Effects of Rubus coreanus-Cheonggukjang on Bone Mineral Density and Bone Mineral Content in Growing Rats

  • The purpose of the present study was to investigate the bone-conserving effects of Rubus coreanus-Cheonggukjang (RC-CGJ) supplemented with more intensified phytochemicals compared to general Cheonggukjang (CGJ) in growing rats. Eighteen rats were divided into 3 treatment groups (Control, CGJ, and RC-CGJ) and were given experimental diets for 9 weeks. All of the rats in this study were fed a AIN-93G-based diet. Both CGJ groups were fed with 33.1% CGJ and RC-CGJ powder, respectively. The results of this study indicate that weight gain, mean food intake, and food efficiency ratio were not significantly different by the experimental diets among all groups. Spine bone mineral density (BMD) and femur BMD were not significantly different by the experimental diets. Spine bone mineral content (BMC) was significantly higher in the RC-CGJ and CGJ groups than in the control group, regardless of CGJ type. The femur BMC of the CGJ supplemented group was significantly higher compared with the control group and the RC-CGJ group. Compared with the control group, spine BMD and femur BMD per weight were markedly increased in the RC-CGJ and CGJ group regardless of CGJ type. Also, spine BMC per weight was significantly higher in the RC-CGJ group than in the CGJ group. However, femur BMC per weight was significantly higher in the CGJ group than in the RC-CGJ group. It can be concluded that RC-CGJ and CGJ supplemented diets have more beneficial effects on spine and femur peak bone mass in growing rats.

난소를 절제한 나이가 다른 흰쥐에서 식이 칼슘 수준이 골격 대사에 미치는 영향 (The Effects of Dietary Ca Levels on Ca and Skeletal Metabolism in Ovariectomized Rats of Different Age)

  • To investigate the effect of dietary Ca levels on metabolic changes of Ca and skeleton in postmenopausal women, 10-month-old ovariectomized female rats were compared with 2 month old rats. The rats were fed either 0.2% or 1.2% Ca diets for 16 weeks. Food intake and weight gain as higher in rats fed high Ca diets and in ovariectomized rats. Apparent Ca absorption as higher, and Ca balance was lower in the low Ca groups. Vertebrae density was higher in old rats or those fed a high Ca diets. The old rats and ovariectomized rats showed decreased bone formation, increased bone resorption and kidney function deterioration resulting in increased urinary Ca excretion. Contradictory to the above observation, old rats and ovariectomized rats still showed higher bone mass and bone ash content. Therefore aging was not fully onging in 10-month-old rats. Bone weights, mineral contents, and mineral/wt ratio were lower in ovariectomized rats. Dietary Ca level did not affect urinary Ca excretion, urinary protein excretion, GFR, serum alkaline phosphatase, or urinary hydroxyporline excretion. This means that dietary Ca level did not influence kidney function or bone turnover. However Ca content and the ash content of femur, 4th vertebra, and scapula were increased in high Ca groups. Therefore, it is considered that decreased bone formation and accelerated bone resorption may account for the increased osteoporotic risk in women in menopause after middle age. However, Ca metabolism can be improved and bone components can be maintained if Ca is supplemented.

한국 사춘기 소녀들의 골밀도와 환경요인들과의 관계(I) (The Relationship Between Bone Mineral Density and The Environmental Factors in Korean Pubescent Girls)

