Effects of Photoperiod Exchanges on the Growth of two Common Live Food Organisms (rotifer, Brachionus rotundiformis and Harpacticoida Copepoda, Tigriopus japonicus) in the Combination Cultures (Brachionus rotundiformis (rotifera)와 Tigriopus japonicus(copepoda; harpacticoida)의 혼합배양에 있어서 광주기 변화가 두 동물먹이생물의 증식에 미치는 영향)

  • Two zooplankton species Brachionus rotundiformis (rotifera) and Tigriopus japonicus (copepoda; harpacticoida) were commonly used as live food organisms in the marine larval rearing centers. The combination culture method of two live food organisms (B. rotundiformis and T. japonicus) was well known as very valuable for stable and mass cultures. In this study, we investigated the effects of photoperiod exchanges on the growth and interspecific relationship in the combination culture of two species. The results showed that, photoperiod condition can change copepod (and rotifer) density under the two species combination cultures. There is 200% higher maximum rotifer density on the 24L:0D photoperiod culture condition compare to 12L:12D condition. However, maximum density of copepod is observed highest on the 0L:24D photoperiod culture condition. In addition, it's differ in the culture densities of nauplii, copepodites and female carrying eggs on the each three photoperiod types.

Ecological Effects of Slag Extracts on the Initial Life Cycle of the Rotifer Brachionus plicatilis and Benthic Copepod Tigriopus japonicus (윤충류 Brachinus plicatilis와 저서성 요각류 Tigriopus japonicus의 초기생활사에 미치는 슬래그 추출액의 생태 영향)

  • In this study, the marine ecological impacts of dephosphorized slag and steel slag on the initial life cycle of the rotifer Brachionus plicatilis and benthic copepod Tigriopus japonicus (in marine trophic structure as a first consumer) exposure to slag extracts have been considered using a marine ecotoxicological assessment. In the results of a screen test on slag extracts, the pH of an undiluted solution was measured to have high alkalinity (pH 8.89-12.16), but a toxic reaction to this undiluted solution before and after aging was divided according to test species. For non-aged slag, the toxic effect ($LC_{50}$) of neonate on B. plicatilis was seen to be severe with dephosphorized slag (20.8 %) than steel slag (63.8 %) with under pH-uncontrolled conditions. The toxic values of dephosphorized and steel slag were estimated to be 35.3 % and 36.0%, respectively, for nauplius with T. japonicus. However, the toxicity of slag extracts before and after aging were different for T. japonicus than for B. plicatilis based on the characteristics of the test materials, with pH-controlled conditions. In conclusion, the results of this study suggest that slag can be relatively stable after aging and may not be likely to influence marine environments, even given repetitive extracting under pH-uncontrolled conditions. This study confirms that a marine ecotoxicological assessment method applied to mechanically activated samples can give an idea of the resistance a marine environment has against the introduction of hazardous materials due to precipitation and weathering.

Dietary Value of Neonates from Rotifer, Brachionus plicatilis and B. rotundiformis Resting Eggs for Flounder and Parrot Fish Larvae (넙치 및 돌돔 자어 사육에 있어서 Brachionus plicatilis와 B. rotundiformis 내구란에서 갓 부화한 rotifer의 먹이효율)

  • The dietary value and fatty acids composition for the hatched neonates from the resting eggs of marine roofers, Brachionus plicatilis and B. rotundiformis were compared with that for mass-cultured rotifers (control) by feeding them to larvae of flounder (Paralichthy olivaceus) and parrot fish (Oplegnathus fasciatus). Resting eggs were mass-produced in $1~4m^3$tanks by feeding Chlorella sp. and baker's yeast. The B. plicatilis and B. rotundiformis eggs were preserved at $5^{\circ}C$ in darkness for 3 and 5 months, respectively, and hatched at $28^{\circ}C$ under continuous light. The hatched neonates from the resting eggs and mass-cultured rotifer, which was used as a rontrol were fed to fish larvaes. The growth and survival rates of parrot fish larvae fed on the neonates from the resting eggs of B. rotundiformis were similar to those of fish larvae fed on the control rotifer. And the growth and survival rates of the flounder larvae with neonates from the resting eggs of B. plicatilis were similar or higher than those fed the control rotifer. Also the fatty acids composition of the neonates from the resting eggs of B. plicatilis and B. rotundiformis were similar to those from the control rotifers. This results showed that the hatched neonates from resting eggs of rotifer could be used as an effective diet for flounder and parrot fish larvae.

