The Clinical Results of Giant Cell Tumor in Tendon Sheath (건초에 생긴 거대세포종에 대한 임상적 연구)

  • Giant cell tumor of tendon sheath is a slow-growing, unilateral and solitary lesion that is most commonly seen in the digits of the hand and knee, but occasionally occurs in the hips, ankles, toes and wrists. The lesion is asymptomatic or mildly painful, especially if it is diffuse and located in a major joint. The concepts concerning about the pathogenesis of these lesion have undergone constant revision, which include neoplastic process, inflammation and lipid metabolism. Authors analysized 20 patients with giant cell tumor of tendon sheath about the etiology, clinical findings, pathologic findings and treatment results. The results were as follows: 1. 13 cases were females (65%) and 7 cases were males (35%), and the range of age was from 9 years old to 60 years old. 2. The hand was most frequently involved site in which 14 cases (70%) were included, and the foot was involved in 5 cases (25%). 3. Solitary lesions were 15 cases and multiple lesions were 5 cases. 4. Radiographically, in 3 cases bony erosion was seen. 5. All cases were treated by surgical excision and presented no recurrence. In conclusion, the giant cell tumor of tendon sheath, which has been considered to be benign tumorous conditions appeared to be necessary for complete surgical excision to prevent recurrence.

In Vivo Tumor Cell Distribution of Antibody-Endostatin Fusion Protein for Tumor-Specific Targeting and Pharmacokinetics (암세포 표적지향화를 위한 항체-엔도스타틴 융합단백질의 체내동태 및 종양으로의 이행성)

  • A novel antitumor agent, antibody-endostatin fusion protein $(anti-HER2/neu\;IgG3C_H3-Endostatin,\;AEFP)$ formed by genetic engineering procedure from antibody (Ab) which specifically targets to tumor cells ad angiogenesis inhibitor, endostatin (Endo) that has excellent antitumor effect, minimizes the toxicity of normal cells and selectively kills only tumor cells. The purpose of this study is to evaluate the phamacokinetic parameters and to analyze the localization of AEFP. After an intravenous injection of $150\;{\mu}l\;(5\;{\mu}Ci)\;[^{125}I]Ab,\;[^{125}I]AEFP$ to mice, blood was collected though retroorbital plexus from 15 min to 2880 min. Following the jugular vein injetion of $150\;{\mu}l\;(10\;{\mu}Ci)\;[^{125}I]Endo$, blood was collected by the use of carotid artery cannulation from 0.25 min to 30 min. Consequently, Endo was very rapidly removed from plasma compartment within 30 min. On the other hand, AEFP similar to Ab was slowly cleared from plasma. Also, Endo was metabolized about 40% within 30 min. However, AEFP was shown to metabolize less than 10% within 2880 min. The organ distribution of Endo was in order kidney, lung, spleen. Both Ab and AEFP were localized in order spleen, kidney, liver. Futhermore the tumor/blood distribution ratio of AEFP at 96 hours after injection is about 20 times higher than it of Endo at one hour after injection. In conclusion, these studies demonstrate that the anti-cancer or suppression of angiogenesis effect of Endo may be improved by the use of AEFP because the longer half life and stability of AEFP is able to selectively target antigens expressed on tumors.

Bioavailability of plant pigment phytochemicals in Angelica keiskei in older adults: A pilot absorption kinetic study

  • BACKGROUND/OBJECTIVES: Angelica keiskei is a green leafy vegetable rich in plant pigment phytochemicals such as flavonoids and carotenoids. This study examined bioavailability of flavonoids and carotenoids in Angelica keiskei and the alteration of the antioxidant performance in vivo. SUBJECTS AND MATERIALS: Absorption kinetics of phytochemicals in Angelica keiskei were determined in healthy older adults (> 60 y, n = 5) and subjects with metabolic syndrome (n = 5). Subjects consumed 5 g dry Angelica keiskei powder encapsulated in gelatin capsules with a low flavonoid and carotenoid liquid meal. Plasma samples were collected at baseline, 0.5, 1, 2, 3, 4, 5, 6, 7, and 8 h. Samples were analyzed for flavonoids and carotenoids using HPLC systems with electrochemical and UV detection, respectively, and for total antioxidant performance by fluorometry. RESULTS: After ingestion of Angelica keiskei increases in plasma quercetin concentrations were observed at 1-3 and 6-8 hr in the healthy group and at all time points in the metabolic syndrome group compared to baseline (P < 0.05). Plasma lutein concentrations were significantly elevated in both the healthy and metabolic syndrome groups at 8 hr (P < 0.05). Significant increases in total antioxidant performance were also observed in both the healthy and the metabolic syndrome groups compared to baseline (P < 0.05). CONCLUSIONS: Findings of this study clearly demonstrate the bioavailability of phytonutrients of Angelica keiskei and their ability to increase antioxidant status in humans.

