Subpopulation of lymphocytes in Korean native cattle infected with enzootic bovine leukosis

  • Enzootic bovine leukosis(EBL) is chronic disease caused by bovine leukemia virus(BLV), retroviridae. The characteristic feature of this disease is proliferation of lymphocytes in circulating blood or lymphoid tissues. Because EBL concern lymphocytes, immunological disorder or alteration in the lymphocyte subpopulation is suggested. In this study, we investigated the changes of the lymphocyte subpopulation in the circulating blood of Korean native cattle infected with bovine leukemia virus. (omitted)

  • To elucidate pathogenesis of bovine leukemia virus(BLV) in Korean native goats, the goats experimentally infected with BLV were studied especially for the aspects of infectivity and hematological changes. The experimental goats were examined for 27 months by agar-gel immunodiffusion(AGID) test and syncytium formation assay. During this period, changes of total leucocyte, absolute Iymphocyte and atypical Iymphocyte were examined, and the distribution of surface immunoglobulin ( sIg ) -bearing cells and rosette forming cell (RFC) in the peripheral Iymphocyte were also investigated. By indirect immunofluorescence (IFA) and complement dependent antibody cytotoxicity (CDAC) assay using monoclonal antibody(Mab) against bovine leukosis tumor-associated anti-gen(BL-TAA), changes of BL-TAA positive Iymphocyte in peripheral blood were measured. The results obtained through the experiment were summarized as follows. 1. Antibody titers were measured by AGID using gP51 and P24 antigens. The animals were serologically converted at 2 months post-inoculation(pi) in gP51 antigen, whereas sero-converted at 4 months pi in P24 antigen. In comparison with antibody titers for gP51, P24 antigen showed lower titers throughout the trial period. 2. The peripheral lymphocytes from all of the infected goats, as co-cultivated with F8l cells manifested syncytial formation at 4 months pi. 3. On counting total leucocyte, Iymphocyte and atypical Iymphocyte, two out of four infected goats showed normal distribution, while No 2 of the remaining two revealed temporal and No 3, Persistant increasing number of the cells. 4. The optimal condition of rosette formation of the peripheral Iymphocyte of normal Korean native goats was shown in the sheep erythrocyte treated with 0.1M AET for 30 nun at $37^{\circ}C$. When the Iymphocytes were treated in nylon wool column, the number of sIg-bear-ing cell were increased in the nylon wool adherent cells, but RFC was increased in the non-adherent cells. Of the infected goats, No 2 and No 3 showed significantly increasing number of sIg-bearing cells at 18 months pi. 5. The Iymphocytes of No 2 and No 3 goats reacted positively in IFA using Mab against BL-TAA at 12 months pi and 18 months pi, respectively. In CDAC test, all of four infected goats revealed positive reaction at 24 months pi. The higher positive rates were observed in No 2 and No 3 as compared with the remainders.

Identification of pol Gene Mutation among BLV Proviruses Found in the Southern Province of Korea

  • Bovine leukemia virus (BLV) is an etiological agent of chronic diseases in cows worldwide. The BLV is one of retroviruses that contain a multi-functional enzyme, reverse transcriptase produced from the pol gene in its genome. We have sequenced some regions in the pol gene of BLV proviruses found in the Southern province of Korea from samples that turned out to be BL V positives by a PCR analysis. On the 5' side of the BLV pol gene (polymerase region), it was found that there were four leucines located at every 7 amino acids. They can form a leucine zipper motif that was not same as the pol gene of Japanese BLV isolate. The sequencing result of the proviral pol gene in Korean-type BLV also revealed some mutations leading to amino acid changes such as $CCT(Pro){\to}CTC(Leu)$, $AAT(Asn){\to}AAA(Lys)$, and non-sensible variations i.e., $TCT(Ser){\to}TCC(Ser)$, $ATT(Ile){\to}ATC(I1e)$ and $ACG(Thr){\to}ACA(Thr)$. On the 3' side of the pol gene (integrase region), some nucleotide sequences were mutated and led to amino acid changes. Among them, a mutation, $GAA(Glu){\to}GAC(Asp)$ occurred in many Korean-type BLV proviruses was very interesting because the amino acid was regarded as one of the most conserved amino acids in the retroviral integrase. It was also notable that the mutation on any leucine residue did not occur, in spite of its frequent appearance.

