Seasonal Variation of Phytoplankton and Environmental Factors in Kyungpo Lake (경포호의 식물플랑크톤과 환경요인의 계절적 변동)

  • In order to elucidate chatracteristics of water quality and seasonal dynamics of phytoplankton community in Kyungpo lake, the weekly variation of algal population and 11 environmental factors were investigated from March 1998 to February 1999. A total 121 species of phytoplankton belonged to 57 genera, 4 genera 7 species of the Euglenophyceae, 1 genera 2 species of the Chrysophyceae, 2 genera 3 species of the Cryptophyceae, 1 genera 1 species of the Dinophyceae, 6 genera 9 species of the Cyanophyceae, 28 genera 78 species of the Bacillario-phyceae and 15 genera 21 species of the Chlorophyceae, was identified. Cryptomonas sp., Chamydomonas sp., Nitzschia frustulum, Oscillatoria sp., Lyngbya sp, and Dictyosphaerium pulchellum occurred successively as dominant phytoplankton species from March 1998 to February 1999. Density of phytoplankton cell number showed the peak in early April when Chlamydomoas sp. was dominated. The diversity index of phytoplankton community showed low values in the dominating period of Cryptomonas sp. and Chlamydomonas sp. The concentration of chlorophyll a fluctuated between 1.09 and 107.7$\mu\textrm{g}$ㆍ1$^{-1}$. Water temperature, salinity, DO, conduc tivity and SS were ranged 0.5-27.8$^{\circ}C$, 2.8-25.2 ppt, 4.62-19.21 mgO$_2$ㆍ1$^{-1}$, 5.33-34.76 mS/cm and 0.017-0.184 mgㆍ1$^{-1}$, respectively. Annual means of Secchi disc transparency was 36cm and thai of pH value was 8.5. The concentrations of PO$_4$-P was ranged 0.0012-0.077 mgㆍ1$^{-1}$ , and showed high value from August to October. The concentrations of NO$_2$-N, NO$_3$-N and NH$_4$-N was ranged 0.0011-0.1 mgㆍ1$^{-1}$, 0.074-3.33mgㆍ1$^{-1}$ and 0.0024-0.259 mgㆍ1$^{-1}$, respectively. Among environmental factors, water temperature was negatively correlated with DO, salinity and conductivity, DO was positively correlated with salinity and conductivity. Chlorophyll a, one of the indicies of eutropication in lake, was negatively correlated with NH$_4$-N. The cell density of Cryptomonas sp., which was the most dominant species in this lake, was negatively correlated with transparency. Effects of Ojawhan on the ethanol-induced erectile dysfunction in rats (오자환(五子丸)이 Ethanol로 발기부전을 유도한 흰쥐의 성기능 개선에 미치는 영향)

  • Objectives : Ojawhan was formulated to contain various natural products known to cure erectile dysfunction. This study was aimed to investigate the effects of Ojawhan on the nitric oxide synthase(NOS) activity, nitrite level, antioxidation and erectile responses in rat's corpus cavernosum penis. Methods : Ojawhan was washed, dried in the shade and crushed. The crushed Ojawhan ,was extracted 3 times, each time with 3 volumes of methyl alcohol at $60^{\circ}C$ for 24 h. The extract was filtered and evaporated under a reduced pressure using a rotary evaporator to yield 62g. Ojawhan extract oral-administered 100 mg per 1 kg of body weight for 30 days. First, samples were treated with Ojawhan, then ethanol-treated rats and L-N-Nitroarginine methyl ester(L-NAME) treated rats were put with the samples. Result : The level of urethral lipid peroxide in the ethanol-Ojawhan double administered rats was decreased as low as in the normal group, while the one in the ethanol-treated group was increased. The urethral NOS activity, the level of urethral nitrite, the level of testosterone and the erectile response to cavernous nerve stimulation in the ethanol-Ojawhan double administered rats were increased as high as in the normal group while the one in the ethanol-treated group was decreased. The electile response to cavernous nerve stimulation and the level of nitrite in L-NAME ($10^{-4}$)-treated rats was restored by the administration of Ojawhan as high as in the normal group. Conclusions : Ojawhan was effective in restoring the ethanol-induced or L-NAME-induced erectile dysfunction in rats.

