Development of a Sequence Characteristic Amplified Region Marker linked to the L4 Locus Conferring Broad Spectrum Resistance to Tobamoviruses in Pepper Plants

  • To develop molecular markers linked to the $L^4$ locus conferring resistance to tobamovirus pathotypes in pepper plants, we performed AFLP with 512 primer combinations for susceptible (S pool) and resistant (R pool) DNA bulks against pathotype 1.2 of pepper mild mottle virus. Each bulk was made by pooling the DNA of five homozygous individuals from a T10 population, which was a near-isogenic $BC_4F_2$ generation for the $L^4$ locus. A total of 19 primer pairs produced scorable bands in the R pool. Further screening with these primer pairs was done on DNA bulks from T102, a $BC_{10}F_2$ derived from T10 by back crossing. Three AFLP markers were finally selected and designated L4-a, L4-b and L4-c. L4-a and L4-c each underwent one recombination event, whereas no recombination for L4-b was seen in 20 individuals of each DNA bulk. Linkage analysis of these markers in 112 $F_2$ T102 individuals showed that they were each within 2.5 cM of the $L^4$ locus. L4-b was successfully converted into a simple 340-bp SCAR marker, designated L4SC340, which mapped 1.8 cM from the $L^4$ locus in T102 and 0.9 cM in another $BC_{10}F_2$ population, T101. We believe that this newly characterized marker will improve selection of tobamovirus resistance in pepper plants by reducing breeding cost and time.

Changes in Amylopectin Structure and Pasting Properties of Starch as Affected by Different Transplanting Dates in Rice

  • Three different transplanting dates for two rice cultivars grown in Daegu, Republic of Korea, were examined to identify the changes in growth, rice quality, and characteristics of amylopectin. An early transplanting date caused a reduction in the number of panicles in both rice cultivars compared to that in the optimal and late transplanting dates. The 1000-grain weight in the two rice cultivars was significantly increased in the late transplanting date. The rice cultivar tolerant to high temperatures, Donganbyeo, exhibited the lowest milky grain rate in the late transplanting date. The highest rate of head grain was observed in the late transplanting date in both rice cultivars. Regarding the pattern of pasting properties, peak viscosity increased with delayed transplanting dates. With respect to changes in the amylopectin branch-chain length distribution, the amylopectin structure of the translucent Dongan rice cultivar transplanted on April 25 was characterized by a significant increase in A chains with DP > 12, and a decrease in long chains $DP{\geq}37$ compared to that transplanted on June 25. In contrast, the amylopectin structure of the chalky Dongan rice cultivar transplanted on April 25 exhibited further decrease in 13 < DP < 19 than that transplanted on June 25. In the Ungwang rice cultivar, the amylopectin structure of the translucent Ungwang rice cultivar transplanted on April 25 and June 25 was characterized by a significant decrease in the lengths of total amylopectin chains. Furthermore, the amylopectin structure of the chalky Ungwangbyeo rice cultivar transplanted on April 25 was characterized by a significant increase in 7 < DP < 30, while a significant decrease was observed in that transplanted on June 25. These results indicate that the amylopectin structure is altered by different transplanting dates depending on the characteristics of the rice cultivar.

Investigation of Cytoplasmic Male Sterility and Development of Maintainer and Restorer Lines in Rye (Secale cereale L.)

  • Rye has been a major winter forage crop in Korea. Varietal improvement of rye has been practiced either by hybrid or population breeding systems. Hybrid breeding offers important advantages over population breeding since it is normally a cross-pollinated crop. The hybrid breeding in rye has been possible since cytoplasmically inherited forms of male sterility (CMS) and corresponding nuclear restorer genes were found. The objectives of this research were to develop the maintainer and restorer lines of Korean inbred lines and to estimate the effect of 'Pampa' type of CMS cytoplasm on yield and its related characteristics. For easy discrimination of male-sterile status of plants, anther scoring and the restore index system in which seed-setting and pollen quantity of viability were taken into account were established. High significant correlation between pollen quantity and pollen viability was found. For "Pampa" cytoplasm, four of 14 Korean inbred lines tested turned out to be a maintainer but no restorer was found. But for "235b" CMS cytoplasm, seven inbred lines acted as complete restorers. The Korean inbred rye lines acted mainly as maintainers in "Pampa" cytoplasm but acted mainly as restorer in "235b" cytoplasm. The 'Pampa' cytoplasm inducing male sterility reduced cohn length and plant height and increased the number of tiller, so forage yield and grain yield were enhanced. Breeding of Orange Rose "Peach Pudding" with Good Color and Shape (화형, 화색이 우수한 오렌지색 절화용 스탠다드 장미 "피치푸딩" 육성)

  • A new standard rose cultivar "Peach Pudding" was bred from the cross between deep red standard cultivar 'Red Sandra' and light pink standard cultivar 'Dolores' at the National Horticulture Research Institute. The cross was made in 2000 and "Peach Pudding" was finally selected in 2006 after investigating characteristics for three years from 2002 to 2006. 'Peach Pudding', a pink standard cultivar has good flower shape and fashionable flower color. The major characteristics of this cultivar are 113.1 stems/$m^2$/year in yield, 60.6 cm in length of cut flower, 8.7 cm in flower diameter, 38.6 in petal number, and 12.8 days in vase life. This cultivar can be propagated by both cutting and grafting. The consumer's preference of this cultivar is relatively higher than that of control cultivar, 'New Miracle'.

