젖소의 신성수정란의 동결수정란의 비외과적 이식에 관하여 (Nonsurgical transfer of fresh and frozen embryos of dairy cattle)

  • Sixty Four fresh and 142 frozen embryos of dairy cattle were transferred to synchronized dairy, beef or Korean Native Cattle nonsurgically at National Animal Breeding Institute from 1985 to 1990. The results obtained were as follows ; 1. The pregnancy rate of fresh embryos(39.1%) was higher than that of frozen embryos(32.4%) and average pregnancy rate was 34.5%. 2. The pregnancy rate of grade 1 embryos was higher than that of grade 2 embryos for both fresh(41.3% vs 33.3%) and frozen embryos(35.4% vs 25.6%). 3. The pregnancy rate according to development stage of fresh embryos was increased with maturity as 29.2%, 33.3%, 50.0% and 54.5% for morula, early blastocyst, blastocyst and expanded blastocyst, respectively. For frozen embryos, the pregnancy rate of blastocyst(44.4%) was higher than those of morula(31.3%) and early blastocyst(28.0%). 4. The pregnancy rate according go recipient-donor synchrony for fresh embryos was higher when the recipients exhibited estrus 1 day earlier than the donors(43.8%) than when the recipients exhibited estrus 1 day later than the donors(38.1%) or when the recipients and donors exhibited estrus at the same time(37.0%). For forzen embryos, the pregnancy rate was decreased when the recipients and donors exhibited estrus at the same time(37.9%), when the recipients exhibited estrus 1 day later than the donors(32.0%) and when the recipients exhibited estrus 1 day earlier than the donors(23.5%), in sequence. 5. The pregnancy rate of heifers was higher than that of cows for both fresh(50.5% vs 37.9%) and frozen embryos(39.7% vs 25.7%). 6. The pregnancy rate according to recipient breed for fresh embryos was higher in dairy cattle(42.1%) and beef cattle(40.%) than in Korean Native Cattle(33.3%). For frozen embryos, the pregnancy rate was decreased beef cattle(39.1%), dairy cattle(30.3%) and Korean Native Cattle(14.3%), in sequence. 7. The pregnancy rate according to equilibrium steps of glycerol and freezing rate was higher when transferred after 3-steps equilibrium and freezing by the rate of $0.3^{\circ}C$/min from $-6^{\circ}C$ to $-35^{\circ}C$ and $0.1^{\circ}C$/min to $-38^{\circ}C$(39.4%) than when transferred after 6-steps equilibrium and freezing by the rate of $0.5^{\circ}C/min$ from $-6^{\circ}C$ to $-30^{\circ}C$(30.3%).

Kinetic Responses of Soil Carbon Dioxide Emission to Increasing Urea Application Rate

  • BACKGROUND: Application of urea may increase $CO_2$ emission from soils due both to $CO_2$ generation from urea hydrolysis and fertilizer-induced decomposition of soil organic carbon (SOC). The objective of this study was to investigate the effects of increasing urea application on $CO_2$ emission from soil and mineralization kinetics of indigenous SOC. METHODS AND RESULTS: Emission of $CO_2$ from a soil amended with four different rates (0, 175, 350, and 700 mg N/kg soil) of urea was investigated in a laboratory incubation experiment for 110 days. Cumulative $CO_2$ emission ($C_{cum}$) was linearly increased with urea application rate due primarily to the contribution of urea-C through hydrolysis to total $CO_2$ emission. First-order kinetics parameters ($C_0$, mineralizable SOC pool size; k, mineralization rate) became greater with increasing urea application rate; $C_0$ increased from 665.1 to 780.3 mg C/kg and k from 0.024 to 0.069 $day^{-1}$, determinately showing fertilizer-induced SOC mineralization. The relationship of $C_0$ (non-linear) and k (linear) with urea-N application rate revealed different responses of $C_0$ and k to increasing rate of fertilizer N. CONCLUSION(s): The relationship of mineralizable SOC pool size and mineralization rate with urea-N application rate suggested that increasing N fertilization may accelerate decomposition of readily decomposable SOC; however, it may not always stimulate decomposition of non-readily decomposable SOC that is protected from microbial decomposition.

