양돈 슬러리에 첨가된 발효탄수화물의 종류별 악취물질 농도 비교 (The Effect of the Addition of Carbohydrate on the Concentration of Odorous Compounds in Pig Slurry)

  • Slurry treatments included peanut shell, palm golden fiber, almond hull, which was added 2% of the amount of slurry, and non-treatment control (n=4 each group). Levels of odorous compounds were measured from the liquid slurry incubated in $20^{\circ}C$ for 2 wk in chamber whose structure is similar to slurry pit. Concentration of phenols and indoles was higher (p<0.05) in control (48.4, 4.0 ppm) compared to almond hull (31.5, 1.4 ppm) or palm golden fiber (29.1, 1.6 ppm) group. Short chain fatty acid (SCFA) level was lowest (p<0.05) in control (2,121 ppm) but highest in peanut shell group (3,640 ppm). Branched chain fatty acid (BCFA) concentration was highest (p<0.05) in peanut shell (296 ppm), but lowest in almond hull (90 ppm). Taken together, concentration of odorous compounds was decreased by addition of almond hull in pig slurry by which crude fiber and non-digestible fiber (NDF) may act as a carbon source.

Effect of Dietary Supplementation of Sodium Salt of Isobutyric Acid on Ruminal Fermentation and Nutrient Utilization in a Wheat Straw Based Low Protein Diet Fed to Crossbred Cattle

  • The effect of dietary supplementation of sodium salt of isobutyric acid in low protein (10% CP) wheat straw based diet on nutrient utilization and rumen fermentation was studied in ruminally fistulated male crossbred cattle. The study included a 7 day metabolism and a 3 day rumen fermentation trials. The cattle were distributed into two equal groups of 4 each. The animals of control group were fed a basal diet consisting of wheat straw, concentrate mixture and green maize fodder in 40:40:20 proportion whereas branched chain volatile fatty acid (BCFA) supplemented group received a basal diet + isobutyric acid at 0.75 percent of basal diet. The duration of study was 36 days. The feed intake between experimental groups did not differ significantly and the average total DMI (% BW) was 2.01 and $2.28kg\;day^{-1}$ in control and BCFA supplemented diets. The dietary supplementation of BCFA improved (p<0.05) the DM, OM, NDF and cellulose digestibility by 4.46, 6.63, 10.57 and 11.31 per cent over those fed control diet. The total N retention on BCFA supplementation was improved (p<0.01) due to decreased (p<0.05) urinary N excretion. The concentrations of ruminal total N was 37.07 and $34.77mg\;100ml^{-1}$ in control and BCFA fed groups, respectively. Dietary supplementation BCFA significantly (p<0.01) reduced the ruminal ammonia N concentration as compared to control and the mean values ($mg\;100ml^{-1}$) were 13.18 and 9.42 in control and BCFA fed groups. The total VFA concentration was higher (p<0.01) in BCFA supplemented group (101.14 mM) than the control (93.05 mM). Among the VFAs, the molar proportion of acetate was higher (p<0.01) in BCFA supplemented group (71.07 mM) as compared to control (64.98 mM). However, the concentration of propionate and butyrate remained unchanged. Amino acids composition of bacterial hydrolysates was similar in both the groups. Ruminal outflow rate of liquid digesta was higher (p<0.01) in BCFA fed group ($67.56l\;day^{-1}$) than control ($52.73l\;day^{-1}$). It is concluded that the dietary supplementation of Na-salt of isobutyric acid in low protein diet improved the nutrient utilization and ruminal fermentation characteristics.

