Model for Cancer Cachexia using C26 Adenocarcinoma-Induced Wasting Syndrome for Newer Therapeutic Approach (새로운 치료 방법 접근을 위한 C26 선암세포 기반의 Cancer Cachexia 동물모델 수립)

  • Background: Cachexia is a multi-factorial syndrome presenting with chronic illness, decreases in body weight, and loss of adipose tissue and skeletal muscle, mostly in patients with advanced cancer and chronic wasting disease. Even after years of intensive researches, there remains no convincing therapy to prevent cancer cachexia. Methods: In this in vivo study, we have established C26 adenocarcinoma-induced cancer cachexia model in mice to explore the underlying core changes in cytokine, signal transduction, and muscle wasting. The ultimate aim of establishing animal model is to find optimal therapeutics to mitigate cancer cachexia. Results: We have administered C26 adenocarcinoma cells onto BALB/c mice and observed 4 weeks to assess the progression of cancer cachexia. Significant loss of weight accompanied with loss of appetite was noted. As C26 adenocarcinoma xenograft progressed, mortality was started from 3 weeks, accompanied with significant sarcopenia and decreased mice movement. Surges in TNF-α and IL-6 were noted with the commencement of cancer cachexia. Conclusion: Using C26 adenocarcinoma cancer cachexia model, we can screen the optimal therapeutics to mitigate cancer cachexia, in which agents to modulate IL-6, TNF-α, and NF-κB were essential.

Therapeutic Potency of N-(Phosphonacetyl)-L-Aspartic Acid in Liposome in Established Tumor Bearing Mice (진행된 암 동물모델에서의 리포좀 포집 PALA의 항암 치료 효과)

  • Previously, we have reported an antitumor efficacy of liposomal N-(phosphon-acetyl)-L-aspartic acid (or PALA) in C-26 tumor bearing Balb/c mice, where PALA in liposome was administered one day after tumor inoculation. In this report, we have investigated the therapeutic potency of liposomal formulation of PALA, which was administered eight days after tumor inoculation in the same C-26 tumor bearing mice. The C-26 murine colon tumor inoculated mice were randomized for the in vivo therapy and the survival was measured after a single intraperitoneal injection of the drug. When the therapy was initiated eight days after tumor inoculation, DSPC-PALA at 150 mg/kg resulted in a significant increase in median survival time (MST) of 56% over the control group which received MES/HEPES buffer alone. However, none of the free PALA and DSPG-PALA liposome doses caused a statistically significant increase in MST over control group at the 95% confidence level. At 750 mg/kg dose, free PALA caused a marginally significant improvement in MST by 34%, but both 375 mg/kg and 150 mg/kg doses of free PALA caused only a 2% and a 4% increase in MST, respectively. These results show that PALA in neutrally charged liposome can exhibit considerably greater potency than free PALA in established C-26 tumor bearing mice.

The effect of post-incubation period and temperature treatment on the uniform primordia formation of Pleurotus ostreatus (느타리버섯 후배양 기간과 온도처리가 발이 균일도에 미치는 영향)

  • The study was conducted to investigate the effects of post-incubation period and temperature treatment conditions during incubation on the uniform primordia formation and cultural characteristics of oyster mushroom (Pleurotus ostreatus). Three kinds post-incubation period; 25, 30, 35 days and control were applied for 30 days while two kinds incubation room temperature $23^{\circ}C$ and $26^{\circ}C$ and control were used $20^{\circ}C$. The substrate temperature during pre-incubation was of 'Suhan No. 1' and 'Gonji No. 7'. Oyster mushroom varieties tended to increase between $24^{\circ}C$ to $26^{\circ}C$ at 11 to 15 days after inoculation and then they were maintained in treatment temperature during post -incubation period. The $CO_2$ occurrence was at the highest at 6,500 ppm for 'Suhan No. 1' and 5,800 ppm for 'Gonji No. 7' at the time of the highest temperature increase. The ratio of un-uniformal primordia formation and the ratio of non-commercial fruit body were reduced by 40%, 10.5%, respectively compared to control for 'Suhan No. 1' when in the post-incubation temperature was $26^{\circ}C$, and incubated for 10 days and 15 days treatment. Also, 'Gonji No. 7' was reduced by 19%, 9.5%, respectively when in the post-incubation temperature was $26^{\circ}C$, and incubated for 10 days treatment. Therefore, the higher post-incubation temperature of room and longer post-incubation period resulted in the higher percentage of primordia formation of two cultivars.