무지개 송어의 유전 육종학적 연구 VII. 동결보존시킨 정자와 신선한 난모세포의 수정 및 미세구조적 변화 (Studies on Genetics and Breeding in Rainbow Trout(Oncorhynchus mykiss) VII. Fertilization of Fresh Egg with Co-Preserved Sperm and Ultrastructural Changes)

  • 건국대학교 축산대학 부설 양어장에서 사육중인 50마리의 무지개 송어 수컷에서 채취된 정액을 각각 1989년 12월부터 1990년 12월까지 $-20^{\circ}C$$-40^{\circ}C$에서 1년 1989년 12월부터 2월까지 2개월동안 $-196^{\circ}C$인 액체질소탱크내에서 그리고 1990년 12월부터 2월까지 $-20^{\circ}C,\;-40^{\circ}C,\;-70^{\circ}C$에서 2개월 동안 동결보존후, $-13^{\circ}C$ 냉수에 해동시켜 20분간 원심분리기에서 정자를 분리시킨 다음 Tyrode 용액으로 50배 정도 희석시킨 후의 정자의 생존율 및 운동성과 신선한 난자와 인공수정시킨 후의 수정률 및 부화율을 조사한 결과는 다음과 같다. 동결보존전과 해동후의 정자의 생존율을 비교해 볼 때 전반적으로 생존전과 큰 차이 없이 생존율이 높게 나타났다. 3가지의 항동해제로 동결보존시킨 후 해동된 정자가 포함된 희석액으로 인공수정시킨 후의 수정률은 모두 $99\%$ 이상으로서 이러한 수치는 $99\%$ 이상을 나타내는 대조구와 차이가 없었고, $-20^{\circ}C,\;-40^{\circ}C,\;-70^{\circ}C$\;-196^{\circ}C와 같이 서로 다른 온도에서도 수정률은 대조구와 큰 차이 없이 나타났다. 1M의 DMSO의 항동해제로 $-196^{\circ}C$에서 2개월 동안 동결보존시킨 후 해동된 정자가 포함된 희석액으로 인공수정시킨지 24일 이후의 수정률 및 부화율은 BSA가 포함된 희석액이 각자 $96\%$$8\%$ 이고, 포함되지 않은 희석액은 각각 $98\%,\;10\%\;$로서 BSA가 포함되지 않은 경우에서 부화율이 약간 높게 나타났다. 1M DMSO의 항동해제로 2개월동안 동종보존시킨 후 해동된 정자가 포함된 희석액으펄 인공수정 시킨지 24일 이후의 부화율은 각각 $-196^{\circ}C$에서 $10\%,\;-40^{\circ}C$에서 $34\%,\;-70^{\circ}C$에서 $35\%$로 나타났다. 1M의 methanol의 항동해제 동결보존시친 후 해동된 정자가 포함된 희석액으로 인공수정시킨지 24일 이후의 부화율은 각각 $-20^{\circ}C$에서 $22\%,\;-70^{\circ}C$에서 $28\%$로 나타났다. 1M glycerol의 항동해제로 동결보존시킨 후 해동된 정자가 포함된 희석액으로 인공수정시킨지 24일 이후의 부화율은 각각 $-20^{\circ}C$에서 $22\%,\;-70^{\circ}C$에서 $33\%$로 나타났다. 1.5M DMSO의 항동제로 2개월동안 동결보존 시킨 후 해동된 정자가 포함된 희석액으로 인공수정시킨지 24일 이후의 부화율은 각각 $-20^{\circ}C$에서 $27\%,\;-40^{\circ}C$에서 $36\%,\;-70^{\circ}C$에서 $35\%$로 나타났다. 1.5M glycerol의 항동해제로 2개월동안 동결보존시킨 후 해동된 정자가 포함된 희석액으로 인공수정시킨지 24일 이후의 부화율은 각각 $-20^{\circ}C$에서 $34\%,\;-40^{\circ}C$에서 $31\%,\;-70^{\circ}C$에서 $31\%$로 나타났다. 1.5M methane의 항동해제로 2개월동안 동결보존시킨후 해동된 정자가 포함된 희석액으로 인공수정시킨지 24일 이후의 부화율은 각각 $-20^{\circ}C$에서 $28\%-40^{\circ}C$에서 $29\%-70^{\circ}C$에서 $28\%$로 나타났다. 신선한 정자의 중편부는 2본의 중앙섬유소, 9본의 외측조대섬유, 미토콘드리아초로 구성되어 있다. 동결보존시 정자는 편모의 구부러짐, 정자경부의 핵막의 이탈, 핵내용물이 유출되는 손상을 입었다.

