Change of Germination Rate for Chili Pepper and Chinese Cabbage Seed in Relation to Packaging Materials and Storage Conditions over 10 Years (보관용기 및 저장조건에 따른 고추 및 배추종자의 10년간 발아율 추이)

  • Seed viability is affected by storage conditions and rapid loss of viability in storage is the major cause for low germination. This study was carried out to examine the effect of packaging materials and storage temperature on seed germination rate over 10 years in two species (Capsicum annuum L. and Brassica rapa L. ssp. pekinensis) and determine effective storage conditions for maintaining seed viability. Seeds were packaged in aluminum poly pouches under vacuum, polyethylene bottles, and paper bags containing silica gel and stored under one of two controlled conditions ($15^{\circ}C$, RH 40% or $5^{\circ}C$, RH 30%) or at ambient condition. Seed germination was recorded at 6-month intervals for 10 years. The seeds of both species showed no decline in viability until 6.5 years at 15 or $5^{\circ}C$, irrespective of packaging materials. However, under ambient conditions, the seeds of chili pepper and Chinese cabbage in paper bags lost viability after 4 and 5 years, respectively. By contrast, seeds of both species in vacuum-aluminum poly pouches exhibited a 99% germination rate after 6 years under ambient conditions. Pepper seeds in the vacuum-aluminum poly pouches maintained a 93% germination rate after 10 years in ambient conditions. These results indicated that a special seed storage facility for maintaining viability of chili pepper and Chinese cabbage seed might not be essential and seed testing would not be necessary for 10 years, if chili pepper and Chinese cabbage seeds were packed in ambient/vacuum-aluminum poly pouches or $5^{\circ}C$/vacuum-aluminum poly pouches.

  • In Korea, Brassica species planted in summer remain in rosette and do not flower. Seed production of vernalized rape seedlings grown in summer was evaluated to establish method of generation shortening and genetically pure seed production without isolation of seed fields. When vernalized rape seedlings of different maturity groups were transplanted in summer, all plants bolted and flowered, and average days from sowing to bolting and flowering was 37 and 58. respectively. It took 85days for early maturity group, 88 days for medium maturity group, and 108 days for late maturity group to mature. Seed yield of vernalized plants grown in summer was 8.8 gr per plant and 53kg per l0a which was 23% of seed yield of rape planted in autumn. However, enough seeds were produced to test oil composition and cake and to plant for a next generation. If seedlings are raised, they can be transplanted in more than 1, 000 times of original seed field, indicating that genetically pure seed can be produced without caging seed fields with nets and isolation of seed fields since other Brassica species do not flower in summer.

Analysis of Characteristics and Test of Combining Ability in Leaf Mustard Allies (갓의 국내 수집종 및 도입종의 형질분석 및 $F_{1}$ 조합능 검정)

  • To examine taxonomic relationships among 24 leaf mustard allies (Brassica juncea) derived from domestic collections and introduced species, principal component analysis (PCA) and cluster analysis on 16 morphological characters were carried out. Of 16 characters, total fresh weight, total dry weight, shoot fresh weight and leaf length were useful characters for the understanding of taxonomic relationships among them. Cluster analysis using scores of the principal components indicated that 24 leaf mustards could be grouped into domestic collections and introduced species at 1.0 of average distance in UPGMA. Moreover, in experiment of $F_{1}$ recombination test, heterosis appeared greatly in Sanchiohbachirimen takana A ${\times}$ Akaohba takana, Goheung namyang ${\times}$ Sanchiohbachirimen takana B, and Goheung namyang ${\times}$ Akaohba takana cross combinations. Sanchiohbachirimen takana A and Akaohba takana have a high parent combining ability for breeding cultivars using cytoplasmic male sterility.

Biochemical Changes in Brassica Seedlings Due to Cold Acclimation Treatment (Brassica속 작물 유묘에서 장기 저온 순화처리에 따른 생화학적 변화)

  • This study was aimed for determining the biochemical mechanism of cold tolerance in crops and for searching the biochemical genetic marker related with cold tolerance by the analysis of isozyme pattern. We investigated various biochemical changes induced by the long-term cold acclimation in cold sensitive rape (B. napus) and in cold tolerant 'Sandongchae'(B. campestris) seedlings. The cold shock after long-term cold acclimation to B. napus and B. campestris greatly increased the activities of peroxidase 157% and 50% in root fraction and, 201% and 205% in hypocotyl, respectively. Simultaneously, the activity of superoxide dismutase was largely increased in hypocotyl fraction, too. Protein contents of hypocotyl fractions in B. napus and B. campestris were also increased by 11.4% and 57.8%, respectively. The band of pl 6.4 among peroxidase isozymes newly biosynthesized during long-term cold acclimation was emerged in the hypocotyl fraction of cold tolerant B. campestris as well as in the root of both species. From above and previous results, we presented a model of interconversions of molecular oxygen species due to the cold injury and biochemically inferred the mechanism of cold tolerance in crops.

