Interaction of Pyrazole - and Chloroacetamide Herbicide Combinations in Control of Echinochloa crusgalli (Pyrazole계(系)와 Chloroacetamide계(系) 제초제(除草劑)들의 혼합처리(混合處理)가 피(Echinochloa crusgalli)의 살초효과(殺草效果)에 미치는 상호작용(相互作用))

  • Three pyrazole-herbicides, pyrazolate, pyrazoxyfene and benzophenap, were evaluated for their interaction in controlling barnyardgrass (Echinochloa crusgalli) with two chloroacetamide-herbicides, butachlor and pretilachlor. Percent inhibition of barnyardgrass growth by pyrazolate, pyrazoxyfene, and benzophenap was 44%, 64%, and 0%, respectively, when each was applied at the 1.5 leaf-stage of barnyardgrass at a rate of 3㎏ ai per ㏊ as single treatment, and the benzophenap showed 60% inhibition when it was applied at the coleoptile stage. While the lowest rate controlling the 1.5 leaf-stage barnyardgrasses by 98 to 100% of the butachlor and pretilachlor was 1.5㎏ and 200g per ㏊, respectively. All of the combinations of pyrazolate with butachlor, pyrazoxyfene with pretilachlor, and benzophenap with butachlor have shown synergistic interaction in controlling barnyardgrass on the Chisaka's isobole of 90% growth inhibition as well as on the Colby`s interaction efficacy data; synergism indices were 2.44, 1.62 and 1.52 in order. The dose combinations shown the maximal synergism were 1870g of pyrazolate with 140g of butachlor (1:0.075), 33008 of pyrazoxyfene with 338 of pretilachlor (1:0.01), and 3350g of benzophenap with 520g of butachlor (1:0.15) on the ai/㏊ basis.

  • This experiment was conducted to evaluate the effect of gibberellin biosynthesis retardants as used by rice lodging inhibitors on the gibberellin antagonism, auxin interation, ethylene evolution and growth of second crops. Results obtained can be summarized as follows. Inabenfide, paclobutrazol and uniconazole markedly inhibited the epicotyl elongation of mung bean. Inhibiting effect of epicotyl by these chemicals was markedly stimulated by gibberellic acid, thus showing clear antagonism between these chemicals and gibberellic acid. Significantly large number of roots were formed in the mung bean cuttings which were rooted in the paclobutrazol and uniconazol of 1 ppm. The higher the concentration, the more the number of roots forms. It was guessed that these effect was closely related with auxin. Ethylene evolution was a little stimulated in the leaf of rice under the treatment of inabenfide, paclobutrazol and uniconazole at earlier stage(5 DAT), however it was suppressed at later stage(10, 30 DAT) at higher concentration. The effect of gibberllin biosynthesis inhibitors to second crops retarded tomato plants without influencing the height of barley. The treatment of paclobutrazol and uniconazol which is triazole-type more severely inhibited than that of inabenfide which is isonicotinanilide-type. The more the concentration, the less the height of tomato plants.

  • Recently, the formation of the complexes between macrocyclic polyethers and alkali metal salts have been determined by several methods. It has been suggested that the specificity of the complexation be due to the nature of the surrounding solvent molecules. The formation constant values ($K_f$) of $K^+$ are shown to be the largest among the other alkali metal cations because the ionic diameter of $K^+$ is approximately the same with the hole size of 18-crown-6. In this study the formation constants of the 1 : 1 complexes of 18-crown-6 with potassium p-substituted phenoxide are calculated by the conductance measurement in organic solvents. As a result, the $K_f$ value series among organic solvents are given in the order of $CH_3$OH > DMF > DMSO. It seems that the donor number of the solvent is a main factor in the formation of the complex between $K^+$ metal ion and 18-crown-6 molecules. At the same time, the formation constants increase with increasing the electron-withdrawing power of substituents because the phenoxide ion is stabilized by the charge dispersion.

