Studies on the Antimicrobial Activities of Prunus mume (매실의 항미생물 활성 연구)

  • This study was conducted to investigate the antimicrobial activity of Prunus mume extracts. Prunus mume extracts have growth inhibitory activity to various microorganisms. Gram positive bacteria such as Bacillus cereus, Bacillus subtilis, Micrococcus leteus, Staphylococcus epidrimidis were more easily inhibited than Gram negative bacteria tested Vibrio parahaemolyticus, Salmonella typhimurium, Escherichia coli, Proteus vulgaris and Yeasts by Prunus mume extracts. Among several fractions of methanol extract, EtOAc & BuOH fractions were showed strong antibacterial activities, but those fractions were not showed on fungi.

Free Radical Scavenging, Cytotoxic Effects, and Flavonoid Content of Fractions from Leaves of Lycopus lucidus Turcz. (택란 잎 추출물의 라디칼 소거활성과 세포독성효과 및 플라보노이드 함량)

  • The free radical scavenging, cytotoxic effects, and flavonoid content of fractions from Lycopus lucidus Turcz leaves were here investigated. The flavonoid contents of 85% methanol (MeOH) and n-butanol (BuOH) fractions of the leaves were 41.5 mg/g and 77.2 mg/g, respectively. In DPPH and ABTs+ assays, 85% MeOH and n-BuOH fractions from the L. lucidus Turcz leaves had a greater scavenging effect (p<0.05). The n-BuOH fraction (0.5 mg/ml concentration) had scavenging effects of 88% and 92% in the DPPH and ABTs+ assays, respectively (p<0.05). Cell viability tests showed that treatment with L. lucidus Turcz leaf fractions caused cytotoxicity in the growth of AGS, HT-29, and HT-1080 cancer cells. Of the different fractions, the 85% MeOH sample displayed the highest cytotoxic activity; the $IC_{50}$ values of this fraction against AGS, HT-1080, and HT-29 cancer cells were 0.03 mg/ml, 0.14 mg/ml, and 0.16 mg/ml, respectively. These biological results indicate that the n-BuOH fraction was more effective in anti-oxidant activity while the 85% MeOH fraction was stronger in cytotoxic effects, and they suggest that these two fractions from L. lucidus Turcz leaves may contain valuable bioactive compounds, such as flavonoids.

  • This research was performed to identify the anti-inflammatory constituent from the herb of Lycorus lucidus (Lamiaceae). The MeOH extract of this plant material and its two fractions, the lipophilic- ($CHCl_3$ fraction) and the hydrophilic fraction (BuOH fraction), were prepared to test anti-inflammatory activity. For this purpose, the inhibition rate on inducible nitric oxide synthase (iNOS) activity was assessed by determining nitric oxide (NO) formation in lipopolysaccharide (LPS)-induced macrophage 264.7 cells. The $CHCl_3$ fraction that greatly inhibited nitric NO formation was chromatographed to lead the isolation of ursolic acid. Since ursolic acid inhibited NO formation dose dependently in this study, this compound was considered as one of the active constituent responsible for anti-inflammation. However, the activity of rosmarinic acid isolated from the BuOH fraction was weak.

Anti-wrinkle effects of extracts and solvent fractions from Nymphoides peltata on CCD-986sk (CCD-986sk 세포내에서의 노랑어리연꽃 추출물 및 분획물의 주름개선 효과)

  • The purpose of this study was to investigate the antiaging effect in the fibroblasts by using the extracts and fractions of Nymphoides peltata for the development of new functional cosmetic materials. The anti-aging effects of the extracts and fractions of N. peltata were measured using CCD-986sk cells. Ethyl aceate (EA) fraction inhibited UVB-induced reactive oxygen species expression by more than 50%, and n-butyl alcohol (BuOH) fraction increased expression of pro-collagen mRNA by more than 50% at $25{\mu}g/mL$. In addition, EA and BuOH fractions inhibited MMP-1 protein and mRNA expression in a concentration-dependent manner, BuOH fraction inhibited MMP-1 mRNA expression by more than 40% at $10{\mu}g/mL$. Especially, the high efficacy of EA and BuOH of the N. peltata was confirmed. Therefore, the EA and BuOH of N. peltata are expected to be useful as anti-aging materials in cosmetics field.

Antioxidant Activity of N-hydroxyethyl Adenosine from Isaria sinclairii

  • The antioxidant activity of Isaria (Paecilomyces) sinclairii was determined by measuring its radical scavenging effect on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radicals. The n- BuOH extract of P. sinclairii showed strong scavenging activity to DPPH. The anti-oxidant potential of the individual fraction was in the order of ethylacetate> n- BuOH>chloroform>n-hexane. The n-BuOH soluble fraction exhibiting strong anti-oxidant activity was further purified by repeated silica gel column chromatography. N-(2-Hydroxyethyl)adenosine (HEA) was isolated as one of the active principles from the n-BuOH layer. The n-BuOH layer, particularly HEA, did not increase the level of nitric oxide (NO) production in vascular endothelial cells that might be related to vasorelaxation such as the action of viagra. In addition, the vascular endothelial growth factor (VEGF) levels showed little or no increase compared with control group with the treatment of I. sinclairii.

