Identification and Characterization of Calcineurin Targets in Cryptococcus neoformans

  • Calcineurin governs stress survival, sexual differentiation, and virulence of the human fungal pathogen Cryptococcus neoformans. Herein, we identified and characterized calcineurin substrates in C. neoformans by employing phosphoproteomic $TiO_2$ enrichment and quantitative mass spectrometry. The identified targets include the zinc finger transcription factor Crz1 and proteins whose functions are linked to P-bodies/stress granules (PBs/SGs) and mRNA translation and decay, such as Pbp1 and Puf4. We show that Crz1 is a bona fide calcineurin substrate, and localization and transcriptional activity of Crz1 are controlled by calcineurin. Several of the calcineurin targets localized to PBs/SGs, including Puf4 and Pbp1, and are required for survival at high temperature and for virulence. Genetic epistasis analysis revealed that Crz1 and the novel targets Lhp1, Puf4, and Pbp1 function in a branched calcineurin pathway that orchestrates stress survival and virulence. These findings propose that calcineurin controls thermal stress and virulence at the transcriptional level via Crz1 and post-transcriptionally by regulating target factors involved in mRNA metabolism.

세신 정유 추출물의 피부진균 및 기회감염진균에 대한 항진균 효과 (Antifungal Activity of Essential Oil from Asarum sieboldii against Epidermal and Opportunistic Pathogenic Fungi)

  • 세신은 한약재로써 오랫동안 사용되어 왔으며 해열, 진정, 진통, 항염증, 면역억제작용등을 나타낸다. 세신에서 증기증류법으로 추출한 정유를 이용하여 이의 항진균 활성을 조사하였다. 세신의 정유는 Aspergillus niger, A. fumigatus, Candida albicans 그리고 Cryptococcus neoformans 등의 피부진균 및 기회감염균에 대하여 높은 항진균 활성을 나타내었다. C. albicans에 대한 최소억제 농도는 약 5 ul/ml로 매우 적은 양으로 높은 억제 효과를 보였으며 이를 바탕으로 천연항균제 또는 천연방부제로 식품 및 화장품에 첨가제로 사용할 수 있는 가능성을 보여주었다.

  • Cryptococcus neoformans causes life-threatening meningoencephalitis in humans, but the treatment of cryptococcosis remains challenging. To develop novel therapeutic targets and approaches, signaling cascades controlling pathogenicity of C. neoformans have been extensively studied but the underlying biological regulatory circuits remain elusive, particularly due to the presence of an evolutionarily divergent set of transcription factors (TFs) in this basidiomycetous fungus. In this study, we constructed a high-quality of 322 signature-tagged gene deletion strains for 155 putative TF genes, which were previously predicted using the DNA-binding domain TF database (http://www.transcriptionfactor.org/). We tested in vivo and in vitro phenotypic traits under 32 distinct growth conditions using 322 TF gene deletion strains. At least one phenotypic trait was exhibited by 145 out of 155 TF mutants (93%) and approximately 85% of the TFs (132/155) have been functionally characterized for the first time in this study. Through high-coverage phenome analysis, we discovered myriad novel TFs that play critical roles in growth, differentiation, virulence-factor (melanin, capsule, and urease) formation, stress responses, antifungal drug resistance, and virulence. Large-scale virulence and infectivity assays in insect (Galleria mellonella) and mouse host models identified 34 novel TFs that are critical for pathogenicity. The genotypic and phenotypic data for each TF are available in the C. neoformans TF phenome database (http://tf.cryptococcus.org). In conclusion, our phenome-based functional analysis of the C. neoformans TF mutant library provides key insights into transcriptional networks of basidiomycetous fungi and ubiquitous human fungal pathogens.

Comparison of Chemical Compositions and Antimicrobial Activities of Essential Oils from Three Conifer Trees; Pinus densiflora, Cryptomeria japonica, and Chamaecyparis obtusa