  • The present study was carried out to investigate the effects of cryoprotectants, equilibration step, freezing rate, culture condition following in vitro fertilization, and age and development stage of embryo by freezing with conventional slow freezing and vitrification on survival of frozen-thawed Korean native cattle(KNC) blastocysts produced in vitro. The KNC blastocysts produced in vitro were equilibrated in 1.8M ethylene glycol or 1.4M glycerol and cooled from -6$^{\circ}C$ to -35$^{\circ}C$ at -0.3$^{\circ}C$ or -O.6$^{\circ}C$ /minute. When equilibrated in 1.8M ethylene glycol, survival rate of fiozen4hawed blastocysts was sarne in both -0. 3$^{\circ}C$ /min and -0.6$^{\circ}C$ /min cooling rate(71.4%). With the equilibration in 1.4M glycerol, survival rate was higher in -0.3$^{\circ}C$ /min(63.6%) than in -0.6$^{\circ}C$ /min cooling rate(53.8%). For vitrification of the KNC blastocysts produced in vitro, they were equilibrated in 2-step or 3-step exposure to vitrification solution(25% ethylene glycol + 25% glycerol). Survival rate was sirilar in both 2-step(45.0%) and 3-step exposure(47.4%). According to culture condition following in vitro fertilization, higher survival rate was obtained for blastocysts co-cultured with bovine oviductal epithelial cell(BOEC, 77.3%) than for those cultured with epidermal growth factor(EGF, 65.7%) or for those co-cultured with BOEG + EGF (54.8%). According to embryo age and development stage, higher survival rate was obtained for 7-day ernbryos(70.0%) than 8-day(56.8%) or 9-day(20.0%) for blastocyst stage and obtained for 8-day embryos(74.3%) than 7-day(62.5%) or 9-day(42.9%) for exponded blastocyst. In surnmary, higher survival rate of frozen4hawed KNC blastocysts produced in vitro were obtained by using ethylene glycol for cryoprotectant and -0.3$^{\circ}C$ /min for cooling rate. And higher survival rate were obtained with co-culture with BOEC for culture condition following in vitro fertilization and with 7-day blastocyst or 8-day expanded blasto cyst for embryo age and development stage.

동결속도와 침지온도가 Hamster 수정란의 생존성에 미치는 영향 (Effects of Cooling Rates and Plunging Temperatures on Survival of Hamster Embryos)

  • This experiment was carried out to investigate the optimal cooling rate and the plunging temperature into liquid nitrogen of the 8-cell hamster embryos. The female hamsters were induced to superovulate by intraperitoneal injection of 30 i.u. PMSG. Embryos were flushed from oviduct and uterine horn. Embryos were frozen and incubated with a modified Dulbecco's phosphate buffered saline, and equilibrated with 1.5M-dimethyl sulfoxide by a 3-step procedure. The cooling rate of samples was 1$^{\circ}C$/min from room temperature to -5$^{\circ}C$ and the samples were seeded at -5$^{\circ}C$. The plunging temperatures into liquid nitrogen were -20, -25, -30, -35, -40, -45, -50 and -55$^{\circ}C$ at 0.3$^{\circ}C$/min, 0.5$^{\circ}C$/min and 1$^{\circ}C$/min cooling rates, respectively. This mean numbers of ovulation points and recovered embryos were 59.4 and 48.4 appearing 81.6% recovery rate. The percentage of 8-cell embryos in recovered embryos was 68.2. The survival rates of embryos plunged at -45$^{\circ}C$ were 73.5% at 0.3$^{\circ}C$/min, 44.8% at 0.5$^{\circ}C$/min and 30.3% at 1$^{\circ}C$/min cooling rates, respectively. This mean numbers of ovulation points and recovered embryos were 59.4 and 48.4 appearing 81.6% recovery rate. The percentage of 8-cell embryos in recovered embryos was 68.2. The survival rates of embryos plunged at -45$^{\circ}C$ were 73.5% at 0.3$^{\circ}C$/min, 44.8% at 0.5$^{\circ}C$/min and 30.3% at 1$^{\circ}C$/min cooling rates, respectively. The survival rates at 0.3$^{\circ}C$/min were significantly high. Under the condition of 0.3$^{\circ}C$/min cooling rate, the survival rates of embryos according to the plunging temperature were 70.0% and 73.5% at -40 and -45$^{\circ}C$, and those were higher than other plunging temperatures. Under the condition of 0.5$^{\circ}C$/min and 1$^{\circ}C$/min cooling rates, the survival rates according to the plunging temperatures were lower than the cooling rate of 0.3$^{\circ}C$/min, showing the similar tendency at all the plunging temperatures. In conclusion, 8-cell hamster embryos showed the best survival rates at 0.3$^{\circ}C$/min cooling rate and -45$^{\circ}C$ plunging temperature.

