Removal Characteristics of Ethyl Acetate and 2-Butanol by a Biofilter Packed with Jeju Scoria

  • The removal characteristics of ethyl acetate and 2-butanol were investigated in a bench-scale down-flow biofilter packed with Jeju scoria medium. Various inlet concentrations and gas flow rates were tested. The adaptation times of microorganisms to the change of the influent concentration of ethyl acetate and 2-butanol gas were found to be about 3 days. At the inlet concentration of 300 ppmv and empty bed contact time (EBCT) of 15 see, the removal efficiencies of the biofilter for ethyl acetate and 2-butanol were above $99.9\%$. The maximum removal capacity of the biofilter for ethyl acetate was $316-318\;g/m^3/h$ and that for 2-butanol was $245-251\;g/m^3/h$. Overall, the removal capacity of the biofilter for ethyl acetate was $50-70\;g/m^3/h$ larger than that for 2-butanol. During the operation of 65 days, the pressure drop through the biofilter column was maintained below $13\;mmH_{2}O/m$. Although the pH in the drain water decreased from 7.2 to 5.0, the pH drop did not affect the removal of the gases. From the above results, the biofilter using Jeju scoria as a packing material seemed to very effectively treat waste gases such as ethyl acetate and 2-butanol.

Taheebo 분획물의 의염 및 위궤양에 대한 효과 (Antigastritic and Antiulcer Actions of the Fraction of Taheebo)

  • In a preliminary screening of plant extracts for the antigastritic and antiulcer actions in rats, the methanol extracts of Taheebo showed positive activity in HCI . ethanol-induced gastric lesion. Among the systematic fractions of hexane, chloroform, butanol and water, the most potent $H_2O$ fraction reduced significantly HCI . ethanol-induced gastric lesion at the oral dose of 300 mg/kg. In pylorus ligated rats chloroform and butanol fraction showed decreases in the volume of gastric secretion and acid output of which effects were stronger in chloroform fraction. Further assays with hexane butanol and $H_2O$ fraction disclosed that it significantly suppressed the aspirin-induced ulcer. The butanol fraction reduced significantly acetic acid induced ulcer at the dose of 400 mg/kg. The butanol and $H_2O$ fraction reduced the malondialdehyde level in HCI . ethanol-induced gastric lesion. In pylorus ligated rats, chloroform and butanol fraction reduced the malondialdehyde level and in aspirin-induced ulcer, chloroform fraction reduced that levle. These results might suggest that the butanol and $H_2O$ fraction of Taheebo had inhibitory action in gastric lesion and ulceration through inhibition of gastric acid secretion and the decrease malondialdehyde level.

마우스의 아세트알데히드 대사에 미치는 인삼 부탄올 분획의 영향 (The Effect of Ginseng Butanol Fraction on the Acetaldehyde Metabolism in Mice)

  • The present study was undertaken to investigate the possible effect of ginseng butanol fraction on the hepatic acetaldehyde metabolism. Experimental animals were used for the subject of the study. When, in case of mitochondrial aldehyde dehydrogenase (Ald DH), ginseng butanol fraction was added, enzyme activity was increased in a small dose, while, in a large dose, it showed inhibitory effect. In terms of kinetic aspect, ginseng butanol fraction has the effect to decrease the Km values of Ald DH. In vivo studies, the activity of Aid DH increased by induction of acute intoxication of ethanol was further increased through pretreatment with ginseng butanol fraction. When ginseng butanol fraction was given to mice fed with 5% ethanol instead of water for 60 days, the activity of Ald DH in mitochondrial fraction decreased to about 35% in chronic alcoholism, but after pretreatment of ginseng butanol fraction the activity was restored to the control level. By the pretreatment with disulfiram, the Ald DH activity was inhibited in normal and alcohol-treated groups, but after the treatment with ginseng butanol fraction the activity was restored to the control level. The results suggest that ginseng butanol fraction enhance the Ald DH activity inhibited by the treatment of disulfiram with no relation to NAD. It was observed that ginseng butanol fraction markedly decrease the acetaldehyde levels in plasma and liver. All these observations suggested that reduction of acetaldehyde in blood and liver should be dependent upon increased activity of mitochonclrial Ald DH. It is concluded that the recovery from alcohol intoxication should be prompted by treatment with ginseng.

부자(附子) Butanol fraction의 강심작용(强心作用)에 관한 연구(硏究) (Studies of the actions of Aconiti tuber butanol fraction on the mechanical and electrical properties of isolated rabbit atrium)

  • In Chinese medicine, it is said that Aconiti tuber has cardiotonic, diuretic and analgesic effects. Kim et al reported that alkaloid free part of Aconiti tuber, $CHCI_3$ insoluble fraction, showed inotropic effect on isolated frog heart and inotropic effect is potenciated by n-butanol fractionation. To investigate the effect of Aconiti tuber butanol fraction on the mechanical and electrical properties of heart, change of active tension, excitability and refractory period of isolated rabbit atrium in the presence of butanol fraction were measured and the comparison with that of ouabain and quinidine was done. The observed results are as follows. 1. $5{\times}10^{-4}g/ml$ concentration of Aconiti tuber butanol fraction showed approximately same effect with therapeutic concentration of ouabain on the increment of contractile force, and the effect of $2{\times}10^{-3}g/ml$ was greater than that of $1{\times}10^{-5}g/ml$ of ouabain. 2. Acceleration of rate of contractile force increment in the presence of Aconiti tuber butanol fraction was greater than in ouabain, and the time to maximum tension was shorter in Aconiti tuber butanol fraction than in ouabain. 3. The excitability of isolated atrium was slightly increased at low concentration of Aconiti tuber butanol fraction, while decreased at higher concentration. 4. Aconiti tuber butanol fraction slightly prolonged refractory period of isolated right atrium at the concentration of $2{\times}10^{-3}g/ml$.