Interaction of physical trainings and coffee intakes in fuel utilization during exercise in rats

  • This study investigates the impact of exercises, coffee intakes, and physical trainings on fuel utilization in rats. Ninety-six rats were fed a control diet with either water (C) or coffee (CF; 0.12 g freeze-dried instant coffee/100 g body weight/d). Additionally, the animals go through physical training (TC and TCF) or no training (NTC and NTCF) for 4 weeks. For physical training, animals have to exercise on treadmills for 30 minutes (5 d per week, $15^{\circ}$ incline, 0.5-0.8 km/h). At the end of week 4, the animals in each group were subdivided into three exercise groups: before exercise (BE), during exercise (DE), and after exercise (AE). The DE rats exercised on treadmills for 1 hour immediately before being sacrificed. Hemoglobin, hematocrit, glucose, glycogen, protein, triglyceride (TG), and free fatty acid (FFA) levels in the plasma, liver, and skeletal muscle of the rats were compared accordingly. Organ weights were also measured. Coffee-training interaction had a significant impact on heart weight, visceral fat, hemoglobin, hematocrit, liver glycogen in DE and AE, and liver triglyceride in DE and AE. Exercise (meaning exercised on a treadmill for 1 hour immediately before being sacrificed) training interaction was significant in liver glycogen, muscle glycogen in control diet and control diet with coffee, FFA and muscle TG levels at control diet with coffee group. Exercise-coffee interactions significantly influenced the FFA with no training groups. Exercise-coffee-training interaction significantly effects on FFA, Liver TG and Muscle TG. Coffee intakes can increase lipolysis during exercising but coffee consumptions delay the recovery of liver glycogen levels in trained rats after exercising. Coffee can be an effective ergogenic aid during exercise for physically trained rats.

Luteolin attenuates migration and invasion of lung cancer cells via suppressing focal adhesion kinase and non-receptor tyrosine kinase signaling pathway

  • BACKGROUND/OBJECTIVES: Non-small cell lung cancer is mostly recognized among other types of lung cancer with a poor prognosis by cause of chemotherapeutic resistance and increased metastasis. Luteolin has been found to decrease cell metastasis. However, its underlying mechanisms remain unresolved. The objective of this study was to examine the effect (and its mechanism) of luteolin on the migration and invasion of human non-small cell lung cancer A549 cells. MATERIALS/METHODS: Cell viability was investigated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. Wound healing and transwell assays were evaluated to assess migration and invasion, respectively. Western blot analysis and immunofluorescence were further performed to investigate the role of luteolin and its mechanisms of action. RESULTS: Administration with up to 40 μM luteolin showed no cytotoxic activity on lung cancer A549 cells or non-cancer MRC-5 cells. Additionally, luteolin at 20-40 μM significantly suppressed A549 cells' migration, invasion, and the formation of filopodia in a concentration-dependent manner at 24 h. This is similar with western blot analysis, which revealed diminished the phosphorylated focal adhesion kinase (pFAK), phosphorylated non-receptor tyrosine kinase (pSrc), Ras-related C3 botulinum toxin substrate 1 (Rac1), cell division control protein 42 (Cdc42), and Ras homolog gene family member A (RhoA) expression levels. CONCLUSIONS: Overall, our data indicate that luteolin plays a role in controlling lung cancer cells' migration and invasion via Src/FAK and its downstream Rac1, Cdc42, and RhoA pathways. Luteolin might be considered a promising candidate for suppressing invasion and metastasis of lung cancer cells.

