Quantitative analysis of the marker compounds in the decoctions of Coptis chinensis-Scutellaria baicalensis at different proportion produced by 'Mixed decoction' and 'Single decoction mixture' (배합 비율에 따른 황련과 황금의 혼합 전탕액 및 개별 전탕 혼합액 내 성분 함량 분석)

  • Objective : The present study aimed to evaluate the change of the content of 7 active components in decoctions produced by various proportional pairs of Coptis chinensis Franch and Scutellaria baicalensis Georgi in 'Mixed decoction (MD)' and 'Single decoction mixture (SDM)'. Methods : The samples of MDs were prepared by decocting C. chinensis : S. baicalensis with the ratios of 10 g:10 g, 10 g:20 g, and 20 g:10 g. Those of SDMs were prepared by blending each single decoction from C. chinensis and S. Baicalensis with the ratios of 1:1, 1:2, and 2:1. The samples were evaluated by high-performance liquid chromatography with statistical analyses. Results : The analytical methods, which were optimized and validated, were reliably applied to present research. The content of all components in both MDs and SDMs at C. chinensis : S. baicalensis = 1:1 ratio were reduced compared with single herb decoction. The components from each compositional herb in MDs were proportionally increased with the ratio of original herb increased, but inversely proportional to paired herb. The contents of components in MDs were significantly lower than those in SDMs at all ratios, except for high content of baicalin at C. chinensis : S. baicalensis = 2:1. Conclusion : It was concluded that MDs and SDMs as well as the proportions of herbs could affect the contents of the components from original herbal medicines. These results provide the information for the quality control of herbal medicine combined C. chinensis with S. baicalensis.

Gomisin N and Wuweizisu C, new Inhibitors for Chitin Systhase II from Schizandra chinensis (오미자에서 분리한 새로운 Chitin Synthase II 저해제인 Gomisin N 과 Wuweizisu C)

  • To search for new chitin biosynthesis inhibitors from natural sources, several higher plants were examined the inhibitory activity against chitin synthase IIby enzymatic assay. Among them, the extract of Schizandra chinensis strongly showed the inhibitory activity against chitin synthase II. Gomisin N and wuweizisu C were isolated from Schizandra chinensis and showed $IC_50$ value of $62.4{\;}\mu\textrm{g}/ml$ and $19.2{\;}\mu\textrm{g}/ml$, respectively. Activities of these compounds were more stronger than that of polyxin D. However, gomisin N and wuweizisu C showed weakly antifungal activities against various human pathogens.

Effects of Temperature and Body Size on the Clearance Rates of a Tidal Flat Bivalve, Coecella chinensis (Deshayes)

  • To know the effects on temperature and body size on the clearance rate (CR) of a small tidal flat bivalve, Coecella chinensis, laboratory experiments were performed with 20 individuals of different sizes (ranging from 7 to 25 mm) at 3 different temperatures (10, 15, $20^{\circ}C$). The relationship between body size and CR was determined by an allometric equation. The CR of C. chinensis varied greatly ranging from 0.003 to 0.103 L/individual/hr. Both temperature and body size affected significantly on the CR of C. chinensis. The CR at $20^{\circ}C$was 1.5 times higher than that at $15^{\circ}C$ and 2.8 times than $10^{\circ}C$. The temperature coefficient ($Q_10$) between 10 and $15^{\circ}C$ was higher than that between 15 and $20^{\circ}C$, which indicates that C. chinensis changes its CR more rapidly in lower temperature range. As body size increased, the CR increased more than 10-fold at all temperatures. The CR relative to flesh dry weight (FDW) were fitted well to the power function: CR = a ${\times}$ $(FDW)^b$. The exponent value (b) of the fitted equation ranged from 0.64 to 0.70, which are similar to those of other bivalves. The weight-specific CR ($CR_w$) was still affected by body size (p < 0.05). This implies that smaller individuals require more energy per unit biomass for growth, and the energy requirement for growth decreases as body size increases.

