Galangin 및 양강추출물의 $KBrO_3$ 유도 DNA 및 염색체 손상에 대한 보호효과 (Protective Effect of the 70% Ethanolic Extract of Alpinia officinarum and Galangin Against $KBrO_3$-induced DNA and Chromosomal Damage in Mice)

  • The aim of this study was to evaluate the in vivo effect of galangin and the 70% ethanolic extract of Alpinia officinarum (AO) toward $KBrO_3$-induced DNA and chromosomal damage in mice. Galangin and AO inhibited the formation of 8-hydroxy-2'-deoxyguanosine (8-OH2'dG) as an indicator of DNA oxidative damage in the liver cell. Galangin and AO showed the inhibitory effect on the formation of DNA single strand break in the splenocyte by single cell gel electrophoresis (SCGE) assay and also inhibited micronucleated reticulocyte (MNRET) formation of peripheral blood in tail blood of mice. Vit-E revealed antigenotoxic effects in DNA and chromosome levels, but galangin was more potent active compound compare to vit-E under our experimental conditions. The results suggest that the extract of Alpinia officinarum containing galangin can modify the oxidative DNA and chromosomal damage and may act as chemopreventive agent against oxidative stress in vivo.

음용수의 전기분해 소독과정에서의 소독부산물 생성특성 (Characteristics of the Disinfection Byproducts Formation in Electrolysis Disinfection of Drinking Water)

  • This study was conducted to examine the byproducts formation characteristics at the water treatment plants which applying electrolysis as a disinfection process in Gangwondo, Korea. Total of forty plants located in Gangwon Province, Korea were selected for the study. Correlation between dissolved organic carbon(DOC) and $SUVA_{254}$ was not clear. Among the species of the disinfection byproducts(DBPs), chlorate and trihalomethanes(THMs) accounted for 76% and 14% of DBPs, respectively. The effect of DOC or $SUVA_{254}$ on DBPs formation was not clearly demonstrated. The increased amount of THMs due to the raw water bromide content was found primarily in the form of chloroform, and the percent fraction of BDCM(bromodichloromethane) and DBCM(Dibromochloromethnae) was relatively insignificant. When the value of $SUVA_{254}$ was greater than $2L/mg{\cdot}m$, the percent fraction of BDCM and DBCM decreased while percent fraction of CF(chloroform) increased.

Antioxidative Constituents from Paeonia lactiflora

  • The ethanol extract of the peony root (Paeonia Lactiflora Pall, Paeoniaceae) as well as its major active components including gallic acid and methyl gallate were evaluated for their protective effects against free radical generation and lipid peroxidation. In addition, the protective effects against hydrogen peroxide-induced oxidative DNA damage in a mammalian cell line were examined. The ethanol extracts of the peony root (PREs) and its active constituents, gallic acid and methyl gallate, exhibited a significant free radical scavenging effect against 1,1-diphenyl-2-picryl hydrazine (DPPH) radical generation and had an inhibitory effect on lipid peroxidation, as measured by the level of malondialdehyde (MDA) formation. The PREs did not have any pro-oxidant effect. They strongly inhibited the hydrogen peroxide-induced DNA damage from NIH/3T3 fibroblasts, as assessed by single cell gel electrophoresis. Furthermore, the oral administration of 50% PRE (50% ethanol extract of peony root), gallic acid and methyl gallate potently inhibited the formation of micronucleated reticulocytes (MNRET) in the mouse peripheral blood induced by a $KBrO_3$ treatment in vivo. Therefore, PREs containing gallic acid and methyl gallate may be a useful antigenotoxic antioxidant by scavenging free radicals, inhibiting lipid peroxidation and protecting against oxidative DNA damage without exhibiting any pro-oxidant effect.

오존처리에 의한 의약품류의 제거와 미생물의 불활성화에 대한 연구 및 고찰 (Study on the Removal of Pharmaceuticals and Personal Care Products and Microorganism Inactivation by Ozonation)

  • 미량오염물질의 산화 및 대체 소독제로 각광받는 오존처리의 하수 2차 처리수중에 잔류하는 의약품류에 대한 제거 성능을 검토하였다. 또한, 의약품류의 제거를 목적으로 한 오존처리에 의한 미생물의 불활성화에 대하여 고찰하였다. 본 연구에서는 시험수로써 하수 2차 처리수를 이용하였으며, 오존처리는 2 mg/L, 4 mg/L, 6 mg/L의 오존 주입량으로 행하였다. 오존처리에 의해 시험수중에서 검출된 37종의 의약품류를 효과적으로 제거하기 위해서는 6 mg/L의 오존 주입량 (오존 소비량 : 4.4 mg/L)이 요구되었다. 동일한 오존처리 조건하에서는 대장균군 및 enteroviruses에 대해 약 3 log의 불활성화가 달성가능할 것으로 고찰되어, 잔류 의약품류의 제거 뿐만 아니라 병원성 미생물에 대해서도 효과적인 소독효과를 달성할 수 있을 것으로 판단되었다. 반면, 6 mg/L의 오존 주입량을 이용한 오존처리시, 처리수중의 용존오존농도가 약 1.8 mg/L까지 증가하여, 발암성 물질인 브로메이트의 생성가능성이 높아질 것으로 예상되었다. 이러한 브로메이트의 생성을 억제하기 위해서는 오존처리와 UV 또는 $H_2O_2$와의 조합공정인 고도산화처리공정에 대한 검토가 필요할 것으로 판단되었다.
