Synthesis and Micellar Characterization of CBABC Type PLGA-PEO-PPO-PEO-PLGA Pentablock Copolymers

  • Poly(lactic-co-glycolic acid) (PLGA) were grafted to both ends of Pluronic$^{(R)}$ F68 ($(EO)_{75}(PO)_{30}(EO)_{75}$) triblock copolymer to produce poly{(lactic acid)$_m$-co-(glycolic acid)$_n$}-b-poly(ethylene oxide)$_{75}$-b-poly(propylene oxide)$_{30}$-b-poly(ethylene oxide)$_{75}$-b-poly{(lactic acid)$_m$-co-(glycolic acid)$_n$} (PLGA-F68-PLGA) pentablock copolymers. Molecular weights of PLGA blocks were controlled and five kinds of pentablock copolymers with different PLGA block lengths were synthesized using in-situ ring-opening polymerization of D,L-lactide and glycolide with tin(II) 2-ethylhexanoate ($Sn(Oct)_2$) catalyst. PLGA-F68-PLGA pentablock copolymers were characterized by $^1H$- and $^{13}C$-NMR, GPC, and TGA. The numbers (2m, 2n) of repeating units for lactic acid and glycolic acid inside PLGA segments were obtained as (48, 17), (90, 23), (125, 40), (180, 59), and (246, 64), with $^1H$-NMR measurement. From NMR data, the resultant molecular weights were determined in the range of 12,700-29,700, which were similar to those obtained from GPC. Polydispersity index was increased in the range of 1.32-1.91 as the content of PLGA blocks increased. TG and DTG thermograms showed discrete degradation traces for PLGA and F68 blocks, which indicate the weight fractions of PLGA blocks in pentablock copolymers can be calculated by TG profile and it is possible to remove PLGA block selectively. Hydrodynamic radius and radius of gyration of pentablock copolymer micelle were obtained in the range of 46-68 nm and 31-49 nm, respectively, in very dilute (i.e. 0.005 wt %) aqueous solution of THF:$H_2O$ = 10:90 by volume at $25^{\circ}C$.

복숭아명나방과 사과애모무늬잎말이나방에 대한 환경조절열처리를 이용한 사과 수확 후 처리기술 (Post-harvest Treatment on the Peach Pyralid Moth and the Small Tea Tortrix Moth Infesting Apples Using Controlled Atmosphere and Temperature Treatment System)

  • 국내 사과를 외국으로 수출하기 위해서는 수입국의 검역 대상 해충을 완전히 제거하는 것이 필수적이다. 이를 위한 수확 후 해충 사멸 기술로서 환경조절열처리(controlled atmosphere and temperature treatment system: CATTS) 기술이 개발되고 있다. 본 연구는 상이한 가해 습성으로 사과 과실에 피해를 주는 두 해충에 대해 CATTS의 사멸조건을 결정하였다. 사과애모무늬잎말이나방(Adoxophyes paraorana)은 사과 과실 표면을 가해하는 해충인 반면 복숭아명나방(Dichocrocis punctiferalis)은 과실의 내부를 가해한다. 여러 가해 발육태 가운데 두 곤충 모두 5령 유충이 열에 대한 내성이 가장 높았다. 고농도(15%)의 이산화탄소, 저농도(1%) 산소 및 $46^{\circ}C$ 온도 조건에서 1 시간 동안 처리하는 CATTS 처리조건은 사과애모무늬잎말이나방 5령 유충을 100% 사멸시켰다. 그러나 동일한 CATTS 조건에서 복숭아명나방 5령 유충은 88%의 사멸 효과를 보였으며, 100% 사멸 효과를 나타내기 위해서는 2 시간의 열처리를 요구했다. 이를 바탕으로 사과를 가해하고 있는 두 종의 5령 유충 3,000 마리 이상에 대해서 각각 CATTS 처리 효과를 확증하였다. 본 연구는 완전 사멸을 위해서는 심식충이 비심식충에 비해 장기간 CATTS 처리가 요구 된다는 것을 보여 주었다.