The effect of extended lactation on parameters of Wood's model of lactation curve in dairy Simmental cows

  • Objective: This study was focused on the estimation of parameters of Wood's model and description of the lactation curve using the cows which were lactated over 24 months on the first lactation. Methods: The database included 1,333 pure-bred dairy Simmental primiparous cows which lactated for 24 months (732 days). The initial dataset entering the procedure of assessment of parameters of Wood's function included 35,826 milk yield records. Milk yield was recorded throughout lactation, with the earliest record taken on day 6 and the latest on day 1,348 of lactation. This dataset was used for the assessment of parameters a, b, c of Wood's model using the non-linear statistical procedure. These parameters were estimated for different length of lactation. The assessed parameters were used for calculation of some characteristics of lactation curves. Results: The lowest value of a parameter (15.2317) of Wood's model of lactation curve was found out in lactations up to 305 days long, contrary to b and c parameters which were highest in those lactations (0.1029 and 0.0015, respectively). The maximum value of a parameter (17.4329) was found out in lactations up to 640 days long, unlike b and c parameters which were minimal in those lactations (0.0603 and 0.0010, respectively). Conclusion: It can be concluded that the parameters of Wood's model and the shape of lactation curve are changing with the growing number of milk yield records. Also, the assessed parameters revealed a significant milk production potential after 305 days of lactation.

Breeding of Velvety Red Poinsettia 'Heidi' (벨벳질감의 적색 포인세티아 'Heidi' 육성)

  • A new poinsettia cultivar 'Heidi' was bred by the National Institute of Horticultural and Herbal Science (NIHHS) in 2004. A cross was made between 'Freedom Rose', a variety with vigorously growing deep pink color, and 'Red Velvet', a variety with dark green leaves and red bracts in 2001. 'Heidi' was finally selected in 2004 after the investigation of the growth and flower characteristics from 2002 to 2004. It has velvety red colored elliptic bracts and transitional leaves, and medium intensity of rugosity between bract veins. Leaf blade is dark green, ovate shape and lobes are shallow. Petiole length is relatively short and stem color is reddish. 'Heidi' has free-branching ability and plant height is short. The bracts and transitional leaves are fully colored 7.5 weeks after short day commencement.

Breeding of Pink Rose 'Pink holic' with Powdery Mildew Resistant (흰가루병에 강한 분홍색 장미 '핑크홀릭' 품종 육성)

  • A new standard rose cultivar 'Pink holic' was bred from the cross between pink standard cultivar 'Lyvia' and red standard cultivar 'Sioux' at the National Horticulture Research Institute. The cross was made in 2002 and 'Pink holic' was finally selected in 2007 after investigating characteristics for three years from 2005 to 2007. 'Pink holic', a light pink standard cultivar grows vigorously and has powder mildew resistant. The major characteristics of this cultivar are $185.4stems/m^2/year$ in yield, 83.9cm in length of cut flower, 11.0cm in flower diameter, 42.9 in petal number, and 22.4 days in vase life. This cultivar can be propagated by both cutting and grafting. The consumer's preference of this cultivar is relatively higher than that of control cultivar, 'Moon light'.

  • A new spray rose cultivar 'Morning Shine' was bred from the cross between light pink spray cultivar 'Charming' and white spray cultivar 'Princess' at the National Horticulture Research Institute. The cross was made in 2000 and 'Morning Shine' was finally selected in 2006 after investigating characteristics for three years from 2003 to 2006. 'Morning Shine', a light yellow spray cultivar grows vigorously and has high productivity. The major characteristics of this cultivar are $159.7 stems/m^2/year$ in yield, 74.3 cm in length of cut flower, 7.2 cm in flower diameter, 35.6 in petal number, and 13.0 days in vase life. This cultivar can be propagated by both cutting and grafting. The consumer's preference of this cultivar is relatively higher than that of control cultivar, 'Sunny Lady'.

A New Double-Petaled Pasqueflower (Pulsatilla koreana) Variety, 'Gyeobi' (겹꽃 분화용 할미꽃 신품종 '겨비')

  • A new pasqueflower (Pulsatilla koreana) variety, 'Gyeobi,' was derived from a mutation of native P. koreana at the Jeollanamdo Agricultural Research and Extension Services (JARES) in Naju, Korea. The 'Gyoebi' was obtained by treating seeds with 3 mM of a chemical mutagen, sodium azide, for 16 hr in 2001. The variety was established in 2005 after two years of selective breeding. 'Gyeobi' is characterized by reddish purple flowers with 12 double petals. The inherent characteristics of the variety are deep yellow anthers and reddish purple stigmas. The agronomic characteristics of the variety are 6.3 flowers per plant, 26.4 cm in flower height, 14.2 cm in leaf length, 7.5 cm in flower width, and 11.0 cm in bract width.

'Pink Bell', a pink color standard cultivar grows vigorously and has a high yield. The major characteristics of this cultivar are $162stems/m^2/year$ in yield, 69.8 cm in length of cut flower, 10.4 cm in flower diameter, 44.5 in petal number per stem, and 12.0 days in vase life. This cultivar can be propagated by both cutting and grafting. The consumer's preference of this cultivar was relatively higher than that of control cultivar, 'Nobless'.
