소 미성숙 난포란의 체외성숙시 ${\beta}-Mercaptoethanol$의 첨가가 체외성숙, 체외수정 및 Glutathione 수준에 미치는 영향 (Effect of ${\beta}-Mercaptoethanol$ Supplement during In Vitro Maturation on IVM, IVF and Glutathione Level in Bovine Oocytes)

  • 본 연구는 소의 미성숙 난포란의 체외성숙시 ${\beta}-mercaptoethanol({\beta}-ME)$의 첨가가 체외성숙, 체외수정 후 웅성전핵의 형성 및 세포질 내의 GSH 수준에 미치는 영향을 알아보고자 실시하였다. 체외성숙시 $25\;{\mu}M$$50\;{\mu}M$${\beta}-ME$를 첨가한 경우 대조구에 비하여 성숙율이 증가하는 것으로 나타났으며(p<0.05), 모든 실험구에 있어서 12시간 체외성숙보다 24시간 체외성숙에서 높은 성숙율을 나타냈다(p<0.05). 체외수정 후 웅성전핵 형성에 있어서는 $25\;{\mu}M$$50\;{\mu}M$ 농도의 ${\beta}-ME$ 첨가구에서 대조구에 비하여 높게 나타났으나(p<0.05), $25\;{\mu}M$$50\;{\mu}M$ 농도구와의 유의적인 차이는 없었다. GSH의 수준은 체외성숙 후 $50\;{\mu}M$${\beta}-ME$ 첨가구가 다른 처리구에 비교하여 높게 나타났으며(p<0.05), 체외수정 후 웅성전핵이 형성된 다음 세포질 내 GSH 수준 역시 $50\;{\mu}M$${\beta}-ME$ 첨가구에서 가장 높은 결과를 나타냈다(p<0.05).

  • Chimerism has become an important tool for investigating fundamental aspects of early embryonic development and differentiation in mammals for producing transgenic animals. The objective of this study was to evaluate the developmental capacity of chimeric embryos reconstructed with parthenotes and IVF bovine embryos into empty zona pellucida. The MII oocytes were activated by two treatment groups [Group 1, 5 μM inomycin, 5min, + 10 ㎍/㎖ cycloheximide (CHX)/5 ㎍/㎖ cytochalasin B (CCB), 3 h; Group 2, 5 μM ionomycin, 5 min + 1.9 mM 6-dimetylaminopurine (6-DMAP), 3 h].   • In vitro embryo culture techniques provide significant contributions not only for a basic research of fertilization and early embryogenesis, but also for a low cost mass production of bovine embryos for transfer, embryo diagnosis, nuclear cloning and the production of transgenic cows. This presentation introduces newly developed serum-free media (IVD101 and IVMD101) that are effective far high yields of transferable embryos of excellent quality from in vitro-matured and fertilized oocytes. Both serum-free media are superior to a conventional serum-containing medium on the increased rates of blastocyst formation, post-thaw embryo viability, and pregnancy after transfer. Furthermore, reduced risks of calf mortality and large calf syndrome are also observed for the serum-free-derived embryos. Serum-derived embryos contain a large number of lipid droplets and immature mitochondria in their cytoplasm that may account for the lower production of transferable embryos and poor embryo quality.

  • 소 난자의 시험관내 수정시 이성체인 1, 2 propanediol과 1, 3 propanediol과의 동결보호제의 이용에 대한 독성효과를 조사하였다. 프랑스에 있는 도축장의 난소에서 채취한 미성숙 난자 212개를 시험관내 성숙과 heparin(10$\mu$g/ml)첨가에 의한 수정 및 배양을 실시하였다. 소 난자의 시험관내 성숙과 수정시 propanediol의 독성효과를 시험한 결과 1,3 propanediol은 80%의 정상 난할분할율과 5.7%의 낮은 다핵분할구율을 나타내었다. 이것은 1,2 propanediol의 73.9%, 10.3%와 미처리대조구의 73.9%, 7.7%와 비교되었으나 처리간의 유의성은 없었다. Heparin 처리구는 1,2 propanediol의 첨가 유무에 관계없이 미처리구에 대하여 고도의 유의성 차이가 인정되었다. (P<0.05)

  • This study was carried out to evaluate the effect of culture methods on development of embryos with each developmental stage after heat shock in bovine oocytes. The results obtained were as fellows. 1. The culture method after heat shock on development of embryos was better drop-culture than co-culture. 2. The medium without amino acids were not effect of heat sock on development of embryos but it was in need of amino acid during formation of blastocyst.

  • Bovine embryos produced by in vitro maturation, feretilization and development was examined for presevation and transfer. The fertilization medium used BO medium with 5 mM/$m\ell$ caffeine and 10$\mu\textrm{g}$/$m\ell$ heparin and adjusted to a pH of 7.2 to 7.4. The final concentration of spermatozoa was adjusted to 1$\times$$10^{6}$ cells/$m\ell$ motile sperm during fertilization in vitro. At 8~10 hrs after insemination, the oocytes were transferred into CR1aa medium and cultured for 7 days. Embryos were preserved by vitrification method for transfer. When the embryos of early, blastocyst and expanded blastocyst stages were frozen-thawed, the proportions of embryos with normal morphology 83.6, 88.1 and 85.2%. (중략)

  • Mainly due to deficiencies in nuclear reprogramming, gene expression and DNA fragmentation, which result in early and late embryonic losses, the overall success rate achieved by cloning techniques to date is low. This present study compared the incidences of DNA fragmentation during development of IVF, parthenotes (PT), nuclear transfer (NT) and transgenic (TG) embryos. Terminal deoxynucleotidyl transferase (TdT) nick-end labelling (TUNEL) with propidium iodide counter staining was used for determination of DNA fragmentation and total number, respectively. TG and NT donor cells were fetal fibroblasts with or without transfection with EGFP, and cultured in DMEM+15% FCS until confluent, for 5 days. At 19 h post-maturation (hpm), enucleated oocytes were reconstructed with donor cells and activated at 24 hpm with the combinations of ionomycin (5 M, 5 min) and cyclo-heximide (10 g/ml, 5 h) after electric fusion by a single DC pulse (1.6 KV/cm, 60 sec). 소 동결 미성숙 난자의 생존율과 체외수정율에 관한 연구 (Studies on the Survival and In Vitro Fertilization Ratesof Frozen-Thawed Bovine Immature Oocytes)

  • 본 연구는 소 미성숙 난자의 동결에 있어서 미성숙 난자의 1, 2, 4, 8 시간 성숙배양한 다음 동결 융해 후 체외배양하였을 때 체외수정율과 생존율을 조사하고저 수행하였다. 미성숙 난자들 1∼8시간 성숙배양후 동결융해 및 체외수정시켰을 때 수정율은 12.8∼30.9%로서 단시간의 체외성숙 배양이 높게 나타났으며(23.8∼28.8%), 또한 체외배양시 생존율은 11.8∼29.9%를 나타냈다.