A Comparison Study of Heat Loss Characteristics in Monofilament and Multifilament Superconducting Coils Driven with AC Currents (단일필라멘트와 다중필라멘트 초전도 코일의 교류 전류에 의한 발열 특성 비교 평가)

  • Since superconducting wires have no resistance, electromagnets based on the superconducting wires produce no resistive heating with DC current as long as the current does not exceed the critical current of the wire. However, unlike resistive wires, superconducting wires exhibit AC heat loss. Embedding fine superconducting filaments inside copper matrix can reduce this AC loss to an acceptable level and opens the way to AC-capable superconducting coils. Here, we introduce an easy and accurate method to measure AC heat loss from sample superconducting coils by measuring changes in the rate of gas helium outflow from the liquid helium dewar in which the sample coil is placed. This method provides accurate information on total heat loss of a superconducting coil without any size limit, as long as the coil can fit inside the liquid helium dewar. With this method, we have evaluated AC heat loss of two superconducting solenoids, a 180-turn solid NbTi wire with 0.127 mm diameter (NbTi coil) and a 100-turn filamented wire with 1.4 mm diameter where 7 NbTi filaments were embedded in a copper matrix with copper to NbTi ratio of 6.7:1 (NbTi-Cu coil). Both coils were wound on 15 mm-diameter G-10 epoxy tubes. The AC heat losses of the NbTi and NbTi-Cu coils were evaluated as $53{\pm}4.7\;{\mu}W/A^2Hzcm^3$ and $0.67{\pm}0.16\;{\mu}W/A^2Hzcm^3$, respectively.

Optimum Combination of Pickup Coil Type and Magnetically Shielded Room for Maximum SNR to Measure Biomagnetism (생체신호 측정을 위한 최대의 신호 대 잡음비를 가지는 검출코일의 형태 와 자기차폐실의 최적 조합)

  • We have investigated the optimum combination of the environmental noise condition and type of SQUID pickup coil in order to obtain maximum signal-to-noise ratio (SNR). The measurement probe consists of 1st order gradiometer with pickup coils of 100 mm, 70 mm, and 50 mm baseline length, a 2nd order gradiometer with 50 mm baseline, and a magnetometer. The pickup coils are fabricated by winding Nb wire on a bobbin with 200 mm diameter. Noise and heart signal of a healthy male were measured by various SQUID sensors with different types of pickup coils in various magnetically shielded rooms (MSR), and compared to each other. The shielding factors were found to be 43 dB, 35 dB and 25 dB at 0.1 Hz for MSR-AS, MSR-BS, MSR-CS, respectively. White noises were $3.5\;fT/Hz^{1/2}$, $4.5\;fT/Hz^{1/2}$ and $3\;fT/Hz^{1/2}$ for the 1st order gradiometers, the 2nd order gradiometers, and magnetometer for all MSRs. SNR of the magnetometer was up to 56 dB in MSR-AS, while the 1st order axial gradiometer with 70 mm baseline length was up to 54 dB in MSR-BS. The 2nd order axial gradiometer with 50 mm baseline length of pickup coil was found to be up to 40 dB in MSR-CS.

Natural Dyeing of Chitosan Crossinked Cotton Fabrics - Gromwell - (키토산 가교 처리된 면직물의 천연염색에 관한 연구 - 자초를 중심으로 -)

  • The purpose of this study is to investigate the dyeing property of gromwell on modified cotton fabric by chitosan. Modified cotton fabrics were manufactured by crosslinking agent epichlorohydrin in the presence of chitosan. Gromwell colorants were extracted with methanol. Modified cotton fabrics dyed using gromwell were post-mordanted using Al, Fe and Cu. The dyeability (K/S) and color factors (L, a, b, ${\Delta}E$ and h) of modified cotton fabrics were measured by computer color matching. Additionally the fastness to washing and light were also investigated. The dye-uptake of modified cotton fabrics increased with the dyeing time. The saturated dyeing time was about 10minutes at $50^{\circ}C$. The dyeability (K/S) was remarkably increased with increasing content of chitosan because of having a amine group of chitosan. Modified cotton fabrics were dyed yellowish red by non and Fe mordanting, blueish red by Al and Cu mordanting, respectively. The washing fastness of non, Al, Fe and Cu mordant in the presence and absence of chitosan were increased $1{\rightarrow}2$, $3{\rightarrow}4$, $4{\rightarrow}4-5$ and $4{\rightarrow}4-5$ respectively. And light fastness of non, Al, Fe and Cu mordant in the presence and absence of chitosan were increased $1{\rightarrow}1-2$, $1{\rightarrow}1-2$, $1.2{\rightarrow}2.3$ and $1-2{\rightarrow}2$ respectively.

