A Consideration on the Lactation Persistency Evaluation in Korean Holstein Dairy Cattle (국내 홀스타인 젖소의 비유지속성 평가에 대한 고찰)

  • The characteristics of lactation persistency was investigated for new evaluation trait using 4,366,900 milk yield records from 436,690 heads of Korean Holstein dairy cattle. The average lactation persistencies of first parity, second parity and over third parity were 97.5%, 95.1% and 94.6%, respectively and there was a trend that after the peak yield, lactation persistency decreased collectively. The average days of peak milk yields after calving was about 50 days, but only 33.2% of cows reached peak yields at 36~66 days (second test day). Also, there was a difference between the milk yield of cows which reached peak yields at first test day by lactation days and that of cows which reached peak yields at second to fourth test day. The estimates of heritabilty and repeatability for mean lactation persistency were 0.16 and 0.35, respectively. The genetic correlation between cumulative lactation persistency from third to tenth test day and that from third to seventh test day was 0.91 and while it increased in later test day, it decreased sharply in earlier test day. The breeding value correlations of Data II and III for Data I were 0.80 and 0.72, respectively, while the rank correlations were 0.78 and 0.71, respectively. Based on the results, the breeding value and rank correlations decreased when more data were added.

Marine Bacteriological Quality and Dynamics in Tongyeong Coastal Area, Gyung-nam, Korea (통영연근해역의 해양세균학적 수질 및 동태에 관한 연구)

  • A bacteriological study of sea water and oyster in Tongyeong coastal area was conducted to evaluate sanitary conditions of the bay and compliance of waters with the recommended bacteriological criteria fur the designated area of shellfish cultivation. The Samples were collected at 5 zone, 34 sampling stations(Fig. 1) established once a month from September 1997 to August 1998. During the study period, temperature ranged from 6.9 to 23.6$^{\circ}C$, transparency ranged from 2.6 to 6.2 m, chemical oxygen demand ranged from 1.35 to 1.82 mg/ι, dissolved oxy-gen ranged from 5.0 to 9.9 mg/ι, dissolved nitrogen ranged from 1.60 to 8.17 $\mu\textrm{g}$-at/ι, phosphate ranged from 0.14 to 1.21 $\mu\textrm{g}$-at/ι, Chlorophyll-a ranged from 2.03 to 69.9 mg/㎥, respectively. The coliform group and fecal coliform MPN's of sea water were ranged from <3.0~1,600 and <3.0~540, respectively. The coliform group and fecal coliform MPN's of oysters were ranged from <18~16,000 and <18~2,200, respectively. The viable cell counts in oyster ranged from $1.5\times$10$^2$to 8.2$\times$10$^3$. The coliform stoup, fecal coliform, classification of coliform group with IMViC reactions and pathogenic vibrios were analyzed. 437 strains that were obtained from Tongyeoung coastal area seawater samples represented E. coli group 47.5%, C. freundii group 14.8%, K. aerogenes 10.9%, unknown 26.8%, respectively. During the study period, infectious bacteria such as Vibrio ohoEerae, Salmonella sp. and Shigella sp. were not detected from the samples, but detection ratios of Vibrio parahaemolyticus and Vibrio vulnificus were 12~21% in summer months.

Effect of Maternal Undernutrition during Late Pregnancy on Growth and Development of Ovine Fetal Visceral Organs

  • This study investigated the effect of maternal undernutrition during late pregnancy on the growth and development of ovine fetal visceral organs. One hundred Mongolian ewes were mated at a synchronized oestrus and divided into three groups and offered 0.175 MJ ME $kgw^{-0.75}\;d^{-1}$ (Restricted Group1; RG1), 0.33 MJ ME $kgw^{-0.75}\;d^{-1}$ (Restricted Group2; RG2) and ad libitum access to feed (Control Group; CG) during late pregnancy (90 days). Selected animals in each group were slaughtered immediately at d 90 of pregnancy and after parturition (neonatal lambs), and major visceral organs were removed and weighed separately. The results indicated that the weights of lung (p<0.01), spleen (p<0.01), heart (p<0.05), liver (p<0.05) and abomasum (p<0.01) in RG1 were significantly lighter than those of CG. For RG2, only the weights of the lung (p<0.05) and spleen (p<0.01) were significantly lighter than those of CG; when expressed as a percentage of body weight, significance was retained in the spleen (p<0.01) for both restricted groups, but the percentage of brain in RG1 was significantly higher than that in CG (p<0.01). For lung and spleen, the amount of DNA was significantly lower (p<0.01) in both groups of restricted neonatal lambs compared to CG; however, there was a significant difference only between RG1 and CG for protein: DNA ratio (p<0.01). The DNA content of kidney, abomasum and jejunum were decreased (p<0.05) in RG1 neonatal lambs, but protein: DNA ratio in the liver was decreased compared with that of CG (p<0.05). The plane of maternal undernutrition during late pregnancy had a significant effect on the growth and development of fetal visceral organs, which altered ontogeny of fetal organ growth and development. These perturbations in fetal visceral development may have significant implications on postnatal growth and adult health.

