Effects of Cooling Rate and Equilibration Time on the Survival and Development of Frozen-thawed Bovine Immature Oocytes (동결속도와 평형시간이 소 미성숙 난자의 동결 융해후 생존율에 미치는 영향)

  • The present study was undertaken to investigate the effects of cooling rate and equilibration time on the survival, in vitro maturation and development to embryos of frozen-thawed bovine immature oocytes(Germinal Vesicle Stage). The cryoprotectants are used 10% ethylene glycol(EG) as permeating cryoprotectant and 0.05M soc.ose(S) or trehalose(T) as low molecular weight nonpermeating cryoprotectants and 5% ficoll(F) or polyvinylpyrrolidone(PVP) as high molecular weight nonpermeating cryoprotectants. Four freezing solution were uysed in this experiment(EFT: 10% EG + 5% F + 0.05M T, EFS: 10% EG + 5% F + 0.05M S, EPT: 10% EG + 5% P + 0.05M T, EPS: 10% EG + 5% P + 0.05M S). The best equilibration time and freezing solution was 15 min in EPT(83% survival rate of frozen-thawed bovine immature oocytes). When frozen-thawed bovine oocytes were cultured following IVM and IVF, there was no significant difference in cleavage and development rates among the EFT, EFS, EPT and EPS solutions. When 9 blastocysts derived from frozen bovine oocytes were transferred to 6 recipients, two recipients were pregnant. And one was aborted at 45 days of pregnancy and the other had a stillbirth.

Late Quaternary Sequence Stratigraphy in Kyeonggi Bay, Mid-eastern Yellow Sea (황해 중동부 경기만의 후기 제4기 순차층서 연구)

  • The Yellow Sea has sensitively responded to high-amplitude sea-level fluctuations during the late Quaternary. The repeated inundation and exposure have produced distinct transgression-regression successions with extensive exposure surfaces in Kyeonggi Bay. The late Quaternary strata consist of four seismic stratigraphic units, considered as depositional sequences (DS-1, DS-2, DS-3, and DS-4). DS-1 was interpreted as ridge-forming sediments of tidal-flat and estuarine channel-fill facies, formed during the Holocene highstand. DS-2 consists of shallow-marine facies in offshore area, which was formed during the regression of Marine Isotope Stage (MIS)-3 period. DS-3 comprises the lower transgressive facies and the upper highstand tidal-flat facies in proximal ridges and forced regression facies in distal ridges and offshore area. The lowermost DS-4 rests on acoustic basement rocks, considered as the shallow-marine and shelf deposits formed before the MIS-6 lowstand. This study suggests six depositional stages. During the first stage-A, MIS-6 lowstand, the Yellow Sea shelf was subaerially exposed with intensive fluvial incision and weathering. The subsequent rapid and high amplitude rise of sea level in stage-B until the MIS-5e highstand produced transgressive deposits in the lowermost part of the MIS-5 sequence, and the successive regression during the MIS-5d to -5a and the MIS-4 lowstand formed the upperpart of the MIS-5 sequence in stage-C. During the stage-D, from the MIS-4 lowstand to MIS-3c highstand period, the transgressive MIS-3 sequence formed in a subtidal environment characterized by repetitive fluvial incision and channel-fill deposition in exposed area. The subsequent sea-level fall culminating the last glacial maximum (Stage-E) made shallow-marine regressive deposits of MIS-3 sequence in offshore distal area, whereas it formed fluvial channel-fills and floodplain deposits in the proximal area. After the last glacial maximum, the overall Yellow Sea shelf was inundated by the Holocene transgression and highstand (Stage-F), forming the Holocene transgressive shelf sands and tidal ridges.

Study on the Freezing of Bovine Embryos II. Bovine embryos survival after freezing and thawing (우수정란의 동결보존에 관한 연구 II. 동결보존후 융해난자의 생존성)

  • This experiment was carried out to investigate the morphology of bovine embryos thawed after deep freezing at -196$^{\circ}C$ and the development of frozen-thawed embryos after in vitro culture in Ham's F-10 medium with 10% NBCS. The results obtained were summarized as follows: 1. The propotion of embryos which a, pp.ared mophologically normal was averaged 77.5% (79/102). 2. The morphologically normal rate of frozen-thawed blastocyst (78.6%) was higher than that of morula (76.7%), but there was no significant difference. 3. Normal development was observed in 20 of 68 embryos cultured for 24-72hr in medium and overall survival rate was 29.4%. 4. Survival rate fo blastocyst (33.3%) was higher than that of morula (25.7%).