Utilization of Biosolid for Enhanced Heavy Metal Removal and Biomass Production in Contaminated Soils (중금속 오염 토양 복원 및 바이오메스 생산량 증대를 위한 biosolid 활용)

  • Cleaning up the landfill soil by phytoremediation in association with biomass production and utilization of biosolid as a soil amendment will be an attractive green technology. In order to examine this integrated green technology, in the current study of pot trial, heavy metal removal rate and biomass production were determined following cultivation of three different plant species in the landfill soil incorporated with biosolid at two different levels (25 ton $ha^{-1}$ and 50 ton $ha^{-1}$). Among the three plant species including Indian mustard (Brassica juncea), giant sunflower (Helianthus giganteus. L), and giant cane (Arundo donax. L), sunflower appeared to produce the largest biomass yield (19.2 ton $ha^{-1}$) and the produced amounts were magnificently increased with biosolid treatment compared to the control (no biosoild treatment). The increased production associated with biosolid treatment was common for other plant species and this was attributed to the biosolid originated nutrients as well as the improved soil physical properties due to the organic matter from biosolid. The elevated heavy metals in soil which was originated from the incorporated biosolid were Cu and Zn. Based on the phytoavailable amount of heavy metals from biosolid, the removed amount by plant shoots were 95% and 165% for Cu and Zn, respectively, when sunflower was grown. This indicated that mitigation of heavy metal accumulation in soils achieved by the removal of metal through sunflower cultivation enables the successive treatment of biosolid to soils. Moreover, sunflower showed heavy metal stabilization ability in the rhizosphere resulting in alleviation of metal release to ground water.

  • A pulse-chase labeling of $^{15}N$ on winter rye (Scale cereale) and forage rape (Brassica napus) grown at $15^{\circ}C$ and $25^{\circ}C$ was carried out to determine the effects of low temperature on the uptake exogenous N and the remobilization of endogenous N. The growth rate of leaves and roots depressed at $5^{\circ}C$. AAer 9 days at $5^{\circ}C$, nitrogen content of leaves decreased to 20% on the average while that of roots increased to 12% compared with the plants grown at $25^{\circ}C$. Total content of $NO_3$- uptake 60m medium was 23.0 and 43.5 mg Nlplant, respectively, for winter rye and forage rape grown at $5^{\circ}C$ during 9 days. These values were corresponded to 59.3 and 26.1% lower uptake than those of $25^{\circ}C$. A large part of 1 5 ~ was distributed into leaves throughout time course in both of two species. The content of $^{15}N$ in leaves of winter rye at day 6 increased to 166 and 296 $\mu^{15}$N/plant compared with the initial value (day 0) in the plants grown at $5^{\circ}C$and $25^{\circ}C$ , corresponding to 90 and 163 $\mu$g N of remobilization h m roots into leaves during the fist 6 days. From 7 to 9 days, 75 and 52 $\mu$gN of outflow 6om leaves were occurred at $5^{\circ}C$ and $25^{\circ}C$. However, little remobilization of endogenous N was estimated in forage rape throughout the entire time course regardless of temperature treatment. Comparing two species studied, winter rye was much sensitively influenced by low temperature on the uptake of exogenous N and the remobilization of endogenous N.

Selection of Plant Species for Phytoremediation of Heavy Metal (As and Cd) Contaminated Soil using Hydroponic Culture (수경재배에 의한 중금속 (As 및 Cd) 오염토양의 식물상 복원공법 적용 식물종 선별)

  • Phytoremediation presents a low-carbon and eco-friendly solution for heavy metal-contaminated soils, which pose great health and environmental risks to humans and ecosystems. A hydroponic culture was used to quantitatively assess the phytoremediation potential of plant species to remediate As or Cd-contaminated soil in field application. This study examined the growth, uptake, and distribution of Cd in the roots and shoots of Phalaris arundinacea and Brassica juncea in hydroponic conditions with Cd concentrations ranging from 0 to 20 mg/L for 10 days. Additionally, Aster koraiensis and Pteris multifida were cultivated in hydroponic conditions containing As concentrations ranging from 0 to 40 mg/L for 10 days. The concentrations of Cd in the above-ground part and root tissues of P. arundinacea and B. juncea reached a maximum of 147.7 and 1926.7 mg/kg-D.W.(Dry Weight), and 351.6 and 11305.5 mg/kg-D.W., respectively. Bioconcentration factor (BCF) for P. arundinacea and B. juncea were 68.9 and 122.3, respectively. Both species exhibited a translocation factor (TF) of less than 0.1, indicating their eligibility for phytostabilization. Aster koraiensis exhibited significant As accumulation of 155.1 and 1306.7 mg/kg D.W. in the above-ground part and root, respectively. However, this accumulation resulted with substantial weight loss and the manifestation of toxic symptoms. P. multifida exhibited higher accumulation of As (345.1 mg/kg-D.W.) in the fronds than in the roots (255.4 mg/kg-D.W.), corresponding to BCF values of 18.6 and 7.6, respectively, and a TF greater than 1.2. A TF value greater than 1.0 indicates that P. multifida is a viable option for phytoextraction.