  • The relationship between bone mineral density and the environmental factors were investigated from the view point of preventing osteoporosis in Korean pubescent girls. The effects of calcium, nutrient intake, physical activity on total bone mineral density, lumbar spine and femoral bone mineral density and total bone mineral content were evaluated 33 healthy pubescent girls aged 14∼16y. A convenient method was used to assess nutritional and energy intake and calcium index was used together. Calcium intake in childhood was estimated by asking whether subjects usually drank milk as children. Eating habits data and history of menstruation were obtained by questionnaire and interview. Average energy expenditure was calculated. Bone mineral density and content were measured by dual energy x-ray absorptiometry using a Lunar DPX+Scanner (Lunar, Madison, WI). The lumbar spine(L2∼L4) and three sites in the proximal femur (femoral neck, trochanteric region, and Ward's triangle)were measured. Height and weight were measured, and the body mass index(BMI) was derived from the formula : BMI=kg/㎡ Statistical analysis was performed by simple correlation using the SAS package. The mean calcium intake (736mg) was below the RDA of 800mg/d. Twelve percent of the total subjects did not drink milk at all because they did not like the taste. Skipping meals, low calcium intake and low energy intake were significantly correlated with the low BMD. Also the data indicate that girls who reported drinking milk with every meal during childhood had significantly higher bone densities than girls who reported drinking milk less frequently. The results suggest that milk consumption in childhood appears to be needed not only for growth and development, but possibly also to assure an optimal peak of bone mass and thus greater latitude for the maintenance or skeletal integrity in the face of bone losses. There was a highly significant correlation between the total BMD and overall level of physical activity. Body weight was a better predictor of total BMD than was and other factor. Simple mechanical loading may explain why body weight, but total BMC was positively relatd to height. Conclusively, increasing calcium intake and physical activity in the pubescent girls could influence BMD.

성장기 수컷 흰쥐에서 Isoflavones의 첨가가 골밀도 및 골함량에 미치는 영향 (The Effect of Isoflavone Suplementation on Bone Mineral Density and Bone Mineral Content in Growing Male Rats)

  • This study was performed to determine the effect of isoflavone on bone mineral density and bone mineral content in growing male rats. Twenty male, Sprague-Dawley rats were assigned to groups, that underwent 9 weeks of experi-ental treatment. Animals were assigned to one of two diet groups (casein group or casein supplemented with isoflavones). During 9 week of the study, food consumption was determined every other day through the measurement of total food given subtracting the food uneaten from original amount given. Rats in two experimental groups had similar initial body weights. At the end of experiment, however, the casein group had significantly greater body weights compared to casein supplemented with isoflavones group. It was also observed that the casein group had greater food intake comared to casein supplemented with isoflavones group. The difference in the final body weights of the groups was therefore due to difference in amount of food ingested, but could be due to the effect of isoflavones. Total BMD, spine BMD, and spine BMC per weight and femoral BMD per weight were significantly greater in casein supplemented with isolaones group than casein group. ALP and osteocalcin were significantly greater in the casein-fed group. Crosslink value was significantly lower in the casein supplemented with isoflavones group, All other variables were statistically similar between two groups. Overall, it can be concluded that casein supplemented with isoflavones beneficial for acquisition of bone mineral density and content on growing male rats.

골감소증 여대생의 영양소 섭취실태와 이소플라본 보충이 골밀도에 미치는 영향 (Nutrient Intake and Effects of Isoflavone Supplementation on Bone Mineral Density in Female College Students with Osteopenia)

  • The principal objective of this study was to assess the effects of soy isoflavone supplementation on bone mineral density in 36 female college students with osteopenia for 12 weeks. The subjects were divided into three groups on the basis of bone mineral density. The experimental groups were provided supplements of either 80 mg of isoflavone (Iso-80) or 40 mg of isoflavone (Iso-40). To the placebo group, 40 mg of powdered glutinous rice was administered. It was determined that many subjects with osteopenia evidenced lower levels of activity as compared to the control group. Isoflavone supplementation was more effective in controlling total cholesterol and LDL-cholesterol than was observed in the placebo group. We noted no significant differences in serum osteocalcin concentration between Iso-40 and the placebo group, but significant differences in osteocalcin concentration were detected between Iso-80 and the placebo group. Bone quality indices (BQI) were correlated positively with mineral content, lean body mass, muscular mass, and blood components including albumin, Ca, Mg, ALPase, and osteocalcin. Both Iso-40 and Iso-80 supplementation for 12 weeks significantly increased protein and mineral content in the body. As lower intakes of Ca and folate were noted in the subjects, emphasis should be given to adequate intakes of these nutrients in the subjects. In conclusion, 12 week isoflavone supplementation in young females with osteopenia exerted positive effects on bone mineral density and bone turnover markers.