Predation of Asterias amurensis and Asterina pectinifera on Valuable Bivalves at Different Water Temperature (유용 양식패류에 대한 아무르불가사리(Asterias amurensis)와 벨불가사리 (Asterina pectinifera)의 수온별 포식 특성)

  • Effect of predation of starfish Asterias amurensis and Asterina pectinifera under different water temperature in the recirculating seawater vessel was conducted at the invertebrate culture laboratory of Yosu National University from February 1 to February 25, 2000. The highest activity of Asterias amurensis and Asterina pectinifera according to water temperature was obtained at 14$\^{C}$ and 22$\^{C}$. The average individual numbers of Anadara broughtonii predated by Asterias amurensis and Asterina pectinifera were 2.20 and 1.60 at 6$\^{C}$ , 3.60 and 2.00 at 10$\^{C}$, 7.20 and 2.50 at 14$\^{C}$, 3.40 and 6.80 at 22$\^{C}$, 0.80 and 3.90 at 26$\^{C}$, respectively.

Physiological Response of parrot fish (Oplegnathus fasciantus) and bivalve (Gomphina melanaegis) by Lowing Water Temperature Exposure (저수온 노출에 따른 돌돔(Oplegnathus fasciantus)과 민들조개(Gomphina melanaegis)의 생리활성 변화 연구)

  • Physiological response of fish and bivalve was estimated to identify the physiological changes of test species by lowering water temperature due to the abrupt stop of cooling water discharge from power plant. The experiment was conducted by two conditions; fall and winter by decreasing water temperature ($2^{\circ}C$/2 days) from $26^{\circ}C$ to $17^{\circ}C$ for fall scenario and from $15^{\circ}C$ to $9^{\circ}C$ for winter scenario, respectively. Test organisms were parrot fish (Oplegnathus fasciantus) and bivalve (Gomphina melanaegis), and end points were mortality for both species, hematocrit and cortisol for fish, and hemolymph and superoxide dismutase(SOD) for bivalve. 48/96hr mortality test revealed no mortality for fish and 47% mortality for bivalve at 96hr/$26^{\circ}C$ only. Significant increases of hematocrit and cortisol were found at fishes exposed to $26^{\circ}C$ (high temperature) and lower temperature ($9{\sim}13^{\circ}C$), respectively. Hemolymph and SOD for bivalve tended to decrease by lowering water temperature from 15 to $9^{\circ}C$ (winter scenario) and no changes from 26 to $17^{\circ}C$ (fall scenario). Fall scenario (from 15 to $9^{\circ}C$) showed more significant changes of physiological response than winter cases (26 to $17^{\circ}C$).

Effect of Ripening Temperatures on Incidences of Postharvest Fruit Rots of Kiwifruits (후숙 온도가 참다래 저장병 발병에 미치는 영향)

  • This study was conducted to identify optimum ripening condition for kiwifruits (Actinidia deliciosa) to prevent postharvest fruit rots caused by Botryosphaeria dothidea, Diaporthe actinidiae and Botrytis cinerea. The optimum temperatures for mycelial growth of B. dothidea, D. actinidiae and B. cinerea were $26{\sim}35^{\circ}C$, $26{\sim}29^{\circ}C$ and $20{\sim}26^{\circ}C$, respectively, and the incidence was closely related with the temperature. Although kiwifruits ripened faster at higher temperatures, the rates of diseased fruits increased with the rates of ripened fruits increased. Optimum conditions for ripening of kiwifruit were 20-day at 17C.