A Study on the Visible Injury of some Herbaceous Plants by $SO_2$ gas (수종(數種) 초본류(草本類)의 $SO_2$ 가스에 의한 가시피해특징(可視被害特徵)에 관(關)한 연구(硏究))

  • $SO_2$ gas injuries for 19 varieties of 10 species of plants were investigated within a environmentally controlled growth chamber. Visible injuries were observed when exposing the plants either 0, 0.2, 0.4, 0.7 or 1.5ppm of $SO_2$ gas for 5 days from 9 : 00 to 17 : 00 everyday. The results obtained were as follows: 1. Light brown spots were shown on Raphanus sativus(Chunchu, Taeback, Jinjudaepyung), Brassica campestris(Manchun, Josaengmiho, Seoul, Jeonseung), and Capsicum annum(Searona, Hongsanho, Hongilpum); brown spots on Sesamum indicum(Pungnyun, Kwangsan); reddish brown shades on Cleosia cristata; and black brown spots on Perilla frutescens, Calendula officinalis, Chrysanthemum morifolium, and Salvia splendens, respectively. 2. The spotty injuries induced by $SO_2$ gas injuries for 19 varieties of 10 species of plants were investigated within a environmentally controlled growth chamber. Study on the Potential of Phytoremediation using Wild Plants for Heavy Metal Pollution (중금속 오염에 대한 Phytoremediation 용 야생식물 연구)

  • The potentials of some Korean wild plants as a phytoremediator for cleaning heavy metal pollution were measured. Several plant species, Ambrosia trifida, Brassica juncea, Rumex crispus, and Abutilon theophrasti screened previously for phytoremediator were treated with cadmium and copper solution. In order to know the growth response to heavy metal stress, the plants were cultivated in hydroponic system containing heavy metals with different concentration. To know the effects of heavy metals on emergence and seedling growth, seeds of 4 species were sown in the pot and watered with heavy metal solution adjusted pH to 6.5, 5.5, and 4.5. A proposed species as potential phytoremediator, A. trifida, showed tolerance to $20{\mu}mol/L$ Cd and $80{\mu}mol/L$ Cu in nutrient solution without apparent growth reduction, and up to $100{\mu}mol/L$ Cd and $400{\mu}mol/L$ Cu without critical visual injury. Up to 311mg/kg of Cd and 369mg/kg were accumulated in dried aerial part in A. trifida. In contrast, A. theophrasti showed injury at $400{\mu}mol/L$ Cu. Significant differences were shown in Cu accumulation among the four species. A. trifida had much higher concentrations of Cd in the shoot, whereas R, crispus accumulated higher concentrations of Cd in the shoot. Testing plant species showed reduced emergence rate with heavy metal treatment. When pH was lowered, the emergence and seedling growth were affected severely with heavy metal. We can suggested that A. trifida was the most proper species for phytoremediation in heavy metal-polluted regions.

Effects of Solvent-extracts Extracted from Coptis chinensis on Seed Germination and Seedling Growth (종자발아 및 유묘생장에 미치는 황련(Coptis chinensis) 추출물의 영향)

  • For searching the natural herbicide-components, the seed germination and seedling growth of receptor plant species (Brassica campestris, Sesamum indicum, Perilla frutescens and Echinochloa crus-galli) were investigated in four solvent-extracts extracted from Coptis chinensis Franch. The seed germination of receptor plant species was largely inhibited in 2,000 ppm of ethyl acetate compared to the control, and it was inhibited in order of P. frutescens, B. campestris, E. crux-galli, and S. indicum. In seedling growth, the shoot and root elongations of receptor plant species were inhibited in order of S. indicum, P. frutescens, B. campostris, and E. crus-galli. Root elongation was remarkably reduced in order of $H_2O$, butyl alcohol, and hexane, ethyl acetate extracts. Of four receptor plant species, seed germination and seedling growth of B. campestris and S. indicum showed the species-specific reaction to the solvent-extracts extracted from C. chinemis. $H_2O$ extract had a natural herbicide potential to the seed germination or root elongation in B. campestris and S. indicum. The result can be provided a basic data f3r the development of natural herbicide.