Effects of Weekly Administration of Implant-type Recombinant Porcine Somatotropin on the Performance and Carcass Characteristics of Finishing Pigs

  • The present study was undertaken to investigate the effects of weekly administration of implant type recombinant porcine somatotropin (rpST) on the performance and carcass characteristics in finishing pigs. A total of 120 crossbred (Landrace${\times}$Yorkshire${\times}$Duroc) pigs were employed for 11 weeks in a growth trial in experiment. A rpST designed to implant every 7 d was used. Forty pigs, each weighing 75 kg, were allocated into three rpST treatments; control (CONT), implant of rpST from 75 kg (TRT1) or 90 kg (TRT2) of body weight. The CONT pig and pigs in TRT2 from 75 kg to 90 kg were treated without rpST but with placebo. In rpST-treated pigs, each 100 mg and 125 mg of the equivalent rpST was implanted from live weight of 75 kg to 90 kg and from 90 kg to market weight, respectively. Half of the pigs from each treatment were marketed at live weight of 110 kg and the rest at 130 kg. All pigs were allowed ad libitum access to a commercial feed containing 0.94% and 0.88% of lysine from 75 to 110 kg, 110 to 130 kg of body weights, respectively. rpST had no effect on daily gain, while feed efficiency was improved by 7 to 13% (p<0.05) in the rpST-treated groups compared with the CONT. Compared with the CONT, backfat thickness was decreased by 12% (p<0.05) in TRT1 at 110 kg of market weight, and by 23 to 32% (p<0.05) in the rpST-treated groups at 130 kg of market weight, respectively. Lean muscle rate tended to be higher in TRT1 at both 110 kg and 130 kg of market weight, and carcass fat percentage in the rpST-treated groups was decreased by 33 to 46% (p<0.05) compared with the CONT.

고무와 화학제품 제조업 산업장의 작업환경실태에 관한 조사연구 (A Study on the Status of Working Environment of Some Rubber and Chemical Products Manufacturing Industries in Busan)

  • This study was conducted in order to investigate the status of harmful working environ ment on twelve rubber and ten chemical products manufacturing industries in Busan area over a period of five months from lune 1 to October 31, 1980. The summarized results were as follows: 1. The highest and lowest mean values of harmful environmental elements in workroom of rubber products manufacturing industries were noted in twisting (98.7dB) and coating department (77.3dB) to noise, molding ($6.43mg/m^3$) and forming ($1.33mg/m^3$) to dust, bonding (toluene 463.7ppm, xylene 457.8ppm and benzene 111.8ppm, respectively) and splicing (toluene 90.0ppm, xylene 83.3ppm and benzene 6.7ppm, respectively) to organic solvents, respectively. Also in chemical products manufacturing, they were noted in grinding (95.1dB) and shining department (76.8dB) to noise, packing ($4.30mg/m^3$) and staining ($3.20mg/m^3$) to dust, shining (393.3ppm and 375.0ppm, respectively) and varnishing(125.5ppm and 121.7ppm, respectively) to toluene and xylene, and scattering (51.8ppm) and mixing (23.9ppm) to benzene, respectively. 2. The mean values of harmful elements in workroom of rubber products manufacturing were 86.3dB to noise, $4.16mg/m^3$ to dust, 258.2ppm to toluene, 230.3ppm to xylene, and 5 4.0ppm to benzene, respectively. Also in chemical products manufacturing, they were 85.2dB to noise, $3.69mg/m^3$ to dust, 227.9ppm to toluene, 213.2ppm to xylene, and 36.3ppm to benzene, respectively. 3. Number of workers exposed to harmful working environment, over TLV, of a total 10,195 workers in rubber products manufacturing were 1,002(9.8%) to noise, 212 (2.1%) to dust, 1,581(15.5%) to toluene, 1,509(14,8%) to xylene, and 1,524(15.0%) to benzene, respectively. Number of workers exposed to harmful working environment, over TLV, of a 1,913 workers in chemical products manufacturing were 112(5.9%) to noise, 132(6.9%) to each organic solvent, respectively. 4. The values of noise and dust of rubber and chemical products manufacturing in 1980 were lower then those in 1977, but the value of organic solvent in 1980 was similar with that in 1977.

