새조개의 효율적인 종묘생산을 위한 생물학적 기초자료를 얻고자 난발생 및 유생사육에 미치는 환경요인으로서 수온의 영향에 대하여 연구하였다. 실험에 사용된 수정란은 산란유도하여 6개의 수온별 실험구 (14, 17, 20, 23, 26 및 $29^{\circ}C$)에 수용하여 발생과정을 관찰하였다. 유생사육의 각 단계에 이르기까지의 수온 (T, $^{\circ}C$) 에 따른 발생속도 (1/h, 시간) 는 수온이 높을수록 빨랐으며 그 관계식은 다음과 같다. 4-cell : 1/h = 0.0067T-0.0879 ($r^2$ = 0.8760) 8-cell : 1/h = 0.0128T-0.1794 ($r^2$ = 0.7589) M (상실기) : 1/h = 0.0234T-0.3274 ($r^2$ = 0.9587) B (포배기) : 1/h = 0.0306T-0.4276 ($r^2$ = 0.9236) T (담륜자기) : 1/h = 0.0442T-0.6224 ($r^2$ = 0.9121) V (D형유생) : 1/h = 0.0864T-1.2850 ($r^2$ = 0.8586) 유생사육시 수온과 난발생 속도와의 관계에서 추정된 D형 유생까지 난발생의 생물학적 영도 및 적산수온은 평균 $0.1^{\circ}C$ 및 $597.3^{\circ}C$였다. D형유생까지 가장 적합한 수온은 $23^{\circ}C$였으며, 발생은 20시간으로 나타났다.

Elsholtzia fruticosa (EF) is present in tropical regions throughout South Asian countries as well as the Himalayas. Although it has been used as a traditional medicine to treat digestive, respiratory, and inflammatory issues, its effect on preadipocyte differentiation is unknown. In this study, we examined the effects of a methanol extract prepared from EF on the differentiation of 3T3-L1 preadipocytes. Cell differentiation was assessed by microscopic observation and oil-red O staining. The expression of adipogenic and lipogenic genes, including PPARγ and C/EBPα, was measured by western blot analysis and quantitative real-time polymerase chain reaction (qRT-PCR), to provide insight into adipogenesis and lipogenesis mechanisms. The results indicated that EF promotes the differentiation of 3T3-L1 preadipocytes, with elevated lipid accumulation occurring in a concentration-dependent manner without apparent cytotoxicity. EF enhances the expression of adipogenic and lipogenic genes, including PPARγ, FABP4, adiponectin, and FAS, at the mRNA and protein levels. The effect of EF was more pronounced during the early and middle stages of 3T3-L1 cell differentiation. Treatment with EF decreased C/EBP homologous protein (CHOP) mRNA and protein levels, while increasing C/EBPα and PPARγ expression. Treatment with EF resulted in the upregulation of cyclin E and CDK2 gene expression within 24 h, followed by a decrease at 48 h, demonstrating the early-stage impact of EF. A concomitant increase in cyclin-D1 levels was observed compared with untreated cells, indicating that EF modulates lipogenic and adipogenic genes through intricate mechanisms involving CHOP and cell cycle pathways. In summary, EF induces the differentiation of 3T3-L1 preadipocytes by increasing the expression of adipogenic and lipogenic genes, possibly through CHOP and cell cycle-dependent mechanisms.
Previously, we reported that vitamin C facilitates the CpG demethylation of Foxp3 enhancer in $CD4^+Foxp3^+$ regulatory T cells (Tregs) by enhancing the activity of a DNA demethylase ten-eleven-translocation (Tet). However, it is not clear whether vitamin C affects other helper T cell lineages like T helper type 17 (Th17) cells which are related with Tregs. Here, we show that the expression of interleukin-17A (IL17) increases with the treatment of vitamin C but not with other antioxidants. Interestingly, the upregulation of IL17 was not accompanied by DNA demethylation in Il17 promoter and was independent of Tet enzymes. Rather, vitamin C reduced the trimethylation of histone H3 lysine 9 (H3K9me3) in the regulatory elements of the Il17 locus, and the effects of vitamin C were abrogated by knockdown of jumonji-C domain-containing protein 2 (jmjd2). These results suggest that vitamin C can affect the expression of IL17 by modulating the histone demethylase activity.
The purpose of this study was to investigate the effect of lactic acid bacteria (LAB) cell wall extract on the proliferation and cytokine production of immune cells to select suitable probiotics for space food. Ten strains of LAB (Lactobacillus bulgaricus, L. paracasei, L. casei, L. acidophilus, L. plantarum, L. delbruekii, Lactococcus lactis, Streptococcus thermophilus, Bifidobacterium breve, and Pedicoccus pentosaceus) were sub-cultured and further cultured for 3 d to reach 7-10 Log colony-forming units (CFU)/mL prior to cell wall extractions. All LAB cell wall extracts failed to inhibit the proliferation of BALB/c mouse splenocytes or mesenteric lymphocytes. Most LAB cell wall extracts except those of L. plantarum and L. delbrueckii induced the proliferation of both immune cells at tested concentrations. In addition, the production of TH1 cytokine (IFN-γ) rather than that of TH2 cytokine (IL-4) was enhanced by LAB cell wall extracts. Of ten LAB extracts, four (from L. acidophilus, L. bulgaricus, L. casei, and S. thermophiles) promoted both cell proliferating and TH1 cytokine production. These results suggested that these LAB could be used as probiotics to maintain immunity and homeostasis for astronauts in extreme space environment and for general people in normal life.