Convergence Study between Helicopter Parenting, Critical Thinking Disposition, and Learning Competence of Nursing Students (간호 대학생 부모의 헬리콥터 양육과 간호대학생의 비판적 사고 성향, 학습역량의 융합연구)

  • This study was conducted to identify a relationship between Helicopter Parenting, Critical thinking disposition, and Learning Competences of Nursing students. This was a descriptive analyzed study, data were collected from April 3 to 14, 2017, study subjects were 208 nursing students in nursing colleges located C and I city, using self-reported questionnaires. The results of the study showed the mean scores of Helicopter parenting and critical thinking were $73.94{\pm}13.88$, $113.30{\pm}12.33$. And learning motivation, cognition, behavior were consisted in learning competence, learning cognition (F=3.62, p=.014) and learning behavior (F=4.07, p=.008) were statistically significant differences in school score. Helicopter parenting and critical thinking were a significant negative relationship (r=-.14, p=.043), according to critical thinking of learning cognition (r=.43, p<.001) and behavior (r=.42, p<.001) were a significant positive relationship. Based on the results, they were taken a baseline data for developing a Parent-Parenting Education Program to enhance on critical thinking and learning competence of nursing students and a Clinical Nursing Empowerment Program.

Bioconversion of Ginsenosides from Red Ginseng Extract Using Candida allociferrii JNO301 Isolated from Meju

  • Red ginseng (Panax ginseng), a Korean traditional medicinal plant, contains a variety of ginsenosides as major functional components. It is necessary to remove sugar moieties from the major ginsenosides, which have a lower absorption rate into the intestine, to obtain the aglycone form. To screen for microorganisms showing bioconversion activity for ginsenosides from red ginseng, 50 yeast strains were isolated from Korean traditional meju (a starter culture made with soybean and wheat flour for the fermentation of soybean paste). Twenty strains in which a black zone formed around the colony on esculin-yeast malt agar plates were screened first, and among them 5 strains having high ${\beta}$-glucosidase activity on p-nitrophenyl-${\beta}$-D-glucopyranoside as a substrate were then selected. Strain JNO301 was finally chosen as a bioconverting strain in this study on the basis of its high bioconversion activity for red ginseng extract as determined by thin-layer chromatography (TLC) analysis. The selected bioconversion strain was identified as Candida allociferrii JNO301 based on the nucleotide sequence analysis of the 18S rRNA gene. The optimum temperature and pH for the cell growth were $20{\sim}30^{\circ}C$ and pH 5~8, respectively. TLC analysis confirmed that C. allociferrii JNO301 converted ginsenoside Rb1 into Rd and then into F2, Rb2 into compound O, Rc into compound Mc1, and Rf into Rh1. Quantitative analysis using high-performance liquid chromatography showed that bioconversion of red ginseng extract resulted in an increase of 2.73, 3.32, 33.87, 16, and 5.48 fold in the concentration of Rd, F2, compound O, compound Mc1, and Rh1, respectively.

Enhancement of the Ionic Conductivity and Mechanical Strength of Micro-porous Separator by Uni-axial Drawing

  • A new porous separator based on poly(vinyl chloride) (PVC)/poly(vinylidene fluoride-co-hexafluoro-propylene) (P(VdF-co-HFP)/poly(methyl methacrylate) (PMMA) was prepared by a phase inversion method. To enhance mechanical property, the membrane was stretched uniaxially at high temperature. Tensile strength and ionic conductivity were measured for various draw ratios. The tensile strength and ionic conductivity were increased with increasing draw ratio. The tensile strength of the separator reached 52MPa after stretching to draw ratio of 5, and the ionic conductivity of the separator was increased from $1.9Xs10^{-4}S/cm\;to\;4.6X10^{-4}S/cm\;at\;25^{\circ}C$. The stretched separator immersed in liquid electrolyte was electrochemically stable up to 4.7 V. The cell based on the stretched separator was maintained at about 99% of the initial discharge capacity after 10th cycle operation at 0.2C rate.