제주산 온주밀감의 CA저장에 관한 연구 (Studies on the controlled atmosphere storage of Unshiu orange)

  • 제주산 온주밀감의 CA저장에 대한 가능성을 검토하기 위하여 일정한 환경가스조성(0,10%, $CO_2$ 2%), 온도 $2{\pm}0.5^{\circ}C$ 및 습도 $86{\pm}3%$의 조건에서 저장중의 과일의 성상 및 성분의 변화를 대조구와 비교 조사하였다. 대조구는 가스대신에 공기를 사용한 것을 제외하면 동일한 조건이었다. 111일 저장후 공기구의 부패율은 12.7%이었는데 비해, CA구에서는 3.9%로 매우 낮은 부패율을 나타냈으며, 과일의 외관도 양호했다. 중량 감소는 공기구에서 16.4%, CA구에서 17.4%로 양자사이에 큰 차이는 없었고, 과육율은 공기구에서는 저장후 62일까지는 거의 변화가 없었지만, 그후 급격히 감소했다. CA구에서는 공기구보다 다소 낮은 과육율을 나타냈으나 저장말기에 약간 증가했다. 저장기간중 과육의 당도변화는 공기구 및 CA구에서 각각 저장후 23일 및 36일에 최고치에 달한후 감소하였고, 유리산함량은 저장기일이 경과함에 따라서 공기구 및 CA구 모두에서 서서히 감소하였다. 저장기간중의 당조성의 변화를 조사한 결과 자당, 과당 및 포도당 중에서 자당의 함량이 월등하게 높았으며 이들 당류의 총량의 변화는 공기구쪽이 CA구보다 다소 높은 수치를 나타했다. 유기산 조성은 구연산이 주된 산이었으며 저장기간이 경과와 함께 감소하였고, 사과산 및 수산 함량의 변화도 구연산과 유사한 경향을 나타냈다. 이상의 결과를 종합하면, 온주밀감의 저온CA저장은 저온공기저장에 비교하여 과육의 당함량이 다소 낮고, 산함량이 다소 높기는 하지만, 부패율의 억제에 효과적이며 과일 외관도 양호하므로 온주밀감의 저장방법으로써 가능성이 높다고 판단된다.

인삼근부병 억제토양 및 유발토양의 근권환경 비교 (Comparison of Rhizosphere Environments in Soils Suppressive and Conducive to Ginseng Root Rot)

  • 인삼 근부병 억제토양 및 유발토양의 추출액 배지에서 병원균인 Fusarium solani, Rhizoctonia solani, Phytophthora cactorum, Sclerotinia sp.의 균계생장과 여기에 영향을 미지는 두 토양의 근권환경을 비교하였다. 4병원균 모두 추출액을 열처리하지 않았을 때 유발토양보다 억제토양 추출액 배지에서 생장이 더 억제되였고, $100^{\circ}C$로 처리 하였을 때는 F. solani와 Sclerotinia sp.는 두 토양간에 차이가 없었으나 R. solani와 P. cactorum은 유발토양에서 균사생장이 더 좋았다. 또 억제토양 추출액의 열처리 온도를 높힐수록 모든 병원균의 생장이 증가되었다. 두 토양의 근권미생물 밀도는 Fusariumtn는 유의차가 없었으나 전세균, 진균은 모두 억제토양에 밀도가 더 높았으며 이 미생물들에 대한 Fusarium밀도의 비율도 억제토양이 더 높았다. 점토함량은 액제토양이, 모래함량은 유발토양이 각각 더 높았고, 화학성분중 Mg, Na 함량은 유발 토양이 더 많았으며 Ca, Fe, $P_2O_5$도 유의성은 없었으나 억제토양 보다 높은 경향이었다.

대구시의 고농도 오존 발생 일에 나타나는 기상학적 특성 (Meteorological Characteristics of High-Ozone Episode Days in Daegu, Korea)

