Effects of Oral Administration of Ligigeopoongsan on Neuropathic Pain and c-Fos Protein Expression in Rats (이기거풍산(理氣祛風散)이 백서(白鼠)의 신경병리성(神經病理性) 동통(疼痛) 억제(抑制) 및 c-Fos 단백(蛋白) 발현(發顯)에 미치는 영향(影響))

  • Objective : We have studied to know effects of Ligigeopoongsan(LGS, 理氣祛風散) on mechanical allodynia, cold allodynia and c-Fos protein expression in the model of neuropathic pain of rats. Methods : The model of neuropathic pain was made by injured tibial nerve and sural never while common peroneal never was maintained. After 2weeks, we performed behavioral test for 7 days to try out mechanical allodynia using von frey filament and cold allodynia using acetone, which are calculated by counting withdrawal response on foot. Rat brains removed and sliced on 8th days. Serial sections were immunohistochemically reacted with polyclonal c-Fos antibody. The numbers of c-Fos protein immunoreactive neurons in the central gray were examined using scion image program. Results : 1. Mechanical allodynia in LGS-2, LGS-3 groups were significantly diminished compared with the control group. 2. Cold allodynia in LGS-3 group was significantly diminished compared with the control group. 3. c-Fos protein expression on the central gray LGS-2, LGS-3 groups were significantly lower than that of control group. conclusions : We have noticed that LGS(理氣祛風散) diminished mechanical and cold allodynia in the model of neuropathic pain compared with the control group. c-Fos protein expression in the central gray of that group was also decreased compared with the control group. Pain control group were LGS was accumulated time goes by. This study can be used as a basic resource on a study and a treatment of pain.

Effects of Herbal Bath on Functional Recovery and c-Fos Expression in the Ventrolateral Periaqueductal Gray Region of the Brain after Sciatic Crushed-Nerve Injury in Rats

  • Peripheral nerve injuries are a commonly encountered clinical problem and often result in chronic pain and severe functional deficits. At the Dept. of Oriental Rehab. Medicine, we have used for pain control a herbal bath containing the following herbs: Harpagophytum radix, Atractylodes japonica and Corydalis tuber. In the present study, we investigated the effects of this herbal bath on the recovery rate of the locomotor function and the expression of c-Fos in the ventrolateral periaqueductal gray (vIPAG) region of the brain following sciatic crushed nerve injury in rats. In the present study, characteristic gait change with decreasing of the sciatic function index (SFI) was observed and c-Fos expression in the vIPAG was suppressed following sciatic crushed nerve injury in rats. Immersion into herbal bath enhanced SFI value and restored c-Fos expression in the vIPAG to the control value. These results suggest the herbal bath might activate neurons in the vIPAG, and could facilitate functional recovery from peripheral nerve injury.

  • PURPOSE: The purpose of this study was to determine the effect of different frequencies (4Hz and 100Hz) of transcutaneous electrical nerve simulation (TENS) on pain relief using c-fos expression in the spinal cord of rat osteoarthritis to investigate the appropriate frequency for pain relief. METHODS: Total of 30 Sprague-Dawley rats was used and randomly divided 2 groups according TENS frequency and applicate the TENS during 3 period (3 days, 7 days, 10 days). The induction of osteoarthritis by 3mg monosodium iodoacetat was injected into the right knee joint of rats. Three days later, commercially available TENS unit was used for stimulation was set to 20minutes on 3, 7, 10 days after surgery. Western blot analysis system was used to detect immunoreactive proteins. The thickness of the bands were photographically measured by Scion Image. RESULTS: When investigating the c-fos expression of TENS on spinal cord in OA knee over 10 days, between-groups differences in c-fos expression reached a significant level by day 10. For within-groups comparisons, the c-fos expression decreased significantly across days in low- and high-frequency TENS groups. CONCLUSION: Whether at low- and high-frequency, the TENS as a therapy obtained beneficial effects of pain relief and TNES at high-frequency is more beneficial effects on the pain relief when TENS applied at injury site.

