Transport properties of carbide superconductor La2C3

  • We investigate the electrical and thermal transport properties of a sesquicarbide superconductor $La_2C_3$, including electrical resistivity, thermoelectric power, and thermal conductivity. The electrical resistivity exhibits a typical metallic character with a saturation behavior at high temperatures. The thermoelectric power shows a metallic behavior with pronounced phonon-drag effect, comparable with pure metals. The broad peak of the thermal conductivity is observed in the superconducting state, which is rapidly suppressed by magnetic fields. These observations suggest that the electron-phonon scattering is significant in $La_2C_3$, which is relevant with the relatively high-$T_c$ in $La_2C_3$ through strong electron-phonon coupling with low frequency phonon modes.

Mutation of a Transposed Amino Acid Triplet Repeat Enhances Coupling of m1 Muscarinic Receptor to Activation of Phospholipase C

  • The C-terminus ends of the second putative transmembrane domains of both m1 and m2 muscarinic receptors contain a triplet of amino acid residues consisting of leucine (L), tyrosine (Y) and threonine (T), This triplet is repeated as LYT-LYT in m2 receptors at the interface between the second transmembrane domain and the first extracellular loop. Interestingly, however, it is repeated in a transposed fashion (LYT-TYL) in the sequence of m1 receptors. In this work we employed site-directed mutagenesis to investigate the possible significance of this unique sequence diversity for determining the distinct differential drug-receptor interaction and cellular function at m1 muscarinic receptor. Mutation of the LYTTYL sequence of m1 receptors to the corresponding m2 receptor LYTLYT sequence, however, did not result in a significant change in the binding affinity of the agonist carbachol or in the affinity of the majority of a series of receptor antagonists which are able to discriminate between wild-type m1 and m2 receptors. Surprisingly, the LYTLYT ml receptor mutant demonstrated markedly enhanced coupling to activation of phospholipase C without a change in its coupling to increased cyclic AMP formation. There was also an enhanced receptor sensitivity in transducing elevation of intracellular Ca$\^$2+/. These changes were not due to alterations in the rate of receptor. desensitization or sequestration, On the other hand, the reverse LYTLYT-LYTTYL mutation in the m2 receptor did not alter its coupling to inhibition of adenylate cyclase, but slightly enhanced its coupling to stimulation of PI hydrolysis, Our data suggest that the LYTTYL/LYTLYT sequence difference between ml and n12 muscarinic receptors is not involved in determining receptor pharmacology. On the other hand, while these differences might play a role in the modulation of muscarinic receptor coupling to PI hydrolysis, they are not important for specifying coupling of various subtypes of muscarinic receptors to different cellular signaling pathways.

Coupling Efficiencies of m1, m3 and m5 Muscarinic Receptors to the Stimulation of Neuronal Nitric Oxide Synthase

  • Through molecular cloning, five muscarinic receptors have been identified. The muscarinic receptors can be generally grouped according to their coupling to either stimulation of phospholipase C (m1, m3, and m5) or the inhibition of adenylate cyclase (m2 and m4). Each m1, m3, and m5 receptors has the additional potential to couple to the activation of phospholipase A$_2$, C, and D, tyrosine kinase, and the mobilization of Ca$\^$2+/. However, the differences in coupling efficiencies to different second messenger systems between these receptors have not been studied well. Ectopic expression of each of these receptors in mammalian cells has provided the opportunity to evaluate the signal transduction of each in some detail. In this work we compared the coupling efficiencies of the m1, m3 and m5 muscarinic receptors expressed in chinese hamster ovary (CHO) cells to the Ca$\^$2+/ mobilization and the stimulation of neuronal nitric oxide synthase (nNOS). Because G protein/PLC/PI turnover/[(Ca$\^$2+/])i/NOS pathway was supposed as a main pathway for the production of nitric oxide via muscarinic receptors, we studied on ml, m3 and m5 receptors. Stimulation of guanylate cyclase activity in detector neuroblastoma cells was used as an index of generation nitric oxide (NO) in CHO cells. The agonist carbachol increased the cGMP formation and the intracellular [Ca$\^$2+/] in concentration dependent manner in three types of receptors and the increased cGMP formation was significantly attenuated by scavenger of NO or inhibitor of NOS. m5 receptors was most efficiently coupled to stimulation of nNOS, And, the coupling efficiencies to the stimulation of neuronal nitric oxide synthase in three types of receptors were parallel with them to the Ca$\^$2+/ mobilization.

