Accumulation of Xanthosine-5'-monophosphate by Adenine-Guanine Double Auxotroph of Brevibacterium ammoniagenes (Brevibacterium ammoniagenes 의 아데닌-구아닌 복영양요구주에(複營養要求株)에 의한 5'-크산틸산(酸)의 축적(蓄積))

  • An adenine-guanine doubless and $\beta$-alanine requiring mutant, D-1550-40, which had been derived from Brevibacterium ammoniagenes ATCC 6872, produced a copious amount of xanthosine-5'-monophosphate (XMP). The optimum concentration of adenine and guanine for maximal accumulation of XMP was about 75 ml/l and 100 ml/l for growth. Concentrations higher than 100 mg/l of adenine and guanine inhibited cell growth and XMP accumulation strongly. The inhibition, however, could be recovered by adding $100{\mu}g$ of biotin per liter or 0.3% of casamino acids to the culture solution. High concentrations of phosphate and magnesium salts (1.0 to 1.5%(w/v) in media) were found to be indispensable for XMP accumulation, and the presence of manganese in the culture medium stimulated both growth of cells and accumulation of XMP leaving 5'-inosinic acid unaffected. The maximal accumulation of XMP reached to 60.5 mg/l after 4 days of fermentation which had been started with a medium containing 100 mg of adenine-guanine, 5 mg of $MnSO_4{\cdot}H_2O$ and $100{\mu}g$ of biotin per liter. The specific XMP synthesis(mg of XMP/mg of cells) was increased with the increase of the cell growth rate.

  • 5'-Xanthylic acid 생성균주인 Brevibacterium ammoniagenes ATCC 21263 R에 XMP에서 GMP로의 전환효소인 GMP synthetase활성을 부여하기 위해 동종간 세포융합을 시도하여 융합균주들을 얻었다. 이들 우량 융합균주들과 융합모균의 GMP synthetase 활성을 측정하여 상호 비교하였으며, pH 변화에 따른 GMP synthetase 활성과 GMP 생성량과의 관계를 검토하였다. 또한 최적 pH에서 균성장에 따른 당소모량과 GMP 생성량을 비교하였다.

  • Growth responses of Brevibadterium ammoniagenes BA 17-2, which had been obtained by the treatment of several mutagens in our previous report, were investigated to select the preliminary optimal concentrations of phosphate, $Mg^{++}$, $Mn^{++}$ and thiamine for the production of 5'-XMP aminase. In this experiment it was shown that the concentration of phosphate in the medium has an important effect on the growth of microorganism. Using the medium containing 0.2% of $MgSO_4{\cdot}7H_2O$, 3mg/l of $MnSO_4{\cdot}H_2O$and $1\;mg/l$ of thiamine-HCl, the maximum cell mass was obtained at the concentration of 0.4% of $KH_2PO_4$ and $K_2HPO_4$, respectively. Above the concentration of these phosphates, cell growth was inhibited as the phosphate concentration increased to 1%, but the inhibition was overcome by the addition of 1% of $MgSO_4{\cdot}7H_2O$ and 3mg/l of thiamine-HCl. The 5'-XMP aminase activity was also influenced by the concentration of phosphate, $Mg^{++}$, $Mn^{++}$, and thiamine. In addition, the optimal culture pH and temperature for the enzyme activity were found to be 6.8 and $32^{\circ}C$, respectively.

  • Searches for the nutrition requirements of three strains of Brevibacterium ammoniagenes reported in the previous paper were carried out with an aim of achieving the striking accumulation of L-glutamic acid and the large multipication of cells. It was recognized that all three strains required both biotin and thiamine, together with amino acids such as histidine or cysteine, for their good growth and extracellular L-glutamic acid accumulation. The quantity of biotin required for remarkable growth of these microorganisms was quite different from that for the maximum production of L-glutamic acid. This result, however, did not apply in the case of thiamine. It was also confirmed that, of 18 amino acids, histidine and cysteine were the msot effective organic nitrogen sources, while the most available inorganic ammonium salt resulting in a large amount of L-glutamic acid-production and considerable cell gorwth was found to be only urea. Maximum accumulation of extracellular L-glutamic acid, more than 50%(w/w) of the initial sugar content, could be obtained from fermentation in the medium containing wheat-bran extract(Brev. ammoniagenes T-1 and Brev.ammoniagenes Y-2) or rice-bran extract(Brev. ammoniagenes YR-2), which confirmed us a possibility that these bacteria might be employed for industrial fermentation of L-glutamic acid.