  • In recent years with development of immuno-electrophoresis, more acurate analysis of the serum protein became possible. However, there is few reports in the literature which investigated the changes of the immunoglobulin compared with electrophoretically fractioned serum thyroglobulin in the patients with various thyroid diseases. The purpose of this report is to investigate the changes of thyroglobulin in various thyroid diseases by the method of immuno-electrophoresis and to compare the results with.serum protein fractionated by the method of agar-gel micro-electrophoresis. Materials and Methods: Sera from 9 patients with diffuse toxic goiter, 2 nodular nontoxic goiter, 2 thyroiditis, 3 hypothy, roidism, 1 thyroid cancer, 7 cystic degeneration of the thyroid gland, and 10 normal subject were taken. All cases were confirmed by various laboratory thyroid function tests and thyroid needle biopsy. Immuno-electrophoretic analysis of the serum were performed by Scheidegger's modified micro-immuno-electrophoretic method. The antiserum was obtained from the Travenol Laboratories International, Hyland Products Division and was rabbit anti-human thyroglobulin. Microscope slide agar-gel electrophoresis for serum protein fractionation was performed at $4^{\circ}C$ using veronal buffer, pH 8.6 and ionic strength 0.05, with 54 volts and 2.8 mA for 60 minutes. The fractionated slide was stained with 0.1% thiazine red. The results were as follows: 1) Increase of immune-globulin macroglobulin (IgM), alphaglobulin, and immune-globulin A (IgA) by 95.8%, 100%, 29.2% respectively was found in the serum from various thyroid diseases. 2) Thyroglobulin fraction was found to be increased in 50%, no change in 41.7%, and no line in 8.3% with all of the various goiter patients. On the other hand, 10 normal control group showed only 2 cases of increase, 5 cases of no change and 3 cases of no line.

  • The high fuel flexibility of gas turbine power system has boosted their use in a wide variety of applications. Recently, the demand for biogas generated from the digestion of organic wastes and sewage waste water as a fuel for gas turbines has increased. We investigated the performance of high pressure biogas compression system and operating conditions for supplying biogas. The total flow per minute of biogas from food waste water digestion tank is $54Nm^3$. The main type of biogas compression system is the reciprocating system and screw type system. The target of biogas mechanical data is the as belows; inlet pressure 0.045bar, supplying biogas temperature is $30{\sim}60^{\circ}C$, and final pressure is above the 25 bar. Also, inlet conditions of biogas consist of CH4 48.5%~83%, $H_2S$ Max. 500ppm, $NH_3$ Max. 1,500ppm and Siloxane 2.7~4.6ppm. The boosting Blower system raises a pressure from 0.045bar to 1bar before main compressor. The main system lay out of reciprocating consisits of compressor driver, filter, cooling system, blowdown vessel, control system and ESD(Emergency Shut Down) system. And an enclosure package needs to be installed for reducing noise up to 75dB. The system driver is the electronic motor of explosion proof type. Forthe compressor system reliable operation, the cleaning system something like particulate filter needs to be set up in the inlet of compressor and Coalescing Filter in the outlet of compressor. Particulate Filter has to be removed above $10{\mu}m$ size of the particles in biogas. The coalescing filter(Micofine Borosilicate Glass Fibers Filter treated phenol acid) also removes moisture and oil of above $0.3{\mu}m$ to be involved in high pressure biogas up to 90%~98%.

  • Three types of proteases (MEF-1, MEF-2 and MEF-3) were purified from the egg cases of Ten-odera sinensis using ammonium sulfate fractionation, gel filtration on Bio-Gel P-60 and affinity chromatography on DEAE Affi-Gel blue gel. The proteases were assessed homogeneous by SDS-polyacrylamide gel electrophoresis and have molecular weight of 31,500, 32,900 and 35,600 Da, respectively. The N-terminal regions of the primary structure were compared and they were found to be different each other. MEFs readily digested the $A\alpha$ - and B$\beta$-chains of fibrinogen and more slowly the ${\gamma}$-chain. The action of the enzymes resulted in extensive hydrolysis of fibrinogen and fibrin, releasing a variety of fibrinopeptides. MEF-1 was inactivated by Cu$^{2+}$ and Zn$^{2+}$ and inhibited by PMSF and chymostatin. MEF-2 was inhibited by PMSF, TLCK. soybean trypsin inhibitor. MEF-3 was only inhibited by PMSF and chymostatin. Antiplasmin was not sensitive to MEF-1 but antithrombin III inhibited the enzymatic activity qf MEF-1. MEF-2 specifically bound to anti plasmin Among the chromogenic protease substrates, the most sensitive one to the hydrolysis of MEFs was benzoyl-Phe-Val-Arg-p-nitroanilide with maximal activity at pH 7.0 and 3$0^{\circ}C$. MEF-1 preferentially cleaved the oxidized B-chain of insulin between Leu15 and Tyr16. In contrast, MEF-2 specifically cleaved the peptide bond between Arg23 and Gly24. D-dimer concentrations increased on incubation of cross-linked fibrin with MEF-1, indicating the enzyme has a strong fibrinolytic activity.ity.