시클로 덱스티린과 그 메틸유도체의 구조분석 (Conformational Analysis of Cyclodextrins and Their Methylated Analogs)

  • ${\alpha}$-와 ${\beta}$-시클로덱스트린과 그의 메틸유도체들을 Chemical shift(${\delta}$)와 Coupling constant(J)를 470 MHz $^1H$ NMR을 이용해서 수용액 안에서 분석하였다. 정확한 ${\delta}$와 J값들을 얻기 위해서 Raccoon spin simulation program을 이용해서 실험 data를 분석하였다. 시클로덱스트린의 $C_{5-}C_6$ bond 주위의 rotamer의 분포를 $J_{56a}$$J_{56b}$값을 이용하여 실험 data를 분석하였다. 위 계산에 의해 ${\alpha}$-와 ${\beta}$-시클로덱스트린에서는 gg conformer가 가장 많이 존재하였고 tg conformer가 가장 적게 존재했다. 그러나 그 메틸 유도체에서는 gg conformer가 더 많이 증가하였고 gt conformer가 가장 적게 존재함을 알았다.

금속 산화물 촉매를 이용한 메탄의 Oxidative Coupling 반응 (Oxidative Coupling of Methane by Metal Oxide Catalysts)

  • 메탄의 직접 전환 기술 중 하나인 OCM(oxidative coupling of methane) 반응을 수행하였다. 사용한 금속 산화물 촉매는 Li/MgO와 Pb/MgO이었다. 온도에 따른 촉매의 반응성을 알아보기 위해 600, 700, $800^{\circ}C$에서 반응을 행하였으며, 반응물 혼합비(메탄:산소)에 따른 반응성, 전환율 및 선택도를 알아보기 위해 $700^{\circ}C$에서 혼합비를 2:1 및 1:1로 하여 실험을 하였다. 그 결과 7wt% Li/MgO 촉매의 경우 반응온도 $700^{\circ}C$이고 혼합비가 2:1일 때 메탄의 전환율과 $C_2$화합물의 선택도가 각각 20%, 65% 정도로서 우수한 반응성을 보임을 알 수 있었다. 7wt% Li/MgO 촉매의 경우 $700^{\circ}C$에서 혼합비가 1:1일 때 메탄의 전환율은 30%로 증가했으나 $C_2$화합물의 선택도는 45%로 감소하였다. Pb/MgO 촉매는 Li/MgO 보다 낮은 선택도를(25%) 나타내었다.

  • NaCl(30wt%)/ZnO(60wt%)/${\alpha}-Al_2O_3$ 촉매상에서 메탄의 oxidative coupling 반응의 속도식을 연구하여 활성 산소종에 관하여 고찰하였다. 반응온도 $650^{\circ}C$에서 $750^{\circ}C$까지 메탄의 전화율 10%미만의 범위에서 메탄과 산소의 분압을 변화시켜 가면서 메탄의 전환속도를 측정하여 속도식을 검증하였다. 제안된 메틸라디칼의 생성반응은 Langmuir-Hinshelwood형 반응기구를 따른다. 촉매표면의 서로 다른 활성점에 흡착된 메탄분자와 산소분자가 반응하여 메틸라디칼이 생성되는 반응이 속도결정단계이며, 이때 활성화 에너지는 약 39kcal/mol이었다. 메탄의 C-H 결합의 해리에 관여하는 산소종은 표면상의 이원자 산소인 $O{_2}{^{2-}}$$O_2{^-}$로 제시할 수 있었다.