난소절제 쥐에서 카페인 첨가식이가 골밀도 및 골함량에 미치는 영향 (Effects of Caffeine on Bone Mineral Density and Bone Mineral Content in Ovariectomized Rats)

  • 본 연구는 0.03%의 카페인 함유 식이를 폐경모델인 난소절제쥐에서 6주간 섭취시켜 골밀도와 골무기질함량에 미치는 영향을 아래와 같이 요약하였다. 1) 체중증가량은 난소절제군이 Sham군에 비해 유의적으로 높았으며 각 군내에서 카페인 섭취에 따른 차이는 없었다. 2) 혈 중 칼슘 농도는 난소절제군내에서 카페인군이 대조군보다 유의적으로 낮게 나타났다. 3) 혈 중 ALP는 Sham군과 난소절제군 모두에서 카페인 군이 대조군보다 높은 경향을 나타내었다. 혈중 Osteocalcin은 Sham 군과 난소절제군, 그리고 각 군내에서 카페인 섭취여부에 따라 유의적인 차이를 나타내지 않았다. 4) 요 중 칼슘 및 인의 농도, 요 중 Deoxypyridinoline(DPD)와 crosslinks value는 Sham 군과 난소절제군, 그리고 각 군내에서 카페인 섭취여부에 따라 유의적인 차이를 나타내지 않았다. 5) 척추골밀도는 Sham군에 비해 난소절제군이 유의적으로 낮았고, 난소절제군내에서 카페인 첨가군과 대조군간에 차이가 없었다. 6) 대퇴골밀도와 대퇴 골무기질 함량은 Sham군과 난소 절제군 간의 차이는 없었고, 각 군내에서 식이에 따른 차이도 없었다. 따라서 카페인 0.03% caffeine 섭취는 난소절제쥐에서 6주간 섭취 시킨 경우 척추와 대퇴골밀도에 부정적 영향을 미치지 않는 것으로 나타났다.

성인여성의 식습관과 영양섭취상태와 골밀도 및 골무기질함량과의 관계 (The Relationship between Food Habit, Nutrient Intakes and Bone Mineral Density and Bone Mineral Content in Adult Women)

  • Bones are important parts in sustaining the shape of the body, but they are also metabolic organs which undergo bone remodeling by constant bone resorption and formation. Osteoporosis, the typical metabolic bone disease, is characterized by a reduction in bone mineral density (BMD). Women more than men are at risk fir osteoporosis-related fractures, especially in the lumbar spine, wrist, and hip region. Risk of fracture depends on one's BMD, which open determined by the peak bone mass value achieved at skeletal maturity and followed by subsequent age-and menopause-related bone loss. Genetic and environmental factors are known to play a key role in bone metabolism and diet is considered as one of the important environmental factors. The purpose of the present study was to assess the status of BMD and bone mineral content(BMC) to clarify the relationships between dietary intakes and the risk of osteoporosis in adult women in Taegu. Subjects were 130 healthy females in between 20 and 69 years of age. BMD and BMC of the lumbar spine(venebrae L2-4) of the subjects were measured by dual energy X-ray absorptiometry. The average age of the subjects was 47.4${\pm}$11.7 years old, the average weight was 57.2${\pm}$8.4kg, the average age of menarche was 16.6${\pm}$1.9 years old and the average age of menopause was 48.4${\pm}$5.3 years old. The nutrient intakes of the subjects measured by the convenient method were generally lower than the level of RDA. The result of nutrient intake assessed has shown that the average energy intake was 1701${\pm}$316kca1 which is 85.1% of the RDA and the average calcium intake was 485.4${\pm}$172.3mg which is 69.3% of the RDA. The intakes of protein, vitamin A, vitamin B$_1$, niacin were greater than the RDA, whereas the remaining nutrient intakes were lower than the RDA. The average BMD of the subjects was shown to be 1.06${\pm}$1.09g/$\textrm{cm}^2$. The highest BMD of 1.24${\pm}$0.14g/$\textrm{cm}^2$ was noticed in the subjects of 30s compared to 20s, 40s, 50s, 60s. The BMD values were compared by the relative body weight(R3W) of the menopause subjects, and it was found that the underweight group had significant lower BMB while the rest of the groups did not have any differences in BMD. The most strongly correlated nutrient with BMD among the menopause subjects appeared to be calcium. The women whose Ca intakes were higher than 500mg showed the significantly higher BMD than those with Ca intakes lower than 500mg. This study suggests that the most effective way to prevent osteoporosis and to reduce the incidences of fractures seems to be minimizing bone loss through the adequate intake of calcium as well as avoiding underweight, especially in menopausal women. The Effect of Water-Soluble Calcium Supplements on Calcium Metabolism and Bone Metabolism of Growing Rats

