Plant Regeneration from Immature Embryo and Bulb Scale Tissue of Hippeastrum hybridum (아마릴리스의 미숙배와 인편조직으로부터 식물체 재분화)

  • Immature hybrid embryos of H. hybridum, 'Picottee', 'White Christmas', 'Eldorado', 'Origin', 'Red Lion', 'elstar', 'Crypsy' were cultured on the MS medium supplemented with various concentrations of 2,4-D, NAA, BA and TDZ. Among the treatments, NAA were more effective for the shoot regeneration and bulblet formation than other treatment. Addition of 0.5 ㎎/L NAA was effective for bulblet induction from explant Shoot regeneration was most effective on the medium with 1.0㎎/L NAA and 2.0 ㎎/L TDZ. The addition of 1.0-2.0㎎/L TDZ induced numerous shoots per explant but strongly inhibited root development when compared to 1.0-2.0㎎/L BA. When bulb scale segments of 'Star Van Holland' was incubated, bulblet formation was the most effective on MS medium with 0.5㎎/L NAA.

Multiple Shoots Regeneration and In vitro Bulblet Formation from Garlic Callus

  • The leaf segments of garlic (Allium sativum L.) were cultured in vitro and determined optimal concentration of plant growth regulators and sugars for callus induction, multiple shoots regeneration and in vitro bulblet formation. Highest yield of callus was observed in the leaf segment culture on Murashige and Skoog (MS) medium supplemented with 1.0 mg/L 2,4-D, 30 g/L sucrose and 8 g/L agar. Regeneration rate of multiple shoots from callus was high in the MS medium supplemented with kinetin 3.0 + NAA 3.0 mg/L or SA 1.0 + NAA 3.0 mg/L, containing 30 g/L sucrose. High rate of bulblet formation was observed as the concentration of jasmonic acid increased from 0.5 to 2.0 mg/L in medium, whereas addition of gibberellic acid significantly suppressed bulblet formation. The rate of in vitro bulbing was as high as $96\%$ in MS medium supplemented with 2.0 mg/L jasmonic acid and 120 g/l sucrose after two month culture at $25{\pm}1^{\circ}C$ under 16 hours day light.

  • The effect of abscisic acid (ABA) and methyl jasmonate (MeJA) on bulblet formation from the culture of Allium victorialis var. platyphyllum Makino was studied. Shoot clumps were cultured on MS medium containing ABA (0, 0.01∼2.0mg/L) and MeJA (0.01∼5.0mg/L). ABA at low concentrations (0.01mg/L) induced shoot proliferation without bulblet formation. However, bulblet formation started on the medium containing MeJA approximately in 4-6 weeks of culture. Furthermore, 1.0mg/L MeJA resulted in bulblet formation at high frequency (100%) as compare to the control (46.1%). Cortical cells of the bulblets enlarged on medium with MeJA had dense protein-like substance in expanded and round cells when examined under the microscope. The data described here show that formation and enlargement of bulblets from Allium victorialis can be improved by addition of appropriate concentration of MeJA.

Stimulation of In Vitro Bulblet Growth by the Addition of Liquid Medium in Lilium Oriental Hybrid 'Casablanca'

  • The bulb scales and shoot sections ($7\;\cal{mm}\;\times\;15\;\cal{mm}$) of Lilium oriental hybrid 'Casablanca' were cultured to compare bulblet growth in vitro. Shoots were induced from in vitro grown bulbscales on MS medium with $1.0\;\cal{mg/L}\;BA,\;0.5\;\cal{mg/L}$ IAA, and 30 g/L sucrose. The regenerated shoots were cut into shoot sections, and cultured on MS medium with $2.0\;\cal{mg/L}\;BA,\;0.5\;\cal{mg/L}$ IAA and 30 g/L sucrose for shoot proliferation. Culture of shoot sections stimulated bulblet growth significantly than the bulb scales on MS medium with 60 g/L sucrose. However, the bulblets from shoot sections did not reach ideal size to produce stems with several leaves. Therefore, liquid medium was added into the same vessels to stimulate bulblet growth further. After shoot sections were cultured on MS medium with 60 g/L sucrose and 2 g/L activated charcoal for two months in dark, $20\;\cal{ml}$ liquid media containing various concentrations of sucrose and MS salts were added. Two months later, the added liquid medium stimulated bulblet growth remarkably as compared to bulblets grown without added liquid medium. The added $25\;\cal{ml}$ liquid medium containing 120 g/L sucrose and double strength of MS salts were the most effective for growth of in vitro bulblets. More than $94\%$ bulblets produced by this method sprouted stems with several leaves after cold treatment at $5^{\circ}C$ for three months.

Effects of Growth Regulators and Sucrose Concentrations on the Bulblet Formation through In Vitro Culture of Scale Segment in Nerine bowdenii (네리네(Nerine bowdenii)의 기내 인편배양시 자구형성에 미치는 생장조절제와 Sucrose 농도의 영향)

  • The twin-scale segments of nerine (Nerine bowdenii) were cultured to investigate the influence of NAA, BA and sucrose concentrations on in vitro bulblet formation. The formation of bulblets from twin-scale segments showed a good response both the percentage of bulblet formation and the number of bulblets per explant on MS medium supplemented with 1mg/L NAA and 2 mg/L BA. Formation of bulblet showed the highest efficiency on medium containing 30g/L, and the formation of bulblets was strongly inhibited on medium containing over 90g/L. When the twin-scale segments formed bulblets were subcultured three times to the same medium by 60 day subculture interval, the number of bulblets per explant was 6.5, 7.3 and 8.2 in order of first, second and third. The bulblets over 3mm in diameter were hypertrophied and rooted after transferring to the hormone-free MS medium. The plantlets over 50mm in height were successfully acclimatized in the soil mixed with the same volume of vermiculite and perlite, and the survival rate was over 95%.