  • Water extracts, ethanol extracts, and juice of Cusuta japonica, C, australis, and C. chinensis were prepared, and their cytotoxicity, antioxidant activity and inhibitory effects on tyrosinase activitiy and melanin biosynthesis were estimated by using melanoma Clone M-3. From this study; the following conclusions were attained. Extracts of Cuscuta japonica, C. australis, and C. chinensis showed noticeable cytotoxicity except ethanol extracts from the stem of C. australis. A maximual cytotoxicity was observed with tile ethanol extract from the seed of C, australis (87.39%). While the ethanol extract from the seed of C. japonica (91.88%) showed the most pronounced inhibitory effect on melanin biosynthesis, the water extract from the stem of C. japonica (1.05%) possessed very little inhibitory effect. The most inhibitory effect on tyrosinase activity was observed with the water extract from the stem of C. australis (76.67%). Howeverr the water extract from the stem of C. japonica showed a very poor effect on the inhibition of tyrosinase activity All the preparations, except extracts from the seed of C. australis were able to remove reactive oxygen species (ROS) in a dose-dependent manner. The juice of C. japonica demonstrated the strongest activity (59.02%).

Effect of Water Extract of Schisandra Chinensis on Osteoclast Differentiation (오미자 물 추출물이 파골세포 분화에 미치는 영향)

  • Bone maintains its homeostasis through balance between bone resorbing osteoclasts and bone forming osteoblasts. Thus, unusual balance between osteoclasts and osteoblasts leads to pathological bone diseases, such as osteoporosis, rheumatoid arthritis, autoimmune arthritis, periodontitis. Schisandra chinensis well known traditional herbal has been used for treatment of diseases in China, Korea, Japan, andothers. Recently, research studies have demonstrated that the lignans found in Schisandra chinensis stimulate osteoblasts and suggest that it may be helpful against osteoporosis. However, the inhibitory effect of water extract of Schisandra chinensis on osteoclast differentiation remains largely unknown. In this study, Water extract of Schisandra chinensis markedly suppressed RANKL-induced osteoclast differentiation in cultures of BMMs without cytotoxicity. The mRNA expression of c-Fos, NFATc1, and TRAP induced by RANKL was inhibited by water extract of Schisandra chinensis. It also suppressed c-Fos and NFATc1 protein expression. Taken together, these results suggest that water extract of Schisandra chinensis has the potential to serve as a treatment of bone disease such as osteoporosis.

A New Species of the Genus Caminus (Astroporida: Geodiidae) from Korea

  • Caminus jejuensis n. sp was collected from depth of 20 m at Geomeunyeo, Seogwipo, Jejudo Island by a SCUBA diving from April 2004 to December 2008. This new species is similar to C. chinensis from China in the composition of spicules except for the spherasters and they differ in spicule size and growth form. This species has longer orthotriaenes and spherules, smaller sterrasters and oxyasters than those of C. chinensis. This species also has many spherasters in choanosome, but C. chinensis lacks. Morever, the new species is a massive shape with wrinkles, whereas C. chinensis is a club shape with smooth surface. Description and figures of the new species are provided.

  • The effects of different night temperatures and NO$_3$$^{[-10]}$ : NH$_4$$^{+}$ ratios in nutrient solution on the growth and quality of Chinese white cabbage(B. chinensis L. var. chinensis) and Chinese flat cabbage(B. chinensis L. var. rosularis) were studied. The results were summarized as follows. 1. Fresh weight was increased higher in night temperature 15$^{\circ}C$ than 5, 1$0^{\circ}C$, but content of vitamin C and dry weight ratio were increased as night temperature was lower. 2, The growth of Chinese white cabbage and Chinese flat cabbage was bad extremely in NO$_3$$^{[-10]}$ : NH$_4$$^{+}$(0 : 8), and the others were little different 3. In nutrient solution, the higher NO$_3$$^{[-10]}$ -N concentration was, the more content of vitamin C, and the higher NH$_4$$^{+}$-N concentration was, the more dry weight ratio.ratio.