  • In order to investigate the fungicidal activities against various plant pathogenes, diminishing effect of plant transpiration, phytotoxicities, vapor effect and the rate of reduction by ultraviolet rays of phenylmercuric 8-oxyquinolinate(P.M.Q), this experiments were undertaken under various laboratory conditions. 1. Inhibitory activity on the spore germination of this chemical was shown less effective than that of P.M.A..(Table 2, Table 3, Table 4, Table 5 and Table 6) Also, P.M.Q. was resulted a somewhat higher inhibitory activity on the hyphae growth than P.M.A. (Table 7). 2. In the diminishing effect of plant transpiration, 8-hydroxyquinoline sulfate(oxine sulfate) was more strong inhibitory at first than P.M.Q., while, at last, P.M.Q. was more strong inhibitory in comparison with oxine sulfate(Table 8, Fig. 1 and Table 9). 3. P.M.Q. was shown less injury on the germination of rice plant seeds and the emergence of their roots than P.M. A.(Table 10). Injuries was not observed on the rice seedlings and soy-bean seedlings sprayed with 40 ppm of this chemical. 4. P.M.A. had more inhibitory effects on the mycelial growth of phytopathogenes than P.M.Q. on the vapor effect (Table 11, Fig. 2). 5. Biological activity and chemical decomposition rate of P.M.A. were greatly reduced by exposure of this compound to ultraviolet rays. But, P.M.Q. was only slightly affected by similar treatment(Table 12, Fig. 3, Table 13 and Fig. 4). From the above results, this chemical will be a promising fungicide adding fungitoxicities against various phytopatho genes, diminishing effect of plant transpiration and physico-stability.

Effects of polysaccharide fractions from phellodendron chinese SCHNEID on tumor progression and immunopontentiation

  • In the previous paper (Kim et al., Glycoconjugate Journal 16, 247-252, 1999), heteropolysaccharides from Korean medicinal plant, Phellodendri cortex (Hwangbek) showed a poten B-Iymphocyte-stimulating activity in a system using polyclonal antibody forming cells in C57BL/6XC3H mice at dosages of 2-10 mg. In a series of biolgical active polysaccharides from natural medicinal plants, the polysaccharide fractions were isolated and purified from Phellodendron chinese SCHNEID, and antitumor activities were examined at dosages of 2, 5 and 10 mg/100 g. F-7 and F-8 showed the highest tumor inhibitory activities (inhibition ratio 96.4 and 98.2% in 2 mg/100 g), and in dose of 5 mg/100 g, the inhibitory ratios were 95.3 and 97.5%, respectively. Furthermore, 10 mg/100 g of intraperitoneal (i.p.) injection gave 97.3 and 98.7% of inhibition. In oral administration, the inhibitory activities were not markedly observed, indicating that the polysaccharides are directly acting to immune system. When the effects on TS and TK activities were determined, TS activities in the F-2 and F-7-treated mice were markedly suppressed to 73.7% and 79.5% of that in the control (p<0.01), while there was little difference in TK activity with a slight decrease in F-2 only. However, in i.p. injection, TS activities in the F-2, F-5, F-7 and F-8-treated mice were markedly suppressed to 83% to 85% of that in the control (p<0.01). Furthermore, there was also significant differences in TK activities in F-2, F-5, F-7 and F-8-treated mice (p<0.05). Therefore, polysaccharide fraction F-8 was further purified to active fractions of F-9 and F-11 by gel permeation chromatography using TSK Gel HW50S. The purified polysaccharides of F-9 and F-11 were composed of GlcNAc (47.3%), Gal (24.7%) and Man (28.0%). These results clearly indicated that the i.p. injection is much effective to suppress tumor growth than oral administration.

