFR) sequences were analyzed. The accumulation of mRNA was nearly completely inhibited as a result of introducing an AS-DFR gene under the control of the 35S CaMV promoter into the red tuber skin Solanum tuberosum L. cv. Desiree. The anthocyanin content of the tuber peels of the transgenic lines was dramatically decreased by up to 70%. The possible production of flavonols in the peels of AS-DFR transgenic potatoes was discussed.

Cryopreservation of Hevea brasiliensis zygotic embryos by vitrification and encapsulation-dehydration

  • The mature zygotic embryos of the Hevea brasiliensis were cryopreserved through the use of the vitrification and encapsulation/dehydration techniques. In all the experiments, the zygotic embryos were pre-cultured for three days in the MS medium supplemented with 0.3 M sucrose before they were used for the cryopreservation technique. In the vitrification procedure, the effect of the plant vitrification solutions (PVS2 and PVS3) and exposure time were studied. The highest survival rate (88.87%) and regrowth (66.33%) were achieved when the precultured zygotic embryos were incubated in a loading solution for 20 minutes at $0^{\circ}C$. They were subsequently exposed to PVS2 for 120 minutes at $0^{\circ}C$ and plunged directly into liquid nitrogen. Cryopreservation by the encapsulation-dehydration method was successfully done by leaving the encapsulated zygotic embryos in a laminar flow for 4 hours prior to plunging into a LN. The survival rate and regrowth of the encapsulated zygotic embryos were 37.50% and 27.98%, respectively. The cryopreserved zygotic embryos were able to develop into whole plants.

A novel WD40 protein, BnSWD1, is involved in salt stress in Brassica napus

  • Genes that are expressed early in specific response to high salinity conditions were isolated from rapeseed plant (Brassica napus L.) using an mRNA differential display method. Five PCR fragments (DD1.5) were isolated that were induced by, but showed different response kinetics to, 200 mM NaCl. Nucleotide sequence analysis and homology search revealed that the deduced amino sequences of three of the five cDNA fragments showed considerable similarity to those of ${\beta}$-mannosidase (DD1), tomato Pti-6 proteins (DD5), and the tobacco harpin-induced protein hin1 (DD4), respectively. In contrast, the remaining clones, DD3 and DD2, did not correspond to any substantial existing annotation. Using the DD3 fragment as a probe, we isolated a full-length cDNA clone from the cDNA library, which we termed BnSWD1 (Brassica napus salt responsive WD40 1). The predicted amino-acid sequence of BnSWD1 contains eight WD40 repeats and is conserved in all eukaryotes. Notably, the BnSWD1 gene is expressed at high levels under salt-stress conditions. Furthermore, we found that BnSWD1 was upregulated after treatment with abscisic acid, salicylic acid, and methyl jasmonate. Our study suggests that BnSWD1, which is a novel WD40 repeat-containing protein, has a function in salt-stress responses in plants, possibly via abscisic acid-dependent and/or -independent signaling pathways.