SEROTYPES AND ANTIBIOTIC SUSCEPTIBILITIES OF STREPTOCOCCUS MUTANS ISOLATED FROM DENTAL PLAQUES OF CHILDREN (치아우식아동과 그의 모친(母親)의 치태(齒苔)에서 분리(分離)한 Streptococcus mutans의 혈청형(血淸型) 분포(分布) 및 약제내성(藥劑耐性)에 관(關)한 연구(硏究))

  • A total of 141 strains of Streptococcus mutans were isolated from dental plaques of 153 subjects. Eighty-two children with caries-experience, 18 caries-free children and their mothers were participated. All isolates were examined for their serotypes by immunodiffusion method with 7 reference sera and their antibiotic susceptibilities to 7 antibiotics by agar dilution method using 7 kinds of antibiotics, and were compared their results by caries experience (DMFT ratio) and by intrafamilial levels. 1. Isolation rate of Streptococcus mutans were greater in samples of carious teeth than those of sound teeth, and in mothers than in children. 2. Multiple serotypes of Streptococcus mutans were occasionally found in a single samples. 3. Of the total 141 isolates (83 isolates from children, and 58 from their mothers), type c isolates were most prevalent (63.8%). Type d,e and f were found, comprizing 14.9%, 10.6% and 6.4% respectively. Serotype g, a and untypable strain were also found but far lower frequencies (2.8-0.7%), and type b was detected. 4. These results suggest that there are no significant correlation among the distribution of serotypes, antibiotic susceptibilities, caries experience and intrafamilial relationships. 5. Most of isolates were susceptible to chloramphenicol (100%), penicillin (95.7%), ampicillin (94.3%), and gentamicin (92.2%), but about one-third isolates were resistant to cephaloridine (27%), streptomycin (33.3%) and kanamycin (47.6%), resulting that 91 strains (64.5%) among 141 isolates were resistant to one or more drugs used. 6. Of the 91 resistant strains, 20 different resistant patterns were observed, and the most frequently encountered patterns were KM, SM and CE.

Porcine lymphotropic herpesvirus (Gammaherpesvirinae) DNA in free-living wild boars (Sus scrofa Linnaeus, 1758) in Brazil

  • Background: Suid gammaherpesvirus 3, 4, and 5 (porcine lymphotropic herpesvirus - PLHV-1, -2, and -3) are viruses that infect domestic and feral pigs. Objectives: This study examined the presence of PLHV DNA in biological samples from free-living wild boars circulating in a Brazilian geographical region with a high density of commercial domestic pigs. Methods: Lung samples of 50 free-living wild boars were collected by exotic wildlife controller agents between 2017 and 2019 in the state of Paraná, southern Brazil. Lung and spleen fragments were obtained from six fetuses collected by hysterectomy post mortem from a pregnant sow. A polymerase chain reaction (PCR) assay using consensus primers (pan-herpesviruses) was performed to detect PLHV DNA. The samples showing positive results for PLHV DNA were submitted to single-round PCR assays with the specific primers for identifying PLHV-1 (213-S/215-As), PLHV-2 (208-S/212-As), and PLHV-3 (886s/886As). The specificity of the species-specific PCR products was assessed by nucleotide sequencing of the amplicons. Results: Forty-eight (96%) of the 50 lung samples analyzed were positive for PLHV by PCR using pan-herpesvirus primers. In 33 (68.75%) of the positive samples, at least two PLHV species were identified simultaneously. The DNA of PLHV-1, -2, and -3 was found in free-living wild boars of all ages, but not in the fetuses, even though they were from a sow that tested positive for all three viruses. Conclusion: These viruses are endemic to the population of feral pigs in the Brazilian region evaluated, as well as in domesticated pigs.