  • 이 연구에서는 1997년부터 1999년 3년 간 대구광역시의 시간별 O$_3$ 농도 자료와 기상자료를 분석하여, 대구시의 고농도 오존 발생 일에 나타나는 기상학적 특성을 알아보고자 한다. 고농도 오존 발생 일의 선정은 우리나라 환경기준치인 1시간 평균 오존 농도 100ppb를 초과한 지점이 대구시의 6개 측정 지점 중 한 지점이라도 있는 경우로 정했다. 고농도 오존 발생 일은 13일이었으며, 5월과 9월이 가장 그 빈도가 높았다. 고농도 오존 발생 일의 하루 평균 최대 오존 농도는 81.6ppb이었으며, 8시간 평균 농도는 58.6ppb이었다. 이는 대구의 오존 오염이 연속적으로 그리고 광범위하게 일어났고 있음을 의미한다. 하루 최고 오존 농도는 일사량, 최고 온도와 양의 상관을 보였으며, 상대 습도, 풍속, 구름양과는 음의 상관을 보였다. 일사량과의 상관계수가 0.45로 가장 높았다. 고농도 오존 발생 일의 기상 값과 그 날을 포함하는 월평균 값과의 차이를 보면, +1.58hPa(해면 기압), +3.45${\circ}$C(최고 기온), -5.69%(상대 습도), -0.46ms$^{-1}$(풍속), -1.79(구름양), +3.97MJm$^{-2}$(일사량)을 각각 보였다. 이는 0700${\sim}$1100LST사이의 높은 일사량, 낮은 풍속, 무강수가 고농도 예측의 중요특징임을 나타낸다. 이는 이 시간의 정체와도 연관이 있다.

Biligrafin 투여 마우스 간세포의 미세구조적 및 세포화학적 연구 (Some Observations on the Organelles Participating in the Biliary Excretion in the Hepatocyte of the Biligrafin Injected Mouse)

  • In this study, an attempt was made to investigate the probable organelles participating in the secretion of biligrafin. The animals (ICR male mice, 25-30gm) were divided into normal control and 6 biligrafin injected groups to which 30% biligrafin (0.006ml/gm b.w.) were injected at 10, 20, 40, 80, 160 and 320 min prior to the sampling. The mice of each group were perfused through the heart with ice-cold 2.5% glutaraldehyde buffered with 0.1M Na-cacodylate (pH. 7.4) under the Na-pentobarbital (Nembtal 0.0015mg/gm b.w.) anesthesia and liver tissues were taken from each group. Some specimens were immersed 1 hr in the same solution used in the perfusion. After an overnight rinse in 0.1M Na-cacodylate buffer containing 10% DMSO and 7.6% sucrose, $75{\mu}m$ fronzen sections were made for cytochemical study. The sections were incubated in thiamin pyrophosphatase (TPPase) and inosine diphosphatase (ID Pase) media for 70 min at $37^{\circ}C$ respectively and acid phosphatase (AcPase) medium for 40 min at $37^{\circ}C$. They were postfixed in 1 % $OsO_4$ for 1 hr. The other specimens were immersed for 8 hrs in the fixative consisting of 2.5% glutaraldehyde and 3.0% paraformaldehyde buffered with Na-cacodylate (pH. 7.4). All of the osmificated specimens were processed for electron microscopy. In both normal and biligrafin injected groups, endoplasmic reticulum (ER), vacuoles, Golgi apparatus and lysosomes were seen in the vicinity of bile canaliculus. In the biligrafin injected groups, however, the Golgi apparatus appeared to be decreased and ER and vacuoles were dilated and increased. The rough endoplasmic reticulum (RER) having a few attached ribosomes appeared to be the round saccule, especially at 20 min after biligrafin injection. Smooth endoplasmic reticulum (SER) seemed to be formed by the detachment of ribosomes at the cisternal end of RER. The cistern of SER showed saccules which probably budded off to form the vacuole. The vacuoles were devoid of visible centents. This finding seemed to be in agreement with the biochemical property of the bile constituents. The fusion between the vacuoles and bile canaliculus were frequently seen in the groups injected with biligrafin. The lysosome did not show any changes in the biligrafin injected groups. Accumulation of some material and lipid droplets were seen at the 40 and 80 min after biligrafin injection, especially at the latter. At 160 and 320 min after biligrafin injections, however, they were decreased successively while the RER stack, free ribosomes and polysomes were increased. Although the reactive products of TPPase and IDPase were observed in the ER saccules and vesicles of the normal control and biligrafin injected groups, the fusion between the bile canaliculus and saccules or vesicles could easily be seen in the latter. The AcPase activity, however, was observed in the cistern at the maturing face of Golgi apparatus and lysosomes in both normal and biligrafin groups. The results suggest that the biligrafin is excreted via the vesicles, vacuoles or sacoules probably derived from the SER without the participation of Golgi apparatus and lysosomes, and the excess amount of material is stored as inclusions during the repairing of the organelles being overactive.