Clinical experience of open heart surgery: 50 cases (개심술 치험 50례)

  • Fifty cases of open heart surgery were done in the Department of Thoracic and Cardiovascular Surgery, Busan National University Hospital during 16 months from July, 1981 to October, 1982. The clinical data were analyzed and summerized as follows. 1. There were 34 cases (68%) of congenital anomalies and 16 cases (32%) of acquired heart diseases. Among the congenital cases, 27 were acyanotic and 7 were cyanotic. All of the acquired heat diseases, 16 cases were valvular diseases and they had valvular replacement surgery. 2. The age distribution of the congenital anomalies ranged from 6 to 27 years with mean age of 14.2 years, and the acquired heart diseases from 18 to 44 years mean age of 27.5 years. The difference of sex distribution was no significance. 3. The clinical minifestations in acyanotic congenital anomalies were exertional dyspnea (81.5%), recurrent respiratory infection (55.6%) and palpitation (22.2%), and in cyanotic congenital anomalies were exertional dyspnea (100%), syncope(57.1%) and growth retardation(57.1%), and in acquired heart diseases were dyspnea(100%), edema (62.5%) and general weakness (62.5%) 4. During the cardiopulmonary bypass, mild to moderate core cooling was performed and added topical cooling for more accurate myocardial preservation. 5. Two kinds of cardioplegic solution used in our institute were Bretschneider solution for the first 7 cases and mixed Harmann's solution 1 L with glucose 5gm, potassium chloride 26 mEq and sodium bicarbonate 24 mEq, making 376 mosmol/L and pH 8.3 at $25^{\circ}C$, for the rest 43 cases. 6. Various kinds of postoperative complications occurred in 14 cases (28%) and showed overall mortality 12%. The mortality along with each disease was 7.4% in congenital acyanotic cases, 42.9% in congenital cyanotic cases and 6.3% in acquired valvular diseases. 7. Pre-and postoperative diagnostic incompatibility was seen in 6 cases (12%). 8. The artificial valves used in the replacement surgery were lonescu-Shiley bovine xenograft in 6 cases and Carpentier-Edwards porcine xenograft in 10 cases.

Properties of Gul Jeotgal (Oyster Jeotgal) Prepared with Different Types of Salt and Bacillus subtilis JS2 as Starter

  • Gul (oyster) jeotgals (GJs) were prepared using different types of salt (23%, w/v): purified salt, solar salt aged for 3 years, and bamboo salt crystalized 3 times. One set of GJs was fermented with Bacillus subtilis JS2 ($10^6CFU/g$), while the other GJ set was fermented without starter. During fermentation for 24 weeks at $15^{\circ}C$, the starter GJs showed 10-fold higher bacilli counts than the no-starter GJs, where the maximum bacilli count was $8{\times}10^3CFU/g$. All 28 bacilli strains isolated from the 6-week GJs were identified as B. subtilis by using a RAPD-PCR, indicating that some of the B. subtilis JS2 cells remained viable. Lactic acid bacteria (LAB) and yeasts were present at low levels, $10^1-10^2CFU/g$. LAB with protease activities isolated from 10-week samples were identified as Enterococcus species. The isolates obtained at 16 weeks were all Staphylococcus species. The GJs with bamboo salt showed higher pH and lower titratable acidity (TA) values than the other GJs due to the strong alkalinity of bamboo salt. The amino-type nitrogen in the GJs increased slowly during the fermentation. At 24 weeks, the GJs with purified salt showed the highest amino-type nitrogen (412-430 mg%), followed by the GJs with solar salt (397-406 mg%) and GJs with bamboo salt (264-276 mg%). Meanwhile, the GJs with bamboo salt showed the highest ammonia-type N (63.67 mg%), followed by the GJs with purified salt (49 mg%) and solar salt (48 mg%).