Establishment of Standard Methods for Marine Ecotoxicological Test (해양생태독성평가를 위한 표준시험방법 개발에 관한 연구)

  • Six standard methods for marine ecotoxicological tests were established(or applicated) using marine decomposer, primary producers and consumers. Development processes referred to the standard methods established by USEPA(United States Environmental Protection Agency), international organizations and European methods. However, the standard test species were selected among the domestic species generally found in the Korean waters and sediments. The test methods provide the culture/maintenance of test species, test methods, reproducibility and quality acceptance criteria etc. A total of nine test species were designated including bioluminescent bacteria(Vibrio fischeri), diatom(Skeletonema costatum), seaweed(Ulva pertusa), rotifer(Brachionus plicatilis), benthic copepod(Tigriopus japonicus), benthic amphipods(Mandibulophoxus mai, Monocorophium acherusicum), and fishes(Oryzias latipes, Paralichthys olivaceus). These test species represent the decomposer, primary producer and consumers in marine trophic system in Korean coastal ecosystems, and we recommend the "battery test" including at least one species from the each trophic level for marine ecotoxicological test.

  • 양식산업에 있어서 어류 자어 시기의 초기 먹이생물로 rotifer가 주로 사용되고 있다. 특히, 담수산 rotifer, B. calyciflorus는 배양이 쉽고 자어의 먹이로 적당한 크기를 가지고 있기 때문에 초기 먹이생물로 적합하다. 최근 rotifer 배양수의 산소 가스 공급과 pH조절, 농축 Chlorella의 공급, rotifer 배양시 누적되는 다량의 유기물 찌꺼기 제거를 위한 filter 사용은rotifer 고밀도 배양을 가능케 했고, 현재 국내 일부 종묘 배양장에서 이러한 고밀도 배양시스템을 이용하고 있다.   • 담수산 rotifer, Braohionus calyciflorus의 내구란은 종의 유전적 보존, 환경 독성학의 실험재료뿐만 아니라, Artemia cyst처럼 간단하게 부화시켜 자어의 먹이로 공급할 수 있는 장점이 있다. 일반적으로 rotifer의 내구란 생산은 배양용기내의 rotifer 밀도가 증가하면서 유성생식 개체가 출현하며 이후 수컷의 수정으로 내구란이 형성되는 일련의 과정을 거친다. 그러나 이러한 과정에 있어서 rotifer의 유성생식 개체의 출현과 수컷의 수정에 영향을 미쳐 rotifer 개체수의 증가와 더불어 용존산소 감소 및 암모니아 증가 둥 배양수의 환경 악화로 내구란 생산과정이 중단되면서 효율적인 내구란 대량생산은 기대하기가 매우 어렵다.   • During a three-year study (2000-2002), dramatic changes in the phytoplankton biomass and high transparency were repeatedly observed during mid-spring in the lower part of the Nakdong River. Rotifers (Brachionus, Keratella, Polyarthra) , sharply increased toward the middle and end of spring. As hydrologic retention time increased (to near 20 days) and water temperature increased from $10^{\circ}C$ to > $20^{\circ}C$ toward the end of spring, small cladocerans noticeably increased. Once phytoplankton biomass passed their peak stage in the mid-spring, a short period (one or two weeks) of relatively low phytoplankton biomass and high Secchi transparencies occurred. Grazing by the zooplankton was highest in spring, thus, it seems that high grazing activities of zooplankton grazing regulated phytoplankton dynamics in the river. The results indicate that the role of zooplankton grazing in controlling the phytoplankton biomass becomes more important during the spring when river water is relatively stagnant.