Quality Characteristics of Extracted Coffee with Cold Water according to Water Quantity and Dilution Ratio (가수량 및 희석 비율을 달리하여 제조한 냉수 추출 커피의 품질 특성)

  • This paper investigated the optimal conditions for coffee extraction and dilution by comparing differences in soluble compounds and sensory characteristics of coffee subjected to cold-water extraction according to water quantity and dilution ratio. For sample preparation, we extracted coffee for 3 to 10 hours by adding 400 mL, 600 mL, 1,000 mL and 1,800 mL of cold water ($20^{\circ}C$) to 100 g of ground coffee, resulting in 200 mL, 400 mL, 800 mL and 1,600 mL portions of extracted coffee, respectively. Following this, the 200 mL, 400 mL and 800 mL portions of extracted coffee were further diluted with 1,400 mL, 1,200 mL and 800 mL of cold water, resulting in ratios of coffee to cold water of 1:7, 1:3 and 1:1, respectively. The 1,600 mL of portion of extracted coffee was not diluted and used as a sample. Undiluted coffee showed the highest values for pH, acidity, reducing sugar, total soluble solids and total phenolic compounds among all sample coffees, whereas the highly diluted coffee with a high ratio of 1:7 showed the lowest values. In a descriptive test, odor, sourness, bitterness and body intensity were also the highest in undiluted coffee due to the overly extracted coffee compounds. Undiluted coffee showed the lowest preference in terms of aroma, aftertaste and balanced coffee taste. On the other hand, diluted coffee at a ratio of 1:7 showed the lowest values for odor, sourness, bitterness and body intensity. Diluted coffee at a ratio of 1:3 showed the highest values for sweetness and acidity and was the most preferred in terms of aroma, aftertaste and balance. As a result, our study concluded that the optimal conditions for extraction and dilution of coffee with cold water are a ratio of ground coffee to cold water of 1:6 (w/v), followed by further dilution of the coffee by cold water at a ratio of 1:3.

Study on the Application of Indigenous Pigmented Rice for Garaedduk Adapted with Mechanically Impacting Technology (기계적 충격기술이 토종 유색미 함유 가래떡 노화지연에 미치는 효과)

  • The present study was performed to expand the application of mechanically impacting technology reported in our previous studies. Several parameters were analyzed to research Korean indigenous pigmented rice. The proximate composition of Korean indigenous rice was the following; 12.0-13.4% moisture, 74.1-77.3% carbohydrate, 6.8-8.7% crude protein, 1.68-3.25% crude lipid, 0.78-1.38% crude ash, with a 6.6-7.07 pH value. The phenolic compound content of red rice was twice that of black rice. Moreover, antioxidative activity was estimated by DPPH radical-scavenging activity (IC50 = 0.15-1.04 mg/mL). Anthocyanin and chlorophyll in each pigmented rice type were detected by spectrophotometer. Garaedduk samples were prepared with non-waxy rice, salt, 24% water, and/or 15% indigenous pigmented rice(red, green and black, w/w of total swelling non-waxy rice) and adapted with or without mechanically impacting technology. Hardness, as a main texture profile parameter of Garaedduk was maintained up to 3 days under storage at $20^{\circ}C$. The results suggest that Korean indigenous pigmented rice can be applied for functional Garaedduk adapted with mechanically impacting technology.

  • 한국천문연구원은 과학기술위성 3호의 주탑재체인 다목적 적외선영상시스템(Multipurpose Infra-Red Imaging System, MIRIS)을 개발하고 있다. 이 연구개발 사업은 2007년 교육과학기술부의 과학위성 3호 사업 주탑재체 공모를 통하여 10여개의 후보 탑재체 제안서 중에서 최종적으로 채택되었고, 2011년 발사를 목표로, 3년 동안의 연구개발 기간을 거쳐 현재 비행모델 (FM, Flight Model) 개발이 진행 중이다. MIRIS는 한국천문연구원이 개발하여 2003년 발사에 성공한 과학위성 1호 주탑재체인 원자외선 영상분광기 (FIMS, Far ultra-violet IMaging Spectroscope)에 이어 국내에서 자체 개발되는 두 번째 우주망원경이다. MIRIS는 우주공간에서 0.9~2 micron 사이 적외선 영역의 파쉔 알파 방출선 (Paschen Alpha Emiision Line)과 광대역 I, H 파장영역을 관측할 예정이다. 주요 과학임무로는 아직까지 국제 천문학계에서 잘 알려지지 않은 우리은하 내부에 분포한 고온 플라즈마 (Warm Ionized Medium, WIM)의 기원 연구와 아울러 우리은하 성간난류(Interstellar Turbulence)의 특성 및 적외선 우주배경복사의 (Cosmic Infrared Background; CIB) 거대구조 등을 관측연구할 예정이다. 특히 MIRIS는 저온상태 (절대온도 77K, 약 $-200^{\circ}C$)에서 우주공간 관측을 수행할 예정이므로, 국내에서는 연구기반이 취약한 극저온 광학계 및 기계부 설계기술, 극저온 냉각기술 및 열해석 설계기술과 적외선 센서기술 및 자료처리 기술 등 관련기술을 개발하고 있으며 이러한 기반기술을 바탕으로, 아직까지 국내에서 시도된 바 없는 적외선우주망원경 개발을 통하여, 우리나라의 관련 우주기술 분야의 기초원천 기술로서 크게 활용될 것으로 기대하고 있다.