Effect of Daekumeumja Herb-acupuncture on c-Fos Expression in Hippocampus of Alcohol Intoxicated Rats (대금음자 약침이 알코올 독성 흰쥐의 해마에서 c-Fos 발현에 미치는 영향)

  • Objectives : The study was conducted to investigate the effect of Deakumeumja herb-acupuncture on c-Fos expression in each area of the hippocampus of Alcohol intoxicated rats. Methods : Experimental groups were divided into five groups ; normal group, the alcohol-treated (control) group, the alcohol- 1 mg/kg Daekumeumja treated (sample A) group, the alcohol- 5 mg/kg Daekumeumja treated (sample B) group, the alcohol- 10 mg/kg Daekumeumja treated (sample C) group (n = 6 for each group). Rats of normal group were injected intraperitoneally with saline once a day for 5 consecutive days, while animals of the alcohol-treated (control) group were injected once a day with 2 g/kg of alcohol for the same duration of time. Animals of the alcohol and Daekumeumja-treated (sample A, B, C) groups were acupunctured at Chung-wan$(CV_{12})$ with 2 g/kg of alcohol and the appropriate amount of Daekumeumja extract once a day for 5 days. Each groups was evaluated by the changes of c-fos-positive neurons in each area of the hippocampus by using an image analyzer and microscope. Results: 1. In the CAI region of the hippocampus, the number of Fos-positive cells in the sample B, C groups were significantly increased compared with the control group. 2. In the CA2-3 regions of the hippocampus, the number of Fos-positive cells of the sample B, C groups were significantly increased compared with the control group. 3. In the Dentate gyrus region of the hippocampus, the number of Fos-positive cells of the sample C group was significantly increased compared with the control group. Conclusion : c-fos expression in each area of the hippocampus was reduced in alcohol-intoxicated groups. Treatment of Daekumeumja increased this reduction. In conclusion, it can be suggested that Daedumeumja possesses protective effects of the amnesia and learning disability in alcoholism.

Effect of Puerariae Radix on c-Fos and c-Jun Expressions in Ischemic Damaged Hippocampus of Rats (갈근이 뇌허혈 손상 흰쥐의 뇌해마 c-Fos와 c-Jun 발현에 미치는 영향)

  • Objective : This study investigated a neuroprotective effect of Puerariae Radix on cerebral ischemia. Method : The global cerebral ischemia was induced by bilateral common carotid arteries occlusion under hypotension (40mmHg) in Sprague-Dawley rats. After the treatment of Puerariae Radix extract, changes of c-Fos and c-Jun expressions, immediate early genes expressed by cerebral ischemia, in the hippocampus were observed immunohistochemically. Result: The results obtained are as follows; The significant increases of c-Fos and c-Jun expressions were observed in the hippocampus of the ischemic damaged rat brains. Then Puerariae Radix treatment demonstrated significant decreases of c-Fos and c-Jun expressions in CA1 region and dentate gyrus as compared with control group. On the upregulated c-Fos expression induced by cerebral ischemia, Puerariae Radix treatment demonstrated significant decreases of c-Fos expressions in CA1 region (P<0.01) and dentate gyrus (P<0.05) as compared to the control group, but there were not a significant changes in CA2 and CA3 regions of the hippocampus. On the upregulated c-Jun expression induced by cerebral ischemia, Puerariae Radix treatment demonstrated significant decrease of c-Jun expression in CA1 region (P<0.05) as compared to the control group, but there were not a significant changes in CA2, CA3, and dentate gyrus of the hippocampus. Conclusion : These results suggested that Puerariae Radix reveals the neuroprotective effect through the reduction of immediate early genes, c-Fos and c-Jun, induced by cerebral ischemia.

Effect of Hyeongbangdojeok-san on Acute Cocaine-induced Behavioral Effect and Immediate Early Gene Expression in Rats. (형방도적산(荊防導赤散)이 급성코카인 투여로 인해 유도된 흰쥐의 행동량과 c-Fos 발현에 미치는 영향)