  • The chemical compositions, and antibacterial and antifungal effects of essential oils extracted from three coniferous species, Pinus densiflora, Cryptomeria japonica, and Chamaecyparis obtusa, were investigated. Gas chromatography mass analysis of the essential oils revealed that the major components and the percentage of each essential oil were 16.66% $\beta$-phellandrene and 14.85% $\alpha$-pinene in P. densiflora; 31.45% kaur-16-ene and 11.06% sabinene in C. japonica; and 18.75% bicyclo [2,2,1] heptan-2-ol and 17.41% 2-carene in Ch. obtusa. The antimicrobial assay by agar disc diffusion method showed that $2.2{\mu}g$ of Ch. obtusa oil inhibited most effectively the growth of Escherichia coli ATCC 33312 and Klebsiella oxytoca ATCC 10031, whereas the C. japonica oil gave weak antimicrobial activity. The minimal inhibitory concentration(MIC) values for bacterial strains were in the range of 5.45-21.8 mg/ml depending on essential oils, but most Gram-negative bacteria were resistant even at 21.8 mg oil/ml. P. densiflora oil showed the most effective antifungal activity and the MIC values for Cryptococcus neoformans B42419 and Candida glabrata YFCC 062CCM 11658 were as low as 0.545 and 2.18 mg/ml, respectively. Cryp. neoformans B42419 was the most sensitive to all essential oils in the range of 0.545-2.18 mg/ml. Our data clearly showed that the essential oils from the three conifers had effective antimicrobial activity, especially against fungi.

양심탕(養心湯)이 스트레스와 면역기능(免疫機能)에 미치는 영향(影響) (The Effects of Yangsimtang on Stress and Immune System)

  • After applying the gravity acceleration stress to a mice, the effect on organ index was examined, and the Con A stimulating proliferation rate of splenocytes, expression of IL-2 receptor and T cell subsets of thymocytes were analyzed and also clearance of C. neoformans was measured. The results were as follows :1. Form finding the organ index after 4 days stress, the indexes of the spleen and thymus were reduced in the res group exposed to the gravity acceleration.2. From finding the proliferation rate by stimulating the splenocytes with Con 4 after 7 days stress, the proliferation rates were all reduced in the stress group, the Yangsimtang group, and the stress and Yangsimtang group. 3. The expression of IL-2 receptor in resting stage was reduced, comparing to the test group, both in the stress group and the Yangsimtang group, however, comparing to the stress group, it was somewhat recovered in the stress and Yangsimtang group.4. To see the IL-2 receptor driven-out after being stimulated by Con-A, the expression of IL-2 receptor was all reduced in the stress group, the Yangsimtang group, and the stress and Yangsimtang group.5. To the rate of T cell subsets of thymus, there's no difference, comparing to the test group, in the Yangsimtang group, however, the rate of $CD4^+CD8^-,\;CD4^-CD8^+,\;and\;CD4^-CD8^-$ cell was significantly reduced in the stress group. And, the $CD4^+CD8^+$ which had been reduced by stress was somewhat recovered in the stress and Yangsimtang group.6. To the effect on the clearance of C, neoformans infection, the numbers of fungi detected at the spleen was, comparing to the test group, increased by 12.6 tines in the Yangsimtang group.

Burkholderia sp. OS17의 항균활성 증진을 위한 배양최적화 (Antimicrobial activities of Burkholderia sp. strains and optimization of culture conditions)

  • 본 연구는 담수환경에서 항균활성을 보유한 미생물을 발굴하고, 활성 증진을 위해 배양조건을 최적화하는 것이다. 상주시 중동면 오상저수지에서 시료를 채취하여 38종의 미생물을 순수분리하였다. 16S rRNA 염기서열 분석에 근거하여 Proteobacteria강(22종), Actinobacteria강(7종), Bacteroidetes강(6종), Firmicutes강(3종)으로 구성되어있는 것을 확인하였다. 메티실린내성 황색포도상구군 등 10종의 유해미생물에 대한 항균활성을 보유한 Burkholderia sp. OS17 균주를 선발하였다. 항균활성 증진을 위한 상용배지, 온도, 초기 pH별 생육 및 항균활성 비교실험을 수행하였다. OS17 균주는 YPD 배지, $35^{\circ}C$, pH 6.5로 배양했을 때 가장 활성이 높았다. LB, NB, TSB, R2A 배지와 $20^{\circ}C$, $25^{\circ}C$ 배양했을 때는 생장은 가능하나 항균활성이 전혀 없었다. 이전결과를 바탕으로 YPD 배지, $35^{\circ}C$에서 배양하면서 5 L fermenter를 이용하여 생육, 항균활성, pH 확인을 통해 배양 48시간을 최적 배양시간으로 선정하였다. 항균활성을 보유한 미생물의 배양 최적화는 항균물질 생산에 영향을 미치고, 이는 상업적 응용에 이점으로 작용할 수 있다.