A reduction in dietary crude protein with amino acid balance has no negative effects in pigs

  • The aim of this experiment was to evaluate the effects of low crude protein (CP) level with essential amino acids (AA) addition on growth performance, nutrient digestibility, microbiota, and volatile fatty acid composition in growing pigs. A total of 160 growing pigs (Landrace × Yorkshire × Duroc [LYD]; average initial body weight 16.68 ± 0.12 kg) were randomly allotted to one of the four treatments on the basis of initial body weight. A randomized complete block design was used to conduct this experiment in the Research Center of Animal Life Sciences at Kangwon National University. There were ten pigs/replicate with four replicates in each treatment. The treatments include; CON (Control, 17.2% dietary CP level), low protein (LP)-1.10 (15.7% dietary CP level + 1.10% lysine level), LP-1.15 (15.7% dietary CP level + 1.15% lysine level), LP1.2 (15.7% dietary CP level + 1.20% lysine level). The pigs fed CON and LP-1.2 diet showed greater final body weight than that of LP-1.1 diet (p < 0.05). Although average daily gain, average daily feed intake, and feed efficiency did not show any difference in phase 2 and 3, average daily gain and feed efficiency was significantly greater in CON and LP-1.20 in phase 1. However, the average daily feed intake did not show any difference during the experimental period. Isobutyric acid and isovaleric acid composition of LP treatments were lower than CON treatment in phase 2. Total branched chain fatty acid composition was significantly lower in LP treatment in phases 1 and 2. However, there was no significant difference among treatments in phase 3. The results of this study underscore the importance of AA supplementation when implementing a low-protein diet during the early growth phase (16-50 kg) in pigs.

배양온도 변화에 따른 Streptomyces viridochromogenes의 지질과 지방산 조성변화 (Changes in Lipids- and Fatty Acids Compositions in Response to Growth Temperature of Streptomyces viridochromogenes)

  • Streptomyces viridochromogenes 야생형과 기중균사 형성 변이주들의 지질과 지방산의 조성을 배양온도와 배양시간에 따라 조사하였다. Triacylglycerol은 기중균사가 형성되는 경우에만 감소되어 기중균사 형성과 연관되어 있음을 나타내었다. 인지질의 경우에는 배양조건에 따른 특이적 변화를 볼 수 없었으나, 기중균사 형성능이 상실된 변이주 BR2가 $20^{\circ}C$에서 배양되었을 매 phosphatidylethanolamine 조성이 특이하게 높게 나타났다. 그리고 변이주 BR2는$20^{\circ}C$$30^{\circ}C$ 배양 시 다양한 $R_{f}$ 간의 동정되지 않은 아미노지질들을 함유하였다. 모든 균주에서 아미노지질의 일종인 ornithinolipid는 배양시간이 경과되면서 증가되는 경향을 나타내었다. 지방산조성은 온도에 민감하여 배양온도가 높을수록 불포화지방산 함량이 낮아졌다. 기중균사 형성은 직쇄형 포화지방산의 함량 감소를 수반하였으며, 기중균사 생성능이 우수한 변이주 M13은 배양초기의 직쇄형 포화지방산 함량이 가장 높았다. 변이주 BR2는 iso형 홀수탄소 지방산의 함량이 특이적으로 높았다.

Different Effects of Acidic pH Shock on the Prodiginine Production in Streptomyces coelicolor M511 and SJM1 Mutants

  • The changes in prodiginines productions caused by pH shock culture of Streptomyces coelicolor strains were estimated. In Streptomyces coelicolor M511, undecylprodiginine and streptorubin B productions increased 1.8-fold (37.22 mg/g) and 2.5-fold (18.61 mg/g), respectively, by pH shock (from 7.2 to 4.0). In contrast, this resulted in the significantly decreased prodigignines production in the redP deletion mutant SJM1; 3.7-fold for undecylprodiginine, 4.4-fold for streptorubin B, 5.2-fold for methylundecylprodiginine, and 6.4-fold for methyldodecylundecylprodiginine, respectively. RT-PCR analyses showed that, during pH shock, expression of redD, the transcription activator gene, was increased while the expression of fabH, the decarboxylative condensation enzyme gene in fatty acid biosynthesis, was decreased in both strains. The enhanced redD expression was in good accordance with the increased total prodiginines production of M511. However, for SJM1 mutant, the decrease of fabH expression occurred more strikingly, such that it became almost completely turned off during acidic pH shock culture. Therefore, a down-regulation of fabH was considered to be the cause of decreased amount of total prodiginines produced, although redD expression was high in SJM1 mutant.