Comparison of Brain Uptakes for Brain Drug Delivery Vector Synthesized by Chemical and Genetical Engineering Method (화학적 및 유전공학적으로 제조한 뇌송달 벡터의 뇌수송량 비교)

  • Drug delivery to the brain may be achieved by producing chimeric peptide, attaching the drug to protein 'vectors' which are transported into the brain from the blood by a receptor-mediated transcytosis through the blood-brain barrier (BBB). Since the BBB expresses high concentrations of transferrin receptor, and it was reported that anti-transferrin receptor mouse monoclonal antibody (OX26) undergoes transcytosis through the BBB, it is logical to assume that a drug delivery system via transferrin receptor-mediated transcytosis is a promising strategy. In the present study, therefore, we tested feasibility of several OX26 based vectors for the brain delivery of a model drug. Avidin-based delivery vectors such as OX26-streptavidin (OX26-SA), OX26-neutralite avidin (OX26-NLA) were chemically synthesized vectors and OX26 immunoglobulin G 3 type $C_{H}3$ fusion avidin $(OX26\;IgG3C_H3-AV)$ was genetically engineered. To improve the efficiency of producing chimeric peptide, we used avidin-biotin technology. Pharmacokinetics of $[^3H]biotin$ bound to OX26-SA, OX26-NLA and $OX26\;IgG3C_H3-AV$ was determined by intravenous injection technique, and their stabilities in plasma were analyzed using HPLC. The brain delivery of $[^3H]biotin$ bound to OX26-SA, OX26-NLA and OX26\;$IgG3C_{H}3-AV$ (expressed as %ID/g brain) was $0.22{\pm}0.01$, $0.18{\pm}0.01$ and $0.25{\pm}0.09$, respectively. The areas under the plasma concentration versus time curve (AUC) for OX26-SA, OX26-NLA, $OX26\;IgG3C_H3-AV$ from time zero to 60 min were $209{\pm}10$, $195{\pm}9$, $134{\pm}29\;%ID\;min/ml$ respectively and their total clearances $(CL_{tot})$ were $1.00{\pm}0.09$, $1.08{\pm}0.07$ and $1.54{\pm}0.29\;ml/min/kg$, espectively. These results showed that these vectors possess preferable pharmaceutical (e.g., resonable stability) and pharmacokinetics (e.g., significant brain uptake and enhanced AUC) for brain delivery. Therefore, these vectors may be broadly useful in the brain delivery of drugs that are not transported into the brain to a significant extent.

Growth and Production of Resting Eggs of Freshwater Rotifer, Brachionus calyciflorus Pallas at the Different Temperatures (수온에 따른 담수산 Rotifer, Brachionus calyciflorus Pallas의 성장과 내구란 생산)

  • The specific growth rate and production of resting ega of the freshwater rotifer, Brachionus calyciflorus Pallas was investigated at the different temperatures ($20\~30^{\circ}C$). The rotifer was cultured in 250 ml flask and fed on concentrated freshwater Chlorella. Specific growth rate of B. calyciflorus showed higher rate at higher temperature, but maximum density was observed higher at lower temperature, expect at $20^{\circ}C$. The production of resting egg with temperature was showed decrease on the basis on $26^{\circ}C$. The highest number of resting egg per ml and rotifer $10^4$ were 157 eggs and 810 eggs at $26^{\circ}C$, respectively. This result shows that the optimum temperature for mass culture and resting egg production of this freshwater rotifer were $30^{\circ}C$ and $26^{\circ}C$, respectively.

  • Emergence and growth of Potamogeton distinctus, Eleocharis kuroguwai, and Sagittaria pygmaea were examined at soil temperatures of 12, 15, 18, 22, 26, and $30^{\circ}C$ and 3 planting depth conditions. Optimum soil temperatures for emergence were 15-$22^{\circ}C$ 22-$26^{\circ}C$, and 18-$26^{\circ}C$ for P. distinctus, E. kurogueai, and S. pygmaea, respectively. Emergence was delayed and growth became poor in three species as planting depths were increased and this trend was more significant in E. kuroguwai.