Properties of the Microinterface formed by Phosphatidylcholine and 1-Butanol as Reaction Media of Hydrolysis of Phosphatidylcholine

  • Microinterface of W/Omicroemulsion prepared by phosphatidylcholine was used as reaction media of hydrolysis of phosphatidylcholine by phospholipaseA$_2$. Phosphatidylcholine was used as an amphiphile and was acted as a substrate. Organic phase of W/Omicroemulsion in this study was prepared by mixed organic solvents i.e. 2,2,4-trimethylpentane (isooctane) as a main solvent and 1-butanol as a co-solvent. The effect of added 1-butanol was remarkable not only on reaction beginning but also on high reaction rate. The hydrolysis reaction was dramatically initiated when 1-butanol was injected into the running isooctane/PC system. The enhancement by 1-butanol addition into single organic solvent was our original finding compare with previous conventional organic solvent. The reaction rate was elevated by the added amount of 1-butanol. The enhanced reaction rate was about 150-folds. This enhancement was speculated as 1-butanol adsorption on the microinterface. The adsorbed 1-butanol improved the properties of microinterface, especially its mobility was increased by difference of the chain length between phosphatidylcholine and 1-butanol. PhospholipaseA$_2$ molecules were located on the microinterface due to modified mobility of microinterface. Located phospholipaseA$_2$ on the microinterface reacted easily with phosphatidylcholine molecule. As a result high reaction rate was obtained. Microinterfacial properties were successfully improved by adsorbed 1-butanol molecule, and were favorable to appear higher reactivity of phospholipaseA$_2$.

  • An extractive fermentation process using pervaporation was studied in a 7 liter fermentor. Pervaporation was performed using a silicone membrane module and a low-acid-producing strain Clostridium acetobutylicu, B18 was used to produce butanol. In batch culture without pervaporation pH 5.5 and initial glucose concentration of 60 g/L resulted in the highest butanol productivity (0.216 g/L$.$h) with butanol yield of 0.261 Butanol flux through the membrane was best at 2.0 L/min-tubing of air flow rate In batch and fed-batch fermentation glucose consumption rate increased by 1.3 times with pervaporation.

오매의 Butanol추출물이 Salmonella typhimurium의 생육에 미치는 영향 (Effect of Butanol Extracts from Prunus mume on the Growth of Salmonella typhimurium)

  • The antimicrobial properties of extracts of Prunus mume were tested on Salmonella paratyphimurium and S. typhimurium. First, the Prumus mume was extracted with methanol at several temperatures, and then fractionation of the methanol extracts from Prunus mume was carried out by using petroleum ether, chloroform, ethyl acetate or butanol. Secondly, absorption column chromatoraphy (using a Diaion HP 20) was conducted to eliminate some water soluble materials that might inhibit the antimicrobial activity of some extracts. The antimicrobial activitry of each of the Prunus mume extracts was determined using a paper disc method against several food-borne pathogens, The growth inhibition curve was determine using butanol extracts of Prunus mume against Salmonella typhimurium. The extraction temperature did not have any significant effect on the yield of the extract or on the level of antimicrobial activity. The butanol extract of Prunus mume showed strong antimicrobial activity against Salmonella paratyphimurium and S. typhimurium; a 1,000 ppm of butanol extract of Prunus mume retared the growth of S. typhimurium up to 36 hours.   • Suitable design and operation of distillation process is very dependent on vapor-liquid equilibrium calculation. The usual calculation method is use binary interaction parameter. Flash points of n-propanol+n-butanol and 2-butanol+n-butanol were measured by Seta-flash closed cup tester. Experimental Flash points were compared with those calculated by the method based on Raoult's law and the optimization method using Wilson equation. The binary interaction parameters obtained by the optimization method are then used to calculate the bubble points of n-propanol+n-butanol and 2-butanol+n-butanol.

  • Objective: The aim of this study is to investigate the effect of Injin butanol fractions with Thin Layer Chromatography on Fas-mediated Apoptosis. Method: Injin-butanol fraction separated by TLC. MIT assay, cell cycle analysis, Caspase-3 protease assay, DNA fragmentation assay and quantitative RT-PCR were performed to evaluate the effects of TLC extraction of lnjin-butanol fraction on cell viability, cell cycle progression and apoptosis. Results: Scopoletin, luteolin, apigenin and unknown powder was isolated by TLC. Fas-mediated apoptosis analysis shows that scopoletin has inhibiting function on apoptosis. Caspase- 3 protease assay analysis shows that scopoletin inhibits activity of caspase-3. Quantitative RT-PCR analysis shows that no activity on caspase-3, but apoptosis inhibition cytokine -Bcl-2- is activated, and apoptosis activating cytokine -Bax- is unactivated. Conclusion: These results show that each fraction of Injin-butanol TLC extraction, especially scopoletin, acts as a protective function on liver cell viability, and inhibitory function on apoptosis.   • The present study was undertaken in order to elucidate the effect of ginseng butanol fraction on ethanol induced hepatic aniline hydroxylase activity in rat. Ginseng butanol fraction increased the hepatic aniline hydroxylase activity which is inhibited by ethanol addition in the enzyme assay system, whereas not shown the ginseng effect in ethanol absence condition in vitro. It was found that ginseng butanol fraction improved the affinity of aniline hydroxylase under presence of ethanol in the reaction mixture. On the contrary ginseng butanol fraction showed significant decreasing effect on aniline hydroxylase activity induced by ethanol administration. These results suggest that ginseng butanol fraction regulate the hepatic aniline hydroxylase activity which is induced by ethanol consumption.