고출력 18650 리튬이온 배터리의 가변전류 열해석 및 추정 (Thermal analysis and estimation of high power 18650 lithium ion battery under varying current condition)

  • 본 논문은 1차 RC 등가회로를 이용하여 리튬이온 배터리의 저항성 발열인 비가역 발열의 파라미터를 제시하였다. 발열 추정을 위해 1 C-rate에서 HPPC(Hybrid Pulse Power Characterization) 실험을 통하여 비가역 발열의 파라미터인 SOC 5%별 내부 저항을 추출하였다. 추출된 SOC 5%별 저항을 이용하여 1C-rate에서 3C-rate로 변화하는 조건에서 열 추정 성능을 확인하였다. 높은 C-rate로 방전 전류가 변화하는 상황에서 발열 시뮬레이션과 실험값을 비교하였으며, 1C-rate의 HPPC 실험에서 얻어진 내부 저항이 부하의 변동에 따른 리튬이온 배터리의 발열 추정 파라미터로써 사용될 수 있음을 검증하였다.

  • 본 논문에서는 원통형 NCA 리튬이온 배터리로 제작된 고출력 직렬조합 배터리팩으로 C-rate에 따른 전기적 특성 실험을 수행하였다. 방전 용량 프로파일 실험을 통해 배터리팩의 충/방전 C-rate가 배터리팩의 내부 파라미터에 어떠한 영향을 주는지 비교 분석한다. 실험을 통해 방전 용량과 전압 편차 파라미터를 측정한다. 전압 편차 그래프는 만방 구간과 만방 이후 휴지 구간의 셀 간 전압 편차를 중심으로 관찰하며 두 구간의 전압 편차를 비교한다. 3가지 파라미터를 비교하여 가장 효율적인 C-rate를 알아보았다.

Modeling for Drying of Thin Layer of Native Cassava Starch in Tray Dryer

  • Purpose: The drying of a thin layer of native cassava starch in a tray dryer was modeled to establish an equation for predicting the drying behavior under given conditions. Methods: Drying tests were performed using samples of native cassava starch over a temperature range of $40-60^{\circ}C$. We investigated the variation in the drying time, dynamic equilibrium moisture content, drying rate period, critical moisture content, and effective diffusivity of the starch with temperature. The starch diffusion coefficient and drying activation energy were determined. A modification of the model developed by Hii et al. was devised and tested alongside fourteen other models. Results: For starch with an initial moisture content of 82% (db), the drying time and dynamic equilibrium moisture content decreased as the temperature increased. The constant drying rate phase preceded the falling rate phase between $40-55^{\circ}C$. Drying at $60^{\circ}C$ occurred only in the falling rate phase. The critical moisture content was observed in the $40-55^{\circ}C$ range and increased with the temperature. The effective diffusivity of the starch increased as the drying temperature increased from 40 to $60^{\circ}C$. The modified Hii et al. model produced randomized residual plots, the highest $R^2$, and the lowest standard error of estimates. Conclusions: Drying time decreased linearly with an increase in the temperature, while the decrease in the moisture content was linear between $40-55^{\circ}C$. The constant drying rate phase occurred without any period of induction over a temperature range of $40-55^{\circ}C$ prior to the falling rate period, while drying at $60^{\circ}C$ took place only in the falling rate phase. The effective diffusivity had an Arrhenius relationship with the temperature. The modified Hii et al. model proved to be optimum for predicting the drying behavior of the starch in the tray dryer.