Biological Activity of Sorghum bicolor M. cv. Bulgeunjangmoksusu Extracts (붉은장목수수 추출물의 생리활성)

  • The objective of this study was to determine the biological activities of Sorglum bicolor extracts. Organic fractions, including n-Hexane, EtOAc, and n-BuOH fractions were obtained from the methanol extract of Sorglum bicolor M.. In DPPH radical scavenging activity, $SC_{50}$ values of methanol extract and EtOAc fraction were exhibited $0.66{\pm}0.26{\mu}g/mL$ and $1.03{\pm}0.02{\mu}g/mL$, respectively. Contents of total polyphenol and flavonoids in EtOAc fraction, which were much higher than those of other fractions, were 58.12 mg/g and 4.79 mg/g respectively. Also, effects of reducing power was strongly showed in EtOAc fraction. ${\alpha}$-Glucosidase and ${\alpha}$-amlyase inhibition activities were showed the higher effect in D.W. fraction ($2.83{\mu}g/mL$, $36.64{\mu}g/mL$). In MTT assay in the AGS, HT29 and HCT116 cell lines were significantly higher in the n-BuOH fraction than in the other fractions at $50{\mu}g/mL$ concentration of extracts.

Variation of Saponin Content in the Decoctions of Platycodi Radix (길경(桔梗)의 추출조건에 따른 사포닌 함량의 변화)

  • We investigated the contents of crude saponin in the decoctions of Platycodi Radix prepared by the different extraction processes. Each lyophilized decoctions prepared by the different protocol were suspended in water and partitioned with ethylacetate (EtOAc) and n-butanol (BuOH), which gave the EtOAc fraction, BuOH fraction and the remaining water fraction, respectively. The content of crude saponin, which was estimated as the amount of BuOH fraction, and the HPLC profiles of the BuOH fraction were analyzed, and compared each others. The crude saponin content in the decoctions were increased proportionally to the increment of the extraction time, amount of water, extraction temperature and the number of repetition for extraction. Consequently, the optimized conditions were deduced to be suitable and recommendable for the preparation of Platycodi Radix ; the water amount needed for the extraction be 15-20 volumes to crude material, temperature for extraction be optimal at $85{\sim}100^{\circ}C$, extraction time be more than 5 hours and the repetition of extraction was 2 times.

The Effects of Alisma canaliculatum Butanol Fraction with Selenium on Glycogen Level, Lipid Metabolism and Lipid Peroxidation in Streptozotocin-Induced Diabetic Rats (택사 Butanol 분획물과 Selenium 보충이 당뇨 흰쥐의 글리코겐 함량, 지질대사 및 지질과산화에 미치는 영향)

  • The purpose of this study was to investigate the effect of butanol (BuOH) fraction of Alisma canaliculatum (Ac) and/or selenium (Se) treatment on glycogen level, lipid metabolism and lipid peroxidation in streptozotocin (STZ)-induced diabetic rats. Male Sprague-Dawley rats were assigned to one of the five groups: normal, STZ-control, and three experimental groups (Ac group, Ac-Se group, and Se group). Diabetes was experimentally induced by intravenous administration of 45 mg/kg of STZ in citrate buffer. The BuOH fraction of Ac (400 mg/kg bw) was orally administered for 3 weeks. The Se group were fed a AIN-93 recommended diet mixed with Na$_2$SeO$_3$ (2 mg/kg diet). The liver glycogen level of Ac and Ac-Se groups were significantly higher, when compared with the STZ-control groups. The muscle glycogen level was not significantly differ among all groups. The levels of liver triglyceride were higher in Ac-Se group than the STZ-control group. Pancreas protein levels were significantly increased in Ac-Se group than STZ-control group. The concentration of liver malondialdehyde (MDA) was significantly decreased in Ac and Se groups and decreased in Ac-Se group. Administration of BuOH fraction of Alisma canaliculatum and selenium supplementation increased the liver glycogen and triglyceride levels, and reduced peroxidative liver damage in STZ induced diabetic rats. These results suggest that treatment with a BuOH fraction of Alisma canaliculatum in combination with selenium has no synergistic antioxidative effect. Selenium supplementation may lead a decrease MDA of liver in diabetic rats.

Anti-inflammatory and Analgesic Activities of the Extracts and Fractions of Cnidii Rhizoma (천궁 엑스 및 분획의 소염.진통작용)

  • Cnidii rhizoma is one of the most important crude drugs used particularly for the treatment of female genital inflammatory diseases in traditional oriental medicine. In this study, its anti-inflammatory and analgesic activities were examined employing animal models. It was found that $H_2O$ extract and n-BuOH fraction inhibited significantly the edema formation after the subplantar injection of carrageenin at oral doses of 100 and 200 mg/kg in a dose-dependent fashion, where MeOH extract showed significant anti-inflammatory activity at a oral dose of 200 mg/kg. In Freund's complete adjuvant-induced arthritis, H2O extract and n-BuOH fraction exerted their significant inhibitory activity on the edema formation at oral doses of 100 and 200 mg/kg. The $H_2O$ extract and n-BuOH fraction also showed significant analgesic activity in a dose-dependent manner in acetic acid-induced writhing test.