Effect of Intraruminal Sucrose Infusion on Volatile Fatty Acid Production and Microbial Protein Synthesis in Sheep

  • Effects of sucrose supplement on the pattern of VFA production and microbial protein synthesis in the rumen were examined in sheep consuming basal diet of grass silage (2.5 kg fresh wt/d) that was provided in 24 equal meals each day by an automatic feeder. Four mature wethers were allocated to four experimental treatments in a 4${\times}$4 Latin square design with periods lasting 14 days. The treatments were (1) the basal diet, (2) supplemented with 150 g sucrose and 7.0 g urea, (3) 300 g sucrose and 13 g urea, and (4) 450 g sucrose and 20 g urea given as a continuous intraruminal infusion for 24 h. All infusions were given in 2 litres of aqueous solution per day using a peristaltic pump. The effect of sucrose level on rumen mean pH was significantly linear (p<0.01). There were not significant differences in the concentration of ammonia-N, total VFA and the molar proportions of acetate, propionate and butyrate with the level of sucrose infusion. The molar proportions of isobutyric acid (p<0.05) and isovaleric acid (p<0.001) were significantly reduced when the infused amount of sucrose was increased. The flow of microbial N was linearly (p<0.001) increased with sucrose and urea level. High levels of readily fermentable carbohydrate in a ration reduced the efficiency of microbial protein synthesis in the rumen. It was demonstrated that of the individual fatty acids, only the molar proportion of isovalerate showed a significant negative correlation (R2=$0.3501^{**}$) with the amount of microbial N produced and a significant positive correlation (R2=$0.2735^{**}$) with the efficiency of microbial growth.

Comparison and analysis on sheep meat quality and flavor under pasture-based fattening contrast to intensive pasture-based feeding system

  • Objective: The objective of this study was to investigate the effect of 4-month intensive feeding on the meat quality, fatty acid profile, flavor, and growth performance of grazing Hulunbuir sheep (HBS). Methods: The HBS were selected 4-months after birth in a pasture rearing system as the experimental animals (n = 44, female, average body weight 23.8±2.2 kg) then divided equally into pasture-based grazing fattening (PAS) and concentrate-included intensive fattening (CON) groups for another 4-month finishing. When finished fattening, all animals were slaughtered to collect musculus longissimus dorsi subcutaneous adipose tissue and to investigate the influences on meat quality, fatty acid profile, flavor and growth performance. Results: The results showed lambs in CON group got significantly higher live weight, hot carcass weight, and dressing percentage. The CON group had significantly higher value of redness (a*), lightness (L*) and water holding capacity (p<0.05), significantly lower value of Warner-Bratzler shear force than the PAS group (p<0.05). The subcutaneous fat from CON group lambs demonstrated a significantly higher content of C18:1 and C18:2 (p<0.05), but lower C14:0 and C16:0, indicating an increased degree of unsaturated fatty acid. The content of 4-methyloctanoic acid, 4-ethyloctanoic acid and 4-methylnonanoic acid had increased 2 to 4 times, representing a more intense odor in the CON group. However, the values were still lower than most sheep breeds reported, indicating the indoor feeding system could not fundamentally deteriorate the excellent meat characteristic of HBS. Conclusion: It was evident that lambs in CON group exhibited a better meat production performance, improved in meat color, texture and healthier fatty acid profile through pasture-weaned concentrate included intensive fattening system, which offers a good alternative regimen for lamb finishing and has a wide prospection in the HBS meat industry.

Influence of Isobutyric Acid Supplementation on Nutrient Intake, Its Utilization, Blood Metabolites and Growth Performance of Crossbred Calves Fed Wheat Straw Based Low Protein Diets