이중상 구조를 가진 보강성 충전제의 물리적 특성 연구 (A Study on the Physical Properties of Reinforcing Fillers with Dual Phase Structure)

  • 본 연구의 목적은 순수 카본블랙과 이중상 구조의 카본블랙 (DPCB)이 함유되어 있는 고무조성물의 물리적 특성에 대하여 조사하였다. DPCB는 FT-IR분석결과 1100~1200 파장에서 실리카 표면에 존재하는 Si-O 피크를 나타냈다. DPCB와 유기 실란커프링제 (Si69)를 함유하는 고무조성물의 가황속도는 순수 카본블랙을 함유한 고무조성물에 비해 지연되었다. Si69를 함유한 DPCB의 300% 모듈러스와 상호작용계수 ($a_f$)는 순수 카본블랙보다 높게 나타났으며, 내마모특성 지표인 피코 무게감소량은 일정하게 나타났다. DPCB의 $0^{\circ}C$에서의 tan ${\delta}$는 DPCB 50 phr 기준 실란커플링제를 2.0% 사용한 수준에서 순수 카본블랙보다 우수하였으며, $60^{\circ}C$에서의 tan ${\delta}$는 실란커프링제의 사용량이 증가 할수륵 감소하였다. 결국 본 연구에 사용된 DPCB는 일정한 마모저항을 유지하면서 회전저항을 개선하는데 적합하다고 생각한다.

  • $PbTiO_3$는 perovskite구조를 가진 강유전체로써 Curie온도가 높고 유전율이 작아 고온. 고주파재료로 주목되어왔으나 curie온도 이하에서 결정이방성(C/a)이 크기 때문에 소결 후 결정립(grain)이 입계(grain boundary)에서 분리되어 치밀하고 기계적으로 강한 세라믹을 얻을 수 없었고, 항전력이 커 분극이 어려위 실용화 되지 않았다. 본 연구에서는 $PbTiO_3$에 Pb일부를 Ca로 치환하고 Mn, Ni, Co및 W의 산화물을 첨가하여 치수가 크고 기계적으로 강한 세라믹을 제조하였고, 그 분극 조건도 PZT의 분극 조건과 같은 정도로 개선하였다. 그리고 결합계수이방성(Kt/Kp)이 $5\sim6$으로 크고, 기계적 품질계수 Qm이 $310\sim480$ 정도로 낮으며, 유전상수가 200대의 값으로 PZT보다 1/5정도 작은 고온, 고주파. 광대역 초음파 발진자 재료로 적합한 압전세라믹을 제조 연구하였다.

Differential Coupling of G$\alpha$q Family of G-protein to Muscarinic $M_1$ Receptor and Neurokinin-2-Receptor

  • The ligand binding signals to a wide variety of seven transmembrane cell surface receptors are transduced into intracellular signals through heterotrimeric G-proteins. Recently, there have been reports which show diverse coupling patterns of ligand-activated receptors to the members of Gq family $\alpha$ subunits. In order to shed some light on these complex signal processing networks, interactions between G$\alpha$q family of G protein and neurokinin-2 receptor as well as muscarinic M$_{1}$ receptor, which are considered to be new thearpeutic targets in asthma, were studied. Using washed membranes from Cos-7 cells co-transfected with different G.alpha.q and receptor cDNAs, the receptors were stimulated with various concentrations of carbachol and neurokinin A and the agonist-dependent release of [$^3H$]inositol phosphates through phospholipase C beta-1 activation was measured. Differential coupling of Gaq family of G-protein to muscarinic M$_{1}$ receptor and neurokinin-2 receptor was observed. The neurokinin-2 receptor shows a ligand-mediated response in membranes co-transfected with G$\alpha$q, G$\alpha$11 and G$\alpha$14 but not G$\alpha$16 and the ability of the muscarinic $M_1$ receptor to activate phospholipase C through G$\alpha$/11 but not G$\alpha$14 and G$\alpha$16 was demonstrated. Clearly G$\alpha$/11 can couple $\M_1$ and neurokinin-2 receptor to activate phospholipase C. But, there are differences in the relative coupling of the G$\alpha$14 and G$\alpha$16 subunits to these receptors.

Dynamic Mechanical and Morphological Properties of Natural Rubber Vulcanizates Containing Fillers

  • In order to examine correlation between silane coupling agent and activator which affects the physical properties of silica filled natural rubber compound, such as heat buildup, rebound property, dynamic mechanical and morphological properties were measured. With incorporation of silane coupling agent, $tan{\delta}$ at $0^{\circ}C$ was increased and at $60^{\circ}C$ was decreased, which resulted in improving of wet grip and rolling resistance. When silane coupling agent which is corresponding to 8~10% of silica was used, most favorable heat build up and $tan{\delta}$ was obtained. And also when activator was used, dispersibility of silica was improved as a result of reduction of reaggregation of silica.