  • Effects of nitrogen sources, supplement of vitamins and minerals on the accumulation of 5'-inosinic acid by an adenineless mutant of Brevibacterium ammoniagenes were examined. Maximal yield of 5'-inosinic acid was attained by the use of yeast extract. as organic nitrogen source Casamino acid supplemented with $Mn^{++}$, $Zn^{++}$ Ca-D-pantothenate and thiamine HCl could be subsitute for it. Subsequent experiment using the defined medium showed that the concentration of these trace element in the medium affected inosinic acid accumulation markedly. And it was found that the simulataneous addition of $Mn^{++}$ (20$\mu\textrm{g}$1). $Zn^{++}$(10$\mu\textrm{g}$1). thiamine Hcl(10mg/1) and Ca-D-pantothenate (5mg/l) to the defined medium stimulated inosinic acid accumulation, from which 7mg/ml of inosinic acid was obtained.

  • Seven new hydroxyamine derivatives of 2,2'-methylene bis(3,4,6-trichloroacetoxy benzene) were synthesized by the Mannich reactions. 13 strains of microorganisms were tested for sensitivity to these derivatives by both paper disk method and tube dilution method. Of these compounds, -NHOH compound displays the most effective antimicrobial activity in vitro against Brevibacterium ammoniagenes, Staphylococcus aureus and Bacillus subtilis. Its minimal inhibitory concentration is $1.6{\mu}g/ml$ for Brevibacterium ammoniagenes, and $5{\mu}g/ml$ for Staphylococcus aureus and Bacillus subtilis.

  • As the first step in the production of nucleic acid derivatives by microorganisms, adenineless mutants were derived from Brevibacterium ammoniagenes ATCC6872. A culture of Br. ammoniagenes was exposed to ultraviolet rays for 120 second and treated with diethylsulfate in phosphate buffer for 2 hours to reach the designed death rate. The yield of mutants induced was 0.28% by the ultraviolet irradiation and 0.66% by the diethylsulfate treatment. By the diethylsulfate treatment. By the treatment of penicillin G in a hypertonic minimal medium, the yield of mutants was increased from 0.28% to 0.54% and from 0.66% to 1.5%, respectively. Thus, in was demonstrated that diethylsulfate treatment was much more efficient than UV irradiation to induce adenineless mutants of the bacteria, and total strains of 120 adenineless mutants were obtained.

  • Three strains which accumulated large quantities of L-hlutamic acid as the chief product of metabolism in the presence of glucose and inorhanic ammonium salt under the aerobic condition sere newly isolated. These strains have the general characters as follows: Gram-positive, short fod-shaped, non-sporulating, non-motile, and facultative anaerobe. A change of cell shates was not almost observed in their life cycles and neither a phenomenon of cell-elongation nor a pleomorphism was recognized in any cases. Together with rigid stability in Gram-stain, these characters as above indicated evidences that newly isolates would belong to genus Brevibacterium clearly differentiated from genus Corynebacterium. At the same time the newly isolates, in addition to nutritional requirement of biotin and thiamine, showed the distinctive character of requirement of special amino acid such as histidine or cysteine for their considerable growth. These physiological characters including nitrates reduction, reaction on milk, and slow formation of acid from sugar also were useful in confirming that these bacteria would be Brevibacterium ammoniagenes.

  • Bacterium strain, K-173-10, which was isolated from waste soil of Korean brewing factories, could grow on acetate as the sole carbone source and accumulate a considerable amount of L-glutamic acid (24g/l) in the liguid culture medium. This strain was named by Brevibacterium ammoniagenes sp. by the standard method of taxonomy procedures given in the Manual of Microbiogical Methods.

Aspergillus niger var. Tieghem, Aspergillus usami Sakakuchi, Penicillium notatum-were tested. 2,2'-methylene-bis [3, 4, 6-trichloro ${{\beta}-(o-nitroanilino)$ propionoxy} benzene] showed a strong antimicrobial activity against Bacilus subtilis Natto and Brevibacterium ammoniagenes.