HCl Removal from Coal-derived Syngas by the Solid Sorbents (고체 흡수제를 이용한 석탄 합성가스 중 HCl 정제)

  • 석탄 합성가스 중에는 $H_2S$, HCl, $NH_3$와 같은 불순물이 포함되어 있다. 이러한 가스들은 오염가스 배출과 관련한 환경기준 준수와 터빈과 같은 설비의 보호를 위해 제거되어야 한다. 석탄 합성가스 중 HCl 농도는 탄종에 따라 다르기는 하지만 많게는 1000 ppmv 수준까지 존재한다. 합성가스를 이용하여 발전을 하는 경우 가스터빈 보호를 위해 HCl은 <3 ppmv 이하로 정제되어야 하고, 합성가스를 연료전지에 사용하고자 하는 경우에는 HCl을 <0.5 ppmv 수준까지, 화학원료로 사용하고자 하는 경우에는 <10 ppbv 수준까지 정제하여야 한다. 또한 HCl은 고온 탈황공정에 사용되는 흡수제의 활성에도 장기적으로 부정적인 영향을 주기 때문에 고온에서 HCl을 정제할 수 있는 흡수제가 필요하다. 본 연구에서는 알칼리금속을 활성물질로 사용하여 분무건조법으로 제조한 HCl 흡수제에 대해 물성 및 HCl과의 반응성을 살펴보았다. $300-500^{\circ}C$ 영역에서 K-계 및 Na-계 흡수제에 대해 고정층반응기에서 HCl 가스를 함유한 모사 합성가스를 이용하여 상압 조건에서 Cl 흡수능을 측정한 결과 15wt% 이상의 흡수능을 나타내었으며 반응온도가 높을수록 흡수능이 증가함을 알 수 있었다. XRD 분석을 통하여 Cl은 K 및 Na와 반응하여 KCl과 NaCl을 형성하면서 흡수됨을 알 수 있었다. 20 bar 조건에서 실험한 결과에서도 동일한 경향의 반응성을 나타내었으며 반응온도가 낮을수록 흡수능은 감소하지만 Cl을 더 낮은 농도로 정제할 수 있었다. 본 실험에 사용된 Na 및 K계 흡수제는 모두 연소 후 배가스 중 $CO_2$를 제거하기 위한 흡수제로 사용되는 고체 흡수제이다. 석탄화력발전소 배가스에 연계되어 $CO_2$ 회수실험에 사용되었던 사용 후 $CO_2$ 흡수제에 대해 HCl 흡수 실험을 수행한 결과에서도 우수한 HCl 제거 성능을 보여 주었다. 이로부터, 폐 $CO_2$ 흡수제의 HCl 흡수제로서의 활용가능성을 확인 하였다.

Peroxynitrite Scavenging Mechanism of Psoralea corylifolia (보골지(補骨脂)의 Peroxynitrite 제거 기전)

  • Objective : Peroxynitrite $(ONOO^-)$, formed from the reaction of $O_2^-$ and NO, is a cytotoxic species that can oxidize several cellular components such as proteins, lipids and DNA. It has been implicated in the aging process and age-related disease such as Alzheimer's disease, rheumatoid arthritis, cancer and atherosclerosis. The aim of this study was to investigate scavenging activities of $ONOO^-$ and its precursors. NO and $O_2^-$ and its scavenging mechanism using fluorescent probes, DCFDA, DAF-2 and DHR 123.. Methods : Psoralea corylifolia was crushed. The crushed Psoralea corylifolia was extracted 3 times, each time with 3 volumes of methyl alcohol at $60^{\circ}C$ for 24 h. The extract was filtered and evaporated under reduced pressured using a rotary evaporator to yield 16g. This was done to investigate scavenging activities of $ONOO^-$, NO, $O_2^-$ and its scavenging mechanism using fluorescent probes, DCFDA, DAF-2 and DHR 123. Results : After Psoralea corylifolia was added authentic $ONOO^-,\;{\cdot}\;O_2^-$ and NO was markedly scavenged. Also, $ONOO^-$ induced by $O_2^-$ and NO (these derived from SIN-1) was inhibited. The data showed a decrease in $ONOO^-$ mediated nitration of tyrosine through electron donation after Psoralea corylifolia was added. Data showed a dose-dependent correlation with inhibition of nitration of bovine serum albumin induced by $ONOO^-$, Furtheremore, LPS-induced ROS and RNS generation was blocked. Conclusions: These results suggest potential for use of Psoralea corylifolia as an effective $ONOO^-$ scavenger to counter the aging process and age-related diseases.
