Transmission of Fusarium oxysporum by the Fungus Gnat, Bradysia difformis (Diptera: Sciaridae) (작은뿌리파리에 의한 Fusarium oxysporum의 전반)

  • The fungus gnat, Bradysia difformis was examined for its ability to transmit Fusarium oxysporum in PDA culture. Larvae and adults of B. difformis were able to transmit the fungus as ingested and sticking. We constructed GFP-expressed mutants with Fusarium oxysporum, then feed it to larvae of fungus gnat, B. difformis. So that mycelia were placed in the alimentary canal of larva.

Biological Control of Bradysia difformis using the Predatory Mite (Hypoaspis aculeifer) in Hydroponically Cultivated Strawberry (수경재배 딸기에서 포식성 천적, 아큐레이퍼응애를 이용한 작은뿌리파리의 생물적 방제)

  • Biological control of the fungus gnat Bradysia difformis by the predatory mite Hypoaspis aculeifer was examined in hydroponic systems of strawberries in Namwon and Hapcheon varieties of Jeollabuk-do and Gyeongsangbuk-do provinces, respectively, from 2012 to 2013. The damage to strawberries caused by B. difformis was examined in seedling stages and during cultivation periods. To achieve environment-friendly management of B. difformis, 60.6 individuals of H. aculeifer were released per square-meter three and four times during cultivations periods of the Namwon and Hapcheon strawberry varieties, respectively. H. aculeifer had better control efficiency against of B. difformis than that observed with conventional cultural practices. Wilt symptoms and damage of strawberries due to B. difformis were decreased by approximately 7.4-10.4%.

Molecular identification of fungus gnats from shiitake mushroom in Korea (국내 표고버섯 주요 버섯파리의 분자생물학적 종 동정 및 발생양상)

  • Fungus gnats are usually found in mushroom farm and have recently become important pest because they can cause severe damage and reduce the production on shiitake mushroom. Usually shiitake mushrooms are cultivated on both oak bed logs and in the artificial sawdust beds in greenhouses. Using yellow sticky trap, the dipteran species in shiitake mushroom farm were collected from May to September in Kyonggi-do and Chungcheong-do in 2013. To identify the main species of fungus gnat on the shiitake farm in Korea, the collected samples were determined the sequence of cytochrome c oxidase subunit I (COI) by DNA barcoding. The phylogeny based on maximum likelihood analyses from COI sequence showed that Bradysia difformis and B. alpicola were main species of shiitake bed log and Scatopsidae sp. and B. difformis were dominant species of sawdust beds.

  • The fungus gnat, Bradysia difformis, has been recognized as an important pest of greenhouse crops. There is a need for research on the control of the fungus gnat. However, it is difficult to obtain many generations of the fungus gnat for several kinds of research. Indoor propagation is a very useful method for obtaining enough individuals in cases where the need is for larvae in soil. This study was conducted to determine the optimum growing media and temperature conditions for rearing the fungus gnat in the laboratory. Under experimental temperature conditions, hatching, pupation, and eclosion rates were the highest at $20^{\circ}C$. The developmental period of the fungus gnat was shortened with higher temperatures. The greatest number of eggs was an average of 144 at $20^{\circ}C$. Using different types of larvae growth media, the highest hatching rates were 84.7 and 84.4% in water agar and potato disks, respectively. The larval period was the shortest, at 14.7 days, when grown on potato disks. The highest pupation and eclosion rates were 85.2 and 82.6% on potato disks, respectively. The highest number of eggs was an average of 125.6 on potato disks. Regarding the effects of different growth media on the eclosion rate of B. difformis, the highest eclosion rate was 88.4% on the soil mix, and was 50% on oatmeal, 25% on shredded potato. The results of four different inoculation levels of larvae on eclosion rate of B. difformis showed that the highest eclosion rate was 84.7% for 1,000 larvae. The eclosion rate was shortened with a higher number of larvae inoculated/cage. In the growth medium used, 3,000 eggs were better for the initial level of inoculation, showing a relatively high emergence rate and short developmental period. Mass rearing procedures were explained in detail.