Efficient Bulblet Regeneration and Growth from Bulb Scale of Hyacinthus orientalis L. cv. Pink Pearl Cultured in vitro (히아신스(cv. Pink Pearl)의 인편 기내 배양시 효과적인 자구의 재생과 생장)

  • The regeneration and growth of bulblets from the bulb scale segments of Hyacinthus orientalis L. cv. Pink Pearl were more efficient in IBA than IAA at the same concentrations (1.0 and 3.0 mg/l). The normal (base-down) orientation of explants was more effective for bulblet regeneration and root growth than the inverted (base-up) orientation. The growth of bulblets and roots was increased higher in the perlite than the agar medium. These results suggested that the alternate culture system, first cultured in the agar medium for bulblet regeneration, and then in the perlite medium for bulblet growth, may be more useful for efficient in vitro culture of hyacinth (H. orientalils) cv. Pink Pearl.

Micropropagation through Stem, Node-bud Shoot Tip and Bulblet Scale Culture in Fritillaria thunbergii Miq. (패모의 줄기, 마디, 정단 및 자구인편 배양에 의한 기내 증식)

  • This experiment was carried out to establish micropropagation system in Fritillaria thunbergii Miq. Through the culture of bulblet scales, stems, node-buds and shoot tips with special reference to the effect of physiological age of explant and plant growth regulators on bulblet formation. Number of formed bulblets was significantly increased in node-bud or stem tissue compared to scals segments and on the medium supplemented with kinetin than BA containing medium. Optimum levels of kinetin for bulblet formation from node-bud taken from above 3 cm shoot length and stem segments excised from below 3 cm shoot length were 5.0 mg /L and $1.0{\sim}3.0\;mg$ /L kinetin, respectively. Interesting phenomenon was observed, the direct formation of bulblets from the axilliary bud of cultured explants. Bulblet forming capacity in stem tissue was depended on stem age, young stem had high regeneration ability compared to old stem taken from above 10 cm shoot length. 1.0 mg /L kinetin was optimum concentration for the formation of bulblets from old stem segments. Stem tissue taken from underground growing plant was promoted coampare to shoot tips or bulb scale segments. Optimum concentration of sucrose was $5{\sim}7%$. Summariged above results revealed that effective explant for micropropagation was stem and /or node-bud tissue excised from less than 3 cm plant height compared to those of bulb scale segments which showed high contamination after culture. Maximum multiplication rate of young stem and /or node-bud segment was about 20 times. Kinetin requirement for stimulation of bulblet formation from cultured explant depended on source of explants but favorable levels of kinetin for organogenesis ranged from 1.0 mg /L to 5.0 mg /L.

Effect of Explant and Cultivars on the Adventitious Shoot Differentiation by Invitro Culture of Narcissus (배양재료와 품종이 수선의 기내배양시 부정아 형성에 미치는 영향)

  • In order to establish a micropropagation system of Naricissus, the ability of bulblet regeneration among propagation materials was compared, and the adequate growth regulators and concentrations for each cultivar were investigated. The inorganic components were also assayed in the parts of propagation materials. In propagation materials, scape with based plates showed hightest rate of bulblet formation and rapid growth of formed bulblets in vitro, comparing to other parts of it. In comparing of varieties, 'Dutch Master' and 'Golden Harvest' showed a high ability for bulblet regeneration. The ability of bulblet regeneration was most favorable in the medium, supplemented with 5.0 mg/L BA and 2.5 mg/L NAA in 'Dutch Master', and 5.0 mg/L BA and 1.0mg/L NAA in 'Golden Harvest', respectively. In inorganic component analysis of propagation materials, the White part of scape contained 1.18 mg/L$P_2O_5$, 2.57 me Ca, 0.94 me Mg and 3.20 mg/L total N. It showed higher levels in concentration of inorganic components as compared to those of the other part of scape. In addition, leaves and yellow part of scape contained significantly high levels of Ca and Mg while scales bulb showed considerably low levels in all inorganic compounds.

High Efficiency Bioreactor Culture System for Mass Proliferation and Bulblet Formation of Allium victorialis var. platyphyllum Makino (산마늘 다신초 증식과 인경형성에 효율적인 생물반응기 배양방식)

  • A suitable bioreactor culture system for shoot proliferation and bulblet formation of Allium victorialis var. platyphyllum Makino was established. Uptake of soluble carbohydrates in different bioreactor culture systems was also analyzed during the entire culture period. Optimal conditions for multiple shoot formation were determined in raft culture (RC) and modified raft culture system (MRC) (13-15 per explant) in which the explants were placed on a net contacting liquid medium. For bulblet formation and enlargement, 93.4% of shoot clumps formed bulblets at the basal part. Furthermore, they were uniform in size when cultured with ebb & flood system (E&FS). Bulblets harvested from RC and MRC showed vigorous rooting, however, their growth was not uniform. Whereas soluble carbohydrate contents in the bulblets cultured in E&FS were low, starch content was high. Sucrose, glucose and fructose concentrations in the medium of E&FS culture system decreased as bulblet formation and enlargement proceeded, suggesting that external sucrose is taken up to by the cells before it is hydrolyzed.

  • This experiment was conducted to clarify the effects of physiological properties in culture media on the proliferation and the acclimation of bulblets of several kinds of lilies cultured in vitro. Proliferation rate of bulblets from bulblet of Oriental hybrid ‘Marco Polo’, ‘Casa Blanca’ and Asiatic hybrid ‘Jolanda’ was higher in solid MS media compared with bulblets from scale Acclimation rate of regenerated bulblets was high in solid media which was pre-cultured in vitro, compared with liquid media. Sprouting rate of bulblets from bulblet was higher than bulblets from scale.