Effect of Lactobacillus buchneri 40788 and Buffered Propionic Acid on Preservation and Nutritive Value of Alfalfa and Timothy High-moisture Hay

  • The effects of Lactobacillus buchneri 40788 and buffered propionic acid on preservation, intake and digestibility of alfalfa (Medicago sativa) and timothy (Phleum pratense) hay were investigated. During baling, forages were treated with L. buchneri 40788 (1.2${\times}$10$^6$ CFU/g) as a liquid (LLB) or as a granular preparation (GLB), with buffered propionic acid (10 mL/kg, BPA), or left untreated (control). Triplicate 500 kg round bales of each treatment were put up at two moisture levels for each forage: 17%${\pm}$0.33% and 20%${\pm}$0.30% for timothy and 17%${\pm}$0.20% and 19%${\pm}$0.27% for alfalfa (mean${\pm}$SD). Bales were sampled for chemical and microbiological analyses after 0, 30 and 60 d of storage. Compared to controls, all preservatives reduced (p<0.05) heating of both forages at all moisture levels with the exception of alfalfa baled at 19% moisture. After 60 d of storage, GLB reduced (p<0.05) moulds in 17% timothy hay as compared to other treatments, but at 20% moisture, moulds were reduced in LLB- and BPA-treated timothy as compared to controls. In alfalfa at 17% moisture, total bacteria were lower (p<0.05) in GLB-treated bales than LLB or control bales, but yeast and total bacteria were only reduced in BPA-treated alfalfa at 19% moisture. In situ DM disappearance of timothy (both moisture levels) and alfalfa (19% moisture level) increased (p<0.05) with LLB treatment compared to control. Digestibility of both forages did not differ (p>0.05) among treatments, however, voluntary DM intake of LLB-treated timothy (1.32 kg/d) was 22.3% higher (p<0.05) than control, and 14.1% higher than BPA-treated timothy. Treating timothy and alfalfa hay with L. buchneri 40788 or buffered propionic acid may improve the nutritive value of the hay when baled at 17 to 20% moisture.

Human metapneumovirus infection in Korean children (소아에서 human metapneumovirus 감염)

  • Purpose : Human metapneumovirus(hMPV) is a respiratory viral pathogen that causes a wide spectrum of illnesses, ranging from asymptomatic infection to severe bronchiolitis. The virus has been identified world widely, but so far it has not been published in Korea. Methods : We obtained clinical samples by nasopharyngeal aspiration from 218 children hospitalized due to acute lower respiratory tract infections at Soonchunhyang University Hospital in Cheonan from October, 2004 to April, 2005. We designed specific primers from conserved region of fusion glycoprotein of hMPV. Total RNA was extracted and RT-PCR was performed, and single specific 423 bp product was obtained. The PCR product was confirmed to be fusion glycoprotein RNA by sequencing. Results : We detected hMPV in 15(6.9 percent) of the 218 hospitalized children. The infected children comprised nine boys and six girls; their mean age was 2.8 years(5 mo-12 yrs) and they were diagnosed with pneumonia(60 percent), bronchiolitis(33.3 percent), croup(6.6 percent). The number of cases of detected hMPV in Korea increased dramatically during the period from March to May 2005. Conclusion : hMPV is circulating in Korean children and is associated with respiratory tract infection. Additional studies are required to define the epidemiology and the extent of diseases in the general population caused by hMPV.

Influence of polymer-coated slow-release urea on total tract apparent digestibility, ruminal fermentation and performance of Nellore steers