Preparation of Calcium Sulfate α-Hemihydrate from FGD Gypum in the Autoclave (가압반응기를 이용한 배연탈황석고로부터 α형 반수석고의 생성)

  • This study was performed to investigate the formation process of calcium sulfate ${\alpha}$-hemihydrate from FGD gypsum produced at thermal power plant burning bituminous coal. The experimental results showed that calcium sulfate $\alpha$-hemihydrate with a large aspect ratio was produced in the temperature range of $120^{\circ}C$ and $140^{\circ}C$ in the absence of additives through dissolution-recrystallization mechanism. It was also observed that addition of Na-succinate as a catalyst changed crystal shape from acicular to prismatic, resulting in decreased water/powder ratio down to 33%. Optimum concentration of Na-succinate was 20mM. It was confirmed that the optimum moulding pressure and moisture content of moulded body from FGD gypsum were $30kg_f/cm^2$ and between 10% and 15% respectively, which prevent moulded body from collapsing and maximize the capillary effect by given pore volume while autoclaving.

A Comparative Analysis of Basal Body Temperature to Ultrasound, as a Method of Ovulation Detection in Induced Ovulatory Menstrual Cycles (배란유도주기에 따른 초음파검사와 기초체온표의 비교분석)

  • Four points on the basal body temperature (B.B.T.) curve was correlated with the estimated time of ovulation, as determined by serial ultrasound in 50 induced menstrual cycles from 22 subjects. The time of ovulation was estimated by measuring the maximal diameter of follicles and observing the morphologic changes within the ovary from follicle to corpus luteum. The results were as following; 1. The diameter of the follicle measured at the day before disappearance was 21.1 mm on an average (S.D.: 2.14). The average follicular growth for 4 days before ovulation was measured at a rate of 2.8 mm/day, and rapid growth of follicle was observed 3.1 mm/day at the day before. 2. The changes associated with rupture of the follicles were the followings, in order of frequency; decrease in size(94%), disappearance of follicles(64%), fluid in the Cul-de-Sac(26%) and increased internal echoes(16%). 3. Only 20 of 50 cycles, exhibited a BBT dip and correlated with the estimated time of ovulation by ultrasound in 2 of which cases(10%). BBT nadir, 30 of 50 cycles, correlated in 5(16.7%). The first day of hyperthermic plateau(FDHP) and BBT coverline was exhibited in all cycles, correlated in 41(82%) and 35(70%) cases. 4. The relationship between the diameter of dominant dominant follicle, measured by ultrasound, and the basal body temperature curve were as following. During cycles in which dip was observed on the BBT curve, the follicular diameter were 10.5${\pm}$2.12 mm on 4 days prior to the point (D-4), and 12.5${\pm}$2.12 mm (D-3), 15.5${\pm$2.12 mm (D-2), 17.0${\pm}$1.41 mm (D-1) and 21.5${\pm}$2.12 mm just prior to the dip (D-0). In the nadir; 9.6${\pm}$1.67 mm (N-4), 12.8${\pm}$1.79 mm (N-3), 16.2${\pm}$1.92 mm (N-2), 18.2${\pm}$2.17 mm (N-1) and 21.4${\pm}$2.61 mm (N-0). In the First day of Hyperthemic Plateau (FDHP); 9.8${\pm}$1.36 mm (F-4), 12.4${\pm}$1.41 mm (F-3),15.1${\pm}$1.57 mm (F-2), 18.1${\pm}$1.67 mm (F-1) and 21.2${\pm}$2.25 mm (F-0). In the BBT coverline endopint; 9.9${\pm}$.39 mm (C-4), 12.5 ${\pm}$1.44 mm (C-3), 15.2${\pm}$1.64 mm (C-2), 18.0 ${\pm}$1.69 mm (C-1), and 21.2${\pm}$2.31 mm (C-0). 5. The relationship between the ultrasonographic signs of ovulation and the basal body temperature curve were as following. The BBT dip correlated with the ovulation in 2 cases, which revealed decrease in follicular diameter (100%), fluid pattem in the Cul-de-Sac (1 case, 50%) and complete disappearance of follicle (1 case, 50%). In the nadir (5 cases); the ultrasonographic signs of ovulation were decrease in follicular diameter (5 cases, 100%), fluid pattern in the Cul-de-Sac (1 case, 20%) and complete disappearance of follicle (3 cases, 60%). In the First day of Hyperthermic Plateau (41 cases); decrease in follicular diameter (40 cases, 97.6%), fluid pattern in the Cul-de-Sac (11 cases, 26.8%), appearance of internal echo and thickening of the wall (6 cases, 14.6%) and com plete disappearance of follicle (28 cases, 68.3%). In the BBT coverline endpoint (35 cases); decrease in follicular diameter (33 cases, 94.3%), fluid pattern in the Cul-de Sac (9 cases, 25.7%), appearance of internal echo and thickening of the wall (5 cases 14.3%) and complete disappearance of follicle (20 cases, 57.1%).