These experiments were conducted to investigate the effects of Glycyrrhizae Radix extract(GR) on histamine synthesis, lymphocyte blastogenesis in C57BL/6J mice splenocytes, IL-1 production, $Ca^{2+}$ uptake by macrophage-like P388D$_{1}$ cells and plaque forming cell assay against SRBC. Histamine contents, lymphocyte blastogenesis, IL-1 activity, $Ca^{2+}$ uptake and plaque forming cell were determined by enzyme isotope method, [$^{3}$H]-thymidine incorporation, C3H/HeJ mouse thymocytes proliferation, the addition of 5 $\mu$Ci/ml $^{45}Ca^{2+}$ to P388D$_{1}$ cell suspension and assay to sheep red blood cell, respectively. Cytotoxicity, which was expressed as 50% mortality, was occurred by the addition of GR(10$^{-3}$g/ml). Histamine production in mouse spleen cell culture was significantly increased by 48 hour incubation added 0.25$\mu\textrm{g}$/ml of Con A. Con A-dependent T-lymphocyte proliferation was also enhanced by the addition of 0.25 $\mu\textrm{g}$/ml of Con A. GR depressed histamine contents at 10$^{-9}$~10$^{-4}$g/ml. and Con A (0.25 $\mu\textrm{g}$/ml) dependent T-lymphocyte proliferation at 10$^{-5}$~10$^{-4}$g/ml. IL-1 activity was significantly decreased by 10$^{-8}$~10$^{-4}$g/ml of GR. $Ca^{2+}$ uptake was not changed by GR, but antibody production markedly increased at 10.0~50.0 mg/kg of GR. From the above results, it is suggested that GR have immuno-regulatory action; GR decreased cell-mediated immune response and increased antibody production by B lymphocyte at high doses.
In order to know the inhibitory effect of magnesium carbonate(MgCO3) on cytotoxicity, DNA damage, mutagenicity, and cell transforming ability of nickel subsulfide, the inhibition of cell proliferation, DNA-protein crosslinks formation (DPC), HGPRT point mutation, and cell transformation were evaluated. Nickel subsulfide(Ni3S2) and magnesium carbonate as insoluble compounds were used for this study. BALB/3T3 cell, CHO-K1 cell, and C3H10T1/2 cell were used in this experiment. Exposure concentration of nickel subsulfide was 1 $\mu\textrm{g}$/ml. The concentrations of magnesium carbonate in this study were 0.6 $\mu\textrm{g}$/ml, 1.2 $\mu\textrm{g}$/ml, 2.4 $\mu\textrm{g}$/ml and the molar ratio of magnesium to nickel when exposed simultanously were 0.5, 1.0 and 2.0 respectively. The results were as follows; 1. Magnesium carbonate reduced the inhibitory effect of nickel subsulfide on cell proliferation. 2. Magnesium carbonate also reduced the effect of nickel subsulfide on DNA-protein crosslinks formation. 3. HGPRT point mutagenicity of nickel subsulfide was reduced when magnesium carbonate treated simultaneously. 4. Magnesium carbonate reduced cell transforming ability of nickel subsulfide. Conclusively, nickel subsulfide showed cytotoxicity, cell transforming ability, and mutagenicity strongly and magnesium carbonate may have protective roles in these nickel effects.
Objectives : Hemistepta lyrata Bunge (Bunge) is a wild herb that has been used for managing fever and wound in Korean Traditional Medicine. The present study explored the effects of H. lyrata extract on liver X receptor (LXR) α-dependent lipogenic genes in hepatocyte-derived cells. Methods : After HepG2 cells or Huh7 cells were pre-treated with 1-10 ㎍/mL of H. lyrata extract or its fractionated extract for 0.5 h, the cells were subsequently exposed to LXR ligand for 6-24 h. Cell viability, LXR response element (LXRE)-driven luciferase activity, sterol regulatory element binding protein-response element (SREBP-RE)-driven luciferase activity, SREBP-1c expression, and mRNA levels of LXRα and its-dependent target genes were determined. In addition, LC-MS/MS analysis was conducted to explore major compounds in H. lyrata-chloroform fractionated extract #4 (HL-CF4). Results : Of various H. lyrata extracts tested, chloroform extract and its fractionated extract #4, HL-CF4, significantly decreased T0901317-mediated SREBP-1c expression. In addition, HL-CF4 significantly reduced LXRE atransactivation and LXRα mRNA expression without any cytotoxicity. Moreover, HL-CF4 prevented the SREBP-RE-driven luciferase activity and mRNA levels of fatty acid synthase and stearoyl-CoA desaturase-1 induced by T0901317. Results from LC-MS/MS analysis at positive/negative mode indicated that HL-CF4 contained several compounds showing m/z 197.1176 (C11H17O3), 693.2913/227.1069 (C38H45O12/C15H15O2), 203.1797 (C15H23), 181.1225 (C11H17O2), 591.2957 (C35H43O8), 379.1040 (C18H19O9), 409.1509 (C20H25O9), 309.1348 (C16H21O6), 391.1404 (C20H23O8), and 669.2924/389.1248 (C36H45O12/C20H21O8). Conclusion : Based on its inhibition of the LXRα-dependent signaling pathway, H. lyrata chloroform extract and HL-CF4 have prophylactic potentials for managing non-alcoholic fatty liver.