An Annealing Control Primer (ACP) System Used for the Isolation and Identification of Copper-Induced Genes in Alfalfa Leaves

  • Copper (Cu) is a necessary microelement for plants. However, high concentrations of Cu are toxic to plants that change the regulation of several stress-induced proteins. In this study, an annealing control primer (ACP) based approach was used to identify differentially expressed Cu-induced genes in alfalfa leaves. Two-week-old alfalfa plants (Medicago sativa L.) were exposed to Cu for 6 h. Total RNAs were isolated from treated and control leaves followed by ACP-based PCR technique. Using GeneFishing ACPs, we obtained several genes those expression levels were induced by Cu. Finally, we identified several genes including UDP-glucuronic acid decarboxylase, transmembrane protein, small heat shock protein, C-type cytochrome biogenesis protein, mitochondrial 2-oxoglutarate, and trans-2,3-enoyl-CoA reductase in alfalfa leaves. These identified genes have putative functions in cellular processes such as cell wall structural rearrangements, transduction, stress tolerance, heme transport, carbon and nitrogen assimilation, and lipid biosynthesis. Response of Cu-induced genes and their identification in alfalfa would be useful for molecular breeding to improve alfalfa with tolerance to heavy metals.

Effects of Albizzia Julibrissin on Chronic Ethanol-treated Erectile Dysfunction in Rats (Ethanol 에 의해 발기부전을 유도한 흰쥐의 성기능 개선에 미치는 합환피(合歡皮)의 영향)

  • Objectives : Albizzia Julibrissin was formulated to contain various natural products known to cure erectile dysfunction. This study was aimed to investigate the effects of Albizzia Julibrissin on the nitric oxide synthase (NOS) activity, nitrite level, antioxidation and erectile responses induced by ethanol in corpus cavernosum penis of rats. Methods : The crushed Albizzia Julibrissin was extracted 3 times, each time with 3 volumes of methyl alcohol at $60^{\circ}C$ for 24 h. The extract was filtered and evaporated under a reduced pressure using a rotary evaporator to yield 45.3 g. Albizzia Julibrissin extract was oral-administered 100 mg per 1 kg of body weight for 20 days, while the normal group was administered only with a saline. The efficacy of Albizzia Julibrissin against erectile function was examined as described in the text. Results : The level of urethral NOS activity and nitrite were increased by Albizzia Julibrissin. The level of lipid peroxide was decreased by Albizzia Julibrissin. The level of urethral lipid peroxide in the ethanol-Albizzia Julibrissin double administered rats was decreased as low as in the norma! group, while the one in the ethanol-treated group was increased. The level of urethral nitrite, NOS activity, glutathione and serum testosterone in the ethanol-Albizzia Julibrissin double administered rats were as high as in the normal group, while the one in the ethanol-treated group was decreased. The erectile response to cavernous nerve stimulation in the ethanol-Albizzia Julibrissin double administered rats increased as high as in the normal group while the one in the ethanol-treated group decreased. Conclusions : Albizzia Julibrissin was shown to be effective for the treatment of erectile dysfunction induced by ethanol in rats.

The Effect of Bacillus-based Feed Additive on Growth Performance, Nutrient Digestibility, Fecal Gas Emission, and Pen Cleanup Characteristics of Growing-finishing Pigs

  • Bacillus-based feed additive was evaluated for its efficacy on growth performance, nutrient digestibility, fecal gas emission, and the consumption of time and amount of water for cleaning the pen of growing finishing pigs. A total of 120 growing pigs ($23.59{\pm}1.41kg$) were used in a 16-wk feeding trial. Pigs were randomly distributed into 1 of 2 treatments on the basis of body weight and sex. There were 12 replicate pens per treatment, with 5 pigs (3 barrows and 2 gilts) per pen. Dietary treatments were CON which was basal diet, and T1 which was CON+62.5 ppm microbial feed additive that provided $1.47{\times}10^8cfu$ of Bacillus organisms per gram of supplement. During the weeks 0 to 6, average daily gain (ADG) in T1 treatment was higher (p<0.05) than CON, but no improvement in average daily feed intake (ADFI) and feed efficiency (G:F) was noted. During 6 to 16 weeks, no difference (p>0.05) was noted in growth performance. However, ADG was improved (p<0.05) and overall ADFI tended (p = 0.06) to improve in T1 compared with CON. At week 6, the co-efficient of apparent total tract digestibility (CATTD) of dry matter (DM) nitrogen (N) was increased (p<0.05) in T1 compared with CON. Fecal $NH_3$ emission was decreased (p<0.05) in T1 compared with CON, at the end of 6th and 15th weeks. The time and water consumed for washing the pens were decreased (p<0.05) in T1 compared with CON. In conclusion, supplementation with Bacillus-based feed additive could improve the overall growth performances, increase the CATTD of DM and decrease the fecal $NH_3$ content and the time and water consumed in washing the pens for growing-finishing pigs.