온실멜론의 암면재배에 있어서 수분기의 양분공급제한이 생육 및 양분흡수에 미치는 영향 (Effects of the Limited Nutrient Supply at the Pollination Stage on the Growth and Nutrient Uptake of Muskmelon Grown in Rockwool)

  • This experiment was carried out to investigate the effects of limited nutrient supply during 21 days before and after pollination stage on the growth, fruit quality and nutrient uptake of muskmelon in rockwool culture. Muskmelon, cv. Earl's Favorite seeds sowed on rockwool cube and transplanted on rockwool slab($90\times15\times7.5cm$) when 2 to 3 true leaf appeared on Sep. 6, 1991. Three kinds of nutrient composition recommended by Shizuoka university, combinated with the composition of Otsuka house A and composition Shizuoka III. One half of calcium nitrate(Ca(NO$_3$)$_2$.4$H_2O$) for limiting nitrogen supply during 21 days was treated and then fertigated the nutrient composition recommended by Shizuoka university up to harvest time. Trickling nozzles(Netafim Co. Israel) were used for fertigation of nutrient solution and noncirculating system was employed. Temperature was maintained $18^{\circ}C$ in night but 23 to $25^{\circ}C$ for 10 days after pollination for softening the fruit. The drainage ratio of nutrient solution was adjusted 20 to 30 percent. Fertigated and drained amount, and the pH and EC of nutrient solution were recorded. The concentrations of mineral elements including N, P, K, Ca, and Mg were analyzed and compared among treatments. In both autumn and winter cultivation, the limitation of nutrient supply by adjustment of nutrient composition(NO$_3$-N : 8meㆍ$\ell^{-1}$) caused the nutrient deficiency in muskmelon plant due to the limited nutrient supply. After pollination nutrient limitation by the lowering the nitrate retarded the over thickening of upper leaves of muskmelon but plant height and fresh weight of fruit were higher in the plot of nonlimited nutrient supply. The phenomena were attributed to the differences of the amount of nutrient uptake due to the limited time of nutrient solution, duration of nutrient supply and concentration of nutrient solution. These results suggested that increasing nutrient supply in the pollination stage was favorable for better appearance of fruit and improving fruit quality. Further trials would be required for the incre-ment of sugar degree of muskmelon grown in rockwool.

Characteristics of Soil Chemical Properties in Abandoned Coal Mine Forest Rehabilitation Areas in Boryeong City, Chungcheongnam-do

  • The objectives of this study were to investigate soil chemical characteristics for forest rehabilitation and suggest management in abandoned coal mine areas in Boryeong City, Chungcheongnam-do. Total study sites were seven sites, and soil properties analyzed were soil pH, total organic carbon (TOC), total-N, C/N ratio, and available $P_2O_5$ (A.v. P). Average soil pH (range) was 5.9 (4.5~7.0). Three study sites (Samgwang, Shinsung1, and Shimwon1) showed lower soil pH than standard (pH 5.6~7.3) of Korea Industrial Standards (KS) for abandoned coal mine forest rehabilitation. Average contents of TOC, and total-N were 1.5% (0.1~4.7%), and 0.10% (0.03~0.23%), respectively. Five study sites where the collapsed time was less than 10 years (Wangjashingang, Wonpoong, Samgwang, Shinsung1, and Shinsung2) showed lower TOC level than standard of KS (more than 1.2%). Wangjashingang, Wonpoong, Samgwang, and Shinsung1 showed lower level of total-N than standard of KS (more than 0.09%). C/N ratio of six study sites except Shimwon1 was out of proper range (15:1~30:1). Average A.v. P (range) was $20.7mgkg^{-1}$ (4.8~63.1), less than other abandoned coal mine fores rehabilitation areas in Mungyong City, and Hwasun-gun. TOC, total N and A.v. P increased with elapsed time from forest rehabilitation, while other soil properties did not show distinct pattern. Betula platyphylla was planted in Samkwang and Sinsung where soil pH was less than KS standard. Because the growth of Betula platyphylla can be limited in acid soil, it is necessary to neutralize soil pH to proper level with some soil amendment such as lime or shell of oyster. Furthermore, TOC, total-N and A.v. P in early stage of forest rehabilitation showed lower level than proper to vegetation growth. Therefore it needs continuous monitoring of soil characteristics and fertilization for vegetation growth and influx from surrounding forest in early stage of rehabilitation.