In view of the growing medicinal importance of pyrazole and its derivatives, the single crystal X-ray diffraction study was carried out for the potential active 2-Benzyl-3-[3-(4-bromo-phenyl)-1-phenyl-1H-pyrazol-4yl]-4,6-dioxo-5-phenyl-octahydro-pyrrolo[3,4-C]pyrrole-1-carboxylic acid ethyl ester ($C_{37}H_{31}BrN_4O_4$, H2O). In the title compound are two molecules exist in the asymmetric unit. It crystallizes in the monoclinic space group $P{\hat{i}}$ with unit cell dimension $a=13.361(18){\AA}$, $b=13.424(17){\AA}$ and $c=21.649(2){\AA}$ [${\alpha}=80.745(9)^{\circ}$, ${\beta}=79.770(10)^{\circ}$ and ${\gamma}=60.788(6)^{\circ}$]. The pyrazole ring adopts planar conformation. The sum of the bond angles at nitrogen atom of the pyrazole ring indicates the $Sp^2$ hybridized state. The crystal structure is stabilized by intramolecular C-H...O hydrogen bond interaction.

Exposure to cold may affect growth performance in accordance with the metabolic and immunological activities of animals. We evaluated whether ambient temperature affects growth performance, blood metabolites, and immune cell populations in Korean cattle. Eighteen Korean cattle steers with a mean age of 10 months and a mean weight of 277 kg were used. All steers were fed a growing stage-concentrate diet at a rate of 1.5% of body weight and Timothy hay ad libitum for 8 weeks. Experimental period 1 (P1) was for four weeks from March 7 to April 3 and period 2 (P2) was four weeks from April 4 to May 1. Mean ($8.7^{\circ}C$) and minimum ($1.0^{\circ}C$) indoor ambient temperatures during P1 were lower (p<0.001) than those ($13.0^{\circ}C$ and $6.2^{\circ}C$, respectively) during P2. Daily dry matter feed intake in both the concentrate diet and forage groups was higher (p<0.001) during P2 than P1. Average daily weight gain was higher (p<0.001) during P2 (1.38 kg/d) than P1 (1.13 kg/d). Feed efficiency during P2 was higher (p = 0.015) than P1. Blood was collected three times; on March 7, April 4, and May 2. Nonesterified fatty acids (NEFA) were higher on March 7 than April 4 and May 2. Blood cortisol, glucose, and triglyceride concentrations did not differ among months. Blood CD4+, CD8+, and CD4+CD25+ T cell percentages were higher, while CD8+CD25+ T cell percentage was lower, during the colder month of March than during May, suggesting that ambient temperature affects blood T cell populations. In conclusion, colder ambient temperature decreased growth and feed efficiency in Korean cattle steers. The higher circulating NEFA concentrations observed in March compared to April suggest that lipolysis may occur at colder ambient temperatures to generate heat and maintain body temperature, resulting in lower feed efficiency in March.

In order to establish effective procedures for chromosome manipulation in Haliotis gigantea and H. discus, which are of enormous aquacultural potential, temperature-dependent measures of mitotic intervals ($\tau$$_{0}$) were determined. Mitotic intervals ($\tau$$_{0}$) in these abalone were determined by averaging the duration of the first and third embryonic divisions over a range of temperatures from 8 to 26$^{\circ}C$. The relationships of each mitotic interval at two cell ($\tau$$_{I}$), four cell ($\tau$$_{II}$ ), eight cell ($\tau$$_{III}$), sixteen cell ($\tau$$_{IV}$ ) and $\tau$$_{0}$, to temperature (T in $^{\circ}C$) in H. gigantea were log $\tau$$_{I}$ : 176.1-28.3T, log $\tau$$_{II}$ : 199.5-12.4T, log $\tau$$_{III}$ = 236.2-12.2T, log $\tau$$_{IV}$ = 269.3-14.lT and log $\tau$$_{0}$ : 83.1-32.8, respectively. The relationships of each mitotic interval at $\tau$$_{I}$, $\tau$$_{II}$ , $\tau$$_{III}$, $\tau$$_{IV}$ and $\tau$$_{0}$, to temperature in H. discus were log $\tau$$_{I}$ = 104.9-13.8T, log $\tau$$_{II}$ : 138.3-10.5T, $\tau$$_{III}$ : 172.4-10.2T, log $\tau$$_{IV}$ : 211.3-12.2T and log $\tau$$_{0}$=85.6-33.3T, respectively. 1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