The calves were divided into two equal groups of 6 each. The animals of the control group were fed a basal diet consisting of wheat straw, concentrate mixture and green oat fodder in 40:40:20 proportion whereas BCFA supplemented group received the basal diet+isobutyric acid at 0.75 percent of basal diet. The duration of study was 120 days. The feed intake between experimental groups did not differ significantly and the average total DMI (% BW) was 1.99 and $1.95kg\;day^{-1}$ in control and BCFA supplemented diets. The dietary supplementation of BCFA improved (p<0.01) the DM, OM, CP (p<0.05), NDF and cellulose digestibilities by 8.50, 9.01, 5.39, 17.78 and 18.44 per cent over those fed control diet. The total N retention on BCFA supplementation was improved (p<0.01) due to the decreased (p<0.05) faecal N excretion. The BCFA supplementation did not alter the blood circulatory levels of glucose, total protein, albumin, urea N and amino acids. However after 120 days of experimental feeding a significant (p<0.05) increase in the concentrations of non-esterified fatty acid was observed in control group. The DCP intake and the DCP content of experimental diets was similar in both groups. However, the TDN content of BCFA supplemented diet was significantly (p<0.01) higher (64.35%) than that of control (59.60%). The total live weight gain in BCFA supplemented diet increased by 15.94% over control. The average daily gain and efficiency of feed conversion were also improved in BCFA fed calves by 13.38 and 26.71% respectively, compared to control. It is concluded that dietary supplementation with isobutyric acid improved the digestibility of nutrients and growth performance of calves.

Function of Global Regulator CodY in Bacillus thuringiensis BMB171 by Comparative Proteomic Analysis

  • CodY is a highly conserved protein in low G+C gram-positive bacteria that regulates genes involved in sporulation and stationary-phase adaptation. Bacillus thuringiensis is a grampositive bacterium that forms spores and parasporal crystals during the stationary phase. To our knowledge, the regulatory mechanism of CodY in B. thuringiensis is unknown. To study the function of CodY protein in B. thuringiensis, BMB171codY- was constructed in a BMB171 strain. A shuttle vector containing the ORF of cry1Ac10 was transformed into BMB171 and BMB171codY-, named BMB171cry1Ac and BMB171codY-cry1Ac, respectively. Some morphological and physiological changes of codY mutant BMB171codY-cry1Ac were observed. A comparative proteomic analysis was conducted for both BMB171codY-cry1Ac and BMB171cry1Ac through two-dimensional gel electrophoresis and MALDI-TOF-MS/MS analysis. The results showed that the proteins regulated by CodY are involved in microbial metabolism, including branched-chain amino acid metabolism, carbohydrate metabolism, fatty acid metabolism, and energy metabolism. Furthermore, we found CodY to be involved in sporulation, biosynthesis of poly-β-hydroxybutyrate, growth, genetic competence, and translation. According to the analysis of differentially expressed proteins, and physiological characterization of the codY mutant, we performed bacterial one-hybrid and electrophoretic mobility shift assay experiments and confirmed the direct regulation of genes by CodY, specifically those involved in metabolism of branched-chain amino acids, ribosomal recycling factor FRR, and the late competence protein ComER. Our data establish the foundation for in-depth study of the regulation of CodY in B. thuringiensis, and also offer a potential biocatalyst for functions of CodY in other bacteria.

Effect of horseradish, mushroom waste and almond hull on the concentrations of odorous compounds in swine manure for spreading on grassland in spring

  • The objective of the current study was to investigate the effect of odor reducing agents on the levels of pH, total carbon, total nitrogen, and odorous compounds [phenols, indoles, short chain fatty acid (SCFA), branched-chain fatty acid (BCFA), and ammonium nitrogen] of swine manure during the spring season (temperature around $20^{\circ}C$). Odor reducing agents included horseradish powder, mushroom waste powder, and almond hull powder. A manure sample (15 L) was taken from the pit under the pens of a swine feeding operation and incubated with 0.03% horseradish powder, 1% mushroom waste powder, and 1% almond hull powder, respectively, in acryl chambers for 14 days. Addition of almond hull powder showed the lowest pH (p < 0.05) and the highest level of total carbon (p < 0.05) among treatments of odor reducing agents. Although addition of odor reducing agents increased the level of phenols (p < 0.05), addition of almond hull powder decreased the level of indoles (p < 0.05). Levels of SCFA and BCFA were higher in almond hull powder than those in control (p < 0.05). Taken together, the results from our current study showed that odor reducing agents can be used for reducing the odor of swine manure by providing fermentable carbohydrates. At $20^{\circ}C$, however, the function of odor reducing agents might be decreased due to lower microbial activity.