Effect of Maternal Under-nutrition during Late Pregnancy on Lamb Birth Weight

  • This study investigated the effects of maternal undernutrition during late pregnancy on lamb birth weight. 45 Mongolian ewes, synchronized for oestrus and then mated, were divided into four groups and offered 0.86 MJME/kgw-0.75d-1 (control group; CG : ad libitum access to feed), $0.44MJME/kgw^{-0.75}d^{-1}$ (Restricted Group 3; R3), $0.33MJME/kgw^{-0.75}d^{-1}$ (Restricted Group 2; R2) and $0.20MJME/kgw^{-0.75}d^{-1}$ (Restricted Group 1; R1) respectively during late pregnancy (90-150 days). During restriction, maternal net body weight loss, insulin and NEFA concentrations and lamb birth weight were measured. The results indicated that loss of maternal body weight in R3, R2 and R1 was 4.42, 7.23, 11.13 kg respectively, which was significantly (p<0.01) higher than that in CG (0.93 kg). Insulin concentrations of the ewes in R1, R2 and R3 were lower and were significantly different (p<0.05) between restricted groups and CG at 124 d of pregnancy. NEFA concentrations in all groups tended to decrease from 90d of gestation to parturition and in R1 were significantly (p<0.05) lower than in CG at 124 d of gestation. Lamb birth weight in R1 was significantly lower than in R2, R3 and CG (p<0.05). In conclusion, with decreasing supply of maternal nutrition, the retardation of fetal growth became worse. When the plane of nutrition was below $0.33MJME/kgw^{-0.75}d^{-1}$, significant effects of maternal undernutrition on lamb birth weight were observed.

Review on The Measurement of Wood Shrinkage (목재수축률 측정에 대한 총설)

  • The goal of this study was to review the wood shrinkage published in Journal of The Korean Wood Science and Technology from 1976 to 2015. Previous studies reported that shrinkage from Larix kaempferi in the tangential, radial, longitudinal directions ranged from 4.21%-9.79%, 2.09%-4.67%, 0.17%-0.33%, respectively. When different drying methods including closed cylinder drying, oven drying, and room temperature drying were used, volumetric shrinkage of Dipterocarpus grandiflorus was different, ranging from 31.6% to 21.0%. With an increment of the drying temperature of $115^{\circ}C$, $120^{\circ}C$, $125^{\circ}C$, the shrinkage of Larix kaempferi did not show a consistent trend. When sample size of Pinus densiflora was increased from $20{\times}20{\times}20mm$ to $100{\times}100{\times}100mm$, the tangential, radial and volumetric shrinkage decreased 2.61%, 1.32%, 0.80%, respectively. When a caliper having a sensitivity of 0.01 mm was used to measure $20mm^3$ specimen from Cryptomeria japonica, the measurement error occurred 1.97% in the radial direction and 35.7% in the longitudinal direction. From the previous studies, wood shrinkage could be influenced by sample size, drying method and measurement technique.