  • Within the elderly population, the use of calcium supplements and the intake of calcium from food are on the rise in order to maintain health. Calcium is absorbed as an ion in vivo, leading to speculation that absorption efficiency is affected by the solubility of the calcium consumed. In our study, the bioavailability of two types of calcium supplements with different solubilities was evaluated. Experimental animals were fed water-soluble or insoluble calcium supplements for 6 weeks. We found that blood alkali phosphatase activity, osteocalcin content, and urine crosslinks values were not different between the groups. Similarly, the degree of apparent calcium absorption between the two calcium supplements was not significantly different. The bone mineral density and bone mineral content of the femur and the tibia increased in the group that consumed insoluble calcium compared with those of the water-soluble calcium supplemented group. However, when considering body weight, the bone mineral density value for all areas, including the spine, was significantly higher in the group that consumed the water-soluble calcium supplement.

한방재료를 첨가한 타조 추출액의 영양학적 특성에 관한 연구 (A Study on the Nutritional Characteristics of Ostrich Extracts Added with Medicinal Herbs)

  • This study was undertaken to analyze chemical composition, macro- and micro-mineral contents of ostrich bone with flesh meat (BF), ostrich bone with flesh meat with herb medicine (BFll) and ostrich bone with internal organs (BI). Crude fat of the BFH was lower, but crude protein and crude ash of the BFH were significantly higher than those of the BF and the BI. Among the minerals, Ca, p, Mg, Fe, Cu, Zn content of the BFH were significantly higher than the content in the BF and the BI. The minerals, Pb, Cd and As were not detected in ostrich and herb medicine extracts. From the sensory test of ostrich and herb medicine, the preferences were in the fellowing order; ostrich bone with flesh meat with herb medicine (BFH), ostrich bone with flesh meat (BF), and ostrich bone with internal organs (BI) in extracts.

Effects of isoflavone supplementation on the bone mineral density of growing female rats

  • This study was focused on whether or not isoflavones affect the increase in bone mineral density of growing females. Female Sprague-Dawley rats ($60{\pm}5\;g$) were randomly assigned to two groups and provided control diets (control group) or isoflavone-supplemented diet (IF group, 57.8 mg isoflavones/100 g diet) for 9 weeks in growing female rats. Measurements of Bone Mineral Density (BMD) and Bone Mineral Content (BMC) on the experimental animals were executed in the $3^{rd}$, $6^{th}$, $9^{th}$ weeks. In result, there was no significant difference in spine BMD between the isoflavones supplemented group and the control group. But, the IF group tended to have higher BMD than the control group in between 3 and 9 experimental weeks, and the striking difference could be shown in the $6^{th}$ week of feeding. In case of femur BMD, the effects of added isoflavones appeared in the $6^{th}$ week of feeding, and it became intensified in the $9^{th}$ week of feeding to the extent that the BMD in the IF group was significantly higher than that of the control group (p<0.05). In conclusion, isoflavone supplementation increased spine BMD per weight in the $6^{th}$ week of feeding, and affected the increase of femur BMD in the $9^{th}$ week. The result of the experiment implies that it affects positively the formation of spine and femur BMD of growing female rats. The study also suggests that the effects of isoflavone on the pattern of BMD formation might differ from the parts of bones.