  • Objective: Two experiments were performed to evaluate the effects of coated slow-release urea on nutrient digestion, ruminal fermentation, nitrogen utilization, blood glucose and urea concentration (Exp 1), and average daily gain (ADG; Exp 2) of steers. Methods: Exp 1: Eight ruminally fistulated steers [$503{\pm}28.5kg$ body weight (BW)] were distributed into a d $4{\times}4$ Latin square design and assigned to treatments: control (CON), feed grade urea (U2), polymer-coated slow-release urea A (SRA2), and polymer-coated slow-release urea B (SRB2). Dietary urea sources were set at 20 g/kg DM. Exp 2: 84 steers ($350.5{\pm}26.5kg$ initial BW) were distributed to treatments: CON, FGU at 10 or 20 g/kg diet DM (U1 and U2, respectively), coated SRA2 at 10 or 20 g/kg diet DM (SRA1 and SRA2, respectively), and coated SRB at 10 or 20 g/kg diet DM (SRB1 and SRB2, respectively). Results: Exp 1: Urea treatments (U2+SRA2+SRB2) decreased (7.4%, p = 0.03) the DM intake and increased (11.4%, p<0.01) crude protein digestibility. Coated slow-release urea (SRA2+-SRB2) showed similar nutrient digestibility compwared to feed grade urea (FGU). However, steers fed SRB2 had higher (p = 0.02) DM digestibility compared to those fed SRA2. Urea sources did not affect ruminal fermentation when compared to CON. Although, coated slow-release urea showed lower (p = 0.01) concentration of $NH_3-N$ (-10.4%) and acetate to propionate ratio than U2. Coated slow-release urea showed lower (p = 0.02) urinary N and blood urea concentration compared to FGU. Exp 2: Urea sources decreased (p = 0.01) the ADG in relation to CON. Animals fed urea sources at 10 g/kg DM showed higher (12.33%, p = 0.01) ADG compared to those fed urea at 20 g/kg DM. Conclusion: Feeding urea decreased the nutrient intake without largely affected the nutrient digestibility. In addition, polymer-coated slow-release urea sources decreased ruminal ammonia concentration and increased ruminal propionate production. Urea at 20 g/kg DM, regardless of source, decreased ADG compared both to CON and diets with urea at 10 g/kg DM.

Study on the Storage of Chestnut (밤 저장(貯藏)에 관(關)한 연구(硏究))

  • A mass production of chestnut necessiates the development of economic long-term storage method. The main objective of this study was to confirm the technical aspect of the chestnut storage method which was developed by two year project and to review the method of commercial application. The chestnut used for the experiments were separated in brine $(5.5{\sim}6.0^{\circ}\:B{\acute{a}}ume)$ into matured and unmatured lots and fumigated with $CS_2$ at a 5 $lb/27\;m^3$ level for $25{\sim}30\;hrs.$ The chestnuts were packed in wooden boxes with sawdust (50% moisture) in the ratio of 1 : 1 by volume. The boxes were stored in the cold room $(1{\pm}1^{\circ}C,\;85{\sim}95%\;RH)$ and the cellar ($0{\sim}10^{\circ}C$, controlled only by circulating night cool air). The results obtained were as follows: 1. Fully matured chestnut could be successfully preserved $8{\sim}9\;months$ at a l0% decay level in the cold room and $4{\sim}5\;months$ months in cellar. 2. Immatured chestnuts wire inferior to the matured in storage stability. At the maximum storage period, its storage life was two months shorter. 3. The heat transfer equation of piled chestnuts with sawdust can be suggested as $T_{\infty}-T_0=(T_{\infty}-T_0){\cdot}10^{-t/320}$ and j and $f_h$ values were 1 and 320 min, respectively. 4. The chestnuts in the package of storage unit had longer shelf life than naked chestnut during the retail distribution at ambient temperature.

Evaluation of color matching ability according to the color temperature and the experience of practitioner (색 온도 및 술자의 숙련도에 따른 비색 능력 평가 원저)

  • Purpose: The aim of this study is to investigate the effect of different experience level and different light source on shade selection ability comparing prosthodontist group and dental student group under 4,000 K and 5,500 K light. Materials and methods: After color difference of Vitapan 3D-master shade guides was measured, 3 sets of 5 shade tabs were selected with similar value but have different chroma (set a, b, c). Also 3 sets of 5 shade tabs were selected with similar chroma but have different values (set d, e, f). Under 4,000 K and 5,500 K light sources, ten prosthodontists and ten dental students were allowed to match in one set of 5 tabs the same shade tab with the tab which was originally selected in the other set of 5 tabs. Color differences of original tab and matched tab were measured by spectrophotometer and the shade selection ability was evaluated with those data. Evaluation of color difference value was performed in regard to different light conditions and different level of experience, followed by t-test with 95% confidence interval. Results: Color difference values under 4,000 K and 5,500 K light source were $1.62{\pm}2.0$, and $1.33{\pm}1.7$ respectively. In addition, color difference values of prosthodontist group and dental student group were $1.34{\pm}1.7$, and $1.61{\pm}2.0$ respectively. Difference of shade selection ability was not found under either different light sources (P=.398), or different experience level (P=.221). Conclusion: Level of experience did not affect on the shade selection ability when prosthodontists and dental students matched the shades with the same shade tab under the same light source.