냉동 및 해동속도가 우육표면 대장균군의 반치사적 손상율에 미치는 영향 (the Effect of Freezing and Thawing Rates on the Percentage of Sub-lethally Injured Total Coliform on Beef Surface)

  • Most of meat spoilage bacteria area Gram negative, which are very sensitive to freezing ; for instance , 90% of E. coli cells are killed or sub-lethally injured by freezing at -3$0^{\circ}C$, and the freeze-injury rate is dependent upon freezing rate. Since the injured bacterial cells are sensitive to selective agents, they fail to multiply in selective media. Injured bacterial cells are, however, capable of spontaneous repair at appropriate environmental and nutritional conditions . Enumeration of injured bacterial cells involves artificial induction of repair at these conditions. Cubic beef samples(3$\times$3$\times$3cm) were frozen at -6$0^{\circ}C$, -4$0^{\circ}C$, or -18$^{\circ}C$. The samples frozen at each temperature were thawed at 4$^{\circ}C$, 2$0^{\circ}C$, or by microwave . After these respective freezing an thawing treatments, the percentage of sub-lethally injured total coliforms out of total surviving ones was measured and compared. The results were as follows: 1. The interaction between freezing and thawing on injury rate was not significant. 2. The injury rates(as means of all three thawing treatments post-freezing) by freezing at -6$0^{\circ}C$, -4$0^{\circ}C$, or -18$^{\circ}C$ were 32.2$^{\circ}C$ and 19.2$^{\circ}C$ respectively . 3. The injury rates(as means of all three freezing treatments)by thawing at 4$^{\circ}C$, 2$0^{\circ}C$, or by microwave were 49.3%, 11.7% and 21.0% respectively. The highest injury rate was caused by freezing at -6$0^{\circ}C$ and subsequent thawing at 4$^{\circ}C$. However since the injury rates by freezing treatment were not significantly different, freezing at -18$^{\circ}C$ and subsequent thawing at 4$^{\circ}C$ can also be recommended , from an economic perspective.

온도, 광세기 및 pH에 따른 Chlorella Vulgaris 증식률 (Effect of Temperature, Light Intensity and pH on the Growth Rate of Chlorella Vulgaris)

  • 본 연구는 온도, 광세기 및 pH가 Chlorella vulgaris (C. vulgaris)의 증식률에 미치는 영향을 알아보고자 하였다. 각 조건에서의 효율을 평가하기 위하여 C. vulgaris (FC-16) ($3{\sim}8{\mu}m$)를 Jaworski's Medium에 증식시킨 뒤, 다양한 온도($10^{\circ}C$, $15^{\circ}C$, $20^{\circ}C$, $25^{\circ}C$, $30^{\circ}C$, $35^{\circ}C$)와 다양한 광세기($100{\sim}800{\mu}Em^{-2}s^{-1}$) 및 다양한 pH (3, 4, 5, 6, 7, 7.5, 9) 조건에서 실험하였다. 실험결과, $25{\sim}30^{\circ}C$ 조건에서는 $10^{\circ}C$ 조건과 비교 시 같은 양의 C. vulgaris를 증식하는데 걸리는 시간은 약 5.2~5.5배 빨랐고, Chlorophyll a는 5~5.5배 높았으며, 단위 면적당 cell volume은 14% 이상 높아 C. vulgaris 최적 배양온도는 $25{\sim}30^{\circ}C$로 조사되었다. C. vulgaris 배양기간의 경우, 증식속도가 5일까지는 느리게 증가했지만, 6일 이후부터 15일까지는 폭발적으로 증가했고, 15일 이후에는 거의 증식이 멈춰 15일 이내가 적당한 것을 알 수 있었다. C. vulgaris 증식을 위한 최적 pH는 pH 7~7.5로 조사되었다.