  • Cytological observations in the seven varieties of Juniperus chinensis L. showed three varieties (J. chinensis var. procumbens, J. chinensis var. kaizuka and J. chinensi.s var. aureo-variegata) were tetraploid with chromosome number, 2n=44, and rest of four varieties (J. chinensis var. horizontalis, J. chinensis var. sargentii, J. chinensis var. gtobosa and J. chinensis var. aureo-globosa) to be diploid, 2n=22. Chromosome configuration and behavior in the meiosis of P.M.C. of three tetraploids appeared to be slightly irregular. These results suggest that triploid tree can be artificially produced with these specific clones.

Changes of Physicochemical Characteristics of Schizandra chinensis during Postharvest Ripening at Various Temperatures (후숙 온도에 따른 오미자의 이화학적 특성 변화)

  • This study was carried out to investigate the changes of physicochemical characteristics of Schizandra chinensis during postharvest ripening for 8 days at various temperatures. The juice yield of S. chinensis, which was 55.7% before postharvest ripening, was unchanged $(55.3{\pm}0.6-56.3{\pm}0.6%)$ at $4^{\circ}C$ storage, but was decreased at the level of 6 and 7% at $25^{\circ}C$ and room temperature (RT), respectively. During storage at $25^{\circ}C$ and RT, the titratable acidities of S. chinensis were the highest at $7.49{\pm}0.03$ and $7.20{\pm}0.03%$ after 6 days of postharvest ripening, respectively. During storage at $25^{\circ}C$ and RT, the soluble solid content of S. chinensis was increased from $8.2{\pm}0.1%$ at initial stage to a peak of $12.2{\pm}0.15%$ at 6-day storage, after which it decreased. L values (lightness) of S. chinensis were increased in all treatments during storage, and a values (redness) of $25^{\circ}C$ and RT treatments were increased from 5.04 initially to 6.77 and 7.65 at 8-day storage, respectively. The major free sugars of S. chinensis were fructose (0.55%), glucose (0.56%), and sucrose (0.50%). During storage at $25^{\circ}C$ and RT, the fructose and glucose contents were continually increased with increasing storage period, while the sucrose contents decreased after 6-day storage. Major non-volatile organic acids of S. chinensis were succinic (1.21%), citric (0.17%), and malic (0.07%) acids. Changes in the organic acids contents of S. chinensis at various temperatures showed a similar tendency to that of the free sugars. We estimated that the best conditions for the postharvest ripening of S. chinensis were 8 days at $4^{\circ}C$ storage, and 6 days at $25^{\circ}C$ and RT.

The Antioxidative Effect and Ingredients of Oil Extracted from Schizandra chinensis Seed (오미자씨에서 추출된 유지의 성분 및 항산화 효과)

  • The objectives of this study were to investigate the antioxidant activity of Schizandra chinensis seed oil and its active ingredients. Schizandra chinensis seed oil content extracted with hexane was 36.06%. Schizandra chinensis seed oil extracted with hexane was purified during 20 min at $85^{\circ}C$ with phosphoric acid 0.15% for degumming and 20 min at $80^{\circ}C$ with 3 M NaOH 1% for deaciding. The purified oil consisted of unsaturated fatty acid (88.7%), fatty acid (9.97%), and so on. The major unsaturated fatty acids of purified oil were linoleic acid (71.1%) followed by oleic acid (15.7%), while the main saturated fatty acid was palmitic acid (6.56%). The purified oil was found that contents of phenolic compounds, vitamin A, and E were 1.45 g/100 g, 1494.86 RE/100 g, and 0.58 mg ${\alpha}$-TE/100 g, respectively. Schizandra chinensis seed oil exhibited strong antioxidant activity (91.7%) as compared to grape seed oil and canola seed oil with 87.4% and 85.1% in the DPPH assays. Present results suggest that Schizandra chinensis seed oil could be potentially used as bioactive source for health and preventing numerous diseases.