갓 정유의 조성 및 항진균작용에 관한 연구 (Studies on Compositions and Antifungal Activities of Essential Oils from Cultivars of Brassica juncea L.)

  • The composition of essential oils in the leaves of three cultivars (Ban-Chung-Gat, Chung-Gat and Dolsan-Gat) of Brassica juncea L. were analyzed and their antifungal activity were investigated in this study. Allyl isothiocyanate, 2-phenyl ethyl isothiocyanate, 4-isothiocyanato-1-butene, 5-methyl isothiazole, benzene acetaldehyde, benzene propane nitrile and beta-ionone have been identified in all of the experimented oils. The main component of the oils from Ban-Chung-Gat and Chung-Gat was 2-phenyl ethyl isothiocyanate while allyl isothiocyanate was the representing compound in the oil of Dolsan-Gat. The antifungal activities of the oils were tested by micro broth dilution method and disc diffusion method. As the result the oils exhibited significant inhibiting activities against Aspergillus niger, A. flavus, Trichoderma viride, Candida albicans, C. utilis, C. tropicalis, Cryptococcus neoformans, Trichosporon mucoides, Trichophyton tonsurans and Geotrichum capitatum.

pH Response Pathways in Fungi: Adapting to Host-derived and Environmental Signals

  • Microorganisms are significantly affected when the ambient pH of their environment changes. They must therefore be able to sense and respond to these changes in order to survive. Previous investigators have studied various fungal species to define conserved pH-responsive signaling pathways. One of these pathways, known as the Pal/Rim pathway, is activated in response to alkaline pH signals, ultimately targeting the PacC/Rim101 transcription factor. Although the central signaling components are conserved among divergent filamentous and yeast-like fungi, there is some degree of signaling specificity between fungal species. This specificity exists primarily in the downstream transcriptional targets of this pathway, likely allowing differential adaptation to species-specific environmental niches. In this review, the role of the Pal/Rim pathway in fungal pH response is discussed. Also highlighted are functional differences present in this pathway among human fungal pathogens, differences that allow these specialized microorganisms to survive in the various micro-environments of the infected human host.

건강인에서 발생한 폐효모균증 1예 (A Case of Pulmonary Cryptococcosis in Healthy Person)

  • 본 환자는 평소 건강하던 38세 남자환자로 기침의 증상과 X-선상 우측폐에 경화소견을 보인 예였다. CT 유도하 세침 흡인 생검법에 의한 세균학적검사와 조직 검사로 C. neoformans에 의한 폐효모균증으로 진단하여, 6주간 fluconazole 사용후 증상은 소실되었고, X-선상 많이 호전되어 현재 추적 관찰중에 있다.

Bacillus amyloliquefaciens IUB158-03이 생산하는 항진균물질의 분리와 항균활성 (Isolation of Antifungal Substances by Bacillus amyloliquefaciens IUB158-03 and Antagonistic Activity against Pathogenic Fungi)

  • 병원성 미생물을 방제하기 위하여 국내의 고추 재배지에서 탄저병균에 감염된 고추로부터 Bacillus속 균주를 분리하였다. 이들 중 고추탄저병균인 C. gloeosporioides 균주와 식물에 회색곰팡이병을 일으키는 Botrytis cinerea 균주에 대한 길항작용이 가장 우수한 Bacillus sp. IUB158-03을 선발하였다. 선발된 균주의 형태적, 생리 생화학적 특성과 MicroLog 방법을 이용하여 동정된된 균주는 B. amyloliquefaciens IUB158-03로 명명하였다. 항진균물질 생산을 위한 최적배지 조건은 2% soluble starch, 3% yeast extract, 0.5% tryptone, 0.5% $NH_4H_2PO_4$, 1% NaCl의 pH 6.0에서 $25^{\circ}C$, 72시간 배양한 추출물에서 가장 높은 항진균활성을 나타내었다. B. amyloliquefaciens IUB158-03이 생산한 항진균물질을 butanol로 추출한 후 silica gel column chromatography, preparative thin layer chromatography, and HPLC을 이용하여 항진균물질을 정제하였다. 정제된 항진균물질은 TLC plate 상에서 $R_f$값이 0.27인 것이 확인되었으며, 이 물질은 식물병원균인 Fusarium solani, Rhizoctonia solani, Botrytis cineria, Alternata alternaria와 인체병원성 진균인 Trichophyton mentagrophytes, Epidermophyton floccosum, Cryptococcus neoformans 등에만 선택적으로 높은 항진균 활성을 보였다.