Control effects of 20 chemical insecticides and new strains of Bacillus thuringiensis against the fungus gnat (Bradysia difformis, Sciaridae, Diptera)

  • The fungus gnat, Bradysia difformis, damages various crops in greenhouses and is recognized as an important pest around the world. Additionally, in the future, many other greenhouse crops will be added to the list of crops damaged by the fungus gnat. In this study, to find effective control methods for the fungus gnat, the insecticidal effect of 20 chemical synthetic insecticides was tested with the potato disc and pot treatment methods; additionally, the control effect of 16 strains of B. thuringiensis was examined with the potato disk method. The fungus gnat larvae were treated for 2 days with each of the synthetic insecticides to determine insecticidal effect using the potato disc method. The results were as follows. Among the highly insecticidal active pesticides, chlorfenapyr exhibited a 100% insecticidal activity, and fenazaquin, acetamiprid, dinotefuran, fenthion and thiamethoxam exhibited more than a 90% insecticidal activity. For the pot treatment method, chlorfenapyr exhibited a 3.3% insecticidal effect, and thiamethoxam, acetamiprid, dinotefuran, fenthion, etc. exhibited an insecticidal effect of less than 10% of the emergence rate to adult fungus gnat after 14 days of treatment. To select the B. thuringiensis strains that have an insecticidal effect on the fungus gnat, 16 strains were biologically assayed using the potato disc method. Among the 16 strains, Bt-3, Bt-8 and Bt-13 had more than a 70% insecticidal effect. The $LC_{50}$ and $LC_{95}$ values of Bt-3, Bt-8 and Bt-13 were $3.7{\times}10^5$ and $4.7{\times}10^8cfu/ml$, $1.4{\times}10^5$ and $1.1{\times}10^7cfu/ml$, and $1.4{\times}10^5$ and $1.3{\times}10^7cfu/ml$, respectively.

Biological control of mushroom flies using the predatory mite Hypoaspis aculeifer in a shiitake cultivation (원목 표고에서 아큐레이퍼응애를 이용한 버섯파리류의 생물학적 방제)

  • The major species of fungus gnats which caused the severe damage in shiitake farm were identified as a Bradysia difformis, B. alpicola, and Camtomyia cortocalis on oak log beds cultivation. The B. difformis occurred early in the middle of March while B. alpicola and C. cortocalis appeared since the beginning of May. The occurrence rate for adults of B. difformis showed highly at the end of July (11.9~1,774.2 in dong-myeon and 0.4~2,583.3 in pungse-myeon) in 2012 and mid-June (10.7~4,650 in dong-myeon and 36.8~4740 in pungse-myeon) in 2013. The counting numbers on the traps for B. alpicola reached highest peak in the middle of June (2.1~63.2 in dong-myeon and 1.0~21.7 in pungse-myeon) and the end of May (0.8~163.7 in dong-myeon and 0.5~280.5 in pungse-myeon) in 2012 and 2013, respectively. The number of C. cortocalis showed high record in the middle of May in 2012 (0.6~4.7) and in the middle of June (2.1~17.3) in 2013 in dong-myeon whereas showed the peaks in the middle of May (0.6~4.7) in 2012 and in the late of May (1.3~17.6) in 2013 in pungse-myeon. The fruiting bodies of shiitake mushroom by fungus gnats were severely damaged from mid-June to late-July and the damage rate were 0.625.5% (2012) and 0.7~30.5% (2013) in dong-myeon and 1.5%~21.6% (2012) and 1.9~36.8%(2012) in pungse-myeon. To investigate the control effect for fungus gnats by Hypoaspis aculeifer, H. aculeifer (30 mixutre of nymph and adult per $m^2$) were treated to oak log beds shiitake cultivation for six times (May 2 and 28, June 25, July 10 and 25 and August 28). The occurrence rate of adults and damage rate of fruiting bodies of 3 major species reduced 79.3% (adult numbers) and 74.8% (fruiting bodies) in dong-myeon and 64.1% (adult numbers) and 65.5% (fruiting bodies) in pungse-myeon, respectively, compared to non-treatment. Accordingly, H. aculeifer effectively controlled the fungus gnats on shiitake mushroom and can be used as good control agent.

Optimal height of installing sticky traps for monitoring fungus gnats (버섯파리류 예찰을 위한 끈끈이트랩의 최적 설치위치)

  • Agricultural sticky traps are used for forecasting flying insects, including fungus gnat adults. Forecasting using sticky trap is an important measure to determine pesticides or environmentally friendly control means. Sticky traps of various colors can be installed easily and quickly. The yellow sticky trap is most effective for forecasting fungus gnat adults. In addition, they are used for control purposes by mass trapping. We tested the optimum installation height of yellow sticky traps to forecast and control the fungus gnat adults effectively in the cultivation of mushroom and horticultural plants. The number of captured fungus gnat adults was highest on the second floor for button mushroom and oyster mushroom, 100 cm above the floor for sawdust shiitake mushroom, 20 cm under the floor for hydroponic strawberry, and 20 cm above the floor for hydroponic tomato. This suggests that sticky trap height should be installed differently depending on the cultivation type or environment for effective forecasting and control of fungus gnat adults.