  • 1. Objectives The present study was designed to investigate the effect of Soyangin Hyeongbangdojeok-san(HBDJS) on acute cocaine-induced behavior effect and gene expression in the rat brain. 2. Methods Experimental animals were composed of saline(SAL), cocaine(COC), HBDJS + COC, HBDJS + SAL group. Rats received HBDJS(100, 200 mg/kg, p.o.) 1 h prior to cocaine hydrochloride(20 mg/kg, i.p.) treatment respectively. After cocaine injection, locomotor activity and rearing were measured in a rectangular container equipped with a video camera above the center of the floor for 60 min. In addiction, c-Fos expression in the rat brain was detected using immunohistochemistry 2 h after cocaine injection. And the effect of HBDJS on acute cocaine-induced pERK, pElk, pCREB upstream of c-Fos expression was detected using western blotting and immunohistochemistry 15 min after cocaine challenge. 3. Results The present results show that HBDJS at dose of 200 mg/kg attenuated cocaine-induced both locomotor activity and rearing. Also HBDJS at dose of 200 mg/kg significantly decreased c-Fos expression in the rat brain(nucleus accumebns and striatum). However HBDJS at dose of 200 mg/kg have no effect on cocaine-induced pERK, pCREB, pElK-1 expression. HBDJS is c-Fos expression through ERK-independent pathway. 4. Conclusions. These results suggest that HBDJS may be effective in suppressing the reinforcing effects of cocaine.

Effects of Rhei Rhizoma on c-fos and c-jun Expressions in the Hippocampus of Old BCAO Rats (대황(大黃)이 뇌허혈 유발 노령(老齡) 흰쥐의 해마 c-fos 및 c-jun 발현에 미치는 영향)

  • Objective : In this study old BCAO rats were observed for effects of 'Dea-Hwang' on brain ischema injury, because risk of stroke increases with age. Method : The brain ischema injury was induced by temporarily closing carotids on both sides in a low blood pressuer state and Dea-Hwang was administered orally to 18 month-old BCAO rats. Results and Conclusions : The ischemically damaged Hippocampus and c-fos and c-jun expression were analyzed by immunohistochemical staining and results are summarized as follows: 1. The c-fos expression after inducing a brain ischema injury in the hippocampus was more inhibited in the dosed group than in the control group. 2. The normalized optical density of c-fos expression was more reduced in the CA1, CA2, and DG areas of the dosed group than in those of the control group. 3. The c-jun expression after inducing brain ischema injury in the hippocampus was more inhibited in the dosed group than in the control group. 4. The normalized optical density of c-jun expression was more reduced in the CAI area of the dosed group than in that of the control group.

Effect of Electrical Stimulation of the Vestibular System on Vestibuloocular Reflex and c-Fos Expression in the Medial Vestibular Nuclei of Unilateral Labyrinthectomized Rats (일측 전정기관 손상 흰쥐에서 전정안구반사와 내측전정신경핵의 c-Fos 단백질발현에 대한 전정기관의 전기자극 효과)

  • Unilateral labyrinthectorny (ULX) causes autonomic symptoms, ocular and postural asymmetries, which disappear over tune in the process of equilibrium recovery known as vestibular compensation. In the present study in order to elucidate mechanisms responsible for the effects of electrical stimulation on vestibular compensation and investigate the relationship between vestibular compensation and c-Fos expression in the medial vestibular nuclei following ULX, we measured spontaneous nystagmus, eye movement induced by sinusoidal rotation and c-Fos expression or to 72 hs after ULX in Sprague-Dawley rats. Experimental animals were divided into two groups: ULX group with ULX only, and electrical stimulation (ES) group with electrical stimulation of $-2{\sim}-5V$, 1.0ms, 100 Hz to the lesioned vestibular system for 4 hs/day. Spontaneous nystagmus following ULX disappeared by 72 hs in ULX group and 36 hs in ES group. In eye movement induced by sinusoidal rotation, normal pattern of eye movement by rotation toward the lesioned side was recovered 24 hs after ULX at rotation of 0.1 Hz and 6 hs after at 0.2 Hz, 0.5 Hz in ULX group. In ES group, the eye movement recovered after 12 hs at 0.1 Hz, 6 hs at 0.2 Hz, and 4 hs at 0.5 Hz. Directional Preponderance which represents the symmetry of bilateral vestibular functions showed significantly early recovery in ES group compared with that of ULX group. Expression or c-Fos immunoreactive cells in the bilateral medial vestibular nuclei was severely asymmetrical till 36 hs in ULX group, and then it became a symmetry and disappeared after 72 hs. However, ES group showed the symmetry of c-Fos expression after 6 hs, which was significantly early recovery in 25 group. All these findings suggest that electrical stimulation ameliorates recovery of vestibuloocular reflex following ULX by the restoration of the balance of the resting activity between bilateral medial vestibular nuclei. In addition, c-Fos expression in the medial vestibular nuclei could be used as a marker of vestibular compensation since c-Fos expression is closely related to the course of recovery following ULX.