홀스타인 젖소의 비유시기별 체세포 수와 우유 성분에 미치는 제요인 (The Effects on Somatic Cell Score and Milk Components by Days in Milk in Holstein Dairy Cows)

  • 본 연구는 낙농가의 소득과 직결된 원유 품질과 관계가 깊은 체세포 수(Somatic Cell Count, SCC)과 주요 우유 성분에 대하여 영향을 미치는 여러 요인 즉 아비 종모우, BLV type의 감수성 여부, 산차, 분만 월, 비유기 등에 대한 효과를 구명하였다. 조사기간은 2001년 1월부터 2004년 3월까지 분만한 홀스타인 젖소로부터 30$\pm$5일의 간격으로 시료를 채취하였으며, 우유성분과 체세포 분석은 Somascope MK2와 Lactoscope FTIR을 이용하여 총 3,897 개의 기록을 분석하였다. 비유기는 비유초기(30일 미만), 최고 비유기(30~60), 비유 중기(60~120), 비유 후기(120~180), 비유 말기(180일 이후)로 편의상 구분하였다. 분석 결과 대부분의 요인들이 모든 비유기에 걸쳐 유의적인 것으로 나타났으나 BLV type은 비유기에 따라서 차이를 나타내었다. 특히, BLV 감수성 여부는 조기에 검사를 통하여 보유 인자를 제거시키는 것이 장기적으로는 축군의 유질의 개선에 도움이 될 것으로 기대된다. 단백질에 대하여 BLV type은 각 비유기에 영향을 미치지 않았으며, 산차는 30일 이상에서 영향을 미쳤고, 분만월은 전 유기에 걸쳐 영향을 미치는 것으로 나타났다. 기타 유성분도 비유기에 따라서 각 각 다른 영향을 받는 것으로 나타났다.

울릉도 소의 전염성 병원체 감염률 조사 (Prevalence of infectious agents in cattle reared in Ulleung island)

  • Prevalence of infectious agents, including Brucella abortus (BA), Mycobacterium bovis (MB), bovine leukemia virus (BLV), M. avium subsp. paratuberculosis (MP), Neospora caninum (NC) and Toxoplasma gondii (TG), was investigated in all the cattle raised in Ulleung island during 2007~2010. For BA, the prevalences in head and farm were 8.1% (44/545) and 5.5% (4/73) in 2007, all negative in 2008~2009, and 0.5% (4/774) and 1.7% (1/58) in 2010, respectively. For MB, no sample was positive by PPD or ELISA in 2007~2010. For BLV and MP, no sample was positive by ELISA in 2007~2009. For NC, seroprevalences in head and farm were 0.2% (1/545) and 1.4% (1/73), respectively, in 2007 and all negative in 2008~2009. For TG, seroprevalences in head and farm were 17.6% (97/552) and 54.8% (34/62) by ELISA in 2009. By regions, the seroprevalences of TG in Ulleung-eup, Seo-myeon and Buk-myeon were 26.0%, 9.8% and 16.7%, respectively, which had significant differences (P<0.0001). Tiger cattle were more resistant to TG infection than Hanwoo. The seroprevalence of TG in summer was higher than in autumn. The seroprevalence of TG in cows was higher than in oxen. The seroprevalence of TG in cattle was increased with age. In conclusion, this study indicates that the prevalences of six infectious diseases, except for TG which are widely spread, are relatively low in cattle reared in Ulleung island.

우백혈병 바이러스감염 림프아구양 B 세포주(BL2M3)의 autocrine 증식 (Autocrine Growth of Bovine Leukemia Virus Infected-Lymphoblastoid B-Cell Line, BL2M3)

  • 우백혈병 바이러스감염 B 세포주(BL2M3 및 BL312)의 배양상층액에 대한 자기세포들의 증식반응을 검토하였다. 그 결과 BL2M3 및 BL312세포의 배양상층액을 자기세포인 BL2M3세포에 첨가했을 때 농도에 비례하여 현저한 증식을 유도하였다. 이것은 배양 4-5일차, 배양상층액의 첨가농도는 50-60%,세포수는 5x$10^4$-5x$10^{5}$ /ml에서 최적의 증식반응을 보였다. 우태아혈청(FBS) 무첨가 BL2M3 및 BL312세포의 배양상층액에 대해서도 BL2M3세포는 동일한 증식반응을 보였다. 한편 BL2M3 및 BL312세포의 배양상층액을 BL312세포에 첨가했을 때는 BL2M3세포의 경우에 비해 현저하지 않았다. 또한 BL2M3 및 BL312세포의 배양상층액은 말초혈액 림프구에 대해서도 pokeweed mitogen(PWM)첨가 유무에 관계없이 증식을 유도하였다. 그러나 PWM자극 말초혈액 단핵세포의 배양상층액은 BL2M3 및 BL312세포에 대해서 전혀 증식을 유도하지 못했다. 이상의 결과로부터 우백혈병 바이러스감염 B세포주 특히 BL2M3세포는 세포자신이 증식인자를 분비하고 그것과 반응하여 증식하는 소위 autocrine growth 양상을 보이는 것으로 판명되었다.