Electrochemical Properties of LiNi1-yMyO2(M=Zn2+, Al3+, and Ti4+) Synthesized by Milling and Solid-State Reaction Method (기계적 혼합과 고상법에 의해 합성한 LiNi1-yMyO2(M=Zn2+, Al3+, and Ti4+)의 전기화학적 특성)

  • By calcining at $750^{\circ}C$ for 30 h in $O_2$ stream after milling, $LiNi_{1-y}M_yO_2(M=Zn^{2+},\;Al^{3+}$, and $Ti^{4+}$, y = 0.005, 0.01, 0.025, 0.05, and 0.1) were synthesized and their electrochemical properties were investigated. All the samples had R3m structure. $LiNi_{1-y}Zn_yO_2$ (y = 0.025, 0.05, and 0.1) contained ZnO anuor $Li_2ZnO_2$ as impurities. Among the samples substituted with the same element, the samples with relatively large value of $I_{003}/I_{104}$ and the smallest R-factor had the largest first discharge capacity and good cycling performance. $LiNi_{0.975}A1_{0.025}O_2$ had the largest first discharge capacity (172.5 mAh/g) and good cycling performance (about $89.4\%$ of the first discharge capacity at the 20th cycle). This sample had the largest value of $I_{003}/I_{104}$ and the smallest R-factor among all the samples. In addition, the particles of this sample were finer and their size was more homogeneous than the other samples. $LiNi_{0.95}A1_{0.05}O_2$ had relatively large first discharge capacity 150.4 mAh/g and good cycling performance.

Methyl Donor Status Influences DNMT Expression and Global DNA Methylation in Cervical Cancer Cells

  • Background: Methyl donor status influences DNA stability and DNA methylation although little is known about effects on DNA methyltransferases. The aim of this study was to determine whether methyl-donor status influences DNA methyltransferase (Dnmt) gene expression in cervical cancer cells, and if so, whether there are associated effects on global DNA methylation. Materials and Methods: The human cervical cancer cell line, C4-II, was grown in complete medium and medium depleted of folate (F-M+) and folate and methionine (F-M-). Growth rate, intracellular folate, intracellular methionine and homocysteine in the extracellular medium were measured to validate the cancer cell model of methyl donor depletion. Dnmt expression was measured by qRT-PCR using relative quantification and global DNA methylation was measured using a flow cytometric method. Results: Intracellular folate and methionine concentrations were significantly reduced after growth in depleted media. Growth rate was also reduced in response to methyl donor depletion. Extracellular homocysteine was raised compared with controls, indicating disturbance to the methyl cycle. Combined folate and methionine depletion led to a significant down-regulation of Dnmt3a and Dnmt3b; this was associated with an 18% reduction in global DNA methylation compared with controls. Effects of folate and methionine depletion on Dnmt3a and 3b expression were reversed by transferring depleted cells to complete medium. Conclusions: Methyl donor status can evidently influence expression of Dnmts in cervical cancer cells, which is associated with DNA global hypomethylation. Effects on Dnmt expression are reversible, suggesting reversible modulating effects of dietary methyl donor intake on gene expression, which may be relevant for cancer progression.