Pharmacopuncture of Anti-inflammatory Herbal Compounds Suppresses Colon Inflammation-induced c-Fos like Protein Expression in Rats (소염(消炎) 약침(藥鍼)이 대장염 유발 흰쥐의 c-Fos 단백 발현에 미치는 효과)

  • Objectives: Colitis is an inflammatory bowel disease characterized by colonic mucosal inflammation and chronic relapsing events represents. The purpose of this study is to evaluate effects of pharmacopuncture of anti-inflammatory herbal compound (AiC) applied to the different acupoints in the acute colitis induced by trinitrobenzenesulphonic acid (TNBS) intracolonic injection in rats. Methods: In Male Sprague - Dawley rats, weighing 250~400g, TNBS (5 mg/kg) was infused intrarectally through a silicon rubber catheter into the anus under isoflurane anaesthesia. Acupoints of LI4 (Hapkok), ST25 (Cheonchu), ST36 (Joksamni), and BL25 (Daejangsu) were intramuscularly injected by AiC, respectively (injection volume & times: 0.2 ml / acupoint, twice times on the 2nd & 3rd day). Expressions of cFos protein in the periaqueductal gray (PAG), locus coeruleus (LC), nucleus of solitary tract (Sol), and the 6th lumbar spinal cord (L6 s.c.) were observed at 24 hr after TNBS induced colitis by immunohistochemistry. Results: The expression of c-Fos protein in the L6 s.c., Sol, LC and PAG increased 24 hr after TNBS injection into colorectum as compared to normal and 50% ethanol treated group. AiC to LI4 inhibited the expression of c-Fos protein in Sol and PAG but not L6 s.c. and LC. AiC to ST36 showed significant inhibition the c-Fos expression in L6 s.c., Sol and PAG. AiC to ST25 only showed the effects in L6 s.c. and PAG. AiC to BL25 inhibited significantly the expression of c-Fos protein all over the areas. To investigate whether or not endogenous opioids are involved, intrathecal injection of naltrexone (30ug/30ul) was applied before the 2nd pharmacopuncture treatment 24 hr after TNBS-induced colitis in rat. Naltrexone reversed the inhibition of c-Fos protein expression in the spinal cord and brainstem. Conclusions: These data show that pharmacopuncture of Aic potently inhibits signal pathways ascending hypersensitivity of colorectum after TNBS induced colitis and depends on the endogenous opioids according to acupoints.

Additive Role of the Vestibular End Organ and Baroreceptors on the Regulation of Blood Pressure in Rats

  • Contribution of the vestibular end organ to regulation of arterial pressure was quantitatively compared with the role of baroreceptors in terms of baroreflex sensitivity and c-Fos protein expression in the rostral ventrolateral medulla (RVLM). Baroreflex sensitivity and c-Fos protein expression in the RVLM were measured in conscious rats that had undergone bilateral labyrinthectomy (BL) and/or baroreceptor unloading. BL attenuated baroreflex sensitivity during intravenous infusion of sodium nitroprusside (SNP), but did not significantly affect the sensitivity following infusion of phenylephrine (PE). Baroreflex sensitivity became positive following sinoaortic denervation (SAD) during infusion of PE and attenuated sensitivity during infusion of SNP. Baroreflex sensitivity also became positive following double ablation (BL+SAD) during infusion of PE, and attenuated sensitivity during infusion of SNP. c-Fos protein expression increased significantly in the RVLM in the sham group after SNP administration. However, the BL, SAD, and SAD+BL groups showed significant decreases in c-Fos protein expression compared with that in the sham group. The SAD group showed more reduced c-Fos protein expression than that in the BL group, and the SAD+BL group showed less expression than that in the SAD group. These results suggest that the vestibular system cooperates with baroreceptors to maintain arterial pressure during hypotension but that baroreceptors regulate arterial pressure during both hypotension and hypertension. Additionally, afferent signals for maintaining blood pressure from the vestibular end organs and the baroreceptors may be integrated in the RVLM.
